Sweet lupines are increasingly used in food production. Cause for concern has been expressed due to the increase in reported lupine-induced allergic incidents and the association between lupine and peanut allergies. In the current study, a polyclonal-monoclonal antibody-based sandwich ELISA for the detection of lupine proteins in foods was developed. The assay was sensitive to both native and processed proteins from Lupinus angustifolius and Lupinus albus and had a detection limit of 1 mug/g. Intra- and interassay coefficients of variation were <5 and <17%, respectively. A selection of 112 food samples, both with and without lupine declaration, was evaluated for their content of lupine. The data showed that the majority were in agreement with the respective labeling. However, some inconsistency was seen, typically in bread/rolls and soy flours.  相似文献

An increasing number of invasive species are changing ecosystems around the world. Road verges have commonly become the first footholds of non-native species in the new environments. Regularly mown road verges also offer habitats for meadow flora and fauna, which in Europe have suffered from the radical decline of semi-natural biotopes due to the agricultural modernization. We studied impacts of an invasive plant Lupinus polyphyllus on the plant and Lepidoptera species composition along road verges. The plant species composition was studied on 15 sites (with 1 m2 quadrats) and butterflies and diurnal moths along 15 transects (with weekly censuses) in SE Finland, each site and transect representing equally lupine invaded verge and an adjacent non-lupine verge. The species richness and diversity of flora and the cover and species richness of low growing (<20 cm) species, in particular, was lower in lupine verges compared to non-lupine verges. Also, the abundance of butterflies was lower in lupine verges. As the lupine cover approached 100%, fewer butterflies were observed in lupine transects compared to the adjacent non-lupine transects and a higher proportion of individuals were flying. Our results suggest that the changes in plant species assemblages and lower plant species richness in lupine invaded areas had “bottom-up” effects on higher trophic levels. Further studies on the control of lupine are urgently needed, but meanwhile we suggest regular mowing before the lupines have shed their seeds, together with the removal of the cuttings, to be the best management option.  相似文献

Pyrrolizidine alkaloid-containing plants are widely distributed throughout the world and are particularly common in the genus Senecio. The structural types and concentrations of the alkaloids vary among plant species. In addition, within a species of plant, concentrations vary with environment and location. Many pyrrolizidine alkaloids are toxic and cause poisoning in livestock and in humans. Rapid, sensitive, and specific diagnostic techniques are needed to identify poisoned animals and to determine the particular plants and conditions under which livestock are likely to be poisoned. In this study, two competitive inhibition enzyme-linked immunosorbent assays for riddelliine, riddelliine N-oxide, and other closely related pyrrolizidine alkaloids were developed using polyclonal antibodies. One assay is class specific toward the free base forms of the pyrrolizidine alkaloids; the other assay showed cross-reactivity to both the free base and N-oxide forms of the alkaloids. The assay with the lowest limit of detection had an I(50) of 803.9 pg with a limit of detection of 47.5 pg for riddelliine. Spike and recovery studies for riddelliine in bovine blood ranged from 45 to 74%. The assay that showed cross-reactivity between the N-oxide and free base forms of the pyrrolizidine alkaloids allowed estimation of the total pyrrolizidine alkaloid content in Senecio riddellii in admixture with alfalfa. These findings suggest that these techniques will be excellent tools to diagnose poisoned animals and identify highly toxic plants.  相似文献

White lupin (Lupinus albus L.) has unexploited potential as a crop plant due to its high seed yield as well as protein and oil content in seeds. Well-characterized collections of gene resources are very important for breeding as a source of genetic variation. This paper presents the results of analyses for total content and qualitative composition of alkaloids in seeds of 367 L. albus accessions from the Polish Genebank. Accessions were divided into four classes of origin: wild collected material, land races, breeding lines, and cultivars. Apart from the expected broad variation as well as strong differentiation in the alkaloid content, a clear influence of domestication was observed. This was shown as an apparent decrease in the alkaloid content in breeding lines and cultivars classes. The total alkaloid content varies from 0.02 to 12.73% of the seed dry weight. Six major alkaloids (abundance >1%) were revealed: lupanine (28.22–94.49%, mean 76.06% in total content), 13-hydroxylupanine (0.10–32.78%, mean 8.23%), multiflorine (0.00–21.67%, mean 5.52%), albine (0.00–18.55%, mean 4.48%), angustifoline (0.24–12.14%, mean 2.07%), 11,12-seco-12,13-didehydromultiflorine (0.00–12.28%, mean 1.74%). Owing to its abundance, lupanine was found to be the most closely correlated to the total alkaloid content.  相似文献

The nutrient composition, toxic factors content, and nutritional and toxicological value of Lupinus splendens, L. rotundiflorus, L. elegans, L. simulans, L. exaltatus, L. reflexus, and L. madrensis species from Mexico were analyzed. The seeds of these species were a good source of protein. All the species showed a high lysine and tryptophan content, though sulfur amino acids were limiting. Cyanogenic glycosides were absent, and lectins, trypsin inhibitors, and tannins were present in low concentrations. Lupanine was the major alkaloid in almost all the samples, although sparteine was the major alkaloid in Lupinus reflexus (26.63 mg/g of sample). Cytisine was not found in any of the studied lupins. L. reflexus showed the highest acute toxicity, and L. elegans exhibited no toxicity as evaluated using a mice model. The alkaloid was reduced by hot-water extraction. The protein efficiency ratio in water-debittered seeds was relatively poor (1.1-1.5). These results suggest that the wild lupins studied represent a potential protein supply, and they could be domesticated and used for animal feed if the alkaloids were eliminated and the protein was supplemented with methionine, or if the lupins were used in mixture with cereals.  相似文献

Willamette Valley upland prairie in western Oregon, USA, has been reduced to less than 1% of its original historic range following European settlement in the 1850s. Lupinus sulphureus ssp. kincaidii (Kincaid's lupine), a threatened species and the primary larval host plant of the endangered Icaricia icarioides fenderi (Fender's blue butterfly), was historically a panmictic metapopulation. Habitat fragmentation may be causing many of the Kincaid's lupine colonies to display typical symptoms of inbreeding depression, such as low seed production. Hand outcrosses on bagged inflorescences significantly increased seed set and seed fitness compared to open pollination and within-colony pollen treatments. Natural seed set was positively correlated with an increase in the number of Kincaid's lupine patches, suggesting that population size limits seed set. An increase in fruit set was positively correlated with Kincaid's lupine raceme number, raceme density, and the number of lupine patches, demonstrating that floral display and population size increase pollinator service. Restoration of Kincaid's lupine populations should consider measures that lessen the effects of inbreeding depression, especially in small, isolated populations, for the long-term persistence of the species.  相似文献

Secondary metabolites (SMs), such as alkaloids and raffinose oligosaccharides (RFOs), play important roles in plant physiology. Although alkaloid and RFO phenotypic variation has been reported for yellow lupin (Lupinus luteus L.), most evaluations have used a reduced number of accessions; thus, limiting the understanding of accumulation patterns and variation ranges. The main goal of this research was to assess alkaloid and RFO content in a diverse L. luteus sample to understand possible SM accumulation patterns across this legume species. Eighteen yellow lupin accessions were analyzed using high performance thin layer chromatography to provide insights on seed and leaf RFO and alkaloid phenotypic variation. Co-dominant markers (170) were used to examine genetic relationships among L. luteus accessions and possible accumulation patterns across closely related genotypes. Significant differences were observed for seed and leaf RFOs. Total seed RFO accumulation ranged from 79.738 to 131.079 mg g?1. Raffinose, stachyose, and verbascose were observed in all genotypes’ seeds, but at different RFO concentrations. Raffinose was the only RFO detected in leaves (2.793–0.4224 mg g?1). Total alkaloid accumulation ranged from 0.22 to 15.12 and 0.00 to 8.007 mg g?1 for seeds and leaves, respectively. Lupinine, sparteine, and gramine were observed in seeds and leaves, and showed a wide range of variation. Neighbor-Joining (NJ) analysis showed an apparent pattern of seed alkaloid accumulation, most likely due to domestication events. However, high RFO accumulating accessions were scattered across the NJ tree. Alkaloid and RFO significant phenotypic variation will not only help to understand the roles of these SMs in L. luteus, but also to uncover the genetic basis behind their accumulation.  相似文献

Neotyphodium species are fungal endophytes best known for their protection of grass hosts and production of bioactive metabolites including ergot alkaloids. Perennial ryegrass-Neotyphodium sp. Lp1 symbiota that have altered ergot alkaloid profiles (resulting from knockouts in two different endophyte genes) were fed, along with controls, to rabbits to test the effects of ergot alkaloids on food preference and satiety. Interestingly, rabbits dramatically preferred plants that were endophyte-infected but free of ergot alkaloids over endophyte-free plants (P = 0.01). Accumulation of ergot alkaloids of the clavine class counteracted the added appeal of endophyte-infected plants. In satiety tests, consumption of ergovaline (the ultimate ergot pathway product in wild-type endophyte), but not of several other ergot alkaloids, during an initial meal had a negative effect on subsequent rabbit chow consumption (P < 0.05). The data indicate that clavines were sufficient to reduce the appeal of endophyte-infected grasses, whereas only ergovaline reduced appetite.  相似文献

Populations of bloodroot [Sanguinaria canadensis L. (Papaveraceae)] are found throughout the eastern forests of North America, with particular abundance in the southern Appalachian Mountains. Increasingly, it is finding use in Europe as a nonantibiotic animal feed supplement to promote weight gain. As efforts to cultivate this herb are underway, there is a need to understand the effect of agronomic permutations on both the dry mass rhizome yield and the yield of benzophenanthridine alkaloids. Month-to-month variability of the concentration of the alkaloids sanguinarine and chelerythrine in both cultivated and wildcrafted bloodroot was examined. The alkaloid yield was consistently higher, but more variable, in wildcrafted plants. On average, cultivated rhizomes were both larger and more consistently sized than those that were wildcrafted. The concentration of a suite of trace elements was measured in soil that was collected concomitantly with each plant accession. Differences in element profiles were compared against alkaloid yields.  相似文献

Cultivation of Crotalaria juncea L. (Sunn Hemp cv. 'Tropic Sun') is recommended as a green manure crop in a rotation cycle to improve soil condition, help control erosion, suppress weeds, and reduce soil nematodes. Because C. juncea belongs to a genus that is known for the production of toxic dehydropyrrolizidine alkaloids, extracts of the roots, stems, leaves, and seeds of 'Tropic Sun' were analyzed for their presence using HPLC-ESI/MS. Qualitative analysis identified previously unknown alkaloids as major components along with the expected macrocyclic dehydropyrrolizidine alkaloid diesters, junceine and trichodesmine. The dehydropyrrolizidine alkaloids occurred mainly as the N-oxides in the roots, stems, and, to a lesser extent, leaves, but mainly as the free bases in the seeds. Comprehensive spectrometric and spectroscopic analysis enabled elucidation of the unknown alkaloids as diastereoisomers of isohemijunceine, a monoester of retronecine with an unusual necic acid. The dehydropyrrolizidine alkaloid contents of the roots, stems, and leaves of immature plants were estimated to be 0.05, 0.12, and 0.01% w/w, respectively, whereas seeds were estimated to contain 0.15% w/w.  相似文献

Selenium has been recognized as essential for all mammals; therefore, its concentration level and speciation are of great concern. Plants are one of the main sources of selenium in the diet. Thus, inorganic selenium uptake and its transformation in different species were evaluated in Indian mustard (Brassica juncea), sunflower (Helianthus annus), and white lupine (Lupinus albus). More than 1.2 g x kg(-)(1) (dry matter) of Se was found in the aerial part of Indian mustard when growing on 1 mg x L(-)(1) of Se as Na(2)SeO(4), and approximately half this amount was determined in the leaves of the lupine, which is still quite high. Selenomethionine was the main selenium-containing amino acid identified in most of the extracts by HPLC-ICP-MS. The higher values were 6.8 and 14.5 mg x kg(-)(1) (expressed as Se in dry matter) in the leaves of lupine and sunflower, respectively. This is of great importance because some authors have considered the combination of this enriched material with non-enriched food as a source of selenium supplementation.  相似文献

A pot experiment was designed with the objective of determining whether the presence of ethylenediaminetetraacetic acid (EDTA) and the resulting mobilization of heavy metals have any affect on: (i) soil microorganisms, (ii) growth of L. albus, and (iii) microbial colonization of roots. There was no effect of the different treatments on the contents of soil microbial biomass C and microbial biomass N. Increasing addition of EDTA to soil led to proportionate increases in extractable C and N, being roughly equivalent to the added amount. Increasing EDTA addition to soil led also to a proportionate increase in mobile heavy metals. Plant height, total amount of shoot and root C were not affected by EDTA addition. Fungal ergosterol in the lupine roots showed a 5- to 8-fold increase in the 0.1 and 0.2% EDTA treatments in comparison with the control. In contrast, EDTA addition did not affect fungal glucosamine or bacterial muramic acid concentrations in the lupine roots. Increasing EDTA addition to soil led also to a proportionate increase in the metal concentrations in the lupine shoots. The concentrations of heavy metals in the lupine shoots and in the NH4NO3 soil extracts were all highly significantly correlated.  相似文献

Plants of the Pilocarpus genus (Rutaceae) are popularly known as jaborandi and are the only source of pilocarpine, an imidazole alkaloid used in eye-drops for the treatment of glaucoma as well as for the stimulation of sweat and lachrymal glands. Alkaloid extracts from leaf samples of seven species of Pilocarpus, from the states of São Paulo and Maranhão in Brazil, were analyzed using HPLC–ESI–MS/MS. The samples contained between 0.88 ± 0.04 and 1.00 ± 0.14% of alkaloids in relation to the dry weight of their leaves, with significant differences in results (P ≤ 0.05) found only between Pilocarpus microphyllus planted in the state of Maranhão and Pilocarpus spicatus, Pilocarpus trachyllophus, Pilocarpus pennatifolius and Pilocarpus jaborandi; as well as between Pilocarpus spicatus and Pilocarpus racemosus. Pilocarpine was not found in P. spicatus, whereas in the other species it ranged from 2.6 ± 0.1 to 70.8 ± 1.2% of total alkaloids. P. microphyllus planted in the state of Maranhão for pilocarpine extraction had the highest total alkaloid content, but it had only 35% of pilocarpine in relation to total alkaloids. Three other species contained more pilocarpine in relation to total alkaloids: P. jaborandi (70.8%), P. racemosus (45.6%) and P. trachyllophus (38.7%); and could be candidates for pilocarpine extraction. Differences in alkaloid content were significant for all these samples (P ≤ 0.05). Imidazole alkaloids were observed and partially characterized based on their retention times and high resolution mass. The seven species analyzed had different imidazole alkaloid profiles, but only one did not present quantifiable pilocarpine contents in its leaves. The Pilocarpus genus shows potential for the prospection of novel alkaloids.  相似文献

The use of lupine in foods has increased considerably during the past decade, reflected by a corresponding increase in reported lupine-induced allergic incidents. Lupine allergy may arise either by primary sensitization or by clinical cross-reactivity in peanut-allergic persons. Detection of lupine proteins in food has previously been based on the use of patient serum. A novel sandwich enzyme-linked immunosorbent assay (ELISA) for the detection and quantification of lupine in processed foods was developed, using a polyclonal rabbit antilupine capture antibody and a biotinylated conjugate of the same antibody for detection. The antibody was highly specific for lupine, apart from minor cross-reactivities to other legumes. The assay had a detection limit of 1 mug/g and was successfully used to quantify lupine protein in various food matrixes. Recoveries ranged from 60 to 116%, while the intra-and interassay coefficients of variation were <6% and <21%, respectively.  相似文献

Prior to extensive European settlement, the fisher (Martes pennanti) occupied most coniferous forest habitats in Washington, Oregon, and California. Human activities since that time have resulted in the apparent extirpation of fishers throughout much of their historical range in the Pacific states. Fisher extirpations in California and Washington have been documented previously, but no comprehensive assessments of the distribution of fishers in Oregon, the history of their translocation into Oregon, or the conservation of fishers in the Pacific states have been conducted. Our objectives are to (1) review historical information on potential causes for fisher population losses in Oregon, (2) document the history of their translocation into Oregon, (3) describe the distribution of fishers in Oregon relative to those translocations and determine if any were successful, and (4) discuss the implications of our findings for the conservation of fishers in the Pacific states. Our results show that extant populations of fishers in Oregon are restricted to two disjunct and genetically isolated populations in the southwestern portion of the state: one in the southern Cascade Range and one in the northern Siskiyou Mountains. In addition, historical changes in the distribution of fisher occurrence records in Oregon and geographic variation in the genetic composition and size of fishers occurring in southwestern Oregon, show that the population in the southern Cascade Range is reintroduced and is descended from fishers that were translocated to Oregon from British Columbia and Minnesota. The loss of fisher populations from central and northern Oregon and throughout Washington has resulted in the isolation of extant populations in Oregon by >650 km from those occurring in southern British Columbia. Our results demonstrate that the historical continuity in fisher distribution that once provided for genetic interchange among fisher populations in the Pacific states no longer exists.  相似文献

The intake of lupin-based foods could imply the exposure of consumers to quinolizidine alkaloids. The objectives of this study were to assess the genetic variation among and within 11 geographic regions of Lupinus albus ecotypes, verify the quinolizidine alkaloids amount of alkaloid-poor L. albus and Lupinus angustifolius varieties, and assess the effect of two climatically contrasting Italian environments on the alkaloid content. The quantitation was performed by GC-MS, and in all samples lupanine was the most abundant quinolizidine alkaloid, followed by albine and 13alpha-hydroxylupanine for L. albus and by 13alpha-hydroxylupanine and angustifoline for L. angustifolius. Some regions tended to have a high (Azores) or low (Egypt, Near East, Maghreb) total alkaloids content, but the variation among ecotypes within regions was larger than that among regions following the estimation of variance components. Alkaloid-poor varieties tended to have higher total alkaloid contents when grown in the subcontinental climate site, exceeding in some cases the limit of 0.200 mg/g.  相似文献

Two Gram-negative bacterial strains capable of using lupanine, the predominant quinolizidine alkaloid in Lupinus albus, as a sole carbon source were isolated from soil in which L. albus and L. luteus had been grown [Santana, F. M. et al. J. Ind. Microbiol. 1996, 17, 110-115]. In the present study, we present results suggesting that these isolates are of potential interest for removing lupanine and other quinolizidine alkaloids (QA) from the effluent resulting from the wet processing of Lupinus seeds, at temperatures within the range 20-34 degrees C. Growth in L. albus aqueous extract was diauxic, with a first period of rapid growth leading to the simultaneous consumption of a significant part of the initial concentration of QA (3 g L(-1), being 2 g L(-1) lupanine) and amino acids (1.5 g L(-1)). This period was followed by a second period of slower growth corresponding to the subsequent partial utilization (25%) of the carbohydrates (initial concentration of 20 g L(-1)) together with further removal of QA and amino acids. Despite the differences detected in the susceptibility of the two strains to lupanine toxicity, in particular at supraoptimal temperatures, and in the efficiency of lupanine catabolism, their performance on L. albus extract did not vary significantly.  相似文献

The Lupinus genus consists of 200–600 species with diversified morphological and biochemical characteristics that provide scope for increasing the value of protein attributes in commercial lupin species through genetic improvement. This study characterised protein polymorphism among 33 selected lupin genotypes representing 19 species from Old World and New World using Matrix-Assisted Laser Desorption Ionization Time-Of-Flight (MALDI-TOF) mass spectrometry with the aim of developing a high throughput screen for assessing variation among samples within a genetic resource. MALDI-TOF mass spectrometry analysis recognized 630 polymorphic protein mass peaks. A total of 19 protein mass peaks were identified as species specific and 23 were recognized to be generally common (observed in more than 10 species). A set of 111 rare protein mass peaks common to 2–3 species were identified to uniquely distinguish the respective species in combination with the species specific mass peaks. Overall, the seed protein profiles showed extensive diversity among the species analysed. Phylogenic analysis based on the protein mass peaks clustered the Old World smooth and rough-seeded and New World lupin species separately, in broad agreement with clustering based on morphological characters, life history, cytogenetics, DNA polymorphism and electrophoreses of seed proteins. Two New World lupin species, L. mutabilis Lindl. and L. succulentus Douglas ex K. Koch were grouped into the smooth-seeded Old World group.  相似文献

The capsule husks of a set of 115 Indian land races of opium poppy Papaver somniferum L. obtained over two cropping seasons were screened for the percent contents of papaverine, reticuline, narcotine, thebaine, codeinone, codeine,morphine and oripavine. Among the accessions screened 36, 7 and 1 didnot accumulate papaverine, papaverine and narcotine and codeine and morphine, respectively. The alkaloid profiles and correlations between alkaloids in all the four classes of accessions showed thatin the Indian genetic resources of P.somniferum (a) morphine is synthesized from codeine rather than oripavine, (b) net alkaloid contents are low under narcotine deficiency, and (c)accumulation of morphine and codeine was limited upstream of codeine and morphine. The accessions identified to be harboring genetic blocks in the phenenthrine and benzylisoquinolinebiosynthetic pathways will be useful for understanding the genetic control of secondary metabolism in opium poppy.  相似文献

Conserving large carnivores throughout the world will often require that they share the landscape with livestock. Minimizing depredations and increasing tolerance by livestock producers will be critical for conservation efforts. To investigate factors influencing calf mortality and producer detection rates (i.e., number of livestock killed by predators, found by producers, and correctly classified as to cause of death), we monitored radio-tagged domestic calves at two sites in the Mexican wolf recovery area (East Eagle [EE] and Adobe Ranch [AR]). Study areas differed in grazing practices, density of predators (mountain lions, black bears, coyotes, and Mexican wolves), and amount of effort spent monitoring cattle. We radiotagged 618 calves over 3.5 years, and 312 calves over 2 years on the EE and AR, respectively. The overall proportion of radioed calves that died was higher on the EE (6.5%) than on the AR (1.9%). Predators (especially mountain lions) accounted for 85% of mortality on the EE and 0% on the AR. Calves selected by predators were on average 25 days younger than the surviving cohort. Our results indicate that year-round calving, especially in areas with high predator densities, are subject to higher losses primarily because calves are exposed to mortality agents for longer periods of time rather than having higher natural rates of mortality. We found a significant difference in producer detection rates between study sites, likely due to differences in the intensity of monitoring cattle between sites. On the EE, the producer detected 77.5% of mortalities and on the AR, the producer detected 33% of mortalities. Our results support changing husbandry practices to limit calving to a seasonal endeavor and that performance payment may be a better compensation strategy than ex post compensation schemes.  相似文献
