Little is known about the transport of microorganisms through freeze-fractured clay soils. Normally consolidated clay (NCC) and compacted clay (CC) columns (representing a natural clay barrier and a compacted barrier, respectively) were exposed to six consecutive freeze–thaw cycles and permeated for 21 days with an Escherichia coli cell suspension (approximately 1?×?107 colony forming units (CFU)/mL) containing a 2.1-mM bromide tracer. An unfractured sand column was also examined for comparison with the clay columns. While no E. coli was detected in the effluent of both untreated NCC and CC control clay columns, a relatively low density of E. coli (between 228 and 270 CFU/mL compared to 1?×?107 CFU/mL in the influent) was first detected in the effluent of the freeze-fractured NCC and CC columns at 0.29 and 0.31 pore volumes (or at 5.4 and 4.1 h), respectively. It took 11 min for a full breakthrough of E. coli through the sand column, but only about 0.1% of the influent E. coli density was detected in the effluents of the freeze-fractured NCC and CC columns at day 21. These observations show that despite the high bacterial retention capacity of the freeze-fractured clay columns, the fractures were large enough for the E. coli to flow through. Based on batch sorption tests and the permeation data, it is estimated that 18%, 7%, and 84% of the freeze-fractured NCC, CC, and sand columns would be exposed to the influent, respectively, under a full E. coli breakthrough condition. Our data show that the high bacterial retention capacity of clay barriers can be compromised by freeze–thaw conditions.  相似文献

Bacterial cells that enter the groundwater system commonly experience desiccation stresses (i.e., bacterial cells are directly exposed to air) when traveling through the unsaturated layer of soil. Little is known about the effects of desiccation on the transport of bacterial cells in the groundwater system. In this research, we investigated the transport of desiccated and non-desiccated Escherichia coli K12 (ATCC 10798) cells through saturated sand packs using laboratory column transport experiments. Cell desiccation was performed at 25°C under relative humidity (RH) levels of 22%, 53%, 75%, and 97%, respectively, and the desiccation duration was 22 h. Our results showed that desiccation reduced the viability of E. coli cells under all RH levels and increased the transport of E. coli cells under ≥75% RH levels. The increase in the transport of the desiccated E. coli cells was not related to changes in cell size or cell zeta potential. Desiccation under high (i.e., ≥75%) RH levels, however, led to lower cell hydrophobicity, which was found to be positively correlated with cell transport.  相似文献

This study reports on the attachment preference of a faecally derived bacterium, Escherichia coli, to soil particles of defined size fractions. In a batch sorption experiment using a clay loam soil it was found that 35% of introduced E. coli cells were associated with soil particulates >2 μm diameter. Of this 35%, most of the E. coli (14%) were found to be associated with the size fraction 15–4 μm. This was attributed to the larger number of particles within this size range and its consequently greater surface area available for attachment. When results were normalised with respect to estimates of the surface area available for bacterial cell attachment to each size fraction, it was found that E. coli preferentially attached to those soil particles within the size range 30–16 μm. For soil particles >2 μm, E. coli showed at least 3.9 times more preference to associate with the 30–16 μm than any other fraction. We report that E. coli can associate with different soil particle size fractions in varying proportions and that this is likely to impact on the hydrological transfer of cells through soil and have clear implications for our wider understanding of the attachment dynamics of faecally derived bacteria in soils of different compositions.  相似文献

For bacterial inoculants to be effective in soil remediation, the bacterial strain must be capable of overcoming any negative effects of soil minerals on cellular processes. One class of minerals commonly encountered by soil bacteria is clays. Thus, the effect of commonly occurring clay minerals in soils on starvation, survival and 2-hydroxypyridine catabolism by Arthrobacter crystallopoietes was evaluated. Stationary phase A. crystallopoietes cells were suspended in 0.03M, pH7.0, phosphate buffer containing no clay or amended with 0.2% (wt/vol) montmorillonite, sodium montmorillonite or kaolinite. Marked effects of clay minerals on both survival rates and catabolic rates of 2-hydroxypyridine were noted. For example, after 14 weeks starvation, 4.6% of the initial cell population was viable with no clay present, compared to 0.8% (montmorillonite), 22.1% (kaolinite) and 54.1% (sodium montmorillonite) in the presence of the clay minerals. Acclimated and nonacclimated cell populations were used to evaluate 2-hydroxypyridine catabolism. Induction of 2-hydroxypyridine metabolism occurred in the unacclimated cells following starvation. Differential impact of the clay minerals on unacclimated cells was detected. Montmorillonite enhanced the capacity for induction of 2-hydroxypyridine catabolism and its decomposition rate after 0–3 days starvation. For acclimated cells, clay did not affect the metabolic activity prior to starvation, but the presence of clay resulted in increased activity during starvation. For example, after 3 days starvation, a nearly two fold increase in metabolism was detected in the presence of clay minerals. These data suggest that some clay minerals in soil alter the survival time and metabolic activity of soil-amended bacteria, thereby affecting the potential for bioremediation success. Summary The leaching of a genetically modified Pseudomonas fluorescens through soil was investigated using intact (undisturbed) soil microcosms over a 2-month period. The microcosms comprised large cylindrical cores of three contrasting soil types (a loamy sand, a sandy loam, and a clay loam) supporting a grass/clover sward. Late log-phase cells of Pseudomonas fluorescens containing lux genes encoding for bioluminescence were applied to the surface of the soil cores. Eighteen hours after application of the inocula, the microcosms were subjected to simulated rain events (9 mm per event) at 3-day intervals and leachates were analysed for the concentration of genetically modified bacteria. The lux-modified pseudomonads were detected immediately in leachate from the clay looam with a steady decline in the concentration of cells with time. Leaching of pseudomonads from the sandy loam and loamy sand only occurred over a few rain events and total recoveries from the leachate were lower than from the clay loam. Leaching patterns are discussed in relation to differences in structure of topsoil and subsoil, which determine the pathways of water flow, and to the matric potential at inoculation, which determines the pore-size classes into which cells were first introduced.  相似文献

The adsorption and binding of plasmid p34S DNA on four different colloidal fractions from a Brown soil and clay minerals in the presence of various Ca2+ concentrations, the ability of bound DNA to transform competent cells of CaCl2-treated Escherichia coli, and the resistance of bound DNA to degradation by DNase I were studied. DNA adsorption on soil colloids and clay minerals was promoted in the presence of Ca2+. Kaolinite exhibited the highest adsorption affinity for DNA among the examined soil colloids and clay minerals. In comparison with organo-mineral complexes (organic clays) and fine clays (<0.2 μm), DNA was tightly adsorbed by H2O2-treated clays (inorganic clays) and coarse clays (0.2-2 μm). The transformation efficiency of bound DNA increased with increasing concentrations of Ca2+ at which soil colloid or clay mineral-DNA complexes were formed. DNA bound by kaolinite showed the lowest transformation efficiency, and especially no transformants were observed with kaolinite-DNA complex prepared at 5-100 mM Ca2+. Compared to organic clays and fine clays, DNA bound on inorganic clays and coarse clays showed a lower capacity to transform E. coli at different Ca2+ concentrations. The presence of soil colloids and minerals provided protection to DNA against degradation by DNase I. Montmorillonite, organic clays and fine clays showed stronger protective effects for DNA than inorganic clays and coarse clays. The protection mechanisms as well as the differences in transforming efficiency of plasmid DNA molecules bound on various soil colloidal particles are discussed. The information obtained in this study is of fundamental significance for the understanding of the horizontal dissemination of recombinant DNA and the fate of extracellular DNA in soil environments.  相似文献

Multiple outbreaks of food poisoning associated with fresh vegetable consumptions have occurred in many countries. Numerous reports have described human pathogenic bacteria, such as Escherichia coli O157:H7 and Salmonella spp., that can internalize into fresh vegetables via root or leaf surfaces. While attempting to obtain the threshold concentration of internalization of E. coli inoculated into hydroponic medium during vegetable cultivation, we observed a rapid decrease in E. coli numbers. In the present study, we determined that the rapid decline in E. coli was not due to a physiological change into a viable but non-culturable (VNC) state. The population crash was instead caused by true bacterial death, as the rapid descent was also confirmed by micro-colony fluorescence in situ hybridization, a culture-independent method that can detect VNC cells. We next monitored the number of E. coli inoculated into intact or filter-sterilized hydroponic medium after cultivation of various types of plants. We found that the number of E. coli in intact hydroponic medium decreased markedly, whereas the level in filter-sterilized hydroponic medium was completely unchanged. This result suggests that biotic factors were present that could be eliminated by filtering. Robust predation of E. coli by protozoa (ciliates and flagellates) was observed using fluorescently labeled bacteria incorporated into the hydroponic medium. Finally, morphological identification of flagellates by scanning electron microscopy revealed the presence of a species of Stramenopiles. These findings suggest the importance of protozoa as bacterial feeders in hydroponic systems and hence the use of these organisms as potential control agents of human pathogenic bacteria.  相似文献

A purified allophane clay fraction increased the respiration of Escherichia coli (ATCC 11303B) by buffering the suspension against decreases in pH. Allophane had no significant effect on respiration in the presence of 0.02 M phosphate buffer. When E. coli was exposed to pH values below 5.0 in the presence of allophane, allophane supernatant (which contains soluble Al), or soluble Al. toxicity was evident even after raising the pH to 7.0.  相似文献

A study is presented of the self-diffusion of sodium in a sub-soil. Impedance factors (fs)associated with the surface phase have been derived, representing the mobility of sorbed sodium ions relative to mobility in an ideal solution. Effects of drying, of cationic com-position and of enrichment with either native clay fraction or ‘pure’ clay minerals were investigated. The liquid phase impedance factor (fL) without added clay was 0.29: clay enrichment affected soil structure and hence fL. Values of fs were in the range 0.19–0.05, and were about one-third of fL. The value off, decreased as clay content increased, being halved when 50% native clay or 10% clay mineral was added. In a sodium saturated soil fs was 0.06, less than half that in a calcium saturated soil.  相似文献

Clay movement and its effect on the differences in texture between the A and B horizons are expressed by mathematical equations. The factors which are considered are the actual amount of clay which has moved (l), the ratio of the thick nesses of the horizons (N), and the original clay content (C). Parent materials are assumed to be homogeneous. Knowing the clay contents of the A and B horizons, it is possible to calculate for given N ratios the theoretical amount of clay which has moved in order to achieve the observed textural difference. The theoretical clay content C'of the parent material can also be computed. All computations assume homogeneous parent materials and closed profiles, i.e. materials that do not collapse, shrink, or swell, and consider all other clay transformations non-operative. An eluviations index k, which indicates the proportion of the original clay that has been translocated, is proposed for evaluating the intensity of clay migration. Examples of practical applications are given: i.e. the required difference in clay content between A and B in the definition of the argillic horizon should not be kept constant in soils which have variable initial clay contents; graphs illustrate the effects of clay migration for constant C and k values (isoparentic and isoluvic lines). Changes in permeability of a soil containing montmorillonite, and the swelling of orientated aggregates of extracted clay have been measured for a range of exchangeable sodium percentages and electrolyte concentrations. The concentrations at which clay moved out of the soil in the percolates have been found and compared with the concentrations at which aggregates dispersed in stationary solutions, and the concentrations at which soil suspensions dispersed when shaken. The results indicate that: (a) permeability begins to decrease at the same concentration as the clay begins to swell, (b) the changes in permeability are directly controlled by the swelling of clay until clay dispersion and movement begins, (c) the concentration at which clay disperses depends on the mechanical stress applied, (d) when small mechanical stresses are applied, the proportion of the clay which swells and disperses depends directly on the exchangeable sodium percentage, and (e) large mechanical stresses may disperse most of the clay even at low exchangeable sodium percentages.  相似文献

Testing compost for pathogens can be very difficult, time consuming and expensive. A method to purify DNA from compost for amplification by polymerase chain reaction (PCR) was sought. The published techniques used to purify DNA from soils or sediments did not work with compost samples in our laboratory. We have devised an extraction protocol specifically for compost samples. In a series of Escherichia coli-spiked compost samples, ranging from 108 to 103 organisms per gram of compost, the removal of substances that inhibit PCR amplification of DNA was accomplished by extracting the organisms from the compost and treating the extract with base resin. After cell lysis, the collected DNA was further purified on a 1 percent low melt agarose gel. A 260 base-pair region of the lac Z gene, unique to E. coli, was successfully amplified by PCR in the DNA extracted from the gel. The technique was sensitive to the level of 104 E. coli cells per gram of compost.  相似文献

用一次平衡实验和柱淋溶实验研究了秸秆生物质炭和铝改性秸秆生物质炭对水中大肠杆菌的吸附量和去除率。结果表明,未改性秸秆生物质炭对大肠杆菌的吸附量很低,铝改性秸秆生物质炭对大肠杆菌有很高的吸附容量和去除率,0.6 mol/L Al(Ⅲ)改性生物质炭对大肠杆菌的去除效果优于0.3 mol/L Al(Ⅲ)改性生物质炭。当大肠杆菌浓度低于0.63 mg/ml时,铝改性大豆秸秆炭对大肠杆菌的去除率达100%,铝改性花生秸秆炭和铝改性稻草炭对大肠杆菌的去除率在96%和90%以上。0.5 mg/ml的大肠杆菌悬液通过装有2 g铝改性稻草炭的淋溶柱,每次收集10 ml淋出液,共收集152次,大部分淋出液中大肠杆菌浓度小于0.02 mg/ml,大肠杆菌去除率在95%以上。因此,铝改性生物质炭可用于水体中大肠杆菌的去除。  相似文献

Fahlerden dominate large areas of the young and old moraine landscapes of N and E Germany. Fahlerden (part of Fahlerden corresponds to Albeluvisols) and their transitional subtypes are supposed to have a higher intensity of clay illuviation than Parabraunerden (Luvisols). Besides this macroscopic feature reflecting periglacial influence, micromorphological features such as lenticular platy microstructure, vesicles, and fragments of clay coatings in Bt streaks document the initiation of Fahlerde genesis, which began in the Late Glacial. A model of Fahlerde genesis chronologically connects sedimentological and periglacial processes, vegetation development, and soil‐forming processes like decalcification, clay illuviation, and humification. The classification criterion of larger differences in clay contents between E and Bt horizons to distinguish Fahlerden from Parabraunerden needs to be reconsidered, because most Fahlerden have developed in stratified parent material in periglacially influenced landscapes. The interpretation of a soil data base listing data of both soil types distributed in Brandenburg demonstrates that the difference in clay contents between E and Bt horizons may even be smaller in Fahlerden than in Parabraunerden.  相似文献

Abstract. This article examines some of the factors that influence the relative risk of Escherichia coli pollution of surface waters from grazing animals compared to cattle slurry application. Drainage water from pipe‐drained plots grazed with sheep (16 sheep + lambs per hectare) from 29 May to 17 July 2002 had average E. coli counts of 11 c.f.u. mL?1 or 0.4% of estimated E. coli inputs over the grazing period. Drainage water from plots on the same site treated with cattle slurry (36 m3ha?1 on 29 May 2002) had lower average E. coli counts of 5 c.f.u. mL?1 or 0.03% of estimated faecal input. Sheep (16 lambs per hectare) grazing under cooler, moister conditions from 24 September to 3 December 2001 gave drainage water with much higher average E. coli counts of 282 c.f.u. mL?1 or 8.2% of estimated input, which is more than twice the average E. coli counts previously reported under such conditions (Vinten et al. 2002 Soil Use and Management 18, 1–9). Laboratory studies of runoff from soil slabs after slurry application showed that the mobility of E. coli in surface soil decreased with time, suggesting that increased attachment to soil or migration to ‘immobile’ water also provides at least part of the physical explanation for the relatively higher risk of pollution from grazing animals compared with slurry. Sampling for E. coli in field drainflow and in streamwater during a storm event in the predominantly dairy Cessnock Water catchment, Ayrshire, Scotland supported the hypothesis that E. coli transport is linked to grazing animals. For a 7‐mm rainfall event, roughly 14% of the estimated daily input from grazing livestock was transported to the river, even though little slurry spreading had occurred in the catchment in the previous month. Spot sampling of field drains in grazed fields and silage fields in the same catchment also showed that grazing animals were the principal source of E. coli and faecal streptococci.  相似文献

Contamination of food and water by microorganisms from animal manure has become an important issue in public health. Escherichia coli O157:H7 is one of several emerging pathogens of concern. In this research, we studied how the self-heating, thermophilic phase of composting influenced laboratory-grown vs. bovine-derived E. coli O157:H7 mortality, specifically the relationship between temperature, time at temperature, and pathogen survival. Composting experiments were conducted in laboratory-scale bioreactors operated in three temperature ranges: 40°C to 50°C, 50°C to 60°C, and greater than 60°C. We measured the effects of temperature and composting time on E. coli O157:H7 mortality. Laboratory-grown E. coli O157:H7, inoculated into the initial compost material, were not detected after approximately 300 degree days of heating. In several experiments where compost temperatures did not rise above 50°C, an initial decline of E. coli O157:H7 with subsequent regrowth was observed. E. coli O157:H7 in compost materials from infected cattle were not detected after approximately 180 degree days of heating. Numbers of total coliform bacteria declined with temperature similarly to those of E. coli O157: H7. The results of this research provide information for reducing or eliminating E. coli O157:H7 in animal wastes.  相似文献

A field study was undertaken to determine the effects of different plant species on soil microbial biomass and N transformations in a well drained silty clay loam (Typic Dystrochrept) and a poorly drained clay loam (Typic Humaquept). The crop treatments were faba bean (Vicia faba L.), alfalfa (Medicago sativa L.), timothy (Phleum pratense L.), bromegrass (Bromus inermis L.), reed canarygrass (Phalaris arundinacea L.), and wheat (Triticum aestivum L.). Measurements of microbial biomass C, denitrification capacity, and nitrification capacity were performed periodically in the top 2–10 cm of soil. On most sampling dates, all three parameters were higher under perennial than under annual species. The nitrification capacity was positively affected by the level of N applied to each species (r=0.65** for the silty clay loam and 0.84*** for the clay loam) and not directly by the plant. The differences found in microbial biomass C were significantly correlated with the water-soluble organic C present under each plant species (r=0.74*** for the silty clay loam and 0.90*** for the clay loam), suggesting differences in C deposition in the soil among plant species. In the silty clay loam, the denitrification capacity was positively related to the amount of organic C found under each plant species, while in the clay loam, it was dependent on the amount of N applied to each species. There was less denitrification activity per unit biomass under legume species than under graminease, suggesting that, depending on their composition, root-derived materials may be used differently by soil microbes.  相似文献

Growth of a nonpathogenic E. coli strain (K12- MG1655, ATCC 700926) in aerated and nonaerated compost teas containing molasses, kelp and carrot juice was examined. Teas were prepared using four different compost types that had undetectable levels of indigenous E. coli. Three of the composts were produced by turn pile windrow composting method using dairy, swine and horse manure as feedstock, while the fourth, a vermicompost, was produced by feeding separated dairy solids to worms Eisenia feotida. Molasses and kelp enhanced the growth of E. coli in inoculated teas and the E. coli density was positively correlated with nutrient concentrations ranging from 0.1 to 8.0 g/L. Irrespective of the presence of molasses and kelp, E. coli was not detected in noninoculated teas. Even though E. coli is a facultative anaerobe, its growth was significantly higher in nonaerated teas than in aerated teas. Without aeration, dissolved oxygen in teas declined rapidly and fell below 0.1 mg/L within 20 h, whereas continuous aeration at 0.8 L/min maintained an aerobic condition (> 5 mg/L dissolved oxygen) in teas during the 48 h brewing period. The pH values of nonaerated teas were significantly lower than those of aerated teas and were always slightly acidic. E. coli growth in different compost types was significantly different. The density of E. coli was lowest in teas made with vermicompost and highest in teas made with swine manure compost. E. coli proliferations in both aerated and nonaerated swine manure compost teas were inhibited by carrot juice. Carrot juice lowered dissolved oxygen in aerated teas. The total bacterial densities in noninoculated compost teas were not reduced by carrot juice.  相似文献

Several studies have investigated the adsorption of pure DNA on soil particles and its transformation ability. However, the presence of not purified (dirty) rather than pure DNA is more common in the extracellular soil environment. For this reason, we have investigated the adsorption and binding of dirty DNA on montmorillonite (M) and kaolinite (K) and their transforming ability in comparison to pure DNA. After lysis of Bacillus subtilis cells, induced by KCl, dirty DNA was characterized by the presence of cellular wall debris (cwd) and other constitutional organic components (coc). The dirty DNA was then divided into two fractions, one with cellular wall debris (DNA +cwd) and the other without cellular wall debris (DNA −cwd). B. subtilis BD 1512 (Cmr) and BD 170 (Cms) were selected as donor and recipient bacteria, respectively, for adsorption and transformation studies. Both cwd and coc seem to facilitate the adsorption of DNA to clay minerals, whereas only cwd promote the DNA binding on clays, protecting also the DNA fragments below 400 bp against nucleases. Both dirty DNA fractions adsorbed and bound on clay minerals were able to transform competent cells.  相似文献

During animal waste agricultural applications, the major concern is pathogen spreading, which may contaminate surface water and groundwater. Among the pathogenic microorganisms found in animal waste, Salmonella typhimurium and Escherichia coli O157:H7 are of particular concern. When transported in sub‐surface agricultural soil, S. typhimurium and E. coli O157:H7 are captured at the air–water–sediment interfaces through physical interactions. Because in situ colloids contribute to the formation of air–water–sediment, their mobilization affects the transport of S. typhimurium and E. coli O157:H7. The impact of irrigation rates on in situ colloid mobilization and S. typhimurium and E. coli O157:H7 transport was investigated in intact soil columns collected from an agricultural site in Gadsden County of Florida, USA. The columns were irrigated with sterilized nano‐pure deionized water to mobilize the colloids in the soil by stepwise increases in flow rate. For each flow rate, after colloids were mobilized and steady state was reached, S. typhimurium and E. coli O157:H7 were introduced. The cumulative amount of released in situ colloids increased linearly with the irrigation rates (R2 = 0.986–0.996) and transport of the bacteria was enhanced after colloid mobilization. Interactions of the bacteria with the sediments and the air‐water interface were characterized: these played an important role in controlling S. typhimurium and E. coli O157:H7 retention in soil.  相似文献
