[Objective] Pharmacognostic identification of Tetracera asiatica was performed. [Methods] Methods of morphological identification,microscopic identification,physical and chemical identification were used. [Results] Wheel stripes were obvious in the transverse section of root; the cortex was narrow and the xylem was broad. It was in radial pattern,the splitting was obvious and there were many ostioles.Needle crystal bundles of calcium oxalate were commonly observed in parenchymal cells. Lignified parenchymal cells could be observed,clear pits were densely distributed. The bordered pit vessels were with diameter of 20- 80 μm. Cork cells were in the shape of polygon and reddish brown. Tannin and flavonoid were contained in tetracera asiatica. [Conclusions] The aforementioned characteristics could provide reference basis for identification of Tetracera asiatica.  相似文献

[Objective] To screen optimum conditions of assay method and thin-layer chromatography was used for qualitative discrimination of astragaloside Ⅳ contained in Jin Weng Zhili particle,which provide basis for quality standard. [Methods] According to method described in Chinese Veterinary Pharmacopoeia,thin-layer chromatography was used for qualitative discrimination of astragaloside Ⅳ contained in Jin Weng Zhili particle by improving extraction method and adjusting the proportion of ingredients in developing solvent. [Results] For identification of astragaloside Ⅳ in Jin Weng Zhili particle using thin-layer chromatography,the results were the optimum after chromatographic condition was improved,ie,extraction,column transferring,developing solvent of trichloromethane-methyl alcohol-water( 10 ∶ 5 ∶ 1) which is better than method described in Chinese Veterinary Pharmacopoeia. [Conclusions] The method is of good feasibility,repeatability and specificity which is benefit to the identification of astragaloside Ⅳ contained in Jin Weng Zhili particle using thin-layer chromatography.  相似文献

[Objective] To research on the protective effects of total flavonoids in Polygonum perfoliatum L. on the acute liver injury of mice induced by CCl4 and its action mechanism. [Methods]Flavonoids were extracted from P. perfoliatum by chemical method. Acute liver injury model of mice was established by intraperitoneal injection of CCl4 peanut oil. The biochemical indexes of ALT,AST,TNF-α,IL-1β and IL-6in mice serum were detected. Histopathological examination of liver tissue was carried out. [Results] Flavonoids in P. perfoliatum restricted the ALT,AST,TNF-α,IL-1β and IL-6 activities in mice serum with acute liver injury( P 0. 01,P 0. 05). Results of histopathological examination also showed that flavonoids significantly improved the liver injury of mice. [Conclusions] Flavonoids in P. perfoliatum showed protective effects of acute liver injury of mice induced by CCl4; and its mechanism might be related with anti-inflammation.  相似文献

[Obejective]To observe the antipyretic effect of total organic acid in Thladiantha Dubia fruit(TOATDF),and explore its mechanism. [Methods]The rat fever models were established by the injection to SD rats with LPS 100 μg /kg of their body weights. The model rats were randomly divided into TOATDF 300,150 and 75 mg /kg dosage groups,aspirin 150 mg /kg control group and model group,and another10 rats were taken as normal control. Body temperature was measured once every one hour after the medicine,and for six times in a row.ELISAE method was used to determine the expression level of PGE2,TNF-α and IL-β in serum and that of PGE2 in cerebrospinal fluid. [Results]The average body temperature of rats at any point of time in TOATDF 300 and 150 mg /kg dose group of rats was lower than that of rats in the model control group(P 0. 05 or P 0. 01); temperature of rats in the 75 mg /kg group 2- 5 h after the drug was significantly lower than that of rats in the control group(P 0. 05); temperature of rats after 1- 5 h in aspirin group after drug was obviously lower than that of rats in the control group. The expression level of TNF-α,PGE2,IL-β in serum and that of PGE2 in cerebrospinal fluid in each dosage group were significantly lower than that in model control group( P 0. 05 or P 0. 01),but higher than that in the normal control group( P 0. 05). [Conclusions]TOATRF has significant antipyretic effect with dose-dependent. Its the antipyretic effect is related to the inhibition of the increase of PGE2,TNF-α and IL-β induced by pyrogen.  相似文献

[Objective] To study the effect of dry methods on the content of ligustilide in Miwu. [Methods]The content of ligustilide in Miwu. dried by different methods was determined by HPLC. [Results]There was a good linear relationship( r = 0. 999 9) between ligustilide and peak area when the sample injection was 0. 019 97- 0. 199 7 μg /mL,the average recovery rate was 100. 2% and RSD was 1. 09%(n =9). Different dry methods had a certain degree of effect on the ligustilide content in Miwu. [Conclusions]Oven-drying after drying in the sun is the best among the three different dry methods.  相似文献

[Objective] To establish the fingerprint detecting standard for RHIZOMA COPTIDIS in Sichuan. [Method] Using berberine hydrochloride as the reference substance, the separation was carried out on a SinoChrom ODS-BP (250 mm×4.6 mm, 5 μm) with the detection wavelength at 345 nm. Following the injection of sample solution, a gradient elution with methanol-0.1% caproic acid and 10 mmol/L ammonium acetate as mobile phase was initiated at a flow rate of 1.0 ml/min. [Result] A total of 14 common peaks were found in the chromatogram of RHIZOMA COPTIDIS, the parameter of which met with the stipulation of Chinese Medicine Injection Fingerprint Technical Requirements. [Conclusion] The method is simple and accurate with a good reproducibility and can be used in quality evaluation and variety discrimination of RHIZOMA COPTIDIS in Sichuan.  相似文献

[Objective] This study was undertaken to extract flavonoids from cherry leaves and to investigate the anti-oxidative activity in vitro. [Methods]The scavenging action on OH·and O-2·and its inhibiting effect on oxidative hemolysis of erythrocyte induced by H2O2and lipid peroxidation of hepatic tissue in mice were investigated in vitro. [Results] The flavonoids showed concentration-dependent scavenging effect on OH·and O-2·. The flavonoids also showed remarkable inhibiting effects on oxidative hemolysis of erythrocyte in mice induced by H2O2. When the final concentration of the flavonoids reached 30%,the inhibition rate was 74. 13%. It could inhibit lipid peroxidation of hepatic tissue in mice induced by vitamin C with low concentration and ferrous sulfate. And the inhibiting effect was concentration-dependent.When the final concentration of the flavonoids reached 30%,the inhibition rate was 68. 33%. [Conclusions]The total flavonoids from cherry leaf have concentration-dependent antioxidant activity in vitro.  相似文献

[Objective]To determine the content of rutin and quercetin in the whole plant of Yantai Bupleurum by HPLC. [Methods] An Agilent C18column( 250 mm × 4. 6 mm,5 μm) was applied with a mobile phase of acetonitrile- 0. 1% phosphoric acid solution,gradient elution was selected with the flow rate of 1. 0 mL / min,the UV detector was set at 360 nm. [Results]The average recovery rates of rutin and quercetin were 100. 47% and 99. 50%,the RSD were 1. 34% and 0. 41%. The average content of rutin and quercetin in the whole plant of Yantai Bupleurum reached 83. 00 and 102. 68 μg / g. [Conclusions]The method is fast,accurate and reliable. It is suitable for content determination of rutin and quercetin in the whole plant of Yantai Bupleurum.  相似文献

[Objective] To detect the 22 inorganic elements in " Zuota" grown in Tibet,Qinghai,Gansu and Sichuan Provinces of China; and to analyze their dissolution situation in artificial gastric fluid and artificial intestinal fluid. [Methods] After processed by microwave digestion and treated in artificial gastric fluid and artificial intestinal fluid,22 inorganic elements in " Zuota" were detected by ICP-OES method. [Results] The 22 inorganic elements in " Zuota" showed good linear relationship within the range of 0- 20 μg / m L(R2 0. 999 4). Hg had the maximum content in " Zuota" among metallic elements. Total Hg had the maximum content in " Zuota" grown in Tibet,followed with Qinghai and Gansu. And total Hg content was the minimum in Aba of Sichuan. Most of the elements had higher dissolution rates in artificial gastric fluid than those in artificial intestinal fluid. Therefore,it could be concluded that the dissolution part in " Zuota" was mainly the stomach. [Conclusions] This method could detect many elements at the same time with wide linear range,which was rapid,accurate,simple and sensitive,and could be used for the detection of 22 inorganic elements in " Zuota".  相似文献

[Objective] To establish the detection method of total flavonoids contents in Rumex acetosa L. by UV spectrophotometry,and to detect the total flavonoids contents in different parts of R. acetosa. [Methods] Effects of different factors on the extraction rate of total flavonoids from R. acetosa were researched by single factor test,which were extraction solvent,extraction method,extraction time,solid-liquid ratio and volume fraction of methanol. The optimal extraction technology of total flavonoids from R. acetosa was screened. Contents of total flavonoids were detected in different parts of R. acetosa.[Results] The optimal extraction technology of total flavonoids from R. acetosa was as follows: 70% methanol,1∶ 40 solid-liquid ratio,and hot reflux extraction for 60 min. Contents of total flavonoids in R. acetosa were the maximum in flowers and fruits.[Conclusions]The detection method for total flavonoids in R. acetosa was simple and accurate. Except the roots of R. acetosa,flowers and fruits also had high contents of total flavonoids,which could be used as the medicinal parts for the extraction of total flavonoids.  相似文献

[Objective]To research on the changes of active ingredients in black beans during vinegar soaking and its antioxidant properties.[Methods]The protein,peptide and polyphenol contents in vinegar-soaked black beans were researched by chemical extraction method. The antioxidant properties of peptide and polyphenol in black beans by different soaking times were detected by establishing DPPH· system,·OH system and reducing power system. [Results]During the process of vinegar soaking,protein content reduced,and decreased by 37. 5% at the21 stday of vinegar soaking. The contents of fat and polyphenol enhanced significantly,which increased by 19. 7% and 72. 4% on the 28 thday,respectively. Changes of peptide content was relatively complex,but showed an increasing trend in general. During the earlier stage of vinegar soaking,clearance effects of peptides and polyphenols in black beans on ·OH and DPPH· enhanced significantly,but became stabilized in the later period. In the three systems,the antioxidant properties of peptides were all stronger than those of polyphenols. [Conclusions]This research provided scientific references for the black bean used as healthcare food.  相似文献

[Objective]To investigate the effect of different temperature on the germination of fire hemp seeds,so as to getting the optimal temperature for the germination of fire hemp seeds. [Methods] By setting 10 temperature gradients between 0- 45 ℃ in constant incubators in laboratory,three indicators including the germination vigor,germination rate and germination index were measured and processed by arcsine transformation for significance analysis. [Results]The results showed that fire hemp seeds germinated at the temperature between 5- 35 ℃.The highest germination rate was 95% at 20 ℃; and the germination vigor and germination index of fire hemp seeds reached the peak of92. 3% and 40. 3% at 25 ℃ respectively. The germination rate descended dramatically with the increase or decrease of treatment temperature.Analysis of variance( ANOVA) showed that there was no significant difference in germination vigor and germination rate between 20 and 25℃,but did in germination index. [Conclusions]The temperature suitable for the germination of fire hemp seeds ranges from 5- 35 ℃,with the optimal one of 25 ℃.  相似文献

[Objective]To explore the radiosensitization and radiation protection effects of aged black garlic extract on colon cancer Ht-29 in mice induced by60Co γ ray and to primarily discuss its molecular mechanism. [Methods] Transplant subcutaneous sarcoma model of colon cancer Ht-29 in mice was constructed. 40 tumor-bearing mice were divided into 4 groups: blank control group,γ ray group,black garlic group and black garlic combined with γ ray group. Mice in these groups were treated with NS,γ ray,black garlic extract and black garlic extract combined with γ ray. The growth rate of transplantation tumor in various groups were observed and also the tumor growth delay of transplantation tumor in treatment groups. Eyeballs were removed to achieve blood and then mice were put to death by break necks. Tumors were obtained and weighed. The growth inhibition ratio of tumor was calculated. The expression of Bcl-2,Bax and Caspase-9 was determined by Real-time-PCR. The validity of SOD and CAT was measured by white blood count and UV spectrophotometry. [Results]Black garlic extract combined with γ ray had growth inhibition effect on transplantation tumor in colon cancer Ht-29 mice. It could prolong the time of tumor growth and increase the radiosensitivity. It could also down-regulate the expression of Bcl-2 in transplantation tumor tissue and up-regulate the expression of Bax. As a result,it could enhance the expression of Caspase-9 protein( P 0. 05). Black garlic extract could increase the number of peripheral white blood cells and enhance the validity of SOD and CAT. [Conclusions]Black garlic extract could increase the radiosensitivity of γ ray on colon cancer Ht-29 transplanted tumor in mice by down-regulation of Bcl-2,up-regulation of Bax and activating the expression of Caspase-9 protein.It could also enhance radiation protection by increasing the number of peripheral white blood cells and the antioxidant ability.  相似文献

[Objective] To establish a rapid analysis method for volatile components in RADIX ADENOPHORAE,to analyze and compare the differences of RADIX ADENOPHORAE in different production areas,and to obtain the characteristic distribution of volatile components and the rapid information identification mode of production areas. [Methods] Volatile components were extracted from RADIX ADENOPHORAE by solid phase microextraction( SPME),and were analyzed by GC-MS. The relative content of each component was calculated by peak area normalization method; and statistical analysis was carried out by SPSS 18. 0 software based on principal component analysis( PCA) and cluster analysis( CA). [Results] A total of 41 chemical components of RADIX ADENOPHORAE were identified from RADIX ADENOPHORAE grown in different areas,mainly including aldehydes,terpenes,organic acids and alcohols. Among them,isovaleraldehyde,hexanal,α-pinene and caprylic aldehyde were common volatile components in RADIX ADENOPHORAE. Besides,RADIX ADENOPHORAE grown in Shennongjia and Jiangsu had abundant terpenoids. [Conclusions]The established method was simple and was suitable for the rapid identification and analysis of RADIX ADENOPHORAE. PCA and CA could effectively distinguish RADIX ADENOPHORAE grown in different areas. The production areas were divided into five categories by cluster analysis. [Objective]To investigate the effects of saikosaponin a(SSa) of RADIX BUPLEURI on the proliferation of human normal liver cell L-02 and its mechanism,and to provide theoretical basis for clinical application of RADIX BUPLEURI. [Methods]Morphological changes were observed and MTT assay was performed to research the toxicity effects of SSa on human liver cell L-02 in different time periods and doses. DNA ladder analysis and Annexin V /PI double staining were both used to detect if cell apoptosis was induced. The contents of SOD,MDA and LDH in supernatant were detected to further research the mechanism of hepatotoxicity induced by SSa. [Results] SSa of different concentrations were all dose dependent to cytotoxicity,but had no relationship with administration time. IC50 values of SSa after administration for 24 and 48 h were 5. 47 and 5. 22 μmol /L,respectively. Annexin V /PI double staining could not detect the early apoptotic cells. Detection results of cell culture liquid showed that SOD activity reduced,while MDA and LDH contents enhanced. [Conclusions] The mechanism of hepatotoxicity in vitro of SSa was not apoptosis induction,but oxidative injury of human liver cell. SSa reduced the activity of SOD,which resulted in cellular membrane injury and finally caused cytotoxicity.  相似文献

[Objective]To determine the content of aesculin in the leaves of Fraxinus chinensis. [Methods]The sample was extracted after it had undergone reflux through absolute methanol,and the filler was the bonding of octadecyl silicon and silica gel; the acetonitrile- 0. 1%concentration phosphoric acid solution(8∶92,V /V) was taken as mobile phase,the wavelength measured was 334 nm,the column temperature was 30 ℃,and the flow velocity was 1. 0 mL /min. The content was determined via high performance liquid chromatography. [Results]The average recovery rate was 96. 59%,the RSD was 2. 39%(n = 6),and the average content of aesculin in the leaves of Fraxinus chinensis was0. 155 9 mg /g. [Conclusions]This method is of high stability,accuracy and reproductivity,and thus suitable for determining aesculin in the leaves of Fraxinus chinensis.  相似文献

A three-dimentional thermal elastoplastic contact FE analysis model of solidifiedshell in moulds for continuous casting of thin slabs is set up, based on the known geometry of theshell and the temperature distribution in the shell. We in turn obtain the stress and Strain distributions of the solidified shells formed in lSP and CSP moulds,the gap distributions between the Shellsand the mould walls,as well as the curves of cracking index [1] in the shells,which provides afoundation for the design and control of technical parameturs for continuous casting of thin slabs.  相似文献

[Objective] To establish a content determination method of gentiopicroside in different parts(roots,stems or leaves) of cultivated G. rigescens,and to provide references for the quality evaluation and medicinal part selection of cultivated G. rigescens. [Methods]Gentiopicroside contents were detected in different parts(roots,stems or leaves) of cultivated G. rigescens by high performance liquid chromatography.Chromatographic column was Hypersil C18column; mobile phase was methanol-water(70∶ 30); column temperature was 30 ℃; flow speed was1 m L / min; detection wavelength was 274 nm; number of theoretical plate was greater than 3 000 peak area; and injection volume was 10 μL.[Results]The regression equation of gentiopicroside was y = 2. 568 8 x + 0. 084 5 with r = 0. 999 4. Gentiopicroside showed good linear relationship within the range of 0. 04- 0. 24 mg / m L. Results of precision test showed that RSD of gentiopicroside was 1. 125%; results of stability test showed that RSD of gentiopicroside was 1. 587%; results of repeatability test showed that the average content of gentiopicroside in roots was 3. 633% with RSD = 1. 486%; results of recovery test showed that the recovery rate was 99. 457% with RSD = 1. 256%. Gentiopicroside contents in roots,stems and leaves of cultivated G. rigescens were 3. 633%,1. 256% and 1. 958%,respectively. [Conclusions]Gentiopicroside content in G. rigescens cultivated in Guizhou exceeded the standard of Chinese Pharmacopoeia(2005 edition)[3]. And cultivated G. rigescens in Guizhou could take the place of wild RADIX ET RHIZOMA GENTIANAE as items for sale.  相似文献
