Vitamin A deficiency is one of the major health problems worldwide. Traditional yellow maize possesses very low provitamin A (proA) concentration in endosperm. The influence of rare alleles of β‐carotene hydroxylase (crtRB1) and lycopene epsilon cyclase (lcyE) genes capable of enhancing proA concentration was studied in four BC2F2 populations generated using subtropical inbreds and CIMMYT‐HarvestPlus lines. The occurrence of severe segregation distortion for crtRB1 gene was observed, while lcyE gene was segregated as per Mendelian ratio. Genotype with favourable allele of crtRB1 (CC) had a significant effect on β‐carotene (BC) (7.9‐fold), β‐cryptoxanthin (BCX) (twofold) and proA (5.5‐fold) accumulation, compared to unfavourable genotype (C+C+). Genotype with favourable allele of lcyE (LL) showed 2.1‐fold, 1.6‐fold and twofold significant enhancement in BC, BCX and proA, respectively, over unfavourable genotype (L+L+) in pooled analysis. Of the nine genotypes, double homozygote (CC/LL) had the highest mean BC (12.60 μg/g), BCX (4.44 μg/g) and proA (14.82 μg/g), and combined effect was significantly better than individual gene effects or any other combinations. The information generated here would be useful in designing strategy for proA enrichment in subtropical maize.  相似文献

Vivek Maize Hybrid 9‐ a popular single‐cross hybrid developed by crossing CM 212 and CM 145 was released for commercial cultivation in India. The parental lines, being deficient in lysine and tryptophan, were selected for introgression of opaque‐2 allele using CML 180 and CML 170 as donor lines through marker‐assisted backcross breeding. The opaque‐2 homozygous recessive genotypes with >90% recovery of the recurrent parent genome were selected in BC2F2, and the seeds with <25% opaqueness in BC2F3 were forwarded for seed multiplication. Vivek Quality Protein Maize (QPM) 9, the improved QPM hybrid, showed 41% increase in tryptophan and 30% increase in lysine over the original hybrid. The grain yield of the improved hybrid was on par with the original hybrid. The newly improved QPM maize hybrid released in 2008 will help in reducing the protein malnutrition because its biological value is superior over the normal maize hybrids. This short duration QPM maize hybrid has been adopted in several hill states of North Western and North Eastern Himalayan regions.  相似文献

Prolificacy assumes significance for development of high‐yielding baby corn hybrids. “Sikkim Primitive” is a native landrace of North‐Eastern Himalaya, and is the highest prolific maize germplasm. So far, the genetics of prolificacy in “Sikkim Primitive” has not been deciphered. Here, a prolific inbred (MGU‐SP‐101) developed from “Sikkim Primitive” was crossed with four non‐prolific inbreds viz., LM13, BML7, HKI161 and HKI1128. Six generations (P1, P2, F1, F2, BC1P1 and BC1P2) of the crosses were evaluated at two locations during rainy season 2018. MGU‐SP‐101 possessed 2.50–3.78 ears per plant compared to 1.06–1.86 among non‐prolific inbreds. The variation for ears was the highest in F2s (1–8), followed by BC1P1 (1–7) and BC1P2 (1–5). The quantitative inheritance pattern of prolificacy with prevalence of non‐allelic interactions of duplicate epistasis type has been observed. Dominance × dominance effect was predominant over additive × additive and additive × dominance effects. Total number of major gene blocks ranged from 0.41 to 2.86, thereby suggesting the involvement of at least one major gene/QTL governing the prolificacy. This is the first report of genetic dissection of prolificacy in “Sikkim Primitive”.  相似文献

Maize is the major staple food in southern Africa with human consumption averaging 91 kg capita?1 year?1, and normal maize is nutritionally deficient in two essential amino acids: tryptophan and lysine. Despite the development of quality protein maize (QPM) with high tryptophan and lysine, stunting and kwashiorkor remain high in sub-Saharan Africa due to lack of high yielding and adapted QPM varieties. This study aimed at evaluating a new generation of QPM varieties for yield and related agronomic traits. Before the QPM varieties were validated on-farm, they were simultaneously selected on-station under five different management conditions. In the 2014/2015 season, 10 elite QPM varieties were selected from on-station trials based on high grain yield and stability, and were compared with the best commercial check varieties on-farm. During the 2015/2016 season, some poorly performing QPM varieties were dropped while new ones were added, resulting in 12 elite QPM varieties being evaluated on-farm. Analysis of variance for the 2014/2015 season showed non-significant hybrid × management condition interaction. Mean grain yields across management conditions ranged from 1.5 to 4 t ha?1 and were higher under mild stress (2.3–5.5 t ha?1) compared to random stress conditions (1.1–2.9 t ha?1). Broad sense heritability estimates were low to moderate (11–69%), and thus could still permit effective selection of better genotypes. Yield advantage ranged from 12 to 25% across the 2 years, suggesting effective genetic gains in QPM breeding. QPM hybrids CZH132044Q, CZH142238Q and CZH142236Q were stable and high yielding. Promotion of such QPM hybrids may help reduce protein energy malnutrition.  相似文献

Modifier loci in QPM play a vital role in achieving acceptable degree of kernel hardness and accumulation of lysine and tryptophan. This study was undertaken to characterize a set of diverse QPM inbreds using SSRs linked to endosperm and amino acids modifier loci for their effective utilization in the breeding programme. Significant variation was observed for endosperm modification (25–100% opaqueness), tryptophan (0.056–0.111%) and lysine (0.223–0.444%). Generally, inbreds with soft endosperm possessed more tryptophan and lysine than inbreds with higher vitreousness. SSRs generated 341 alleles with two to seven alleles per locus. The frequency of unique and rare alleles was more for amino acid modifications, compared to endosperm modifications. Phylogenetic analyses grouped the inbreds into three major clusters, and the study identified suitable crosses for accumulation of endosperm and amino acids modifiers. QPM inbreds with desirable modifications identified here would serve as suitable donor for both opaque2 and modifier loci in the marker‐assisted backcross breeding. Further, contrasting inbreds can be used for generating mapping populations to identify new modifier loci underlying both endosperm and amino acids modifications.  相似文献

The peach root‐knot nematode, Meloidogyne floridensis (MF), infects majority of available nematode‐resistant peach rootstocks which are mostly derived from peach (Prunus persica) and Chinese wild peach (P. davidiana). Interspecific hybridization of peach with its wild relative, Kansu peach (P. kansuensis), offers potential for broadening the resistance spectrum in standard peach rootstocks. We investigated the inheritance of resistance to MF in segregating populations of peach (‘Okinawa’ or ‘Flordaguard’) × P. kansuensis. A total of 379 individuals from 13 F2 and BC1F1 families were challenged with a pathogenic MF isolate “MFGnv14” and were classified as resistant (R) or susceptible (S) based on root galling intensity. Segregation analyses in F2 progeny revealed the involvement of a major locus with a dominant or recessive allele determining resistance in progeny segregating 3R:1S and 1R:3S, respectively. Testcrosses with a homozygous‐susceptible peach genotype (‘Flordaguard’ or ‘UFSharp’) confirmed P. kansuensis as a source of new resistance and the heterozygous allelic status of P. kansuensis at the locus conferring resistance to MF. We propose a single‐locus dominant/recessive model for the inheritance of resistance.  相似文献

Soybean mosaic virus (SMV) can cause serious yield losses in soybean. Soybean cultivar ‘RN‐9’ is resistant to 15 of 21 SMV strains. To well‐characterize this invaluable broad‐spectrum SMV‐resistance, populations (F1, F2 and F2:3) derived from resistant (R) × susceptible (S) and R × R crosses were tested for SMV‐SC18 resistance. Genetic analysis revealed that SC18 resistance in ‘RN‐9’ plus two elite SMV‐resistant genotypes (‘Qihuang No.1’ and ‘Kefeng No.1’) are controlled by independently single dominant genes. Linkage analysis showed that the resistance of ‘RN‐9’ to SMV strains SC10, SC14, SC15 and SC18 is controlled by more than one gene(s). Moreover, Rsc10‐r and Rsc18‐r were both positioned between the two simple sequence repeats markers Satt286 and Satt277, while Rsc14‐r was fine‐mapped in 136.8‐kb genomic region containing sixteen genes, flanked by BARCSOYSSR_06_0786 and BARCSOYSSR_06_0790 at genetic distances of 3.79 and 4.14 cM, respectively. Allelic sequence comparison showed that Cytochrome P450‐encoding genes (Glyma.06g176000 and Glyma.06g176100) likely confer the resistance to SC14 in ‘RN‐9’. Our results would facilitate the breeding of broad‐spectrum and durable SMV resistance in soybeans.  相似文献

Chickpea shows a distinct domestication trajectory vis‐a‐vis pod dehiscence and growth cycle mediated by vernalization insensitivity compared with its companion Near Eastern legumes. Our objectives were: (i) to map the quantitative trait loci (QTLs) associated with vernalization response and seed free tryptophan in domesticated × wild chickpea progeny and (ii) estimate the genetic correlation between vernalization response and free tryptophan content. A domesticated × wild chickpea cross was used to document phenotypic segregation in both traits and to construct a skeletal genetic map for QTL detection. A number of vernalization response and seed free tryptophan content QTLs were documented in both F2 and F3 generations. No significant genetic correlation between these two traits was observed. Epistatic relationship between two free tryptophan loci was documented. It is evident that selection for high seed tryptophan is easier to accomplish relative to selection for vernalization insensitivity. This suggests that the two traits were selected independently in antiquity, thereby corroborating earlier claims for conscious selection processes associated with chickpea domestication.  相似文献

Interspecific hybrids between Trifolium uniflorum and cultivated white clover (Trifolium repens) have highly useful characteristics for temperate pastoral systems derived from both parent species. However, the early hybrids (F1 and BC1) also have unacceptably poor seed production for commerce. This study analysed the basis for the poor seed production and investigated breeding strategies for overcoming the problem. The BC1F1 generation produced lower‐than‐expected numbers of heads per plant and seeds per floret. Backcrossing of selected hybrids to white clover corrected these deficiencies and created new variation. Seed numbers were also returned to near target levels by recurrent selection within the BC1 generation. Thus, it was possible to retain a theoretical average of 25% of T. uniflorum genome and still achieve high seed production per plant. The BC1F2 and BC2F1 generations produced high seed numbers per plant, along with reasonable variation. Both of these second‐generation hybrid forms have high reproductive potential and should be the focus for the selection of the desired combinations of agronomic and seed production traits.  相似文献

Ogura cytoplasmic male sterility (Ogu-CMS) is widely used in the production of commercial hybrids of Brassica oleracea. However, the widespread application of the Ogu-CMS system in B. oleracea has hindered the germplasm innovation of Ogu-CMS resources due to the lack of a natural restorer line. Previously, the Ogu-CMS fertility-restored interspecific hybrids between rapeseed 15Y403 (2n = 38, AACC) and Chinese kale JL1 (2n = 18, CC) have been successfully produced. However, these progenies, which still contained a large proportion of rapeseed genomic components, showed poor fertility and a low seed setting rate under natural pollination. To improve fertility and seed setting, a successive backcross with JL1 was performed to produce BC2 progenies. Screening with the Rfo-specific marker, five individuals harboring the Rfo gene were identified among 98 BC2 progenies. These five individuals underwent background marker screening and an evaluation of agronomic traits and fertility. One individual (code: 15Q23) was identified with higher pollen viability, better seed setting under natural pollination, and a closer genetic background to the parent Chinese kale JL1. Many morphological traits showed no significant differences (P < 0.05) between 15Q23 and the backcross parent JL1. However, the average seed setting of 15Q23 under natural pollination was 0.72 seeds per pod, which was 50 times higher than that of BC1 progenies, and the average pollen viability was 87.07%, which was significantly better than that of the F1 and BC1 plants (P < 0.01). The genetic background of 15Q23 was more closer to the parent JL1 than that of BC1 plants and another BC2 fertility-restored individual, with 82% of the polymorphic alleles being the same as those of the parent Chinese kale JL1. Thus, the individual 15Q23 could be used as a donor plant for further backcrosses. This study lays the foundation for the development of Ogu-CMS restorer material in B. oleracea.  相似文献

Increasing productivity through improvement of photosynthesis in faba bean breeding programmes requires understanding of the genetic control of photosynthesis‐related traits. Hence, we investigated the gene action of leaf area, gas exchange traits, canopy temperature, chlorophyll content, chlorophyll fluorescence parameters and biomass. We chose inbred lines derived from cultivars 'Aurora' (Sweden) and 'Mélodie' (France) along with an Andean accession, ILB 938, crossed them (Aurora/2 × Mélodie/2, ILB 938/2 × Aurora/2 and Mélodie/2 × ILB 938/2), and prepared the six standard generations for quantitative analysis (P1, P2, F1, F2, B1, and B2). Gene action was complex for each trait, involving additive and dominance gene actions and interactions. Additive gene action was important for SPAD, photosynthetic rate, stomatal conductance and Fv/Fm. Dominance effect was important for biomass production. It is suggested that breeders selecting for productivity can maximize genetic gain by selecting early generations for canopy temperature, SPAD and Fv/Fm, then later generations for biomass. The information on genetics of various contributing traits of photosynthesis will assist plant breeders in choosing an appropriate breeding strategy for enhancing productivity in faba bean.  相似文献

Waxy (Wx) protein is a key enzyme for synthesis of amylose in endosperm. Amylose content in wheat grain influences the quality of end‐use products. Seven alleles have been described at the Wx‐D1 locus, but only two of them (Wx‐D1b, Wx‐D1e) were genotyped with codominant markers. The waxy wheat line K107Wx1 developed by treating ‘Kanto 107’ seeds with ethyl methanesulphonate carries the Wx‐D1d allele. However, no molecular basis supports this nomenclature. In the present study, DNA sequence analysis confirmed that a single nucleotide polymorphism in the sixth exon of Wx‐D1 changed tryptophan at position 301 into a termination codon. Based on this sequence variation, a PCR‐based KASP marker was developed to detect this point mutation using 68 BC8F1 plants and 297 BC8F2 lines derived from the cross ‘Ningmai 14’*9/K107Wx1. Combined with codominant markers for the Wx‐A1 and Wx‐B1 alleles, waxy and non‐waxy near‐isogenic lines were distinguished. The KASP marker was efficient in identifying the mutant allele and can be used to transfer waxiness to elite lines.  相似文献

It is crucial to develop a rapid technique for identifying sexuality in the seedling stage of persimmon (Diospyros kaki Thunb.), and the elimination of male progeny has been regarded as an important strategy for enhancing breeding efficiency. In this study, phenotype characterization and genotyping of the male‐linked OGI marker were carried out using 205 accessions, including persimmon cultivars, F1 progeny and nine related Diospyros species. All persimmon cultivars displayed consistent results regarding OGI amplification and sex phenotype. A total of 143 F1 progeny were derived from 11 crosses, among which 95 individuals had flowered. In the flowering full‐sib families, the amplification of the OGI marker in agreement with the sex phenotype was obtained in 85 plants (89.5%). The segregation of OGI in ‘Huashi 1’ × ‘Luotian Tianshi’ and ‘Huashi 1’ × Male 3 F1 populations fit a 1 : 1 ratio. Furthermore, high OGI transferability was observed in nine related species. Overall, the results indicated that the OGI locus could be used to distinguish male from female persimmon plants at an early stage.  相似文献

The number of drought and low-N tolerant hybrids with elevated levels of provitamin A (PVA) in sub-Saharan Africa could increase when PVA genes are optimized and validated for developed drought and low-N tolerant inbred lines. This study aimed to (a) determine the levels of drought and low-N tolerance, and PVA concentrations in early maturing PVA-quality protein maize (QPM) inbred lines, and (b) identify lines harbouring the crtRB1 and LcyE genes as sources of favourable alleles of PVA. Seventy early maturing PVA-QPM inbreds were evaluated under drought, low-N and optimal environments in Nigeria for two years. The inbreds were assayed for PVA levels and the presence of PVA genes using allele-specific PCR markers. Moderate range of PVA contents was observed for the inbreds. Nonetheless, TZEIORQ 55 combined high PVA concentration with drought and low-N tolerance. The crtRB1-3′TE primer and the KASP SNP (snpZM0015) consistently identified nine inbreds including TZEIORQ 55 harbouring the favourable alleles of the crtRB1 gene. These inbreds could serve as donor parents of the favourable crtRB1-3′TE allele for PVA breeding in maize.  相似文献

Heterosis, or hybrid vigour, has been used to improve seed yield in several important crops for decades and it has potential applications in soybean. The discovery of over‐dominant quantitative trait loci (QTL) underlying yield‐related traits, such as seed weight, will facilitate hybrid soybean breeding via marker‐assisted selection. In this study, F2 and F2 : 3 populations derived from the crosses of ‘Jidou 12’ (Glycine max) × ‘ZYD2738’ (Glycine soja) and ‘Jidou 9’ (G. max) × ‘ZYD2738’ were used to identify over‐dominant QTL associated with seed weight. A total of seven QTL were identified. Among them, qSWT_13_1, mapped on chromosome 13 and linked with Satt114, showed an over‐dominant effect in two populations for two successive generations. This over‐dominant effect was further examined by six subpopulations derived from ‘Jidou12’ × ‘ZYD2738’. The seed weight for heterozygous individuals was 1.1‐ to 1.6‐fold higher than that of homozygous individuals among the six validation populations examined in different locations and years. Therefore, qSWT_13_1 may be a useful locus to improve the yield of hybrid soybean and to understand the molecular mechanism of heterosis in soybean.  相似文献

The nutritional value of cucumber (Cucumis sativus L.) can be improved by the introgression of β-carotene (i.e., provitamin A and/or orange flesh) genes from “Xishuangbanna gourd” (XIS; Cucumis sativus var. xishuangbannanesis Qi et Yuan) into US pickling cucumber. However, the genetics of β-carotene content has not been clearly defined in this US market type. Thus, three previous populations derived from a US pickling cucumber (‘Addis’) × XIS mating were evaluated for β-carotene content, from which the high β-carotene inbred line (S4), ‘EOM 402-10’, was developed. A cross was then made between the US pickling cucumber inbred line ‘Gy7’ [gynoecious, no β-carotene, white flesh; P1] and ‘EOM 402-10’ [monoecious, possessing β-carotene, orange flesh; P2] to determine the inheritance of β-carotene in fruit mesocarp and endocarp tissue. Parents and derived cross-progenies (F1, F2, BC1P1, and BC1P2) were evaluated for β-carotene content in a greenhouse in Madison, Wisconsin. While F1 and BC1P1 progeny produced mature fruits possessing white, light-green, and green (0.01–0.02 μg g−1 β-carotene) mesocarp, the F2 and BC1P2 progeny mesocarp segregated in various hues of white, green, yellow (0.01–0.34 μg g−1 β-carotene), and orange (1.90–2.72 μg g−1 β-carotene). Mesocarp and endocarp F2 segregation adequately fit a 15:1 [low-β-carotene (0.01–0.34 μg g−1): high-β-carotene (1.90–2.72 μg g−1)] and 3:1 (low-β-carotene: high-β-carotene) ratio, respectively. Likewise, segregation of carotene concentration in mesocarp and endocarp tissues in BC1P2 progeny adequately fit a 3:1 (low-β-carotene: high-β-carotene) and 1:1 (low-β-carotene: high-β-carotene) ratio, respectively. Progeny segregations indicate that two recessive genes control the β-carotene content in the mesocarp, while one recessive gene controls β-carotene content in the endocarp. Single marker analysis of F2 progeny using the carotenoid biosynthesis gene Phytoene synthase determined that there was no association between this gene and the observed β-carotene variation in either fruit mesocarp or endocarp.  相似文献

Thermotolerance acclimation of photosystem II to heat and drought is well documented, but studies demonstrating developmental impacts on heat tolerance in field‐grown plants are limited. Consequently, climatic variables, estimated canopy temperature, predawn leaf water potential (ΨPD), and the temperature responses of maximum quantum yield of photosystem II (Fv/Fm), variable fluorescence (Fv/F0), quantum yield of electron transport (φEο) and efficiency of PSI electron acceptor reduction (REο/ABS) were characterized for Gossypium hirsutum at three sample times during the growing season (21 June, 2 July and 18 July 2013) under well‐watered conditions. The temperature decreasing a given photosynthetic parameter 15% from the optimum is referred to as T15 and served as a standardized measure of heat tolerance. Ambient and estimated canopy temperatures were well within the optimal range for cotton throughout the sample period, and leaves were verified well watered using ΨPD measurements. However, T15 varied with sample date (highest on July 2 for all parameters), being 2 °C (Fv/F0) to 5.5 °C (φEο) higher on July 2 relative to June 21, despite optimal temperature conditions and predawn leaf water potential on all sample dates. These findings suggest that even under optimum temperature conditions and water availability, heat tolerance could be influenced by plant developmental stage.  相似文献

Using the advanced backcross quantitative trait loci (AB‐QTL) strategy, we successfully transferred and mapped valuable allelic variants from the high β‐glucan (BG) accession IAH611 (PI 502955), into the genome of cultivar ‘Iltis’. By backcrossing one BC1F1 plant to ‘Iltis’, we developed two BC2F2‐6 populations A and B, comprising 98 and 72 F2‐individuals, respectively. Genotyping of BC2F2 individuals with predominantly AFLP markers resulted in 12 linkage groups with a map size of 455.4 cM for Population A and 11 linkage groups with a map size of 313.5 cM for Population B. Both populations were grown at three sites in Germany over a three‐year period. Individuals were then phenotyped for 13 traits including grain yield (YD) and β‐glucan content (BG). QTL analysis via stepwise regression detected a total of 33 QTLs, most of which were clustered in three linkage groups. Two dense linkage groups A1 and B13 were found to be putatively homologous to groups KO_6 and KO_11 of the ‘Kanota’/‘Ogle’ map, respectively.  相似文献

Striga hermonthica can cause as high as 100% yield loss in maize depending on soil fertility level, type of genotype, severity of infestation and climatic conditions. Understanding the mode of inheritance of Striga resistance in maize is crucial for introgression of resistance genes into tropical germplasm and deployment of resistant varieties. This study examined the mode of inheritance of resistance to Striga in early‐maturing inbred line, TZdEI 352 containing resistance genes from Zea diploperennis. Six generations, P1, P2, F1, F2, BC1P1 and BC1P2 derived from a cross between resistant line, TZdEI 352 and susceptible line, TZdEI 425 were screened under artificial Striga infestation at Mokwa and Abuja, Nigeria, 2015. Additive‐dominance model was adequate in describing observed variations in the number of emerged Striga plants among the population; hence, digenic epistatic model was adopted for Striga damage. Dominance effects were higher than the additive effects for the number of emerged Striga plants at both locations signifying that non‐additive gene action conditioned inheritance of Striga resistance. Inbred TZdEI 352 could serve as invaluable parent for hybrid development in Striga endemic agro‐ecologies of sub‐Saharan Africa.  相似文献

In order to introduce the Tomato Spotted Wilt Virus (TSWV) resistance from Nicotiana alata into Nicotiana tabacum, a cytoplasmic male sterility (CMS) line of N. tabacum (N. tabacum L. cv. (gla.) S ‘K326’), was successfully crossed with N. alata. Despite a high DNA content variability, F1 hybrids could be classified in two subgroups, a major one encompassing fertile hybrids morphologically similar to their tobacco maternal parent but TSWV sensitive, and a minor one displaying sterile hybrids showing an intermediate phenotype and TSWV resistant. In order to elucidate the unexpected fertility recovery of the fertile F1 plants, some N. alata fertility restoration ppr genes were cloned and were shown to be differentially expressed between parental lineages as well as between both F1 subgroups, suggesting that N. alata contains fertility restoring allele able to overcome the CMS of N. tabacum.  相似文献
