Olive leaves represent a quantitatively significant by-product of agroindustry. They are rich in phenols, mainly oleuropein, which can be hydrolyzed into several bioactive compounds, including hydroxytyrosol. In this study, water extract from olive leaves ‘Biancolilla’ was analyzed for polyphenol profile, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and protective effect on differentiated Caco-2 cells. The efficacy of two enzymatic treatments in promoting the release of bioactive phenols was investigated: a) enzymatic extract from Wickerhamomyces anomalus, characterized by β-glucosidase and esterase activities; b) commercial β-glucosidase. Composition and bioactivity of the resulting extracts were compared. The results showed that the yeast-treated extract presented hydroxytyrosol content and DPPH radical scavenging activity comparable to those obtained using commercial β-glucosidase; however, it was showed the additional presence of hydroxycinnamic acids. In experiments on Caco-2 cells, the leaf extracts promoted the recovery of cell membrane barrier at different minimum effective concentrations. The high specificity of W. anomalus enzymatic extract may represent an effective tool for the release of bioactive phenols from olive by-products.  相似文献

This study investigated the effect of extracts with time (a range of extraction periods: 0.2 days to maximum 150 days) of rice and rye straw on the growth of Microcystis aeruginosa (a cyanobacterium). The effect was assessed by an effective concentration 50 (EC50) of extracts measured by a carbon (C) content, when 50% normalized maximum growth yield (50% inhibition effect) of M. aeruginosa occurred. We found that extracts of rice and rye straw inhibited the growth of M. aeruginosa. Extracts from the earliest incubation phase (0.2 days) for rice straw and the 40-day extract for rye straw showed the most inhibitive EC50 value of 28.0 mg C L?1 and 18.9 mg C L?1, respectively. The inhibition effect was positively related to the extraction period of the rye straw, but inversely of the rice straw. Our results suggested that the long-term decomposition of extracts during straw extraction be linked with the formation of allelochemicals. These results provided information on how allelochemicals derived from agricultural by-products affect algal growth in aquatic ecosystems.  相似文献

Camellia euphlebia (family, Theaceae) has been used for the prevention and treatment of cardiovascular diseases in Southern China. However, there has been no report on the hypolipidemic activity of Camellia euphlebia flower. This study evaluated the hypolipidemic activity of different preparation of Camellia euphlebia flower extracts using in vivo models. Mice intragastrically administered aqueous extract at 400 mg/kg dose or ethanol extract at 100 and 400 mg/kg doses of Camellia euphlebia flower for 28 days exhibited significant decreases in the levels of total cholesterol, triglycerides and low-density lipoprotein cholesterol, while displaying increased level of high-density lipoprotein cholesterol in the serum. The Camellia euphlebia flower extracts also improved the antioxidant ability of hyperlipidemic mice as well as protecting the animals against liver damage by lowering the level of glutamic-pyruvic transaminase activity. Furthermore, 400 mg/kg ethanol extract effectively down-regulated the mRNA levels of fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and glycerol-3-phosphate acyl transferase, suggesting that Camellia euphlebia flower extract may potentially inhibit lipid accumulation in the liver by regulating the expression of fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and glycerol-3-phosphate acyl transferase. These results provided support for the potential hypolipidemic activity of Camellia euphlebia flower and could partly explain the basis of using Camellia euphlebia for the treatment of hyperlipidemia.  相似文献

During the last few years, numerous attempts were made to identify effective α-glucosidase inhibitors from natural sources in order to develop new alternatives for diabetes management. Smallanthus sonchifolius (yacon) leaves were found to be effective in controlling postprandial hyperglycemia. Enhydrin, a constituent of yacon leaves, was noted for its significant hypoglycemic properties in diabetic rats. These properties were also demonstrated for yacon leaves decoction, which is rich in phenolic compounds such as chlorogenic acid and its derivatives. The purpose of the present study was to evaluate the potential of yacon leaves decoction and the isolated compound enhydrin to inhibit α-glucosidase enzyme, a possible mechanism of the above antihyperglycemic effect. In vitro assays showed that both 10% decoction and enhydrin significantly inhibited the activity of the yeast α-glucosidase enzyme in a dose-dependent manner, IC50 values being 50.40 and 134.17 μg/ml, respectively. In vivo experiments showed a rapid decrease in the hyperglycemic peak after sucrose load (2 g/kg body weight) in normal rats treated with the 10% decoction (140 mg/kg) and enhydrin (0.8 mg/kg). Both treatments caused a significant decrease in blood glucose levels in diabetic rats after sucrose load compared to diabetic control. These results suggest that both products assayed could be effective in the management of postprandial hyperglycemia through inhibition of α-glucosidase in the small intestine.  相似文献

Surface properties of fibrous and ground cotton and linen were investigated by inverse gas chromatography (IGC) and the contact angle with different liquids was also measured on fabrics composed of both fibers. Results proved that dispersion component of surface tension (γ s d ) determined by IGC depends not only on the surface energy, but also on several factors influencing the adsorbability of probe molecules on the cellulosic substrates. For cotton samples, the trapping of n-alkanes among waxy molecules in the outer layer of fibers can be presumed. This effect results in larger γ s d for cotton fibers than for linen in spite of the higher wettability of the linen fabrics. Besides the surface energy and trapping effects, the grinding also influences the γ s d values. Specific enthalpy of adsorption (ΔH A ab ) of polar probes could be determined on all linen samples, but only on the ground cotton sample. Lewis acid-base character calculated for linen and ground cotton samples depends on the same effects as the γ s d does. The similar ΔH A ab values of chloroform (acidic) and THF (basic) measured on each of the samples support the conclusion that the surface character is amphoteric, which is also proved by the high ΔH A ab values of the amphoteric ethyl acetate and acetone probes.  相似文献

Dyeing and antibacterial properties of a natural dye extracted from Liriope platyphylla fruit applied on silk fabrics have been studied. The total phenolic content (1109.13±69.02 mg), total flavonoid content (530.60±89.44 mg), and total anthocyanin content (492.26±77.79 mg) were measured in 100 g fresh weight of L. platyphylla fruits. In addition, ten anthocyanins and four flavanols were identified in L. platyphylla fruits by high performance liquid chromatography with diode array detection coupled with electrospray ionization mass spectrometry (HPLC-DAD/ESI-MS). A broad variation in color shade and color depth can be achieved with mixtures of dye extracts and metal mordants. Purple, blue, and pale green were main color shades of silk fabrics dyed with the extracts. The fastness of dyed silk fabrics except for control dyed fabrics against light, washing, and rubbing were acceptable with at least a grey scale rating of 3. The antibacterial activities of L. platyphylla fruit extracts were retained on dyed silk fabrics even after home washing 30 cycles. Mordanting with metal salt mordant had a positive effect on antibacterial activity of dyed silk fabrics in this study. Among them, aluminum and copper were the most effective mordants for improving antibacterial activity of silk fabrics dyed with L. platyphylla fruit extracts. The costs of natural dyeing of per silk fabrics kg by the extracts from L. platyphylla fruit were also calculated on laboratory scale.  相似文献

Water management is an important factor in regulating soil respiration and the net ecosystem exchange of CO2 (NEE) between croplands and atmosphere. However, how water management affects soil respiration and the NEE of paddy fields remains unexplored. Thus, a 2-year field experiment was carried out to study the effects of controlled irrigation (CI) during the rice season on the variation of soil respiration and NEE, with flooding irrigation (FI) as the control. A decrease of irrigation water input by 46.39% did not significantly affect rice yield but significantly increased irrigation water use efficiency by 0.99 kg m?3. The soil respiration rate of CI paddy fields was larger than that of FI paddy fields except during the ripening stage. Natural drying management during the ripening stage resulted in a significant increase of the soil respiration rate of the FI paddy fields. Variations of NEE with different water managements were opposite to soil respiration rates during the whole rice growth stages. Total CO2 emission of CI paddy fields through soil respiration (total R soil) increased by 11.66% compared with FI paddy fields. The increase of total R soil resulted in the significant decrease of total net CO2 absorption of CI paddy fields by 11.57% compared with FI paddy fields (p < 0.05). There were inter-annual differences of soil respiration and the NEE of paddy fields. Frequent alternate wetting and drying processes in the CI paddy fields were the main factors influencing soil respiration and NEE. CI management slightly enhanced the rice dry matter amount but accelerated the consumption and decomposition of soil organic carbon and significantly increased soil respiration, which led to the decrease of net CO2 absorption. CI management and organic carbon input technologies should be combined in applications to achieve sustainable use of water and soil resources in paddy fields.  相似文献

Background

The rice Pi2/9 locus harbors multiple resistance (R) genes each controlling broad-spectrum resistance against diverse isolates of Magnaporthe oryzae, a fungal pathogen causing devastating blast disease to rice. Identification of more resistance germplasm containing novel R genes at or tightly linked to the Pi2/9 locus would promote breeding of resistance rice cultivars.

Results

In this study, we aim to identify resistant germplasm containing novel R genes at or tightly linked to the Pi2/9 locus using a molecular marker, designated as Pi2/9-RH (Pi2/9 resistant haplotype), developed from the 5′ portion of the Pi2 sequence which was conserved only in the rice lines containing functional Pi2/9 alleles. DNA analysis using Pi2/9-RH identified 24 positive lines in 55 shortlisted landraces which showed resistance to 4 rice blast isolates. Analysis of partial sequences of the full-length cDNAs of Pi2/9 homologues resulted in the clustering of these 24 lines into 5 haplotypes each containing different Pi2/9 homologues which were designated as Pi2/9-A5, ?A15, ?A42, ?A53, and -A54. Interestingly, Pi2/9-A5 and Pi2/9-A54 are identical to Piz-t and Pi2, respectively. To validate the association of other three novel Pi2/9 homologues with the blast resistance, monogenic lines at BC3F3 generation were generated by marker assisted backcrossing (MABC). Resistance assessment of the derived monogenic lines in both the greenhouse and the field hotspot indicated that they all controlled broad-spectrum resistance against rice blast. Moreover, genetic analysis revealed that the blast resistance of these three monogenic lines was co-segregated with Pi2/9-RH, suggesting that the Pi2/9 locus or tightly linked loci could be responsible for the resistance.

Conclusion

The newly developed marker Pi2/9-RH could be used as a potentially diagnostic marker for the quick identification of resistant donors containing functional Pi2/9 alleles or unknown linked R genes. The three new monogenic lines containing the Pi2/9 introgression segment could be used as valuable materials for disease assessment and resistance donors in breeding program.

Raw, skinless peanut kernels from US commercial production lines were dry- and oil-roasted according to standard industrial practices. Eighty percent (v/v) methanolic extracts from the peanut cultivars were prepared and characterized by RP-HPLC: five predominant compounds were found comprising free p-coumaric acid and potential p-coumaric acid derivatives, as elucidated by DAD-UV spectra with comparisons to those of commercial standards. A Spanish high-oleic peanut possessed the greatest naturally-occurring level of p-coumaric acid and its derivatives, followed by a high-oleic Runner, a normal Runner, and a Virginia peanut. Upon thermal processing, p-coumaric acid was liberated at the expense of its derivatives according to the relationship: oil roasting > dry roasting > raw. A high-oleic Runner exhibited the greatest increase (~785%) in free p-coumaric acid levels after oil roasting. For many of the samples from the 2007 crop, processing increased the TPC and antioxidant capacities in the order of raw < dry roast < oil roast, but results were cultivar dependent. Oil-roasted peanuts were more effective at scavenging O2 - than their dry-roasted counterparts, as determined by a photochemiluminescence assay. Overall findings indicate that although thermal processing altered the composition of peanut kernel antioxidants, TPC values and radical-scavenging activities are preserved. Depending on peanut type, cultivar, and harvest date, enhanced antioxidant capacities can result.  相似文献

In the present work, oil adsorption, desorption, and resorption of poly(lauryl methacrylate-co-hydroxyethyl methacrylate) P(LMA-co-HEMA) were evaluated with different oils by a gravimetric method. Adsorption kinetics were modeled using pseudo-first-order and pseudo-second-order equations. Polymer network parameters of P(LMA-co-HEMA) regarding average molecular weight (Mc) between two crosslink piontss can be calculated by oil absorbency at equilibrium (Q e ), the solubility parameter (δ) and polymer-solvent interaction parameter (χ) with Flory-Huggins relation. The results showed pseudo-second-order model has a better fit to the oil adsorption kinetic data The desorption can be analyzed by fitting a prediction of exponential-like decay to the deswelling curves. A typical oil desorption exhibited two stages: a burst release driven by concentration gradient, and a slow release controlled by diffusion and the elastic recovery of polymer networks. For reusability, the resorption behavior of P(LMA-co-HEMA) was also investigated. It was worth noting that oil resorption was faster than the first adsorption due to potential passages. Moreover, the adsorption capacity was not significantly changed after regeneration.  相似文献

Phthorimaea operculella (Zeller) is one of the most common insect pests of cultivated potato in tropical and subtropical regions. In this research, a potential strategy to improve the insecticidal activity of plant essential oils for the effective management of P. operculella was studied. The insecticidal and residual effects of nanofiber oil (NFO) and pure essential oil (PEO) of Cinnamomum zeylanicum were assessed on PTM under laboratory conditions. The nanofibers were made by the electrospinning method using polyvinyl alcohol (PVA) polymer. The morphological characteristics of the nanofibers were evaluated by scanning electron microscopy and Fourier transform infrared spectroscopy. The chemical constituents of cinnamon essential oil (EO) were detected by GC/MS. Fumigant toxicity of NFO and PEO were evaluated on different growth stages (egg, male and female adults) of P. operculella. SEM and FTIR analyses confirmed the presence of EO on the nanofiber structure. The yield of the EO from C. zelanicum on the nanofibers was 1.86%. GC/MS analysis showed that cinnamaldehyde was the primary constituent (69.88%) of cinnamon EO. LC50 values of C. zelanicum EO and NFO were 4.92 and 1.76 μl/l air for eggs, 0.444 and 0.212 μl/l air for female adults, and 0.424 and 0.192 μl/l air for male adults, respectively. Fumigant bioassays revealed that NFO was more toxic than C. zeylanicum oil against at all stages of P. operculella. The residual effect of PEO and NFO was evaluated against the egg stage of the P. operculella. NFO lost insecticidal effectiveness 47 days after application, while the efficacy of PEO decreased 15 days after application. Our results suggest that NFO of C. zeylanicum can be used as an effective new tool for the management of P. operculella.  相似文献

Carotenoids and phenolic profile, antioxidant activity as well as concentrations of selected macronutrients (K, N, Mg, Ca and Na) and micronutrients (Zn, Cu and Mn) in flesh and peel of peach fruit were recorded at two harvest dates. Predominant mineral was potassium, followed by calcium, magnesium and sodium. The concentration of most micronutrients was greater in the peel than in the flesh especially in early season. The concentration of most elements in flesh and peel decreased during fruit maturation. Total carotenoids content varied with respect to the cultivar. β-cryptoxanthin and β-carotene were the major carotenoids in both tissues and flesh contain the lowest amounts. Neochlorogenic acid, chlorogenic acid, catechin, epicatechin, gallic acid, rutin, quercetin-3-O-galactoside, cyanidin-3-O-glucoside, cyanidin-3-O-rutinoside, were detected in both peel and flesh, with chlorogenic acid and catechin being the predominant components. Peel extracts showed markedly higher antioxidant activities, when estimated by ABTS or DPPH assays, than the flesh counterparts, consistent with the observed higher phenolic content. Overall, total phenolics levels increased at full ripening stage in both peel and flesh. The results found herein provide important data on carotenoids, phenolic and macro- and micronutrient changes during fruit growth, and emphases peach fruit as a potential functional food.  相似文献

In order to prepare antimicrobial regenerated cellulose fibers from blended spinning solutions, three non-water soluble polymeric guanidine derivatives, polyhexamethylene guanidine dodecyl benzene sulfonate (PHGDBS), polyhexamethylene guanidine dodecyl sulfate (PHGDSA), and polyhexamethylene guanidine laurylsulfonate (PHGLSO) were synthesized. And the chemical structure of these agents was verified by element analysis, Fourier transform infrared spectroscopy (FTIR), and proton nuclear magnetic resonance (1H-NMR). The antimicrobial activity of the three agents as well as cellulose films containing PHGDBS was also studied. The results showed that the compounds we prepared had strong properties against both bacterial and fungus, including Staphylococcus aureus (S. aureus), Escherichia coli (E. coli), Candida albicans, and Aspergillus niger. Moreover, it was found that three antimicrobial agents were insoluble in water but they can dissolve in solvents of cellulose such as 1-butyl-3-methylimidazolium chloride ([BMIM]Cl) and N-methylmorpholine-N-oxide monohydrate (NMMO·H2O). Meanwhile, it was also proved that [BMIM]Cl had little effect on the antimicrobial properties of these agents. The cellulose films containing only 1.0 wt% PHGDBS showed 99.94 % and 96.95 % bacterial reduction rates for S. aureus and E. coli, respectively. Moreover, still over 91 % of bacterial reduction was maintained after 15 laundering cycles. It suggests that the three agents will be suitable to prepare antimicrobial regenerated cellulose fibers or films.  相似文献

The precise level of environmental control in vitro may aid in identifying genetically superior plant germplasm for rooting characteristics (RC) linked to increased foraging for plant nitrogen (N). The objectives of this research were to determine the phenotypic variation in root morphological responses of 49 Solanum chacoense (chc), 30 Solanum tuberosum Group Phureja – Solanum tuberosum Group Stenotomum (phu-stn), and three Solanum tuberosum (tbr) genotypes to 1.0 and 0.5 N rate in vitro for 28 d, and identify genotypes with superior RC. The 0.5 N significantly increased density of root length, surface area, and tips. All RC were significantly greater in chc than in either phu-stn or tbr. Based upon clustering on root length, surface area, and volume, the cluster with the greatest rooting values consisted of eight chc genotypes that may be utilized to initiate a breeding program to improve RC in potato.  相似文献

Background

Rice plays an extremely important role in food safety because it feeds more than half of the world’s population. Rice grain yield depends on biomass and the harvest index. An important strategy to break through the rice grain yield ceiling is to increase the biological yield. Therefore, genes associated with organ size are important targets for rice breeding.

Results

We characterized a rice mutant gns4 (grain number and size on chromosome 4) with reduced organ size, fewer grains per panicle, and smaller grains compared with those of WT. Map-based cloning indicated that the GNS4 gene, encoding a cytochrome P450 protein, is a novel allele of DWARF11 (D11). A single nucleotide polymorphism (deletion) in the promoter region of GNS4 reduced its expression level in the mutant, leading to reduced grain number and smaller grains. Morphological and cellular analyses suggested that GNS4 positively regulates grain size by promoting cell elongation. Overexpression of GNS4 significantly increased organ size, 1000-grain weight, and panicle size, and subsequently enhanced grain yields in both the Nipponbare and Wuyunjing7 (a high-yielding cultivar) backgrounds. These results suggest that GNS4 is key target gene with possible applications in rice yield breeding.

Conclusion

GNS4 was identified as a positive regulator of grain number and grain size in rice. Increasing the expression level of this gene in a high-yielding rice variety enhanced grain yield. GNS4 can be targeted in breeding programs to increase yields.

Background

The brown planthopper (BPH) has become the most destructive and a serious threat to the rice production in Asia. Breeding the resistant varieties with improved host resistance is the most effective and ecosystem-friendly strategy of BPH biological management. As host resistance was always broken down by the presence of the upgrading BPH biotype, the more resistant varieties with novel resistance genes or pyramiding known identified BPH resistance genes would be needed urgently for higher resistant level and more durability of resistance.

Results

Here, we developed near isogenic lines of Bph9 (NIL-Bph9) by backcrossing elite cultivar 93–11 with Pokkali (harboring Bph9) using marker-assisted selection (MAS). Subsequently, we pyramided Bph6 and Bph9 in 93–11 genetic background through MAS. The resulting Bph6 and Bph9 pyramided line LuoYang69 had stronger antixenotic and antibiosis effects on BPH and exhibited significantly enhanced resistance to BPH than near isogenic lines NIL-Bph6 and NIL-Bph9. LuoYang69 derived hybrids, harboring heterozygous Bph6 and Bph9 genes, also conferred high level of resistance to BPH. Furthermore, LuoYang69 did not affect the elite agronomic traits and rice grain quality of 93–11. The current study also developed functional markers for Bph9. Using functional dominant marker, we screened and evaluated worldwide accessions of rice germplasm. Of the 673 varieties tested, 8 cultivars were identified to harbor functional Bph9 gene.

Conclusion

The development of Bph6 and Bph9 pyramided line LuoYang69 provides valuable resource to develop hybrid rice with highly and durable BPH resistance. The development of functional markers will promote MAS of Bph9. The identified Bph9 containing cultivars can be used as new sources for BPH resistance breeding programs.

Based on data collected from rice fields under drying–wetting cycle condition, the procedure of dual-crop coefficient (K cd) approaches was calibrated and validated to reveal its feasibility and improve its performance in rice evapotranspiration (ET c) estimation. It was found that K cd based on FAO-recommended basal crop coefficients (K cb) underestimated dual-crop coefficients in monsoon climate region in East China. The recommended coefficient (K cp) value of 1.2 was not high enough to reflect the pulse increase of rice ET c after soil wetting. The K cb values were calibrated as 1.52 and 0.63 in midseason and late season, and the K cp value was adjusted as 1.29 after soil wetting in rice field under drying–wetting cycle condition. The dual-crop coefficient curves based on locally calibrated K cbCal and K cpCor matched well with the measured crop coefficients and performed well in calculating rice evapotranspiration from paddy fields under drying–wetting cycle condition. So it can be concluded that the procedure of dual-crop coefficient method is feasible in rice ET c estimation, and locally calibrated K cb and K cp can improve its performance remarkably.  相似文献

Background

The DEFECTIVE IN OUTER CELL LAYER SPECIFICATION 1 (DOCS1) gene belongs to the Leucine-Rich Repeat Receptor-Like Kinase (LRR-RLK) subfamily. It has been discovered few years ago in Oryza sativa (rice) in a screen to isolate mutants with defects in sensitivity to aluminum. The c68 (docs1–1) mutant possessed a nonsense mutation in the C-terminal part of the DOCS1 kinase domain.

Findings

We have generated a new loss-of-function mutation in the DOCS1 gene (docs1–2) using the CRISPR-Cas9 technology. This new loss-of-function mutant and docs1–1 present similar phenotypes suggesting the original docs1–1 was a null allele. Besides the aluminum sensitivity phenotype, both docs1 mutants shared also several root phenotypes described previously: less root hairs and mixed identities of the outer cell layers. Moreover, our new results suggest that DOCS1 could also play a role in root cap development. We hypothesized these docs1 root phenotypes may affect gravity responses. As expected, in seedlings, the early gravitropic response was delayed. Furthermore, at adult stage, the root gravitropic set angle of docs1 mutants was also affected since docs1 mutant plants displayed larger root cone angles.

Conclusions

All these observations add new insights into the DOCS1 gene function in gravitropic responses at several stages of plant development.

The phenolic compounds, color and antioxidant capacity of gluten-free pasta prepared with non-conventional flours such as chickpea (CHF), unripe plantain (UPF), white maize (WMF) and blue maize (BMF) were analyzed. Fifteen phenolic compounds (five anthocyanins, five hydroxybenzoic acids, three hydroxycinnamic acids, one hydroxyphenylacetic acid and one flavonol) were identified in pasta prepared with blue maize, and 10 compounds were identified for samples prepared with white maize. The principal component analysis (PCA) led to results describing 98% of the total variance establishing a clear separation for each pasta. Both the proportion (25, 50 and 75%) and type of maize flour (white and blue) affected the color parameters (L*, C ab *, h ab and ΔE* ab ) and antioxidant properties (DPPH, ABTS and FRAP methods) of samples, thus producing gluten-free products with potential health benefits intended for general consumers (including the population with celiac disease).  相似文献
