Probiotics renowned as valuable microbes serve as a potential alternative to control diseases in aquaculture and are considered as an efficient and environment‐friendly approach to reduce the use of antibiotics. The present study aims at the isolation of putative probiotic bacteria from the intestinal tract of different fish species from the Doaba region of Punjab, India. In this study, isolated bacterial strains were characterized based on their morphological, biochemical and molecular characterization by 16S rRNA gene sequencing, followed by in vitro evaluation of different selection parameters described in FAO/WHO guidelines. A total of 169 different bacterial strains were isolated from the gastrointestinal tract of 52 different fish species. After in vitro evaluation, out of 169 bacterial strains only five bacteria (S3, S7, BDK2', BDK7 and BDK9) identified as Enterococcus and Bacillus species showed antagonistic activity against the fish pathogen Pseudomonas aeruginosa (MTCC 4 673). These isolates were screened based on their response towards bile tolerance, pH tolerance, adhesion and drug susceptibility to different antibiotic discs. And, the in vivo results indicated improved growth and survival against the infection (P. aeruginosa) after oral administration of the probiotics. The observations of in vitro and in vivo evaluation indicate that these isolated probiotic strains serve as effective probiotics and can be used as a novel and safe treatment to cure current issues in aquaculture.  相似文献

Flavobacterium columnare is the causative agent of columnaris disease in diverse fish species worldwide. Although columnaris is an important disease, the antimicrobial susceptibility pattern of F. columnare is not well studied. Thus, the purpose of this study was to test the in vitro antimicrobial susceptibility of 97 F. columnare isolates collected worldwide between 1987 and 2011 from 17 fish species. The broth microdilution technique was utilized for reliable testing of these fastidious organisms. None of the isolates displayed acquired resistance to florfenicol, gentamicin, ormetoprim‐sulfadimethoxine and trimethoprim‐sulfamethoxazole. Acquired resistance to chloramphenicol was detected in 1%, to nitrofuran in 5%, to oxytetracycline in 11% and to enrofloxacin, flumequine and oxolinic acid in 10%, 16% and 16% of the isolates, respectively, as reflected by a bimodal or trimodal distribution of their minimum inhibitory concentrations (MICs). One isolate showed acquired resistance towards several antimicrobial agents including erythromycin. Another isolate revealed acquired resistance towards – amongst others – ampicillin. The isolates displaying acquired resistance originated from ornamental fish species or Vietnamese catfish, except for two isolates coming from wild channel catfish in which acquired resistance was encountered towards oxytetracycline only. Fifty per cent of the resistant isolates from ornamental fish were shown to have acquired resistance against three classes of antimicrobial agents, assigning these isolates as multiple resistant. These data might indicate less prudent use of antimicrobials especially in ornamental fish species.  相似文献

ABSTRACT:   Antibiotic residues in marine sediments of fish farms negatively influence microbial ecologic systems. The microbial degradation of antibiotic residues was experimentally examined in the marine sediments of Uranouchi Bay, to which one of five antibiotics was added. After incubation reducing physical factors, ampicillin, doxycycline, oxytetracycline, and thiamphenicol were significantly degraded, while josamycin maintained most of the initial amounts. The isolates resistant to ampicillin, josamycin, oxytetracycline, or thiamphenicol degraded each antibiotic in wide ranges of degrees, whereas the isolates degrading doxycycline were not obtained. Microbial degradation may contribute to the disappearance of ampicillin, doxycycline, oxytetracycline, and thiamphenicol in the fish farm. In contrast, the disappearance of josamycin would depend on physical factors, but the bacteria degrading josamycin at least exist in the marine sediments. Phylogenetic analysis using 16S rDNA sequences demonstrated that the antibiotic-resistant isolates formed several clusters in the Gram-positive bacterial group, the Flavobacterium–Cytophaga–Bacteroides group, and the proteobacteria subdivisions. The antibiotic-resistant bacterial population would be composed of various species including ubiquitous coastal bacterial groups. Several species of antibiotic resistant bacteria show antibiotic degradation activities, and appear to contribute to the disappearance of antibiotics in Uranouchi Bay.  相似文献

This study evaluated antibiotic resistance and the related genes in total 47 Aeromonas veronii isolates from pet fish, eel (Anguilla japonica) and koi (Cyprinus carpio) in Korea. In comparison with the antibiotic susceptibilities of isolates from eel and koi, those of pet fish were more resistant to ceftiofur, aminoglycosides, tetracycline and nitrofurantoin. And isolates from pet fish showed high prevalences of class 1 integron, quinolones and tetracycline resistance determinants than those from eel and koi. Repetitive‐element palindromic PCR (rep‐PCR) showed larger diversities among A. veronii isolates. Collectively, pet fish may be a reservoir for multiple clones of A. veronii involved in antibiotic resistance. In this aspect, imported fish in the aquaculture trade should be steadily and continually screened for bacterial antibiotic resistance and related genes.  相似文献

The disease outbreaks in aquaculture system of wetlands are the major cause of fish mortality. Among various bacterial septicaemic diseases, fish mortality caused by Acinetobacter spp. is recently reported in different fish species. Fish disease outbreak was investigated in a wetland of West Bengal, India to identify the aetiological factors involved. The moribund fish were examined and subjected to bacterial isolation. Two bacterial causative agents were identified as Acinetobacter junii and Acinetobacter pittii by biochemical characterization and 16S rRNA gene amplification. Both the isolates were oxidase‐negative, nitrate‐negative, catalase‐positive and indole‐negative. The molecular identification using 16S rRNA gene sequencing and phylogenetic tree analysis further confirmed the two Acinetobacter spp. with 97%–99% similarity. The antibiotic resistance patterns of these two bacteria revealed that both of them were resistant to β‐lactam, cefalexin, cephalothin, amoxyclav, cefuroxime, cefadroxil, clindamycin, vancomycin and penicillin. In addition, A. pittii was also resistant to other antibiotics of cephams group such as ceftazidime and cefotaxime. In the challenge experiment, both A. junii and A. pittii were found to be pathogenic with LD50 of 1.24 × 105 and 1.88 × 107 cfu/fish respectively. Histopathological examination of gill, liver and kidney revealed prominent changes supporting bacterial septicaemia. The investigation reports for the first time on concurrent infection by A. junii and multidrug‐resistant (MDR)‐A. pittii as emerging fish pathogens to cause severe mortality in Labeo catla and Hypophthalmichthys molitrix in a freshwater wetland.  相似文献

Since 2012, low‐to‐moderate mortality associated with an Erysipelothrix sp. bacterium has been reported in ornamental fish. Histological findings have included facial cellulitis, necrotizing dermatitis and myositis, and disseminated coelomitis with abundant intralesional Gram‐positive bacterial colonies. Sixteen Erysipelothrix sp. isolates identified phenotypically as E. rhusiopathiae were recovered from diseased cyprinid and characid fish. Similar clinical and histological changes were also observed in zebrafish, Danio rerio, challenged by intracoelomic injection. The Erysipelothrix sp. isolates from ornamental fish were compared phenotypically and genetically to E. rhusiopathiae and E. tonsillarum isolates recovered from aquatic and terrestrial animals from multiple facilities. Results demonstrated that isolates from diseased fish were largely clonal and divergent from E. rhusiopathiae and E. tonsillarum isolates from normal fish skin, marine mammals and terrestrial animals. All ornamental fish isolates were PCR positive for spaC, with marked genetic divergence (<92% similarity at gyrB, <60% similarity by rep‐PCR) between the ornamental fish isolates and other Erysipelothrix spp. isolates. This study supports previous work citing the genetic variability of Erysipelothrix spp. spa types and suggests isolates from diseased ornamental fish may represent a genetically distinct species.  相似文献

Shewanella spp. are Gram‐negative, rod‐shaped, motile bacteria that are widely distributed in marine and freshwater environments. The bacteria are present in the physiological microflora of fish from temperate waters and are known as fish spoilage species. From clinically healthy fish and from fish with skin ulcerations, Shewanella spp. is regularly isolated, indicating a possible role as fish pathogen. In this study, 74 isolates of Shewanella spp. were analysed. For species identification, biochemical techniques, 16S rRNA sequencing, MALDI‐TOF MS and the Sherlock Microbial Identification System (MIS) based on the composition of fatty acid ethyl esters were compared. The phylogenetic relationship, cytotoxicity in vitro and resistance against antibiotics were tested. The most reliable method for species identification was 16S rRNA sequencing. From diseased fish, clinically healthy fish and the aquatic environment, different Shewanella species were isolated. This indicates that Shewanella spp. is widespread in the aquatic milieu and acts as a secondary pathogen. The virulence of Shewanella spp. is probably not depending on the species but on the isolate itself. Many isolates of Shewanella spp. were showing multiresistances against antibiotic substances, especially in samples derived from retailers and in routine diagnostics, all Shewanella spp. should therefore be tested for resistances against antibiotic agents.  相似文献

Ornamental fish production is among the leading cash crops of the United States of America aquaculture economy, and retail value of the fish trade is worth approximately US$l,000 million. However, few studies exist to document this trade. Using import and export documents we report trends in total values for the U.S. trade in ornamental fish. Also, we determined the number and value of the most commonly imported ornamental fish. In 1992, approximately 201 million fish valued at $44.7 million were imported into the U.S. The port of Los Angeles handled 39% of all trade activity, Miami 22%, New York 16%, Tampa 6%, and Honolulu 6%. Freshwater fish accounted for approximately 96% of the total volume and 80% of the value of the imports. Most freshwater ornamental fish were farm-raised and imported from Southeast Asia. Most U.S. exports of ornamental fish were cultured in the state of Florida. Although saltwater fish had a high market value (20% of the declared value of the imports), the volume of these fish was only 4%. Most imported and exported saltwater ornamental fish were collected from the wild. Of the 1,539 species declared as ornamental fish, 32 species dominated the trade. These were all of freshwater origin. The guppy Poecilia reticulala and neon tetra Paracheirodon innesi were the most popular ornamental fish kept in U.S. households. The average prices paid for imported ornamental freshwater fish were 45 cents for egg layers and 22 cents for live bearers. The results of this study document the importance of the ornamental fish industry and identifies the most valuable species in the trade for potential domestic culture and protection in the wild.  相似文献

Antibiotic resistance and the presence of resistance genes (ARGs) were investigated in the bacteria isolated from rainbow trout (Oncorhynchus mykiss) from different trout farms located in Turkey. The most frequent types of antibiotic resistance were towards β‐lactams (cephalothin [70% of bacterial isolates], amoxicillin [63%], ampicillin [62%], ticarcillin [56%], aztreonam [51%]), macrolide [erythromycin, 68%] and sulphonamide [sulphamethoxazole, 51%]. Of bacterial isolates, 51% were multiple drug resistant (MDR), while 35% of the isolates were extensively drug resistant (XDR). None of isolates were pandrug resistant (PDR). The most common ARGs were ampC (36%) and sul1 (24%). The class 1 integron gene cassette was detected in 51% of the bacteria. There was a strong positive correlation between the antibiotic resistance rate and the presence of ARGs (r2 = .932). Gene encodes blaCTX‐M1, one of the extended spectrum beta‐lactamase enzymes, was first described in Aeromonas caviae, Photobacterium damselae, Pseudomonas luteola and Burkholderia cepacia. It was determined that 35% of the bacteria harboured at least one plasmid. Plasmid‐mediated ARGs were identified to be tetracyclines (tetA, tetB, tetC, tetD), sulphonamides (sul1, sul3) and β lactams (ampC, blapse). Thus, results suggest that ARG contamination situation deliberates resistance to tetracycline, aminoglycoside, chloramphenicol and sulphonamide. Therefore, the presence and activity of ARGs in fish and in environmental bacteria may play an important role in the spread of resistance genes among bacteria by transposition or integron gene cassettes.  相似文献

为了更有针对性地防控水产动物细菌性病害的发生和流行,本实验对美洲鳗鲡及其养殖水体耐药细菌的种属特征及耐药情况开展了相关研究。首先采集美洲鳗鲡不同部位(表皮、鳃、肠道)及其养殖水体的样品,经5种抗菌药物平板筛选耐药菌株,然后采用K-B纸片扩散法检测细菌对抗菌药物的耐药性,同时测定耐药菌株的16S rDNA序列,进而分析耐药菌的种属分布和多重耐药性。结果显示,经耐药平板筛选分离纯化得到108株细菌,分别属于气单胞菌属、柠檬酸杆菌属、不动杆菌属等20个属;其中,93.5%的菌株对3种(含)以上的抗菌药物具有耐药性,86.1%的菌株对3类(含)以上的药物具有抗性。对阿莫西林的耐药率高达90.7%,对四环素、利福平以及磺胺类和酰胺醇类药物类的耐药率为60%~80%,对头孢噻肟、新霉素以及喹诺酮类的耐药性弱(低于20%)。美洲鳗鲡肠道(0.40)、表皮(0.41)、鳃部(0.42)及水样(0.47)菌群的多重耐药指数显示各生态样品耐药程度较为严重,尤以水样为最。各菌属中,柠檬酸杆菌属(0.58)和克雷伯菌属(0.61)的多重耐药指数最高,而不动杆菌属(0.21)则相对较低。美洲鳗鲡及养殖水体普遍存在多重耐药菌株,对此应引起足够的重视;水产动物及养殖环境耐药细菌对某些水产用药如诺氟沙星、新霉素等耐药率低,可将其做为水产动物细菌性疾病治疗的首选药物。  相似文献

Carps are the most diversified freshwater fish belonging to family Cyprinidae. Numerous probiotic and pathogenic lactic acid bacteria (LAB) have been characterized from carps. However, the diversity of these ecologically important bacteria is entirely unknown in freshwater fish of Pakistan. The present study aimed to characterize and identify the lactic acid bacteria from two carps viz. Laboe rohita and Cirrhinus mrigala and determine their antagonistic activity. Seventeen bacterial isolates were purified from the gastrointestinal tract and gills of these fish and characterized morphologically. Initially, seven isolates were screened as LAB using agar supplemented with CaCO3. Subsequently, only two isolates CILB2 and RIL10 were selected as LAB after high‐performance liquid chromatography analysis for lactic acid production. Isolates CILB2 and RIL10 were genetically identified as Enterococcus faecalis and Weissella sp., respectively after 16S rRNA gene sequence analysis. Both strains exhibited significant antagonistic activity against common fish pathogens Streptococcus agalactiae, Staphylococcus aureus and Escherichia coli. Enterococcus faecalis CILB2 and Weissella sp. RIL10 were also found negative for haemolysis and gelatinase activities and were sensitive to ampicillin, amoxicillin, doxycycline, erythromycin, chloramphenicol and co‐trimoxazole antibiotics. The identified LAB strains may further be investigated for their potential probiotic application in fish feed and food preservation techniques.  相似文献

A survey was performed on ornamental fish imported into the EU to detect viral agents belonging to the genus Ranavirus. The objective was to gain knowledge of the potential for these systemic iridoviruses to gain entry into the EU via international trade in ornamental fish. A total of 208 pooled samples, representing 753 individual fish, were tested. The samples included 13 orders and 37 families, originating from different countries and continents. Tissues from fish that died during or just after transport were collected and examined by standard virological techniques in epithelioma papulosum cyprini cells, by transmission electron microscopy and by PCR for the detection of the major capsid protein and DNA polymerase gene sequences of ranaviruses. Virus was isolated from nine fish species but ranavirus was not identified in those samples. The results suggest that ranaviruses are not highly prevalent in ornamental fish imported into the EU.  相似文献

Megalocytiviruses have been associated globally with severe systemic disease and economic loss in farmed food fish and ornamental fish. The viruses have been spread internationally by translocation of live fish. In New Zealand, megalocytiviruses are regarded as exotic. A potential pathway for introduction has been identified, namely imported ornamental fish. In the present study, real‐time PCR assays were developed for detection of megalocytiviruses using a conserved major capsid protein gene. A SYBR green assay was developed to target all known megalocytiviruses. A second real‐time PCR assay using a molecular beacon was developed to specifically target gourami, Trichogaster trichopterus, iridovirus, a species of iridovirus previously linked to ornamental fish imports in Australia. The analytical sensitivity for the SYBR green and molecular beacon assays were 10 and 100 fg, respectively. The analytical specificity of the real‐time PCR assays determined using genomic DNA templates from three target viruses, 12 non‐target viruses and 25 aquatic bacterial species were 100%. The intra‐run and inter‐run coefficients of variation of both assays were <5%. The real‐time PCR assays developed in this study provide rapid, sensitive, and specific detection of megalocytiviruses and gourami iridovirus.  相似文献

For the sustainable farming of tilapia, proper maintenance of their health and adequate treatment for infections at appropriate time are inevitable. The indiscriminate use of antibiotics in aquaculture, as a part of treatment and as growth promoters, accelerates antimicrobial resistance (AMR) among the fish pathogens. In the present study, we have isolated diverse aeromonads from Nile tilapia and studied their antibiogram and plasmid profiling. Aeromonas hydrophila, Aeromonas veronii, Aeromonas sobria, Aeromonas dhakensis, Aeromonas caviae, Aeromonas jandaei and Aeromonas aquatica were isolated from infected tilapia (n = 150), and their Shannon wiener diversity index was calculated as 1.926. A. veronii was found to be the most multiple antibiotic‐resistant pathogen with the MAR index of 0.46, and A. aquatica was noticed as the least resistant isolate. The minimum inhibitory concentration of resistant antibiotics was shown as >256 mcg/ml for most of the isolates. The virulent genes such as aerolysin and hemolysin were identified in all the isolates except A. aquatica. The detection of class 1 integrons, plasmid profiling and plasmid curing studies confirmed that AMR exhibited by most of the Aeromonas species is of plasmid mediated. This challenges the risk of wide spread of AMR among the pathogens and subsequent treatment of the infection.  相似文献

Pseudomonas fluorescens are known bacterial pathogens in fish. The P. fluorescens group contains at least nine different bacterial species, although species from fish have rarely been differentiated. Two isolated fish kills affecting wild bluegills, Lepomis macrochirus Rafinesque, and pumpkinseed sunfish, Lepomis gibbosus (Linnaeus), occurred in the spring of 2015 during cool water temperatures (12.5°C–15.5°C). Disease signs included severe bacteraemia with rare gross external signs. Pure bacterial cultures isolated from kidneys of all affected fish were identified as P. fluorescens using the API 20NE system, while no bacteria were isolated from asymptomatic fish. To further identify the species of bacterium within the P. fluorescens complex, genetic analysis of the 16S rRNA, rpoD and gyrB genes was conducted. DNA sequences of bacterial isolates from both mortality events were identical and had close identity (≥99.7%) to Pseudomonas mandelii. Although likely widespread in the aquatic environment, this is the first report of a bacterium closely resembling P. mandelii infecting and causing disease in fish. The bacterium grew at temperatures between 5°C and 30°C, but not at 37°C. It is possible that infections in fish were a result of immunosuppression associated with spring conditions combined with the psychrotrophic nature of the bacterium.  相似文献

Keeping fish is a popular pastime in South Africa and the majority of ornamental fish are imported. A pilot study was initiated to examine the health status of ornamental freshwater fishes imported to South Africa. Four groups of thirty fish each were examined for the presence of external parasites, and processed for virus isolation and for bacterial and mycobacterial culture. The groups consisted of goldfish (Carassius auratus), koi (Cyprinus carpio), guppies (Poecilia reticulata) and cardinal tetras (Cheirodon axelrodi). Mycobacterium fortuitum was isolated from the goldfish and koi. No other significant bacteria were isolated and virus culture was negative for all groups. Skin scrapings and wet gill preparations were made to detect external parasites. Parasites were identified from fixed material. External parasites included Trichodina mutabilis, Ichthyophthirius multifiliis, ciliophorans of the genus Tetrahymena, and monogeneans belonging to the genera Dactylogyrus and Gyrodactylus. This is the first report of Trichodina mutabilis in South Africa. Diseases imported with ornamental fish pose a risk to both indigenous fish populations and the aquaculture industry. To understand the driving environmental factors in changes of bacterial virulence and antibiotic resistance, we determined the prevalence, antibiotic resistance and antibiotic resistance and virulence genes of Vibrio harveyi isolated from diseased marine fish in south coastal China. We isolated 2, 52 and 53 V. harveyi strains from Fujian, Hainan and Guangdong, respectively, and identified them by multilocus sequence analysis of 16S rRNA-toxRVh-rctB. Nine typical virulence genes were represented at a higher average in Hainan (7.39 ± 0.24) than in Guangdong (6.91 ± 0.28). Five atypical virulence genes were detected in some isolates. In particular, flaC and vvh were detected in more than 60% of isolates. Their average number was significantly higher in Hainan (2.30 ± 0.20) than in Guangdong (1.70 ± 0.10). Multidrug resistance was widespread with an average resistance to 4.57 ± 0.18 of 15 antibiotics. Both the average number of antibiotic resistance and antibiotic resistance genes were higher in Hainan (5.25 ± 0.27 and 1.11 ± 0.15, respectively) than in Guangdong (3.87 ± 0.21 and 0.75 ± 0.10, respectively). This study demonstrated that there were more virulence genes and greater drug resistance in Hainan than in Guangdong, suggesting that warmer temperature and antibiotics pollutants probably enhance antibiotic resistance and bacterial infection.  相似文献

从广东省珠海市养殖花鲈(基因鉴定出87株杀鱼爱德华菌()。耐药谱分析显示,杀鱼爱德华菌对利福平(98.85%)、麦迪霉素(96.55%)、红霉素(95.40%)、青霉素(68.96%)、磺胺异恶唑(58.62%)、复方新诺明(28.73%)、阿莫西林(21.83%)、庆大霉素(13.79%)、新霉素(10.34%)、呋喃唑酮(3.45%)、诺氟沙星(2.29%)、氯霉素(2.29%)、多西环素(2.29%)、土霉素(1.15%)、氟苯尼考(1.14%)、恩诺沙星(0%)耐药。杀鱼爱德华菌共有32种耐药谱型且均为多重耐药菌株,多重耐药指数为0.423。斑马鱼致死率结果发现,杀鱼爱德华菌是一株中高毒力菌株;进一步的相关性分析揭示,杀鱼爱德华菌毒力与庆大霉素抗性呈正相关(<0.01)呈负相关。综上所述,花鲈源杀鱼爱德华菌为高毒、多重耐药的菌种,其毒力与耐药性多呈现负相关,推测是由于细菌因获得外源DNA而产生额外的生物成本所致。  相似文献

The occurrence of Mycobacterium spp. in freshwater and marine ornamental fish was studied in Italy from June 2002 to May 2005. Two surveys were carried out, one of aquarium fish sent to the Laboratory for diagnosis, and the other of prevalence of infection by mycobacteria in ornamental fish imported into Italy. Bacterial isolation was carried out from the spleen, kidney and liver, and the isolates were subsequently identified by biochemical tests. In the first survey, 387 fish were examined and Mycobacterium spp. were isolated from 181 (46.8%) fish. In the second survey 127 batches of ornamental fish from different countries were examined. Mycobacterium spp. were isolated from 38 (29.9%) batches. The following species were found: M. fortuitum, M. peregrinum, M. chelonae, M. abscessus, M. marinum, M. gordonae, M. nonchromogenicum and M. interjectum. There was a high prevalence of infection independent of the presence of macroscopic lesions. Mycobacterium fortuitum and M. chelonae were more prevalent than M. marinum in the samples examined.  相似文献

Antibiotic resistance, driven by inappropriate use of antimicrobial agents for fish farming, is an expanding global health threat. Demand of animal protein for human consumption is entraining at remarkable pace which in turn, upturning the non‐judicial use of life‐saving drugs in these modern animal production practices at an incautious rate. Hence, study was devised to observe the antimicrobial resistance (AMR) pattern of human prominence bacteria, against drugs of critically and highly importance of both human and veterinary medicine, circulating over the fish farming in India. One hundred sixty fish faecal samples were obtained from distinct geographical locales of Rajasthan, India. Overall, 202 isolates comprising of Escherichia coli (81, 51%), Salmonella spp. (72, 45%) and Staphylococcus aureus (49, 31%) were isolated to screen for resistance. Antimicrobial resistance has been analysed using broth microdilution method. In case of E. coli and Salmonella spp., highest resistance (>95%) was observed against streptomycin while for S. aureus, it was observed against trimethoprim, along with high resistance to other antibiotics. Majority of the isolates were found to be resistant to more than three antibiotics implying multi‐drug resistance (>89%) relative to the critical and highly important antibiotics. The high AMR is a reflection of misuse of these agents in the fish farming, which is a concern of serious health issues. Our findings persuade the prudent use of antimicrobial agents in fish farming and other aquaculture settings that may assist in drafting of the new policies for the judicial use of these life‐saving drugs in these sectors.  相似文献
