Monogeneans are ectoparasites that may cause losses in production and productivity in the aquaculture of Colossoma macropomum. Chemotherapeutics used in aquaculture usually have major adverse effects on fish; hence, the use of essential oils has been considered advantageous, but these are not soluble in water. Thus, the use of nanostructures to enhance water solubility of compounds and improve bioactivity may be very promising. This study investigated the antiparasitic activity of nanoemulsion prepared with Copaifera officinalis oleoresin (50, 100, 150, 200 and 300 mg/L), against monogenean parasites from the gills of C. macropomum. The particle size distribution and zeta potential suggested that a potentially kinetic stable system was generated. The nanoemulsion from C. officinalis oleoresin achieved high efficacy (100%) at low concentrations (200 and 300 mg/L) after 15 min of exposure. This was the first time that a nanoemulsion was generated from C. officinalis oleoresin using a solvent‐free, non‐heating and low‐energy method. Moreover, this was the first time that an antiparasitic against monogeneans on fish gills, based on nanoemulsion of C. officinalis oleoresin, was tested.  相似文献

Outbreaks of diseases pose a major threat to sustainable aquaculture development worldwide. Application of herbal products to combat parasitic diseases provides an alternative approach for sustainable aquaculture. This study investigated the in vitro antiparasitic effects of an oil‐in‐water nanoemulsion prepared using the essential oil from Pterodon emarginatus, against monogeneans infesting Colossoma macropomum. Gill arches from C. macropomum (47.6 ± 14.5 g and 13.5 ± 1.4 cm) that were naturally parasitized by Anacanthorus spathulatus, Notozothecium janauachensis and Mymarothecium boegeri were immersed in different dispersions of the P. emarginatus nanoemulsions (0, 50, 100, 200, 400 and 600 mg/L). The major compounds presented in the essential oil of P. emarginatus were β‐elemene, β‐caryophyllene and α‐humulene. Characterization of these nanoemulsions showed that they have a small mean droplet size and low polydispersity index, which is concordant with stable systems. In this in vitro trial, the P. emarginatus nanoemulsion concentrations of 100, 200, 400 and 600 mg/L presented 100% helminthic efficacy against monogeneans of the gills of C. macropomum. The highest two concentrations used (400 and 600 mg/L) were seen to immobilize the parasites after only 15 min. Therefore, it would be worthwhile testing these concentrations in therapeutic baths against monogeneans of C. macropomum.  相似文献

This study investigated the efficacy of Lippia grata essential oil (EO) against monogeneans of Colossoma macropomum and effects on the haematology after immersion baths. In the in vitro assays, the efficacy of 100, 250, 350 and 700 mg/L of L. grata EO and two controls were tested, of which one control was with the cultivation tank water and the other was with the tank water and 70% ethyl alcohol. Composition of majority bioactive compounds in the EO was carvacrol (48.12%), p‐cymene (24.39%) and γ‐terpinene (2.49%). Anacanthorus spathulatus, Notozothecium janauachensis, Mymarothecium boegeri and Linguadactyloides brinkmanni obtained of fish gills showed 100% immobilization in the in vitro assays when exposed to 700 and 350 mg/L after 30 min and 2 hr respectively. Fish showed tolerance to 700 mg/L of the EO, which was used in therapeutic in three consecutive baths, which caused lamellar hyperplasia in C. macropomum gills. The 700 mg/L of the EO in the therapeutic baths showed an efficacy of 95.1% against the monogeneans and caused decrease in haemoglobin, whereas control fish showed a reduction in mean corpuscular haemoglobin concentration. Results indicated that 700 mg/L of Lgrata EO is a safe and effective concentration to treat C. macropomum infected with monogeneans.  相似文献

This study evaluated the efficacy of albendazole, ivermectin, levamisole, mebendazole and praziquantel on monogeneans of Colossoma macropomum, based on in vitro and in vivo assays. In vitro assays indicated that albendazole (500, 100, 1,500 and 2,000 mg/L), ivermectin (200, 250, 300 and 350 mg/L) and levamisole (50, 75, 100 and 125 mg/L) were 100% effective against Anacanthorus spatulatus, Notozothecium janauachensis, Mymarothecium boegeri and Linguadactyloides brinkmanni, while mebendazole (125, 150, 175 and 200 mg/L) and praziquantel (5, 10, 15 and 20 mg/L) were ineffective. Fish mortality in 24 hr therapeutic baths with 500 mg/L of albendazole was 6.6%, but the behaviour of the animals remained unchanged, while 200 mg/L of ivermectin caused lethargy, signs of hypoxia and 100% mortality within 2 hr, and 125 mg/L of levamisole caused no mortality. The efficacy of 500 mg/L of albendazole was 48.6% in the 24 hr baths, while that of 125 mg/L levamisole was 88.2%. Although ivermectin showed in vitro efficacy, the lowest concentration used in baths was highly toxic to fish. Therefore, we recommend the use of 125 mg/L of levamisole to control and treat monogenean infestations on C. macropomum in fish farming.  相似文献

In this study, the effect of edible coarse/nanoemulsions of alginate as coating containing Zataria multiflora Boiss essential oil (ZEO) on the microbial quality of fish fillets was investigated at three concentrations (i.e. 0.25%, 0.5% and 1% [w/v]) in 16 days of storage at 4°C. Moreover, a comparison was made between the fabricated coarse emulsion and nanoemulsion coatings by particle size and polydispersity index analyses. Sample analysis was performed with regard to total viable count (TVC), total psychrophilic count (TPC), hydrogen sulphide‐producing bacteria count (HSC) and Enterobacteriaceae count (ENC) on days 0, 4, 8, 12 and 16. According to the findings, all the samples showed increased microbial counts during the storage periods. The highest reduction rates of TVC (about 2 log CFU/g), TPC (about 1 log CFU/g), HSC (about 1.5 log CFU/g) and ENC (about 3 log CFU/g) were observed in the nanoemulsion of alginate containing 1% w/v ZEO (NE 1%) samples, compared with alginate and control samples. The microbial growth was significantly inhibited in all treatments, compared with that in the control (p < .05). Accordingly, it can be concluded that nanoemulsion coating was more effective than coarse emulsion to prolong the durability of fish fillets and revealed a faster and greater inhibition of microbial flora, compared with coarse emulsions during the storage time.  相似文献

The effect of a gradual substitution of dietary fish meal protein by soya meal protein on growth, feed uptake and protein utilization of 1 g Colossoma macropomum (Cuvier) was studied at two different dietary protein levels. Growth rates of fish fed ad libitum 20 and 45% protein diets fluctuated between 41 and 49, and between 60 and 68 g kg-0 8 day-1, respectively. Fish incorporated between 31 and 47% of the dietary protein in their body. Increased amounts of soya meal in the diet led to decreased feed uptake, higher body protein levels, lower ash levels and increased NPU values. If the low ash contents in the fish fed 100% soya diets are not prejudicial for health and growth of the fish in the long term, soya meal must be considered a superior protein source for C. macropomum. The high growth and the efficient use of the dietary protein indicate the C. macropomum is able to utilize soya protein more efficiently than other fish species.  相似文献

Juvenile Colossoma macropomum were fed ad libitum diets containing either 30% or 40% protein, while the dietary lipid level varied between 5% and 20%. Growth and protein utilization efficiency increased with the dietary lipid level. However, the economic feasibility of the addition of extra dietary lipid to C. macropomum diets is questionable, as for every extra gram of protein deposition it was necessary to replace 20–25 g of dietary carbohydrates by lipids. Moreover, increased dietary lipid levels resulted in increased lipid deposition. In C. macropomum, feed uptake was regulated by the dietary protein level and unaffected by the dietary lipid level. Dissection of the body into head, viscera and trunk revealed that 45–48% of the body lipids were stored in the trunk, independent of diet composition. A positive relation between dietary and body protein level was confirmed in this experiment. However, the dissection of the body revealed that the body protein concentrations in head + viscera and trunk are not changed significantly by the dietary protein level. The higher protein contents offish fed higher-protein diets are due to an increase in the relative weight of the trunk (muscle) to the total body weight.  相似文献

Fisheries zoning and the influence of biotic and abiotic factors in the spatial distribution of Colossoma macropomum (Cuvier) in the Piagaçu‐Purus Sustainable Development Reserve, Central Amazon, during 2012–2013 were evaluated. The relative abundance and population parameters of the C. macropomum were compared between conservation (no‐take zone) and fishing (open to fishing zone) lakes. The fish were collected by experimental fishing in 2012 and 2013. A total of 296 individuals were captured, most of which were from the conservation lakes (62.8%). Higher proportions of larger adults and individuals were found in the conservation lakes, although no significant difference in the average C. macropomum catch‐per‐unit‐effort was found between the lake categories. Abundance was positively influenced by zooplankton density (r2 = 0.53, < 0.03). Therefore, it is hypothesised that conservation lakes may serve as refugia for juveniles due to the high availability of planktonic food available during the dry season. Although there were environmental differences between the two lake types, the relative success of management models that use no‐take zones in freshwater habitats was highlighted. Finally, the importance of zooplankton as a factor in the distribution of C. macropomum among floodplain lakes was emphasised.  相似文献

Fish are frequently anesthetized to reduce stress and injury during various procedures, particularly surgery. Nonetheless, the majority of chemical fish anesthetics have some negative effects on both fish and humans. 2-Phenoxyethanol (2-PE), for example, which is normally used as an anesthetic for ornamental fish, can cause neuropsychological syndromes in humans. To reduce toxicity, some herbal essential oils have been applied as an anesthetic to fish. However, essential oils must be dissolved in ethanol, which has side effects such as disrupting gas exchange in fish. This study aimed to develop mugwort (Artemisia vulgaris) oil as a nanoemulsion without ethanol and determine their optimal concentrations to use as an anesthetic for surgery on koi carp (Cyprinus carpio). Gas chromatography-mass spectrometry (GC/MS) was used to examine the chemical compositions of mugwort oil. The droplet size of mugwort oil nanoemulsion was measured by the dynamic light scattering method. The results revealed that mugwort oil consisted of three major active compounds which were b-pinene (11.7%), 1,8-cineole (10.34%), and a-pinene (8.29%). The average diameter of mugwort oil nanoemulsion droplets was 22.08 ± 0.70 nm. The lowest effective concentrations were 0.6 ml/L for mugwort oil and 12.5 ml/L for mugwort oil nanoemulsion.

The amino acid profile of Colossoma macropomum was determined. Indispensable amino acid levels were similar to those reported in other fish species. Suitability of local, Costa Rican, ingredients as protein sources for C. macropomum was evaluated based on their protein content and indispensable amino acid profile. It was calculated that, in a standard diet (40% protein. 25% fish meal), blood meal, soya meal, poultry byproducts, meat and bone meal, and meat meal can substitute more than 80% of the fish meal. More than 40% of the fish meal can be substituted by shrimp meal, pigeon pea, cotton meal, peanut oil cake, cow pea, corn gluten and water hyacinth. As differences between amino acid profiles between fish species seem to be small, promising alternative ingredients for C. macropomum might be equally interesting for formulation of feeds for other species. Soya meal is the currently used ingredient with the highest potential to substitute fish meal completely. Further research is needed to determine if the use of alternative ingredients is limited by constraints such as decreased feed digestibility, presence of anti-nutritional factors, additional production and processing costs.  相似文献

Three size groups of Colossoma macropomum were submitted to a 4-week growth trial. Five nearly isocaloric (18.8-21.0 kJ g?1) diets with protein concentrations ranging between 17 and 64% were administrated at a fixed, near satiation level. Maximum growth was 6.6, 3.6 and 1.9 g protein kg?0.8 day?1 for 5, 50 and 125 g fish, respectively. The protein requirement to achieve maximum growth decreased from 28.9 g protein kg?0.8 day?1 for 5 g fish to 11.7 g kg?0.8 day?1 for 125 g fish. Possibly because of its high growth rate. C. macropomum needs a slightly higher dietary P/E ration (25.4-27.9 mg protein kJ?1) to obtain maximal growth than most other fishes. The relation between protein ration and protein gain was studied by a quadratic regression model. In fish receiving protein rations equal or below rations resulting in maximal growth, protein ration and protein gain were almost linearly related. The model showed that the portion of the dietary protein which is digested decreases with increasing protein ration. Body protein content increased and body lipid content decreased with feed protein level. Fish fed a 17% protein diet deposited as much as 18% lipid.  相似文献

The interaction between cultured barber goby Elacatinus figaro (cleaner fish) and the dusky grouper Epinephelus marginatus, and the efficiency of the cleaner fish in removing ectoparasites were evaluated. When the interaction between these two species was observed, cleaner fish showed a preference for the largest groupers. In a second trial, treatments: TWC (Control) – two groupers without a cleaner fish, T1C – two groupers with one cleaner fish, T3C – two groupers with three cleaner fish and T6C – two groupers with six cleaner fish were tested in four replicates. After 8 days, monogeneans were removed and identified as Neobenedenia melleni. The highest mean abundance of parasites was found on the groupers in the TWC group (37 parasites per host) and the lowest on those in the T6C group (4.1 parasites per host). By increasing the number of cleaner fish, a higher cleaning efficiency was obtained, as observed in T6C, where almost 90% of the parasites were removed. Possibly, this removal would have been complete if the number of cleaners had not been reduced in the treatments due to the mortalities observed. This study demonstrates the possibility of using gobies to remove monogeneans and in improving grouper health.  相似文献

Juvenile, 0.87 g Colossoma macropomum (Cuvier) were submitted to different ad libitum feeding regimes. Feeding frequency ranged from one to five meals per day and intervals between meals from 3 h to 24 h. Higher feeding frequencies resulted in higher feed uptake, a higher growth rate and a decreased feed utilization efficiency. Feed consumption was lower at 0700 h than at 1900 h. The amount of feed from different meals persisting in the digestive tract was determined with chromic-oxide-marked feed. This method revealed that with ad libitum feeding, about 21% of the feed remained uneaten. Feed losses did not differ significantly between feeding regimes. Feed accumulated in the digestive tract in the morning and early afternoon. In the late afternoon most feed rests disappeared from the digestive tract and feed consumption increased. It is suggested that feed uptake and growth of C. macropomum can be improved by extending the feeding period after 1900 h.  相似文献

The efficacy of anaesthetic tricaine methanesulfonate (MS‐222) was evaluated in four freshwater aquarium fish species, Zebrafish (Danio rerio), Guppy (Poecilia reticulata), Discu (Symphysodon discus) and Green swordtail (Xiphophorus helleri). The correct dose of anaesthetic should induce the plane 4 of anaesthesia in less than 180 s, recovery in less than 300 s and must survive when exposed during 30 min to anaesthetic. Fishes were exposed to six concentrations of anaesthetic (75, 100, 125, 150, 200 and 250 mg L?1) and the time of fish reaching plane 4 of anaesthesia, post exposure recovery, and the percentage of survival when fish were subject to 30 min in the anaesthetic were recorded. The optimal doses varied according to the species: D. rerio – 75, 100 and 125 mg L?1, P. reticulata – 125, 150 and 200 mg L?1, S. discus – 75 and 100 mg L?1 and X. helleri – 125 and 150 mg L?1. The induction time generally decreased significantly with increasing concentration of MS‐222 for all of the species evaluated. The recovery time had a tendency to increase with the increase of the MS‐222 concentration for D. rerio, P. reticulata and S. discus. On the other hand, X. helleri recovery time decreased with the increase of MS‐222 concentration. MS‐222 proved to be effective in anaesthesia for all the freshwater ornamental species studied. The main results clearly show that the optimal dose to anesthetize is fish species dependent and it is completely wrong to extrapolate optimal anaesthetic concentrations between different species.  相似文献

Accumulated data from growth experiments on the nutrition of Colossoma macropomum (Cuvier) were analysed using an empirical (linear regression) and an explanatory model called the fish growth simulator (FGS). Both methods gave satisfactory results. Differences between predicted and observed values fluctuated less with the FGS. However, the linear regression method is simple and results are easily translated to practical recommendations. The FGS covered a wider range of experimental conditions than the linear regression method. Both linear regression and the FGS stressed the importance of dietary protein in growth of C, macropomum. Dietary protein concentration is the main explaining variable of ad libitum feed uptake, while protein ration is the main variable determining growth. Dietary protein concentration also affects body protein and body lipid content. Protein utilization efficiency data indicate that substitution of fishmeal by soya meal increases dietary protein quality. Although C, macropomum can thrive under less than optimal conditions, a high-quality, high-protein diet is required for maximal growth and optimal feed utilization.  相似文献

Despite carotenoids and essential fatty acids seem to play important roles in fish reproduction, no studies have yet been conducted to determine the effect of dietary carotenoids on gilthead seabream broodstock performance and their relation as antioxidants with dietary n‐3 highly unsaturated fatty acid (HUFA) levels. In addition, the high cost of synthetic sources of carotenoids is leading to the search for new natural carotenoid sources such as paprika oleoresin. Four experimental diets containing two combined levels of carotenoids from paprika oleoresin (40 and 60 mg kg?1) and n‐3 HUFA (25 and 40 g kg?1) were respectively fed to triplicate groups of gilthead seabream (Sparus aurata) broodfish. Elevation of n‐3 HUFA dietary levels from 25 to 40 g kg?1 significantly improved gilthead seabream broodstock performance in terms of egg viability, hatching rates and fecundity. Besides, it markedly increased egg contents in HUFAs which play important energetic and structural roles and improve embryo development. Both arachidonic acid and eicosapentaenoic acid (20:5n‐3) egg contents were more readily affected by dietary n‐3 HUFA than docosahexaenoic acid. HUFA levels did not caused any negative effect suggesting an optimized content of antioxidants in broodstock diets. Increase in dietary carotenoids from 40 to 60 mg kg?1 increased carotenoid contents in eggs and significantly improved egg fertilization rates suggesting an important sperm cell’s protective role by reducing the risk of lipid peroxidation which is detrimental for sperm motility. The increased inclusion of dietary paprika oleoresin enhanced egg carotenoid deposition and improved fish reproductive performance, denoting the high nutritional value of this product as a source of carotenoids for broodstock of this species.  相似文献

Our study assessed the efficiency of a formulated new extender in maintaining viability and morphological integrity of Colossoma macropomum spermatozoa under chilling storage. Semen was diluted in the test extender and BTS? (Beltsville Thawing Solution) and exposed to a short‐term storage at 4.6 ± 0.6°C for 96 hr. Both extenders were able to maintain 17% ± 8% motile spermatozoa by the end of experiment. Sperm dilution in test extender did not affect the morphologically normal cells (61% ± 6%) up to 48 hr of chilling, being higher than in BTS? (50% ± 6%) (p < 0.05). After 96 hr, samples kept in the test extender had 50% of normal spermatozoa, whereas those kept in BTS? presented only 38% of normal cells. Chilling storage increased the incidence of cells with strongly coiled flagella in BTS?. Our study is the first to evaluate in detail the spermatozoa morphology as indicative of C. macropomum semen viability. The new extender was able to protect the spermatozoa against increase in coiled flagellum injuries.  相似文献

Colossoma macropomum is the second largest scaled fish in the Amazon basin. Reduced harvests in recent decades suggest that populations of this species are already overexploited in some areas. In this study, the population dynamics of C. macropomum was investigated using length and weight data for fish caught in the Manacapuru Lake system, a large ria lake located along the lower reach of the Solimões River. A total of 1270 individuals of C. macropomum with standard lengths varying from 17 to 62 cm and mean length of 31.0 ± 6.7 cm were sampled. Weights varied from 165 to 8195 g, and mean weight was 1148.3 ± 883.8 g. Maximum sustainable yield was estimated at 285.8 g recruit−1, corresponding to a fishing mortality of 0.51 year−1. A scenario analysis based on changes in fishing mortality and age at first capture, two parameters potentially addressed by fishing management strategies, suggested that any improvement in fishing yield depends on increases in age at first capture.  相似文献

Two experiments were carried out to evaluate the effects of palm kernel meal (Elaeis guineensis) as corn (Zea mays) substitutes in tambaqui (Colossoma macropomum) diets based on zootechnical performance, health, characteristics of the muscle, yield and economic efficiency. The first one was carried out in a recirculation aquaculture system for 75 days wherein tambaquis received diets with 0% (T0), 25% (T25), 50% (T50), 75% (T75) and 100% (T100) of corn replacement by palm kernel meal. The second experiment, conducted in ponds for 184 days, tambaquis were fed T0 and T25 diets (selected because showed greater weight gain without health damage). Final weight and weight gain of tambaquis were impaired by the increasing levels of this ingredient on diets, and the feed conversion ratio was worse. The protein efficiency ratio and specific growth rate gradually reduced, while the total number of leucocytes and neutrophils gradually increased. Cholesterol and triglycerides decreased in T75 and T100. Variables analysed in experiment 2 were not affected by treatment, except muscle lipids. Palm kernel meal can replace up to 25% of corn in tambaqui diets since it maintains a balance in economic and zootechnical performance of production.  相似文献

Fish farming has grown rapidly in Brazil over the last two decades, due in part to the availability of water and species with potential for cultivation, such as Colossoma macropomum. This farmed fish, however, can be affected by parasitic infections with high mortality rates (>35%). The main objective of the present study was to describe the first occurrence of the Trypanosoma species in Colossoma macropomum, which occurred in fish cultured in the Brazilian Amazon region, as well as to morphologically characterize the trypomastigotes and the impact of these hemoparasites on the body condition of the hosts. The trypanosomes found in the blood of Colossoma macropomum were morphologically characterized by the size of the trypomastigote form, indicating the presence of only one morphotype. Of the 39 hosts examined from one fish farming, 41.0% were infected by Trypanosoma sp., with low levels of infection (mean blood density of 3.7/μL). The condition factor of the asymptomatic hosts was not affected by hemoparasitism. There was no correlation between the abundance of Trypanosoma sp. and the condition factor and size of the hosts. Finally, our understanding of host-parasite interactions and the detection of emerging diseases are fundamental for aquaculture.  相似文献
