Somatolactin (SL) is a novel pituitary hormone recently characterized in several fish species. Structural analyses have shown that SL belongs to the growth hormone/prolactin family, and that it is a highly conserved protein. SL is synthesized by the periodic acid/Schiff-positive cells in the pars intermedia, but has an as yet unidentified function(s). We have recently developed a homologous radioimmunoassay for coho salmon SL and measured plasma levels of SL during two stages of the coho salmon life cycle, smoltification and sexual maturation. During smoltification, plasma levels of SL changed almost in parallel with plasma levels of thyroxine; levels increased as morphological indices of smoltification appeared and decreased as smoltification was completed. Following this period, SL levels remained low until the spring prior to spawning. In a separate study, plasma levels of SL were measured in sexually maturing coho salmon that remained in fresh water throughout their life cycle. During the year of sexual maturation, plasma levels of SL gradually increased from the spring onward, reaching peak levels at the time of spawning in November and December. These data are similar to those previously reported for sexually maturing coho salmon that were maintained in seawater prior to spawning (Rand-Weaver et al. 1992). Therefore, increases in plasma SL levels occurred in sexually maturing fish irrespective of whether they were maintained in fresh water or seawater. Peak levels at spawning were higher than those observed during smoltification. Possible roles for SL in metabolism and reproduction are discussed.  相似文献

The expression levels of growth hormone receptor (GHR) and insulin-like growth factor (IGF-I) in gonads of maturing coho salmon were measured by real-time PCR. The GHR and IGF-I mRNAs showed different gene expression pattern between male and female. Effects of dietary tetradecylthioacetic fatty acid (TTA) on muscle fat, development of gonads and early sexual maturation in S0 Atlantic salmon during the first year in sea were investigated. TTA (0.5% w/w) was added to the feeds for 8 weeks in the spring. In May, at the end of the TTA‐feeding period, the fish in the TTA group had significantly (P < 0.05) less fat (10.1%) stored in muscle compared with the control group (10.8%). In September, mean male gonadosomatic index (GSI) in maturing fish in the TTA group was found to be lower compared with the maturing fish in the control group (P = 0.05). On the basis of GSI values, male sexual maturation in September was 10.0% vs. 14.4% for the TTA and the control group respectively. Thus, relative to the control group, the incidence of male sexual maturation in the TTA group was reduced by about 1/3 (P = 0.002). Production data was not affected by dietary supplementation of TTA. This study reveals that TTA significantly reduces the incidence of male sexual maturation in S0 Atlantic salmon. A significant elimination model of TTA in fish muscle that takes into account, the growth rate of the fish was further developed in this study.  相似文献

While the pineal organ in poikilothermic vertebrates consistently influences a number of physiological processes, its precise role, and what controlling signal (hormonal and/or neural) is abolished by its removal, is still uncertain. For this reason, the effect of long-term pinealectomy (PINX) on seasonal growth and reproductive development was investigated in Atlantic salmon, Salmo salar, parr. Mean daytime plasma melatonin level in the control, sham-operated and PINX fish was 35 pg·mL–1 in all groups, while mean night-time levels were 296, 255 and 25 pg·mL–1, respectively, indicating that PINX abolished the natural nocturnal rise in melatonin. Pinealectomy did not influence the incidence or timing of early sexual maturation in the male parr. However, pinealectomy significantly affected growth, its effect being strongly seasonally dependent. Compared to sham-operated and control fish, pinealectomized fish showed significantly lower specific growth rates (SGRs) during the period of lengthening photoperiod up until the summer solstice. Thereafter, corresponding to the season of decreasing photoperiods, the pinealectomized fish exhibited higher SGRs. These results suggest a functional relationship between the pineal organ and somatic growth in the Atlantic salmon, although what controlling signal from the pineal (melatonin or neural) is involved has yet to be determined.  相似文献

Precocious sexual maturation of male Atlantic salmon (Salmo salar) was induced by photoperiod manipulation to shorten the time required for optimisation of sex inversion treatments. Exposure to a short-day photoperiod (8L:16D) prior to the summer solstice reduced the time to completion of sexual maturation from 3 years to 8–9 months for the majority of fish.  相似文献

Early sexual maturation of male chinook salmon (maturation 1 to 4 years prior to females in the same age class) results in reduced effectiveness of stock enhancement programs and a financial loss to the salmon farming industry. Previous studies in Atlantic salmon have shown that the age of maturity in males is affected by growth and/or body energy stores, but the relative roles of these two factors are not well understood. Therefore, an experiment was designed to determine when spermatogenesis was initiated, to characterize the endocrine changes during the onset of puberty in male salmon, and to determine if the level of whole-body lipid affects the incidence of early male maturation in a wild stock (Yakima River) of 1+ spring chinook salmon. Fry were fed a commercial diet from February until August and were then divided into groups of 320 fish (mean weight, 5.6 g) and fed one of five experimental diets (two replicate groups/diet) containing 4%, 9%, 14%, 18% or 22% lipid and 82%, 77%, 73%, 69%, or 65% protein for 13 months. Fish were reared on natural photoperiod and ambient temperature (6°C to 16°C), and pair-fed to a level based on the tank with the lowest feed consumption. Fish were weighed monthly and sampled to determine body composition, pituitary follicle-stimulating hormone (FSH) and luteinizing hormone (LH) levels, plasma insulin-like growth factor I (IGF-I) levels, and stage of gonadal development.

Throughout the experimental period the mean fish weight was similar among treatment groups. However, from December through the end of the experiment in the following September, maturing males were significantly larger than nonmaturing fish. Initial lipid levels in 0-age experimental fish were near 6%, which is similar to wild fish of the same stock and age captured in the Yakima River during August. Fish fed diets containing more than 4% lipid increased in whole-body lipid content during the first 2 months of feeding and then maintained at relatively constant levels during the course of the experiment. Whole-body lipid levels for the dietary treatment groups averaged 5.6%, 7.1%, 8.2%, 9.4%, and 9.6% from October through the following September.

Based on histological examination of the testes of experimental fish, type B spermatogonia and primary spermatocytes were first observed in some of the yearling males during November. These were designated maturing males. Pituitary FSH levels were significantly higher in maturing than nonmaturing males at this time and for the remainder of the study. Pituitary FSH levels increased as spermatogenesis proceeded in maturing fish, whereas pituitary LH levels increased in maturing 1+ males only during July and August, when testes were in late stages of spermatogenesis and in September during spermiation. Plasma IGF-I levels were significantly higher in maturing males than nonmaturing fish from December through the end of experiment. Since maturing males were significantly larger than nonmaturing fish of both sexes from December through September, the difference in IGF-I levels could be due to differences in growth or due to maturation.

The percentage of maturing males was significantly influenced by whole-body lipid, increasing from 34% in fish fed the 4% lipid diet to 45% in fish fed the 22% lipid diet. These data suggest that whole-body lipid levels influenced the incidence of maturation of male spring chinook salmon. In addition, both endocrine and histological indicators suggest that maturation was initiated in males approximately a full year prior to the time the fish will spawn.  相似文献

The roles of salmon GnRH (sGnRH) and gonadal steroid hormones in regulation of LH synthesis and release were examined in primary pituitary cell cultures of masu salmon (Oncorhynchus masou). Pituitaries were taken from fish at four reproductive stages: in March (initiation of sexual maturation); May (early maturation); July (pre-spawning); and September (spawning period). Amounts of LHβ subunit mRNA in the pituitary cells were determined by real-time PCR, and LH levels in the medium were determined by RIA. sGnRH and gonadal steroids including estradiol-17β (E2), testosterone (T) and 11-ketotestosterone (11-KT) were added to the cultures to examine their direct effects on LH response. sGnRH had no significant effect on LHβ mRNA levels at any stages, although a stimulatory trend was noted in March. In contrast, E2 and T considerably increased LHβ subunit mRNA levels in March and May during initial stages of maturation, and the effects were less pronounced in July and September. On the other hand, sGnRH stimulated LH release at all stages in the males and the effects were most prominent in July and September. E2 and T also stimulated LH release in July and September, but their effects were weaker than that of sGnRH. The present results indicate that sGnRH and gonadal steroids directly regulate LH synthesis and release in masu salmon pituitary cells: sGnRH mainly stimulates LH release in the late stage of sexual maturation; whereas, E2 and T are effective in stimulating LH synthesis at earlier stages of maturation.  相似文献

The effects of various environmental parameters on sexual maturation of two sea-winter Atlantic salmon (Salmo salar) were tested in two separate experiments. In the first experiment Atlantic salmon with initial mean individual weight 1.5 kg (smolt 13 months before) were reared for 8 months from June to February at different oxygen levels and stocking densities using continuous light. Oxygen levels of 5–7, 7.5–9.5 and 10–12 mg O2 l−1 and stocking densities starting at about 20, 30 and 40 kg m−3 and increasing as the fish grew to 80–90 kg m−3 for the highest densities were tested in a factorial design. Only male fish matured, and incidence of maturation among males varied from 4.1% to 25% between tanks. The highest percentage of mature males was found in the tanks with low stocking density. No clear effect on oxygen level was found.The second experiment lasted 20 months from seawater transfer in May until the fish weighed 3.3–3.5 kg. Two water current speeds (14–16 and 20–24 cm s−1) and two photoperiod regimes (LD 20:4) and continuous light (LL) were tested in a factorial design. Neither swimming velocity nor photoperiod affected growth rate. Continuous light reduced the incidence of sexual maturation. The average proportion of maturation among males was 8% and 25% under the LL and LD 20:4 regimes respectively. The fish reared under the LD 20:4 light regime had a significant lower condition factor and significant larger hearts than the fish reared under continuous light. Swimming velocity had no significant effect on the incidence of maturation. The results indicate that the swimming velocity must be higher than 0.5 BL s−1 in order to influence the energy stores. An important finding in this study is that light cues are not required for gonadal growth. The results also indicate that environmental factors can affect maturation even after the first sea-winter.  相似文献

Plasma somatolactin (SL) concentrations were examined in chum salmon in relation to gonadal maturation; immature salmon in the Bering Sea at various stages of maturation, and mature salmon during upstream migration caught at the ocean, bay and river. Plasma SL concentrations as well as plasma prolactin (PRL) and growth hormone (GH) levels in the immature fish caught in the Bering Sea were maintained essentially at similar levels. Plasma SL in mature salmon increased significantly from the fish in the ocean to the fish in the river in both sexes. Although all the fish had fully developed gonads, females completed ovulation while still in the bay, whereas final spermeation in males was achieved after entry into the river. Thus, no clear correlation was seen between plasma SL levels and final gonadal maturation. On the other hand, plasma PRL concentrations in both male and female fish were higher in the fish in the river than those in the ocean and bay, and plasma GH levels were higher in both sexes in the fish in the bay and river than those in the ocean. Plasma levels of triglycerides, glucose, free fatty acids and ionized sodium and calcium were also examined. Significant-negative correlations were seen between plasma SL and plasma ionized calcium in mature male salmon, and between plasma SL and plasma triglycerides in mature female salmon. Although our findings do not rule out the possibility of the involvement of SL in final maturation, the results indicate that SL seems to be involved at least in energy and/or calcium metabolism during the spawning migration.  相似文献

A major challenge in Clariid catfish seed production is our inability to induce voluntary captive spawning of the brooders. Since voluntary captive spawning had been previously elicited in a Clarias species by simulating natural breeding environment, we speculated that the major obstacle in voluntary spawning might be the absence of an inducing agent that triggers mating response in the brooders. To address the issue, oxytocin was injected to the brooders along with Ovatide (a commercial inducing agent comprising salmon gonadotropin‐releasing hormone analogue and domperidone). Administration of 40 mIU/kg body weight of oxytocin after 12 hr of Ovatide injection resulted in voluntary spawning of the fish (81.5%, i.e. 31 out of 38 trials), with more than 70% hatching rate in simple and low‐cost spawning facilities. Histology of male gonads indicated sustained gamete maturation and the gonads were observed to be visibly turgid in the fish that received both the hormones. In contrast, gamete maturation slackened after 10 hr in the control fish, injected with Ovatide only. Administration of oxytocin along with Ovatide was also observed to induce aggressive sexual behaviour among the males. Neither of the hormones, acting alone, could induce voluntary spawning in the fish or influence sexual behaviour. Our results clearly show that oxytocin plays a significant role in sustaining the male gamete maturation induced by Ovatide and trigger the behavioural mating response, leading to voluntary spawning in C. magur. The results hold potential to make a paradigm shift in the commercial catfish seed production technology.  相似文献

The sex differentiation period of the Siberian sturgeon was investigated through expression profiling of two testicular markers (dmrt1 and sox9). At the molecular level, a clear sexual dimorphism of dmrt1 and sox9 was observed in 3-year-old fish with immature gonads, in which males showed higher expression of these genes. Among 16-month-old sturgeons cultured in Uruguay, gonad morphology analyses showed one group of fish with undifferentiated gonads and a second group which had started their histological differentiation into ovaries or testes. dmrt1 showed a significantly higher expression in testes of recently differentiated fish, but this was not the case for sox9. In undifferentiated fish, we observed two clearly different groups in terms of expression: one group of fish over-expressing male markers (dmrt1, sox9) and another group of fish showing very low expression of these genes. This suggests that fish undergoing male differentiation can be identified by their profiles of gene expression before they undergo morphological differentiation.  相似文献

It was demonstrated that pregnenolone (P5) and 17α-hydroxypregnenolone (17α-OH-P5) were identified as cholesterol metabolites in the brain and pituitary gland of lacustrine sockeye salmon (Oncorhynchus nerka) during sexual maturation. Moreover, 17α-OH-P5 was determined as the major cholesterol metabolite. The conversion of P5 to 17α-OH-P5 in the brain and pituitary gland showed a clear regional difference, and their productivity were dramatically changed during sexual maturation. From these data, it was suggested that steroid converting enzyme in the brain and pituitary gland of lacustrine sockeye salmon might be involved in brain region-dependent roles on sexual maturation.  相似文献

SOX9基因在脊椎动物性别决定和性腺分化中扮演重要的调控作用。从mRNA和蛋白水平分析中华鳖SOX9基因在不同组织中的差异性表达、在胚胎性腺和成年睾丸中的细胞定位以及在性逆转中的表达变化,研究SOX9基因在中华鳖性别分化中的调控作用。Real-time PCR结果显示,SOX9基因在中华鳖雄性性腺中特异性表达。免疫荧光染色分析显示,SOX9蛋白在雄性18期胚胎性腺中开始表达,随着性腺的发育,SOX9蛋白定位于性腺Sertoli前体细胞细胞核中;而在雌性胚胎性腺并未见其表达。此外,在雌激素诱导的雄性向雌性性逆转胚胎中,SOX9基因显著下调,而在芳香化酶抑制剂诱导的雌性向雄性性逆转胚胎中,SOX9基因表达则显著上升。研究表明,SOX9基因为中华鳖雄性特异性基因,参与雄性性腺的发育过程,可能在中华鳖早期性别分化过程中起调控作用。  相似文献

Atlantic salmon (Salmo salar) that mature sexually at 1 year of age (as parr or precocious males), at 4 years of age (after two winters in the sea) and at 5 years of age (after three winters in the sea) were used to study response to individual selection for age at sexual maturity. Six mating groups were made: between fish of the same age (4 × 4 and 5 × 5); reciprocal matings between 4- and 5-year-old fish (4 × 5 and 5 × 4); and matings between 1-year-old precocious males and 4- and 5-year-old females (1 × 4 and 1 × 5). Parental age at sexual maturity was found to influence markedly the offspring age at sexual maturity. The heritability for age at maturity in the sea was estimated at 0.48 ± 0.20 from the offspring—dam regression. It is concluded that there is considerable opportunity to alter the age at sexual maturity in Atlantic salmon through selection. Evidence was found that maturation of parr is a heritable trait and that it is independently inherited from maturation in the sea.  相似文献

A group of Atlantic salmon (Salmo salar) was followed through their first year of maturation and spawning. At monthly intervals, starting with juvenile fish in December, 5–7 fish of each sex were killed, and liver and plasma were sampled. The last sampling point was of spawning fish in November a year later. Variables in the cytochrome P450 (P450) system were studied in hepatic microsomes, and estradiol 17 was measured in the plasma of females to assess the maturational status. The P450 1A1-mediated 7-ethoxyresorufin O-deethylase (EROD) started at high levels in winter, but decreased to non-detectable activities in pre-spawning females. Decreases, but not to the same extent, were also observed during this period in total cytochrome P450, cytochrome b5, NADPH-cytochrome P450 reductase, and in the content of two immunochemically determined P450 isozymes. At the same time, LSI levels increased in maturing females (starting in July), and GSI levels increased in both sexes (starting in May). Sex specific differences were observed in pre-spawning fish in September and October, with levels of total P450, b5, NADPH-cytochrome P450 reductase, EROD and P450 isozymes significantly lower in females. At the same time, plasma estradiol-17 levels reached peak values in females. The results point to the important role of sex steroids such as estradiol-17 as major factors in the regulation of final sexual maturation. However, this study also indicates that there may be estradiol-17 independent events of equal importance in the early stages of gonadal maturation that may involve the P450 system. The changes observed in the P450 system (as a major drug and steroid metabolizing system) of Atlantic salmon during sexual maturation may be of importance both in the endogenous transduction of hormonal signals, and as a pharmacological basis for designing therapeutic treatment of diseases in the aquaculture industry.Parts of this work were presented at the 5th International Symposium on Responses of Marine Organisms to Pollutants, April 1989 in Plymouth, United Kingdom (Larsen and Goks\/oyr 1989).  相似文献

Commercially farmed Atlantic halibut (Hippoglossus hippoglossus) were reared at the Aga Marin's facility located on Dønna (Norway) under ambient environmental conditions in duplicate 15 × 15 × 8 m netpens (May 2004 to May 2005). Twenty fish were sampled five times over a twelve month production period during which time the average body weight increased from 1.26 to 2.08 kg (n = 100 fish). Body mass, fork length (LF), and the number and size distributions of fast muscle fibres were determined in male and female fish. All males matured during the autumn whereas no maturation was observed in females. From the point of maturation females had superior growth performance to males and body mass and the total cross-sectional area of muscle were 1.4-fold (P < 0.05) and 1.3-fold higher (P < 0.01) respectively by May 2005. The total number of fast muscle fibres per trunk cross-section at 0.55 LF was 24.5% higher in females (7.58 × 105) than males (5.80 × 105) prior to sexual maturation. In females, muscle fibre recruitment slowed with short days and low water temperatures (< 6.5 °C), but had increased to 1.01 × 106 by May 2005 (P < 0.001). In contrast, there was no growth and no increase in muscle fibre number in males following the onset of maturation. The distribution of muscle fibre diameters prior to maturation in males was also significantly different between male and female fish matched for LF. The results illustrate a sexual dimorphism of muscle fibre recruitment patterns in Atlantic halibut and highlight the adverse affects of sexual maturation in males on muscle growth.  相似文献

Multiple forms of the gonadotropin-releasing hormone (GnRH) exist in teleost fish. A salmonid fish, masu salmon Oncorhynchus masou has salmon GnRH (sGnRH) and chicken GnRH-II (cGnRH-II). sGnRH neurons were scattered from the olfactory nerve through the ventral telencephalon (VT) and the preoptic area (POA). sGnRH but not cGnRH-II was detected in the pituitary. sGnRH mRNA levels in the VT and the POA increased during gonadal maturation, suggesting that sGnRH neurons in these areas are involved in gonadal maturation. sGnRH neurons were first detected in a cluster near the olfactory epithelium 40 days after fertilization. sGnRH neurons were not detected in the brain by the olfactory epithelia lesion, suggesting that sGnRH neurons are derived from the olfactory epithelium. A pleuronectiform fish, barfin flounder Verasper moseri has sGnRH, cGnRH-II and seabream GnRH (sbGnRH). sGnRH and cGnRH-II-immunoreactive fibers were observed throughout the brain, but not in the pituitary. sbGnRH neurons were located in the POA and sent fibers to the pituitary, indicating that sbGnRH is involved in GTH secretion. Judging from the location of neuronal somata and their projections, it is indicated that three GnRH systems exist in the barfin flounder; the TN-, the MT- and the POA-GnRH system. However, in masu salmon, clear anatomical identification of the TN- and the POA-GnRH system is difficult, because the GnRH neurons located in the ventral forebrain are consecutive and the GnRH form produced in these neurons is the same (sGnRH). Thus, it is suggested in masu salmon that sGnRH neurons are derived from the olfactory epithelium, migrate into the brain and play different roles according to the location in the brain.  相似文献

The significance of the olfactory sense upon sexual maturation in Atlantic salmon was studied. Eighty-one salmon in the salt-water stage were rendered anosmic by cauterizing the olfactory mucosa, a further 81 fishes served as a reference group. All fishes were raised in the same floating net cage. The olfactory mucosa was destroyed once more during the experimental period in the already anosmicized group in order to avoid regeneration. After 5 months, the sexual maturation in both groups was determined by visual inspection of the gonads, and ranged on a six-step scale. The results indicate that the olfactory sense is not mandatory to late development of sexual maturity. No difference was found between the two groups with regard to growth during the experimental period. This finding indicates that the olfactory sense is of little importance to salmonids in the competition for food in a fish-farming environment.  相似文献
