四株鸡传染性法氏囊病病毒毒力的研究   总被引:5,自引:0,他引:5  
对从黑龙江省分离的4株鸡传染性法氏囊病病毒通过易感鸡接种,用病理学技术进行了致病力的研究。结果表明,各毒株毒力有一定差异,基保H3,H4株毒力高于H1,H2株,且有迅速致法氏囊萎缩的特性;H3,H4株引起试验鸡法氏囊指数显著下降(P〈0.05),脾脏指数明显增加(P〈0.05),H3,H4株接种50日龄D78疫苗免疫病,免疫保护指数分别为50%,60%,传统疫苗不能提供以分保护。  相似文献

助阳中药粗提物对粤黄鸡免疫功能的影响   总被引:5,自引:0,他引:5  
将1日龄健康雌性粤黄鸡120只随机均分为4组。Ⅰ组饲喂基础日粮,Ⅱ,Ⅲ,Ⅳ组在基础日粮中分别添加2%淫羊藿粗提物、2%补骨脂粗提物、1%淫羊藿粗提物+1%补骨脂粗提物。在14日龄时给每只鸡接种1次鸡新城疫Ⅰ系疫苗。试验期(8周)末,与Ⅰ组比较,Ⅱ、Ⅳ组胸腺指数显著减小(P<0.05),脾脏指数显著增大(P<0.05),Ⅳ组的法氏囊指数也显著增大(P<0.05);外周血液中T淋巴细胞百分率以Ⅱ、Ⅳ组升高极显著(P<0.01),Ⅲ组升高显著(P<0.05);血清中NDV抗体滴度Ⅱ、Ⅳ组升高显著(P<0.05);血清中T3含量Ⅱ、Ⅲ组升高显著(P<0.05)、Ⅳ组升高极显著(P<0.01);T4含量仅Ⅳ组升高显著(P<0.05),Ⅱ、Ⅲ组升高不显著(P>0.05)。上述结果提示,淫羊藿粗提物和淫羊藿粗提物+补骨脂粗提物均能增强粤黄鸡细胞免疫及体液免疫功能,而补骨脂粗提物只增强细胞免疫功能。  相似文献

本文应用鸡新城疫(ND)Ⅱ系和V4弱毒疫苗分别气室接种不同孵化日龄鸡胚,出雏后计算其孵化率并在不同日龄测定血清HI抗体效价,结果表明,NDⅡ系和V4疫苗1∶10稀释,于18、19胚龄气室接种,对孵化率无显著影响(P>005),两种疫苗间也无明显差异(P>005),均能产生良好的免疫保护力。  相似文献

冻前于牛稀释精液中添加32mmol/L还原型谷胱甘肽(GSH)做为试验组,对照组冻精中无GSH。结果表明:冻精刚解冻后精子活力试验组比对照组高75%(P<001);解冻后稀释精液在37℃水浴中保存3小时后,精子活力和顶体正常精子数量试验组比对照组分别高78%(P<001)和445%(P<005);37℃水浴条件下保存时精子寿命试验组比对照组平均延长32min(P<001);此外,GSH使母牛情期受胎率提高了624个百分点(P<005)。  相似文献

96 头“杜长大”生长猪按饲养试验要求分成 4 组(每组 3 个重复),分别饲 以添加 0、1 000、1 500、2 000 m g/kg甜菜碱的基础日粮,试验期 62 d。结果表明,1 000 m g/kg 剂量组试验猪生长性能最佳,日增重较对照组提高了 1320% ( P < 001), 料重比降低了 793% ( P < 001)。该组猪血清 G H、 I G F I、 T3 、 T4 水平分别升高了 10176% ( P< 001), 4475% ( P < 001), 2653% ( P< 001) 和 1683% ( P< 005); 血清游离丝氨酸含量提高了 1428% ( P < 005); 血清总蛋白上升了 2169% ( P < 001), 血清尿素氮含量降低 了 4767% ( P < 001); 背最 长肌 和 肝脏 中 R N A 含量 及 背最 长肌 R N A/ D N A 比 率分 别升 高了1260% ( P < 005), 1780% ( P < 002) 和 1979% ( P < 002); 肝脏 和腺 垂体 c A M P 含 量提 高了4753% ( P < 001) 和 6521% ( P < 001)。研究结果提示,甜菜碱似作用  相似文献

大麦日粮中添加粗酶制剂对鸡外周血液中代谢激素的影响   总被引:9,自引:1,他引:8  
为探讨粗酶制剂促进鸡生长的作用机制,通过三系列试验研究了粗酶制剂添加于大麦日粮中对不同基因型或生理状况的鸡血液代谢激素的影响。结果表明,粗酶制剂处理,使蛋用雏鸡T3(三碘甲腺原氨酸)、TSH(促甲状腺激素)和T3/T4(甲状腺素)显著提高(P<0.01或P<0.05);21日龄肉鸡T3、Ins(胰岛素)、T3/T4和Ins/Gluc(胰高血糖素)提高(P<0.05);42日龄肉鸡T3、GH(生长激素)、Ins、T3/T4和Ins/Gluc显著提高(P<0.05),而Gluc显著降低(P<0.05);生长后期鸡T3、GH和T3/T4显著升高(P<0.05)。结果提示,粗酶制剂可通过调节代谢激素的分泌而提高鸡的生产性能。  相似文献

鸡传染性喉气管炎病毒SA2疫苗株免疫效力的研究   总被引:1,自引:0,他引:1  
鸡传染性喉气管炎病毒(ILTV)疫苗株尿囊膜乳剂原液经点眼接种2、7和15日龄雏,2日龄雏鸡组在接种后第7天有20%(1/5)鸡出现眼睑肿胀和流泪等眼部反应,7日龄和15日龄雏鸡组无明显临床变化。用该疫苗株乳剂原液点眼接种1月龄以上鸡眼部无明显反应,但通过气管接种后,26.7%(4/15)鸡表现出ILT临床症状。试验用SA2株点眼接种免疫1月龄以上鸡,免疫组保护率为100%(20/20),对照组发病率为100%(10/10)、死亡率为40%(4/10)。2、7和15日龄雏鸡点眼毒力试验的3组鸡,于接种后21~34天用王岗株ILT强毒攻击,100%得到保护,而对照组鸡100%发病。试验结果表明,SA2疫苗株是一株毒力温和、副作用小而且免疫效力好的疫苗用毒株  相似文献

4株不同毒力鸡传染性法氏囊病(IBD)活疫苗,分别接种30dSPF公维,4d后再接种鸡新城(ND)Lasota系活疫苗,同时每组捕杀5只,观察法氏囊损伤情况,另4组用同1天的SPF公无接种ND-Lasotq系疫苗4天后再接种不同毒力的IBD疫苗。试验结果表明,4株IBD活疫苗对雏鸡法氏囊不同程度损伤,并且对ND免疫抗体产生有明显抑制作用。  相似文献

用新城疫病毒( N D V) La Sota 株、鸡传染性支气管炎病毒 ( I B V) M4 1 株接种鸡胚, 收获含毒鸡胚尿液。用传染性法氏囊病病毒 ( I B D V) 地方强毒 H B9 0 9 株接种30 ~40 日龄肉用鸡, 发病后取囊组织 ( 或收集 I B D 典型发病鸡囊组织) 制备成含毒组织悬液。将三种病毒液按一定比例混合, 制备成三联油乳剂灭活苗。三联苗免疫18 日~28 日龄雏鸡, 免疫后21 天攻毒测保护力, 三种传染病保护率均在90 % ~100 % , N D H I抗体≥1∶32 , I B D V A G P阳性率100 % , I B V N 抗体≥1∶8 , 疫苗免疫期可达4 个月以上。疫苗在4 ℃~8 ℃保存期为18 个月, 10 ℃~20 ℃保存为6 个月, 该疫苗在全省不同地区、不同品种鸡应用15 万羽份, 取得理想免疫效果。  相似文献

1 日龄艾维茵肉仔鸡120 只,随机分组,自由采食以葡萄糖- 大豆分离蛋白为主要原料的半纯合日粮,日粮中分别添加 0、11 和440m g/kg 铜,饮用去离子水,分别于 1 日龄皮下接种 H V T 冻干苗、7 和14 日龄滴鼻接种 Lasota 系冻干苗。试验结果表明,日粮中0 或440m g/kg 铜添加组与11m g/kg 铜添加组相比:19、43 日龄体重小( P< 005),0m g/kg 铜添加组0~43 日龄单位增重料耗显著高于11m g/kg 铜添加组( P< 005);胸腺、法氏囊和脾脏不同程度地萎缩( P< 005),显微分析0m g/kg 铜添加组胸腺、脾脏、法氏囊、盲肠扁桃体和哈德氏腺发现,淋巴细胞减少且分布疏松,胸腺淋巴细胞膜和核膜不清晰; T、 B淋巴细胞对丝裂原的反应性降低( P< 005);血清中 N D V、 M D V 抗体 E L I S A 效价降低( P< 005),说明铜对维持机体免疫功能状态必不可少。  相似文献

Five kinds of infectious bursal disease (IBD) immune complex (IC) vaccines were prepared with infectious bursal disease virus (IBDV) BX strain mixed with IBDV hyperimmune serum according to a certain proportion (containing 32, 8, 4, 0.5 and 0.125 units IBDV neutralizing antibody, respectively).One-day old low maternal antibody chickens were vaccinated with IBD IC vaccines 1 to 5 and BX strain live vaccine, respectively, pathological changes of the bursa of fabricius in chickens were observed at the 9th day after immunization.On the day of 28 after immunization, blood samples were taken and the IBDV neutralizing antibody were tested, meanwhile all experimental chickens were challenged with high virulent IBDV, the protective rates of vaccines were calculated.The results showed that at the 9th day after immunization, the bursa of fabricius were normal in IC vaccine 1, 2 and 3 groups, however 2/10, 4/10 and 5/10 of pathological changes of the bursa of fabricius in IC vaccine 4, 5 groups and BX strain vaccine group, respectively.At the 28th day after immunization, the IBDV neutralizing antibodies in IC vaccine 1, 2, 3, 4, 5 groups and BX strain vaccine group were 8.34log2, 9.60log2, 9.21log2, 7.88log2, 9.50log2 and 9.12log2, while 90%, 100%, 100%, 80%, 90% and 80% protection rates were provided, respectively.The results showed that IBD IC vaccines 2 and 3 (containing 8, 4 units IBDV neutralizing antibody, respectively) had the best immunity effect on one-day old low maternal antibody chickens, protection rates were both 100%.  相似文献

将鸡传染性法代囊病病毒(IBDV)BX株抗原和IBDV高免血清按一定比例混合,初步制备了无菌的鸡传染性法氏囊病免疫复合物疫苗1~5(分别含有32、8、4、0.5及0.125单位IBDV中和抗体),用鸡传染性法氏囊病免疫复合物疫苗1~5及BX株活疫苗分别免疫1日龄低母源抗体水平雏鸡,免疫后9 d观察法氏囊病变,免疫后28 d采血测定IBDV中和抗体,并用强毒攻击,计算各组疫苗保护率。结果显示,免疫后9 d,复合物疫苗1~3免疫组鸡法氏囊正常,复合物疫苗4、5免疫组和BX株活疫苗免疫组分别有2/10、4/10和5/10的试验鸡法氏囊出现了病变;免疫后28 d,复合物疫苗1~5免疫组和BX株活疫苗免疫组IBDV中和抗体效价分别为8.34log2、9.60log2、9.21log2、7.88log2、9.50log2和9.12log2,攻毒保护率分别为90%、100%、100%、80%、90%和80%。试验结果表明鸡传染性法氏囊病免疫复合物疫苗2、3(分别含8、4单位IBDV中和抗体)免疫1日龄低母源抗体水平的雏鸡效果最好,攻毒保护率能达到100%。  相似文献

对IBDV疫苗株:B87的鸡胚传代毒E2和E6的VP2片段进行测序和分析,发现两个代次毒株VP2序列存在7个氨基酸的差异,即S76G、L217S、Q253H、D279N、A284T、I294.L、S330R。将E2和E6接种CEF细胞,E2不能在CEF'上增殖,而接种E6的CEF细胞可以产生明显的CPE,说明两个毒株对CEF的嗜性不同。分别将E2和E6接种3周龄SPF鸡,对鸡的毒力进行比较,接种后第7天开始,E2出现囊体比下降,BB指数明显低于E6,相应的法氏囊切片经HE染色后,接种E2的法氏囊滤泡萎缩严重,结构松散,E6只有部分滤泡出现萎缩,滤泡间隔基本正常,说明E2,E6对SPF'鸡法氏囊毒力存在明显差异。根据两个毒株的VP2序列差异以及相关研究,推测Q253H/D279N/A284T三个位点突变可能是IBDV毒力和细胞嗜性差异的分子基础,但另外4个位点也可能对IBDV毒力差异产生影响。  相似文献

Studies with specific-pathogen-free chickens revealed that chicks hatching from eggs inoculated at the 18th day of embryonation with infectious bursal disease (IBD) vaccine viruses of low virulence (isolates TC-IBDV and BVM-IBDV) developed antibody against IBD virus (IBDV) and resisted challenge with virulent IBDV at 3 weeks of age or older. Embryo vaccination did not adversely affect hatchability of chicks or survival of hatched chicks. Chicks embryonally vaccinated with TC-IBDV had transient histologic lesions in the bursa of Fabricius at hatch. Similar but milder lesions were also noted in chickens that received TC-IBDV at hatch. The level of protection following embryo vaccination with TC-IBDV and BVM-IBDV was similar to that following vaccination with the same vaccines at hatch. Vaccine viruses of moderate virulence (isolates BV-IBDV and 2512-IBDV) were not suitable as vaccines in embryos lacking maternal antibody to IBDV, because the vaccinated chicks developed acute IBD after hatch. Isolate 2512-IBDV was not pathogenic for embryos bearing maternal antibody to IBDV. Maternal antibody against IBDV interfered with efficacy of embryo vaccination with BVM-IBDV but not with 2512-IBDV. Embryo vaccination with a mixture of vaccines against IBD and Marek's disease resulted in protection of hatched chicks against challenge with virulent IBDV and Marek's disease virus.  相似文献

The evolution of very virulent (vv) infectious bursal disease virus (IBDV) has led to significant economic losses in many poultry-producing areas. Despite vigorous vaccination strategies, IBDV has been difficult to control. The protective efficacy of IBDV vaccines is traditionally evaluated in specific pathogen-free (SPF) chickens. But under field conditions, residual maternal antibody (mAb) levels may interfere with vaccine efficacy. In this study, commercial broilers with various levels of maternally derived antibodies were vaccinated with IBDV vaccines of different virulence (vaccines 1-3, intermediate; vaccine 4, intermediate plus). At an average maternal virus-neutralizing antibody (mAb) level of log2 10.8 (range 7.6-11.6) at day of vaccination, only the intermediate plus vaccine induced IBDV antibodies after 18 days, while the other intermediate vaccines did not. At average mAb levels of log2 6.7 (range 5.6-8.6) at day of vaccination, all vaccines induced circulating antibodies, although the onset of antibody production differed significantly between strains (P < 0.05). While the intermediate plus vaccine induced enzyme-linked immunosorbent assay antibody levels already at 14 days postvaccination (PV), the intermediate vaccines induced significant antibody levels 28 (vaccines 1, 2) and 35 (vaccine 3) days PV. The time of IBDV antibody induction correlated with the onset of bursa lesions. The severity of lesions was comparable between vaccines 1, 3, and 4 (lesion score 4), while vaccine 2 induce only mild lesions of score 1 in 23% of the tested birds. Despite the induction of antibodies, none of the tested vaccines fully protected against challenge with vvIBDV. All challenged birds had either significantly higher bursal lesion scores or a higher IBDV antigen load in the bursa or sometimes both in comparison with nonchallenged birds (P < 0.05). Our study demonstrates that the evaluation of IBDV-vaccine efficacy is difficult in commercial broilers. For the first time, it was shown that the onset of bursa lesions and recovery of IBDV-vaccinated broilers is delayed in the presence of mAb in comparison with SPF chickens but not suppressed as previously assumed. At the time of challenge, vaccinated birds may still have significant bursa lesions and may lack target cells for IBDV-challenge virus. To be able to evaluate vaccine efficacy in commercial broilers, parameters such as intrabursal IBDV-antigen load should also be considered in conjunction with bursa lesion scores.  相似文献

以传染性法氏囊病病毒(IBDV)VP2蛋白基因表达质粒DNA为免疫原,以CpG的寡核苷酸(CpG-0DN)为免疫佐剂,肌肉注射于14日龄SPF鸡,1周后加强免疫1次,2次免疫后15d和21d分别测定血清ELISA抗体效价,并于免疫后21d用IBDV99儿强毒株攻毒和进行病理学观察。结果显示,(1)VP2基因重组质粒DNA与CpG共同免疫组的ELISA抗体水平明显高于VP2重组质粒免疫组;(2)IBD弱毒苗与VP2重组质粒免疫组抗体水平明显高于VP2重组质粒免疫组,且比VP2基因重组质粒DNA与CpG共同免疫组略高;(3)VP2基因重组质粒DNA与CpG共同免疫组及IBD弱毒苗与VP2重组质粒免疫组可明显降低IBDV强毒攻击后引起的急性发病率和死亡率。由此表明,CpG寡核苷酸对IBDV VP2蛋白基因真核表达质粒免疫具有明显增强作用,有很大的应用前景。  相似文献

【目的】 研究1株传染性法氏囊病病毒(Infectious bursal disease virus,IBDV)广西分离株的毒力特征及其与VP2基因序列特征的关系,为IBDV的流行病学研究和疫病防控提供参考。【方法】 通过PCR、鸡胚传代培养、DF-1细胞的适应培养及琼脂扩散试验等方法分离鉴定病毒,并扩增其VP2基因,利用Mega 7.0、MegAlign软件将其与其他不同毒力IBDV毒株进行核苷酸及氨基酸序列比对分析,并进行SPF鸡致病性试验,用实时荧光定量PCR方法测定IBDV拷贝数变化情况。【结果】 成功分离到1株IBDV毒株,命名为GX20210126株。该毒株接种SPF鸡胚可导致鸡胚出现出血、矮化和肝脏发黄、出血及针尖状坏死;且GX20210126对DF-1具有良好的细胞适应性,可引起显著细胞病变。琼脂扩散试验显示,该毒株具有良好的抗原性,病毒液能与IBDV阳性血清之间形成白色沉淀线。VP2基因进化分析显示GX20210126株与国际标准超强毒株在同一个大的分支上,氨基酸序列相似性在86.8%~99.6%之间,其中与UK661株相似性最高,为99.6%,仅有2处氨基酸位点发生改变,即D279N和I272T。以EID50为10-3.8/0.1 mL,0.5 mL/只的剂量点眼、滴鼻接种8日龄SPF鸡,攻毒后鸡出现羽毛蓬松、精神萎靡、拉白绿色水样粪便等临床症状,剖检可见肌肉、肝脏出血,肾脏尿酸盐沉积,法氏囊萎缩,IBDV拷贝数在感染后第5天达到峰值。【结论】 IBDV广西流行株GX20210126具有超强毒株基因序列特征,人工感染鸡群可导致IBDV典型临床症状和病理变化。  相似文献

为了解IBDV流行强毒HQ-b株囊毒与其细胞适应毒间生物学特性差异及2毒株毒力变化与VP2、VP5基因变异的关系,对2毒株的细胞适应性、致病性等进行比较,同时对其VP2、VP5基因序列进行分析。结果表明,HQ-b株囊毒对CEF、CEK、CELi、DF-1和Vero均不适应,而细胞适应毒HQ株仅不能适应Vero细胞、且批内及批间毒价稳定。致病性结果显示HQ-b株对4周龄SPF鸡致死率高达80%,是真正的超强毒,而细胞适应毒致死率已降为0%。对VP2基因高变区研究表明,HQ-b株具备IBDV超强毒株的分子特征,即222A、256I、294I和299S;其细胞适应毒HQ株除222A→P、256I→V、294I→L和299S→N外,在VP2公认的毒力位点253(Q→H)、279(D→N)、284(A→T)位氨基酸也发生改变,导致细胞适应毒具备经典弱毒株的分子特征,即222P、256V、279N、284T、294L和299N。对VP5基因研究表明:流行强毒HQ-b株也具有IBDV超强毒株的分子特征;其细胞适应毒VP5基因有12个位点碱基突变并导致9处氨基酸变异,尤其是ORF区第2个碱基由"T"突变为"C"后,导致细胞适应毒VP5的N端丢失了4个氨基酸,这种突变与现有的疫苗毒完全一致。本研究提供了超强毒HQ-b培育、驯化后致病性和细胞适应性转变的分子机理,也丰富了IBDV分子流行病学的理论。  相似文献

One-day-old moderate maternal antibody Jinghong chickens and SPF chickens were vaccinated with infectious bursal disease (IBD) immune complex (IC) vaccine and BX strain live vaccine,respectively,pathological changes of bursa of fabricius in chickens were observed at the 9th day after immunization.On the 28th day after immunization,blood samples were taken and the IBDV neutralizing antibody were tested,meanwhile experimental chickens were challenged with high virulent IBDV,the protective rates of experimental groups were calculated.The results showed that at the 9th day after immunization,the bursa of fabricius in SPF and Jinghong chickens were normal in IC vaccine group,however atrophic changes of bursa of fabricius in BX strain live vaccine group were shown.At the 28th day after immunization,the IBDV neutralizing antibody of IC vaccine group and BX strain live vaccine group in SPF and Jinghong chickens were 9.2log2,8.5log2,7.2log2 and 4.4log2,respectively,while 100% protection rates were provided.The results provided that IC vaccine had better immunity effect on one-day-old moderate maternal antibody chicken,high levels of neutralizing antibodies against IBDV were produced and protection rates were 100%.  相似文献
