The concentration of condensed tannins (CT) in sulla (Hedysarum coronarium L.) is moderate and overall regarded as beneficial. However, the intake of this forage can reduce diet digestibility, particularly during flowering phase. An experiment was run to assess the effect of CT on feeding behaviour, intake, diet digestibility and performance of dairy sheep rotationally grazing sulla at flowering phase. Twenty-four late-lactating sheep were blocked in two homogeneous groups and submitted to the following treatments: i) twice daily drenching with 200 g/day of a 50% w/v water solution of an anti-tannic substance, polyethylene glycol, group PEG; ii) twice daily drenching with 200 g/day of water, group CON (Control group). All the sheep rotationally grazed as a flock two sulla plots from April to June (8 weeks in total). Sward height, herbage mass, botanical and chemical composition of the herbage were measured at the beginning and the end of each grazing period. The feeding behaviour (3 sheep per group) was continuously monitored for 24 h in 6 weeks using the IGER behaviour recorders. Herbage DM intake (DMI), dietary DM digestibility (DMD) and apparent CP digestibility (CPD) were estimated on 8 sheep per group by the n-alkane method. On average, PEG group had longer total grazing (503 ± 12 vs 460 ± 12 min, P < 0.05) and eating time (425 ± 13 vs 391 ± 13 min, P < 0.07) than CON group. Moreover PEG group showed shorter inter-meal intervals (41 ± 3 vs 52 ± 3, min, P < 0.05) and higher number of daily meals than CT-exposed group (24 ± 1 vs 19 ± 1 min, P < 0.01). The herbage DMI was not affected by the treatment whereas DMD (74.60 ± 3.48 vs 58.30 ± 3.01%), and CPD (60.14 ± 4.83 vs 38.21 ± 4.83%) were both increased by PEG administration (P < 0.05) confirming the negative effect of sulla CT on these variables. Milk yield tended to be higher in PEG than CON (1331 ± 45 vs 1205 ± 59 ml, P < 0.11). Milk protein content was similar between groups while milk fat content was higher in CON than PEG ewes (6.61 ± 0.15 vs 6.11 ± 0.15%, P < 0.05), being the reverse true for milk urea (46.04 ± 1.27 vs 53.04 ± 0.76%, P < 0.001). To conclude, this experiment shows that when sulla is grazed at flowering as monoculture, dietary CT can exert negative effects on DM and CP apparent digestibility, in this study partially counterbalanced by a better metabolic utilisation of the nutrients up-taken.  相似文献

The effects of host age and immune suppression on abomasal parasitic infection in sheep were investigated following single experimental oral infections with MHco3 (ISE), MHco4 (WRS) and MHco10 (CAVR) strains of Haemonchus contortus in naïve 5-month-old crossbred lambs (n = 1 per group) and 15-month-old Greyface sheep treated with methyl prednisolone acetate (n = 2 per group) or without corticosteroid treatment (n = 2 per group). Adult female H. contortus in 5-month-old lambs (n = 1 per group) shed on average 6.5, 3.1 and 8.0 times more eggs than in 15-month-old sheep (n = 4 per group) following infection with MHco3 (ISE), MHco4 (WRS) and MHco10 (CAVR) strains of H. contortus, respectively, over a period of 28 days following the commencement of patency. There was no obvious effect of age of sheep or corticosteroid treatment on the abomasal establishment of H. contortus or on in vitro assays for egg hatching or larval feeding at different concentrations of anthelmintics, although statistical analysis could not be performed due to the small group sizes.  相似文献

The aim of the study was to evaluate the protection generated in mice against Toxoplasma gondii brain cyst burden by vaccination with T. gondii cytoskeleton proteins using Lactobacillus casei as adjuvant. One hundred and sixty-eight NIH mice were randomly allocated into eight groups of 21 mice each. Animals were immunized as follows: in group 1 with Toxoplasma lysate antigen (TLA) in Freund's modified adjuvant, containing L. casei (FMA), in group 2 with Toxoplasma cytoskeleton proteins (TCPs) in FMA, in group 3 with FMA, in group 4 with phosphate buffered saline (PBS), in group 5 with L. casei dead by heath (Lc), in group 6 with Freund's complete adjuvant (FCA), in group 7 with TLA in FCA, and in group 8 with TCP in FCA. Mean brain cyst burden (±S.E.M.) was assessed in mice 8 weeks after challenge with T. gondii Me49 strain (20 cysts per mouse). The percentages of reduction in cyst burden per brain (P < 0.01) as compared with the group 4 (control: mean 3181 ± 97.5) were 77.25% (724 ± 98) in group 1, 88.02% (381 ± 97.5) in group 2, 38.92% (1943 ± 130.3) in group 3, 44.31% (1771.4 ± 102) in group 5, 59.28% (1295.2 ± 99.1) in group 7 and 55.69% (1409.5 ± 89.9) in group 8. In order of importance, the best protection was obtained in groups 2, 1, 7, 8, 5 and 3. Noticeably the mice inoculated with L. casei alone showed a significant reduction in T. gondii brain cysts (P < 0.01), while those animals treated with FCA alone did not. Additionally, IgM anti-T. gondii antibody levels, as determined by ELISA 2 weeks after challenge, were highest in group 2 (P < 0.01) than in the other seven groups. Results suggest that T. gondii cytoskeleton proteins with L. casei as adjuvant constitute a good anti-toxoplasmosis vaccine candidate.  相似文献

The aims were (a) to quantify the number of Duddingtonia flagrans chlamydospores per gram of faeces (CPG) recovered from sheep administered with different oral doses and, (b) to describe the relationship between CPG and eggs per gram of faeces (EPG) on the efficacy to reduce Haemonchus contortus infective larvae. Three doses of chlamydospores per kg BW were orally administered during seven days: (T1) non treated control group, (T2) 1 × 106, (T3) 2.5 × 106 and (T4) 5 × 106. Three lambs, infected with H. contortus, were used per group. Faeces were obtained from the rectum of each lamb during the fungal administration period (days 0–6) and for six days after that period. Four coproculture replicates were made from each animal in days 2, 4, 6, 8 and 10. A higher chlamydospore dose produced higher CPG in faeces (p < 0.05), but a clear dose dependent effect was not found either in the larvae reduction or in the CPG:EPG ratio. When ratios were re-analyzed, independently of the treatment groups of origin, a better efficacy was obtained with a ratio from 5 to 10 CPG:EPG and a higher ratio (>10 per egg) showed a lower reduction efficacy (p < 0.05). The binomial analysis showed that for each unit of increment in CPG:EPG ratio there was a reduction of larvae number until a point (between 5 and 10 CPG:EPG) where no further reduction was detected. The surface response test indicated that the number of larvae was reduced by CPG until possible saturation. The highest CPG:EPG ratios did not necessarily improve efficacy of D. flagrans.  相似文献

The benefit of long-term feeding of fresh or ensiled cassava foliage on gastrointestinal parasite in goats was evaluated. Eighteen male goats (15.15 ± 2.83kg and between 4–6months) were randomly allocated into three treatments supplemented with 200g of wheat bran head−1 day−1. All groups were fed ad-libitum on either grass (CO), fresh cassava (CaF) or ensiled cassava foliage (CaS). At the beginning of the trial, each goat was inoculated with 3000 L3 containing ≈50% Haemonchus contortus. Individual LWt, FEC and PCV were measured at weekly intervals for 10weeks. At the termination of the experiment all goats were slaughtered for worm recovery and enumeration. The goats in CaF and CaS had similar weight gains while those in CO lost weight (p < 0.05) through the trial. FEC in CaF and CaS were lower (p < 0.05) than CO during the patency of parasite infections, but there was no difference between CaF and CaS goats. PCV of all groups decreased from above 30% to around 25% at the end of the trial. The compositions of established worm burdens were mainly H. contortus (19–40%) and Trichostrongylus colubriformis (55–76%). TWB did not differ among the groups, however, CaS significantly reduced H. contortus burdens, as compared to CaF and CO (p ≤ 0.005). Thus, ensiled cassava foliage reduced the H. contortus population while the fresh foliage only reduced worm fecundity.  相似文献

Clinical, parasitological and biochemical parameters were evaluated in Corriedale and Crioula Lanada sheep after a single experimental infection with Haemonchus contortus. Ten 4-month-old worm-free lambs, of each breed, were infected with 200 L3 H. contortus per kg live weight and four uninfected animals of each breed were used as controls. Every week, the animals were weighed and blood and faecal samples were collected for measurement of packed cell volume (PCV), total serum protein (TSP) and albumin (ALB), and the number of eggs per gram of faeces (EPG), respectively. Twelve weeks after infection, the animals were slaughtered. The worm burden was determined and samples of the abomasal mucosa were processed for determination of the number of eosinophils, mast cells and globule leukocytes. No significant differences in PCV, TSP, ALB, parasite burden or the cell populations of the abomasal mucosa were observed between breeds, but Crioula lambs had a lower EPG count. The comparison of the infected groups with their respective controls revealed significant alterations in PCV, TSP and ALB in the Corriedale lambs and in PCV, TSP, ALB and the density of eosinophils and mast cells in the Crioula lambs.  相似文献

Mycoplasma synoviae and Mycoplasma gallisepticum are major poultry pathogens, but their strains differ significantly in invasiveness and pathogenicity. Recent studies have demonstrated that M. gallisepticum invades chicken erythrocytes (CER) and chicken embryonic fibroblasts. The aim of this study was to determine whether M. synoviae also invades chicken cells. Using the gentamicin invasion assay, relative invasion frequency (RIF) of four M. synoviae strains was determined for CER, chicken embryonic cell line (CEC-32) and/or primary chicken chondrocytes (CCH). All tested strains of M. synoviae were capable of invading chicken cells within 24 h after infection. The type strain WVU 1853 showed significantly higher invasiveness in CER (RIF 7.5 ± 1.5%) and CEC-32 (RIF 7.0 ± 0.3%) than field strain ULB 02/T6 and M. gallisepticum strain Rlow. Surprisingly, WVU 1853, which is capable of causing synovitis and arthritis in chickens, was less invasive for CCH with a RIF (1.2 ± 0.3%) similar to that of Rlow (1.1 ± 0.1%). This is the first study documenting the invasiveness of M. synoviae strains for non-phagocytic chicken cells.  相似文献

A comparative slaughter trial was conducted to assess the net requirements for gain of Ca, P, Na, K and Mg of bulls, steers and heifers of Nellore and Red Angus crossbreds. Twenty seven F1 Nellore and Red Angus crossbred calves (9 steers, 9 bulls, and 9 heifers), averaging 274 kg BW, were used. At the beginning of the trial, three animals from each gender were slaughtered to determine the initial body composition. The remaining 18 animals (3 animals of each gender) were randomly assigned to two treatments: fed 0.75 or 1.5% of BW of concentrate. The diets were based on corn silage and were isonitrogenous (2% N, DM). After three growing periods of 28 d, all animals were slaughtered. The cleaned gastrointestinal tract, organs, carcass, head, hide, tail, feet, and tissues were weighed to determine the empty BW (EBW). These parts were ground separately and subsampled for chemical analyses. The log of the contents of each mineral in the empty body was regressed on the log of the EBW to estimate the net requirement for each mineral per kg of empty body gain (EBG). There were no differences (P > 0.05) in the net requirements for growth of all macrominerals among genders. The equations of the pooled data of the net requirements for growth (g/kg EBG) were: 332.6 × EBW − 0.6367 for Ca, 112.1 × EBW − 0.5615 for P, 10.85 × EBW − 0.3992 for Na, 4.01 × EBW − 0.153 for K, and 3.589 × EBW − 0.462 for Mg. Our findings indicated that retained Ca and retained P were poorly related to the retained protein.  相似文献

The effect of transportation during the hot season (42 °C) and low voltage electrical stimulation on physiological, histochemical and meat quality characteristics of Omani sheep was studied. Forty intact male sheep were divided into two equal groups: 3 h transported or non-transported. The non-transported group remained in holding pens for 48 h prior to slaughter, while the transported group was transported 300 km (approximately 3 h) in an open truck under solar radiation on the day of slaughter. Blood samples were collected from the animals before loading and prior to slaughter. Fifty percent of the carcasses from each group were randomly assigned to low voltage (90 V) at 20 min postmortem. Temperature and pH decline of the left longissimus thoracis muscle were monitored. Ultimate pH, WB-shear force, sarcomere length, myofibrillar fragmentation index (MFI), expressed juice, cooking loss and color L, a, b were measured on samples from both sides muscles collected at 24 h postmortem at 3–4 °C. The transported sheep had significantly higher plasma cortisol (P < 0.01), adrenaline, nor-adrenaline and dopamine concentrations (P < 0.05) than non-stimulated animals. Electrical stimulation resulted in a significantly (P < 0.05) more rapid pH fall in muscle during the first 12 h after slaughter. Muscles from electrically-stimulated carcasses had significantly (P < 0.05) lower pH values, longer sarcomere length, lower shear force value, higher expressed juice, MFI and lighter L than those from non-stimulated ones. The muscle samples from the transported sheep had significantly (P < 0.05) smaller and lower proportion of Types I and IIA fibers than those from the non-transported group. This study indicated that pre-slaughter transport at high ambient temperatures can cause noticeable changes in physiological and muscle metabolism responses in sheep. Electrical stimulation improved meat quality characteristics, which indicate that meat quality of transported sheep can be improved by electrical stimulation.  相似文献

The climate in the UK is changing, with a trend towards increased rainfall in the autumn and winter and warmer average temperatures throughout the year. There has also been a 4-week extension of the herbage growing season over the past 40 years. These changes may have implications for the epidemiology of sheep helminth parasites. Here, we describe production-limiting disease outbreaks caused by Haemonchus contortus, Nematodirus battus, Teladorsagia circumcincta and Fasciola hepatica in sheep flocks in south eastern Scotland. The occurrence and timing of these disease outbreaks could not have been predicted in this region highlighting changes in the epidemiology of helminth infections from the patterns historically described. These cases are used to introduce discussion regarding the potential effects of climate change on the epidemiology of helminth parasites and the implications for sheep farming in the UK.  相似文献

An experiment was conducted with eight ruminally-cannulated cows using a crossover design with 2 periods to determine the effects of yeast supplementation on rumen fermentation. Holstein dairy cows in late lactation were either supplemented with 0.5 g/hd/d of Saccharomyces cerevisiae, an active dry yeast (CNCM-1077, Levucell SC20 (r) SC, Lallemand Animal Nutrition) or not supplemented (control). A basal diet consisting of 60% forage and 40% concentrate (DM basis) was fed once daily to both groups of cows throughout the entire experiment. Ruminal pH was measured continuously every 22 min using a pH probe that was placed in the ventral rumen sac for 6 consecutive days. Volatile fatty acid and ammonia N concentrations in the rumen were measured on days 5 or 6 of the 12-d period for each cow and DM intake was monitored throughout the experiment. Data were analyzed using a mixed-effects model with repeated measures. There were no differences in dry matter intake between treatments. Mean ruminal pH was greater (P < 0.05) when yeast was supplemented (6.53 ± 0.07) compared with the control (6.32 ± 0.07). Average maximum and minimum ruminal pH were also greater (P < 0.05) when yeast was supplemented (7.01 ± 0.09 and 5.97 ± 0.08, respectively) compared with the control (6.80 ± 0.09 and 5.69 ± 0.09, respectively). Time spent under the subacute acidosis threshold, pH less than 5.6, was lower (P < 0.05) with yeast supplementation compared with control cows. No difference was observed for ruminal ammonia N concentrations (mean = 14.0 ± 1.2 mg/dL) between treatments. Total VFA concentration (mM) in the rumen tended to be lower (P = 0.10) in the yeast-supplemented cows (107.3 ± 6.35) than in the control cows (122.4 ± 6.35), which could be related to the greater pH observed with yeast supplementation. Supplementing dairy cows with active dry yeast in the current experiment increased the mean, minimum and maximum ruminal pH; decreased time spent in subacute rumen acidosis, and tended to decrease total VFA concentration in the rumen compared with control cows.  相似文献

Between autumn and spring 2006, a coprological survey was performed in two wildlife reserves located in the north of Argentine Patagonia to determine the prevalence of Fasciola hepatica and the number of parasite eggs per gram (epg) of feces in wild guanacos (Lama guanicoe), coypus (Myocastor coypus), and locally born and raised goats and sheep. Snails of the Family Lymnaeidae were collected in freshwater habitats, identified taxonomically and analyzed parasitologically.Prevalence of patent infection was 100% in sheep (n = 69) and coypus (n = 9), 84% in goats (n = 20) and 0.5% in guanacos (n = 224). No significant differences in epg were found among animals, but the median epg of coypus (160) and sheep (160) was higher than that of goats (80). For guanacos and goats, a negative binomial model estimating the population egg-count frequency could be fitted, while for coypus and sheep parasite egg-count frequencies trended toward a normal distribution, indicative of a more even, and much less aggregated distribution across sampled hosts. All snails (n = 175) were Lymnaea truncatula and none of them was found infected. This is the first report of fascioliasis in free-ranging guanacos in Argentina. Coypu appears to be a major wildlife reservoir of F. hepatica, which was presumably introduced locally by livestock.  相似文献

The chemical composition, the fatty acids profile, and conjugated linoleic acids content in Longissimus muscle (LM) of steers have been determined. For such, 18 steers (6, Nellore, NEL) and their Simmental (6, NSI), and Santa Gertrudes (6, NSG) crossbreds finished in pasture system with Brachiaria brizantha cv. marandu for about 3 months with approximate weight at slaughter of 480 kg at average age approximate of 25 months. The lipid content increased in the following order influenced by genetic groups: Nellore, F1 Nellore × Simmental and F1 Nellore × Santa Gertrudes crossbreds. The lipid content increased while moisture, ash and protein contents decreased. The content of saturated fatty acids (SFA) was affected by genetic groups. The conjugated linoleic acids contents (CLA) in fat were similar in the genetic groups, but the quantity of CLA concentrations in muscle lipids of steers with larger total lipid was higher. The predominant CLA was CLA cis-9, trans-11.  相似文献

The circadian body (vaginal) temperature rhythm was measured in 36 dairy cows: Holstein–Friesian (HF, n = 12), Jersey (J, n = 12) and their crossbreed, Holstein–Friesian × Jersey (HF × J, n = 12) at six different periods (5-7 days in duration) during the year that could potentially be a seasonal and physiological stressor. These were a) early winter (late pregnancy), (b) early spring (early lactation), (c) late spring (before mating at mid-lactation), (d) early summer (after mating at mid-lactation), (e) late summer (late lactation, early pregnancy) and, (f) early autumn (drying-off at the end of lactation, mid-pregnancy). Live weight and body condition score was recorded on the first day of each measurement period. Live weight differed (P < 0.001) with breed with HF cows heaviest and J cows lightest (HF: 525 ± 51, HF × J: 456 ± 56, J: 401 ± 30 kg). HF cows tended (P = 0.099) to have a higher body condition than either J or HF × J cows although the overall change in body condition during the experiment was similar (P = 0.961) amongst breeds. The circadian body temperature rhythm differed markedly between each measurement period, being flatter and having a less prominent morning nadir and afternoon peak during early winter compared to spring and summer. The timing of peak body temperature was 4-5 h earlier in early autumn in HF and HF × J cows relative to J cows. There was no breed difference (P ≥ 0.162) in daily mean body temperature except for a tendency (P = 0.075) for mean body temperature to be moderately higher in J cows (38.5 °C) during late summer compared to HF and HF × J cows (38.4 °C), primarily due to an increase (P = 0.001) in daily minimum body temperature (38.0 vs. 37.8 °C). The daily range of oscillation (maximum–minimum) in the body temperature rhythm was highest during late summer, particularly in HF and HF × J cows. Minimum body temperature was also higher (P = 0.033) for J cows than HF and HF × J cows during early autumn (37.9 vs. 37.8 °C). In conclusion, season and physiological status of cattle had a profound effect on the circadian body temperature rhythm but there was little difference between the breeds of dairy cows studied.  相似文献

The present study was conducted to investigate the effects of Acanthopanax senticosus extract (ASE) as a dietary additive on gut health in weanling piglets by examining diarrhea frequency, intestinal microbiota and morphology. A total of 96 Duroc× (Landrace × Yorkshire) piglets weaned at 21 days of age with an average initial body weight (BW) of 5.6 ± 0.4 kg were randomly assigned to 3 treatment groups with 4 duplicates of 8 piglets each. The piglets were fed basal diet to which had been added 0 or 1 g/kg of ASE, or 0.7 g/kg antibiotics, respectively. Fecal consistence was monitored twice daily and the frequency of diarrhea was calculated. On day 21 after the initiation of supplementation, 8 piglets were randomly selected from each treatment group (2 piglets per pen) and slaughtered. The jejunum, ileum, colon and cecum were then excised and fixed in 10% neutral formalin solution to determine villus height and crypt depth, after their contents were collected to determine microbiota. The results showed that dietary supplementation with ASE increased (P < 0.05) the density of bacterial populations that co-migrated with Lactobacillus amylovorus, Lactobacillus salivarius, Bacillus subtilis, and Clostridium lituseburens, but decreased (P < 0.05) those co-migrating with Staphylococcus aureus, Salmonella typhimurium, Ruminococcus forques, and E. coli O157:H7 in the PCR-DGGE profiling analysis when compared with the control group. The villus height of the duodenum, jejunum and ileum increased (P < 0.05) by 14.8, 13.7 and 10.0%, while the crypt depth decreased (P < 0.05) by 17.9, 9.1 and 12.1%, respectively, in response to dietary ASE supplementation. Additionally dietary supplementation with ASE or an antibiotic decreased (P < 0.05) the frequency of diarrhea by 55.6 and 52.2%, respectively, compared with the control group. In conclusion, these findings suggest that dietary supplementation with ASE could regulate the microbiota composition and maintain a normal morphology of gut mucosa in weanling piglets, thereby decreasing diarrhea that resulted from weaning stress.  相似文献

The present study describes the development of a specific Mycoplasma mycoides subsp. mycoides Small Colony (MmmSC) monoclonal antibody (MAb), 6E3, and its application in a sandwich ELISA (sELISA) format. Mab 6E3 reacted only to the 12 MmmSC within the 32 M. mycoides cluster strains and 12 representative strains of other bovine, ovine and caprine associated mycoplasmas examined. A capture/enrichment format of the sELISA that combined MAb 6E3 with a previously developed MAb 3H12 that cross reacted with Mmm Large Colony [Rodriguez, F., Ball, H.J., Finlay, D., Campbell, D., Mackie, D.P., 1996. Detection of Mycoplasma mycoides sub-species mycoides by monoclonal antibody-based sandwich ELISA. Veterinary Microbiology 51, 69–76], retained MmmSC specificity and improved the sensitivity from the 1.2 × 107 cfu/ml for a standard 2 h capture stage sELISA down to as low as 2 cfu/ml for a 72 h capture. A low level of false positives (1%) was observed when this assay was applied to 200 bovine respiratory and milk samples submitted for diagnostic investigation. This simple and specific sELISA provides a suitable assay for screening large numbers of samples for CBPP.  相似文献

This laboratory study investigates the sub-adult developmental cycle of field collected Culicoides (Avaritia) imicola Kieffer (Diptera; Ceratopogonidae). The period required from blood-feeding field-collected females to the production of progeny adults occupied 34–56 days at 20 °C, 15–21 days at 25 °C and 11–16 days at 28 °C, demonstrating clear temperature dependence. When reared at 28 °C, C. imicola demonstrated higher variability in fecundity (between 2.4 and 20.6 eggs/female) and lower hatching rates (50.0–62.2%), although larval survival rates to pupation were low at all temperatures (20–30%). Similarly, the mean emergence rate from pupae was the highest at lower temperatures. These results highlight the difficulty in establishing and maintaining a laboratory colony of this species from field-collected material and results are discussed in reference to future research directions that may aid this process.  相似文献

Detection methods for Mycobacterium avium subsp. paratuberculosis (MAP) are imperfect, yet crucial for diagnosis of Johne's disease. Our purpose was to test for significant and biologically relevant changes in Johne's ELISA results associated with how field-collected blood samples were transported to the laboratory, prepared and stored prior to testing, while removing potential confounding by test kit and laboratory variables. Blood samples were collected from 21 cows that previously had MAP ELISA scores ranging from negative to highly positive. Samples for immediate laboratory processing were subjected to different transportation temperatures (on ice, 26 °C) and preparation methods (serum separated, hemolyzed and serum separated, clotted whole blood), but were tested using the same ELISA kit in the same laboratory. Samples for laboratory processing after one week of storage were subjected to different storage temperatures (4 °C, −20 °C) and preparation methods (serum separated, hemolyzed and serum separated, clotted whole blood), and again were tested using the same ELISA kit in the same laboratory. Finally, samples were evaluated by time to processing (one day, one week) and storage temperature (4 °C, −20 °C). Data were checked for normality and analyzed with repeated measures ANOVAs. Significantly (P = 0.027) higher MAP ELISA scores were recorded for whole blood and hemolyzed samples transported at 26 °C than serum separated samples. Sample storage for one week at −20 °C resulted in significantly (P < 0.001) lower MAP ELISA scores, regardless of handling method, compared to samples stored at 4 °C for one week. Method of sample preparation, as well as transportation temperature and medium-term storage temperature, affects MAP ELISA results. Such discrepancies will inevitably result in improper classification of MAP-infected cattle, impeding both biosecurity measures on uninfected farms and MAP control programs.  相似文献

This research assessed the potential use of a low input forage containing a high amount of condensed tannins (CT) to reduce foodborne pathogens prior to slaughter of meat goats. In a completely randomized design, twenty Kiko × Spanish intact male kids (BW = 19.2 ± 0.74 kg) were fed ground sericea lespedeza [SL; Lespedeza cuneata (Dum-Cours) G. Don; 2 pens], a high-CT legume, or bermudagrass hay [BG; Cynodon dactyon (L.) Pers.; 2 pens], at 75% of daily intake with a corn-based supplement (25% of intake) for 14 weeks (n = 10 goats/treatment). At the end of the feeding trial, the animals were slaughtered using standard procedures. Immediately after evisceration, rumen and rectal samples were collected to assess bacterial loads and volatile fatty acids in the rumen. Concentrations of rumen volatile fatty acids were significantly different between dietary treatments. Goats fed SL hay had higher (P < 0.05) contents of butyric (8.66 vs 7.16 mM), isobutyric (1.94 vs 1.44 mM), isovaleric (3.03 vs 2.13 mM), and valeric (1.43 vs 1.07 mM) acids than those fed BG hay; however, the content of acetic acid (78.6 vs 64.4 mM) was higher (P < 0.05) in the BG-fed groups than in SL-fed groups. Escherichia coli (2.33 vs 1.13 log10 CFU/g) counts of rumen contents were higher (P < 0.05) in the SL-fed group compared with the BG-fed group. However, E. coli counts in feces were not different (P > 0.05) between dietary treatments. The high-CT influenced (P < 0.05) total plate counts in the feces; and the total plate counts in feces of SL- and BG-fed goats were 4.95 and 6.57 log10 CFU/g, respectively. The results indicated that high CT in the diet might influence rumen volatile fatty acid composition, but might not reduce the bacterial loads in gastrointestinal tracts of meat goats.  相似文献
