Relationship between ultrastructure and specific functions of macrophages   总被引:1,自引:0,他引:1  
The main function of the macrophages, which is to ingest and degrade any foreign molecules or particles penetrating the organism, appears in the development of the different structures implicated in endocytic activity. The macrophage's high endocytic property first appears in its irregular shape and the large number of extensions of the cell membrane, allowing the rapid capture of extra-cellular material. Adhesion between macrophage cell surface and molecules or particles is greatly enhanced by the presence of varied kinds of receptors: lectin-like receptors which bind specific sugars or highly specific receptors such as Fc and C3b receptors, which increase phagocytosis of opsonized microbes. The microbicidal properties reside in part in the production of superoxide anions which result from the activity of a NAD(P)H oxidase. This enzyme is located in the plasma membrane. Its activity could be demonstrated with a cytochemical method, on the cell surface and along the phagosome membrane. It is, however, very weak in resident macrophages and increases after stimulation or activation. The second kind of bactericidal property corresponds to cationic proteins located in lysosomes. After fusion between lysosomes and phagosomes, they contribute to microbe killing by permeabilizing microbe envelopes. Lysosomes, which contain diverse acid hydrolases and are responsible for the degradation of ingested material, play a crucial role in macrophage endocytic activity. Their number increases in parallel with endocytic activity during macrophage differentiation and is particularly high after ingestion of degradable material. Contrary to polymorphonuclear leukocytes, macrophage is very poor in granules containing peroxidase. The latter, which are rather abundant in monocytes, disappear during macrophage maturation. They do not seem thus to be implicated in macrophage microbicidal activity. Endocytosis is accompanied by rapid and intense exchanges between the different membrane compartments of the cell (plasma membrane, pinosomes or phagosomes, endosomes, lysosomes, Golgi apparatus, etc.). These exchanges seem to occur by transitory fusions between vesicles coming from different compartments, rapidly followed by their recycling to their original compartment. This system of membrane shuttle has been clearly observed after formation of phagosomes or pinosomes in which the internalized plasma membrane is recycled back to the cell surface within a few minutes after their formation. The capability of activated macrophages to kill tumor cells in vitro is now well documented. The tumoricidal activation of macrophages against intestinal tumor cells by different agents is described and the main hypothesis on the mechanisms of tumor cell killing in vitro are discussed. These in vitro results suggest that the macrophage can constitute an efficient effector cell in the defense against intestinal tumors. The distribution and ratio of macrophages in normal intestine and intestinal tumors is described. At the moment, potent activators of macrophages studied in vivo on experimental and human intestinal tumors give poor results or even enhance the growth of tumors. Macrophages may also interfere with the specific immune response in two directions by enhancing the immune response or decreasing it by elaboration of mediators such as prostaglandins.  相似文献

Respiratory burst is due to the activation of a membrane bound NADPH oxidase induced by perturbation of the plasma membrane during phagocytosis or following interaction between the cell surface and a number of environmental stimuli. It refers to the increase in the non-mitochondrial O2 consumption with a concomitant production of different reactive species (superoxide anion, hydrogen peroxide, hydroxyl radical, singlet oxygen ...). The effects of the respiratory burst depend on the intensity and combination of the different actions which are defensive, toxic, activatory and modulatory of the inflammatory process.  相似文献

Following a presentation of different methods used to collect alveolar macrophages by lung washing performed on killed or anaesthetized animals, several main features of these cells are described: in vitro adherence, enzymatic properties and morphology, phagocytosis. Studies of postnatal development show that swine alveolar macrophages appear during the first week of age. Finally, the alveolar macrophage immunological behaviour (surface receptors, cytotoxicity, co-operation with lymphocytes, activation) and the complex micro-organisms-macrophages interrelationships are discussed.  相似文献

This paper describes the handled walking and trotting kinematics (linear, temporal, and angular traits) of 35 Menorca Purebred (MEN) stallions, and the relationships among these variables is presented for the first time, along with a discussion of the influence of the hind limb pastern angle on kinematic variables at both gaits. For data collection, all animals, aged between 3 and 10 years old and belonging to 28 different studs, were recorded under the same experimental and environmental conditions, using a three-dimensional (3D) semiautomatic movement analysis system. A total of 24 kinematic variables (temporal, linear, and angular) at the walk and the trot and a morphometric variable measured at the mid stance position of walking (hind pastern angle) were included in this analysis. Angle-time diagrams of the hind pastern angle while walking and trotting normalized to stride duration were also obtained. Generally the MEN stallions' forelimb movements closely resembled the movement characteristics of other European dressage performance breeds, while the hind limb locomotion showed a greater likeness to Iberian dressage Purebreds. Despite this, their ability in collection and propulsion at the walk and the trot was relatively low. The hind limb pastern conformation was partially connected to the hind limb movements for both gaits, with an apparently negative effect of excessively upright pasterns on the amplitude at the trot, which indirectly reduced collection ability.  相似文献

Changes in shape of the equine thorax during locomotion are not well defined, although it has been shown recently that the transverse hemi-diameter changes its dimension by up to 80 mm on the side of the trailing forelimb during gallop, despite minimal change in thoracic circumference. The aim of this project was to analyse transverse and dorso-ventral changes in shape of the thorax simultaneously, and to determine if leading limb, treadmill slope and speed have an effect on these shape changes. Reflective markers were placed on the horse’s hemi-thorax and movement of the markers was recorded using a motion capture system while the horse trotted and cantered on a treadmill. Treadmill speed and slope, and the lead the horse cantered on were varied to determine the effects these had on transverse hemi-diameter and dorso-ventral diameter.There was a negative correlation between transverse and dorso-ventral changes in thoracic dimension, the strength of which increased with speed on the trailing limb side. On the leading side, the relationship was either weakly negative or positive. The changes in dimension of the hemi-thorax were significantly greater on the trailing side compared to the leading side. Speed had small effects on thoracic shape changes, but inclined exercise caused an increase in transverse hemi-diameter on the trailing side of the thorax. The changes in thoracic shape are unlikely to contribute substantially to ventilation and rib motion is likely to be due to protraction and retraction of the forelimbs. However, it may reflect asynchronous ventilation of lung lobes and partially explain the prevalence of exercise-induced pulmonary haemorrhage.  相似文献

A cell line of bovine endothelial cells (E5), infected with 3 different stocks of Cowdria ruminantium, was used as antigen in an indirect fluorescent antibody test for the serodiagnosis of heartwater. These antigens were compared to peritoneal macrophages from mice infected with the Kümm stock and to caprine neutrophils in primary cultures from goats infected with 4 different stocks of Cowdria. The use of endothelial cell cultures proved to be superior in all respects. The antigens can be produced in large quantities at a low cost, contrary to the other types. The reaction is easily and quickly read, compared to the laborious reading of neutrophil or macrophage antigens which often contain few and small colonies of Cowdria. Moreover, not all stocks are suitable for the preparation of neutrophil antigens, while macrophage antigen can only be obtained with the Kümm stock. Endothelial cell antigens also distinguish serotypes in C. ruminantium, but these differences seem to be less pronounced than those found with neutrophil antigens. Finally, the specificity of endothelial cell antigens appears to be better than that of Kümm antigen and comparable to that of neutrophil antigens. The use of Kümm antigen may have been responsible to a large extent for past unexplained positive serological results on certain Caribbean islands where it has not been possible to isolate Cowdria and where no clinical evidence of the disease has been found.  相似文献

Forty six spring-calving Holstein–Friesians (12 primiparous, 34 pluriparous) were block-paired (expected calving date, parity, body condition score and genetic merit) and allocated to either a PASTURE or HOUSED system for a full production cycle (− 40 to 305 days relative to calving). Both hind claws were inspected on six occasions (− 40, 10, 35, 85, 120 and 210 days relative to calving) to determine the severity of 5 disorders (sole and white line area haemorrhages, white line disease, heel horn erosion, digital dermatitis and other lesions). Six aspects of locomotion ability (tracking, spine curvature, speed, head bobbing, general symmetry and abduction/adduction) were assessed from 1 (normal) to 5 (abnormal) every 2 weeks. Throughout the study records of clinical lameness were kept for all animals. Lying times of 26 block-paired cows (PASTURE n = 13, HOUSED n = 13) were recorded automatically every 5 min for 48 h at 33, 83 and 193 days post-calving. Data were analysed using mixed models for repeated measures, logistic regression and survival analysis, as appropriate. The severity of hoof disorders was lower for PASTURE compared to HOUSED cows from 85 days post-calving onwards (P < 0.05). HOUSED cows had a greater hazard ratio (P < 0.01) of presenting an abnormal (i.e. scoring ≥ 3) tracking (2.8), spine curvature (2.3), head bobbing (3.6), general symmetry (3.0), abduction/adduction (4.2) and for the average (3.9) of all locomotion aspects investigated (i.e. abnormal locomotion). Furthermore, HOUSED cows had a greater odds ratio (6.5, P < 0.01) of clinical lameness from day 180 post-calving onwards. Mean total lying times per 48 h period were shorter (P < 0.001) for HOUSED compared to PASTURE cows (18.1 h, SE 0.71 vs. 20.5 h, SE 0.73). In summary, from day 85 post-calving to the end of the production cycle PASTURE cows had less severe hoof disorders, better locomotion ability and reduced likelihood of clinical lameness compared to similar cows in a HOUSED system. The PASTURE system also facilitated longer, undisrupted lying times that have beneficial implications for lameness. A PASTURE system therefore improved cow welfare in terms of lameness compared to a HOUSED system.  相似文献

为探究体况评分(Body Condition Score,BCS)和步态评分(Locomotion Score,LS)对奶牛泌乳性能的影响,本试验在2016年7月至8月通过对北京地区7个奶牛场近5 000头泌乳牛进行体况和步态评分并收集相关的DHI数据,采用固定模型分析BCS和LS的影响因素及场、胎次、泌乳阶段、LS和BCS对日产奶量、体细胞数及乳成分的影响。结果表明:BCS和LS对日产奶量均有极显著影响(P0.01),BCS对乳蛋白率有显著影响(P0.05);胎次对LS有极显著影响(P0.01),而对BCS无显著影响,场-测量人、泌乳阶段对BCS和LS均有极显著影响(P0.01);LS与BCS之间回归关系显著,LS较高的个体BCS较低。在生产上可以综合运用BCS和LS加强管理,提升产奶量,改善乳品质量,使牛场获得更大的经济利益。  相似文献

It is well established that T cells cannot be activated by antigen alone but only if antigen is presented in context with I region associated (Ia) determinants. As a matter of fact, antigen-presenting cells or accessory cells, which are obligatory for the induction of any type of immune response, all share the same major characteristic of Ia expression. Thus, there seems to be a direct correlation between accessory cell function and Ia expression. Originally, Ia determinants were only detected on a few cell types, B cells and macrophages being the first. However, during the course of time, more and more cells were found to be Ia positive (Ia +) and it is possible that most cells can express Ia, if appropriately induced. The regulation of Ia expression has been best studied in macrophages, where it has been found that positive induction elements include phagocytosis and gamma-interferon, while prostaglandin E and alpha-fetoprotein tend to down-regulate the expression of Ia. The regulation of Ia expression on accessory cells is thus an integrated part of immune regulation. It is highly likely, although not yet directly proven, that the Ia molecules are the products of the immune response (Ir) genes located within the major histocompatibility complex. They may even be the mediators of the Ir genes which determine whether an immune response can take place at all and/or the extent of the response. 本研究利用2007年间武汉惠尔康扬子江乳业有限公司武湖牧场887头母牛运动评分的鉴定记录,配合单性状动物模型,利用MTDFREML软件,采用REML方法估计了相关的参数。结果表明,运动评分受泌乳期和饲养方式的影响,运动评分随泌乳期的增加有增加的趋势,栓系式饲养奶牛相对于散栏式饲养奶牛有更高的运动评分。运动评分的遗传力为0.09,为低遗传力性状。  相似文献

Various leukocytes are involved in the reaction to counter Salmonella infection in chicken. The various leukocyte types react differently after an infection, since some clear the infection while others may cause dissemination of Salmonella throughout the chicken. Therefore, we investigated in vitro the entry and survival of Salmonella enterica serotype Enteritidis in chicken cell lines of various cell types, including two macrophage cell lines, HD11 and MQ-NCSU (NCSU), two B-cell lines LSCC-1104-X5 (1104) and LSCC-RP9 (RP9), and a T-cell line MDCC-MSB-1 (MSB-1). The macrophages were able to internalize high numbers of S. Enteritidis. In contrast and as expected, cells of the T-cell line MSB-1 and the B-cell line RP9 internalized bacteria at a much lower level. After S. Enteritidis entered the macrophages, the number of intracellular S. Enteritidis decreased over time, so that after 48h no more than 20% of the bacteria, which had entered, survived intracellularly. In contrast to macrophages, the number of S. Enteritidis in cells of the T-cell line MSB-1 and the B-cell line RP9 increased rapidly within 12h post-inoculation. Thereafter the number of intracellular S. Enteritidis decreased only slowly. In conclusion, all three different cell types were able to control and to start clearing S. Enteritidis, although macrophages were far more effective compared to T- and B-cells. However, none of the cell lines were able to clear S. Enteritidis fully within 48h. These results suggest that the three cell types play an important but different role in the dissemination and elimination of S. Enteritidis throughout the animal.  相似文献

Locomotion scoring, lying behaviour and lesion recording during hoof trimming are all ways of evaluating hoof health in dairy cows. The objective of this study was to evaluate the relationship between these measures in a random sample of 1340 cows from 42 Danish dairy herds. The hypothesis was that locomotion scoring and/or the monitoring of lying behaviour could be used as tools to identify cows with hoof lesions, either of the horn or of the skin. Cows were locomotion scored, lying behaviour recorded and data on hoof lesions seen during hoof trimming collected. The results were analysed using logistic regression with hoof lesion as the outcome and locomotion score (1-5), mean duration of lying bouts, parity and lactation stage as explanatory variables. This analysis was undertaken for all types of lesions, for hoof horn lesions only and for skin lesions only. Odds of all hoof lesions and of skin lesions increased with increasing locomotion score and increasing mean duration of lying bouts. Odds of horn lesions also increased with increasing locomotion score, but there was no significant association between horn lesions and the mean duration of lying bouts. It was concluded that locomotion scoring and duration of lying bouts may be used as tools in the management of hoof health in dairy herds.  相似文献

The detrimental effects of the pro-inflammatory cytokine TNF-alpha during equine acute abdominal disease are well known. Its pivotal role in many human diseases has led to various in-depth studies regarding its release mechanism, in particular by TNF-alpha converting enzyme (TACE). In this study we investigated the inhibitory effect of a TACE-inhibitor on cytokine release (TNF-alpha, IL-1alpha and IL-6) in three different cell models, including U937 cells, a recently established equine macrophage cell line, known as eCAS, and primary equine PBMC. The aim was to show the similarity of TNF-alpha release through the TACE mechanism in human and equine cells after stimulation with LPS. Results indicate that, by using a TACE-inhibitor, TNF-alpha, IL-1alpha and IL-6 release can be reduced in both equine cell models and achieved comparable results in the human U937 cells. These results suggest that equine TNF-alpha, like its human homologue, is released from its membrane-bound position by TACE.  相似文献

The anatomy of the sternum in birds varies according to their habitats and type of locomotion. In particular, the carina sterni manifest different shape variations. In this study, the shape of the carina sterni was investigated by means of geometric morphometrics. Birds of different types of locomotion were used in the study: flying, swimming, and terrestrial. Ducks and chickens show a wider variety of shapes. Pigeons are the species with the least differences. The margo cranialis carinae in a turkey is the flattest compared to other species. In chickens, the apex carinae is more caudally than the base of the carina sterni. The margo cranialis of the carina sterni in ducks is concave. The differences in centroid size and shape differences between species collectively are statistically significant (p < 0.0001). The most distinct shape contrast is between the duck and turkey (p: 0.0003). Form differences between the ducks and geese as well as between the chicken and turkey are statistically insignificant. There is less variation in the shape of the carina sterni among avian species representing the same type of locomotion. Although there are many comparative morphological and morphometric studies of birds, shape analysis studies revealing the interspecific differences and similarities of the sternum are very limited. Morphology of the carina sterni can be useful in taxonomic investigations.  相似文献

There are currently eight native horse populations in Japan, namely, Hokkaido, Kiso, Noma, Taishu, Misaki, Tokara, Miyako, and Yonaguni horses. Since locomotion traits, including gaitedness, are important for riding and packing horses, the genetic properties associated with these traits could be informative for understanding the characteristics and history of these horses. In this study, we investigated the distribution of the mutant allele of DMRT3 gene (DMRT3:p.Ser301Ter) associated with ambling gaits in the Japanese native horse. We also examined haplotypes of SNPs in the 83‐kb region including DMRT3 gene by genotyping four SNPs in this region. The results revealed the presence of DMRT3:p.Ser301Ter in the Hokkaido and Yonaguni populations at allele frequencies of 0.18 and 0.02, respectively, and the observed haplotype associated with DMRT3:p.Ser301Ter was estimated as the most common haplotype in the horses in the world. Since DMRT3:p.Ser301Ter has been hypothesized to spread across Eurasian continent from Medieval England after 850 to 900 CE, our findings of the presence of DMRT3:p.Ser301Ter with the common haplotype in the Japanese native horses will provide a new insight into the history of the Japanese native horse, such as considerable level of gene flow from Eurasian continent after 850 to 900 CE.  相似文献

Attachment, a normal behavior among social animals, is quite significant since owners worry about their pets and take care of them because of this affective connection. There are not enough research studies that focus on attachment between owners and their cats. The general objective of this study was to identify attachment behaviors, directed toward their owners, in cats of different body types, age groups, and sexes in an experimental situation.Twenty-eight cats, ranging from 1 to 7 years of age and having different body types, were used in the study without taking into account sex or reproductive status. These cats underwent an Ainsworth’s Adapted Strange Situation Test. Event frequencies and behavioral state durations in individual type behaviors such as exploration/locomotion, alertness, and inactivity were registered using direct focal sampling. For data analysis, cats were divided by body type, sex, and reproductive status. Analysis of variance (ANOVA) of locomotion/exploration revealed a statistically significant difference (N = 28, F = 13.55, P < 0.001) between the episodes with the owner, alone, and with a stranger with cats spending more time engaged in locomotion/exploration while accompanied by their owner. On the alert behavior event frequency, difference (ANOVA, F = 7.44, P < 0.05) was found, which showed a higher frequency while in the company of a stranger. Last, in the inactivity time ratio, a significant difference was found (ANOVA, F = 18.55, P < 0.001), where the time spent on this behavior was considerably higher when the animal was alone.These results are consistent with the ones obtained by Ainsworth in children attached to their mothers; therefore, it can be said that cats can manifest attachment behaviors toward their owners. Further studies are indicated to see whether cats can develop separation anxiety.  相似文献

Background

The effects of lameness on fertility have been documented frequently but few data are available from seasonally breeding, pasture-based herds (such as those used in Ireland) where cows are housed during the winter months but managed at pasture for the remainder of the year. This study determined the prevalence of lameness in a group of 786 cows in 10 pasture-based Irish dairy herds before, during and after the breeding season and assessed the relationship between lameness and the reproductive performance in these herds through serial locomotion scoring during the grazing period.

Results

Lameness prevalences of 11.6 % before, 14.6 % during and 11.6 % after the breeding season were found and these compared favourably to results from housed cattle and are similar to other studies carried out in grazing herds. A Cox proportional hazards model with locomotion score as time varying covariate was used. After controlling for the effect of farm, month of calving, body condition score at calving, body condition score loss after calving and economic breeding index, cows identified as lame during the study were less likely to become pregnant. Cows lame before the earliest serve date but no longer lame during the breeding season, cows becoming lame after the earliest serve date and cows identified lame both before and after this date were respectively 12 %, 35 % and 38 % less likely to become pregnant compared to cows never observed lame during the study. However, these findings were only significant for cows becoming lame after the earliest serve date and cows lame both before and after the start of breeding.

Conclusions

This study found that the reproductive efficiency was significantly (p < 0.05) lower in cows becoming lame during the breeding season and cows lame before and during the breeding season compared to non-lame cows. Cows no longer lame during the breeding season had a lower Submission Rate to first serve within 3 weeks of earliest serve date. However, the Pregnancy Rate was not significantly (p > 0.05) lower in these animals compared to cows never diagnosed as lame. In addition to lameness status, nutritional status and genetics were found to influence the reproductive performance in pasture-based Irish dairy herds.  相似文献

The direct effects of four non-steroidal anti-inflammatory drugs (NSAIDs) on equine polymorphonuclear (PMN) and mononuclear (MN) leucocyte movement were investigated using two in vitro assay systems. The Boyden chamber microfilter technique measures both chemokinetic and chemotactic locomotion, and the agarose microdroplet assay measures solely chemokinesis. Zymosan-activated plasma (ZAP) and the synthetic peptide N-formyl-methionyl-leucyl-phenylalanine (FMLP) were used as standard chemoattractants for PMN and MN leucocytes, respectively. The actions of six concentrations of each NSAID, indomethacin (50 microM-10 mM), phenylbutazone (10 microM-1 mM), oxyphenbutazone (2.5 microM-500 microM) and flunixin (0.1 microM-50 microM), in suppressing cell movement induced by ZAP and FMLP were investigated. All four drugs exerted inhibitory effects on induced movement of both cell types in the Boyden chamber assay, usually in a concentration-dependent manner, although oxyphenbutazone action on PMN cells occurred only at the highest concentration tested. Significant inhibition of PMN and MN cell locomotion was produced by indomethacin, flunixin and oxyphenbutazone, and inhibition of PMN movement by phenylbutazone occurred in the agarose microdroplet assay. Flunixin was the most potent of the four drugs investigated in both assay systems. The findings may be of importance to the use of phenylbutazone and flunixin as NSAIDs in equine medicine, since the concentrations used were similar to concentrations of both drugs and the phenylbutazone metabolite oxyphenbutazone previously reported to occur in equine plasma and inflammatory exudate.  相似文献

Cells recovered by bronchoalveolar lavage from the lungs of specific pathogen-free (SPF) cats from birth to maturity and from adult conventional cats were enumerated and identified. The predominant cell type recovered was the pulmonary alveolar macrophage from all ages of both SPF and conventional cats. Other cell types included eosinophils, neutrophils and lymphocytes. Lavage of conventional cats yielded significantly more eosinophils and neutrophils than were recovered from SPF cats.  相似文献
