Inter-section hybrids were obtained in the reciprocal crosses between Primula filchnerae (2n = 2x = 24) of Sect. Pinnatae and P. sinensis ‘Fanfare’ (2n = 2x = 24) of Sect. Auganthus by rescuing ovules on half-strength (1/2) Murashige and Skoog's (MS) medium supplemented with 50 g l−1 sucrose, 2.5 g l−1 gellan gum, 0.1 mg l−1 α-naphthaleneacetic acid (NAA), 0.1 mg l−1 6-benzyladenine (BA) and 50 mg l−1 gibberellic acid (GA3). In ovule culture, germination occurred with radicle elongation but no plumule was observed. The radicle kept on the initial medium showed root proliferation with callus formation. When the calluses were transferred to (1/2)MS media containing 30 g l−1 sucrose and 3 g l−1 gellan gum, without plant growth regulators (PGRs) or with 1 mg l−1 zeatin and 0.1 mg l−1 NAA, plantlets were regenerated. The plants thus obtained were confirmed to be hybrids through flow cytometry (FCM) and random amplified polymorphic DNA (RAPD) analyses. The hybrid obtained when P. filchnerae was used as the maternal parent was diploid, whereas hexaploid hybrid was obtained when using P. sinensis as the maternal parent. The hexaploid hybrid might be produced through chromosome doubling of a triploid originated from the fertilization of P. sinensis with unreduced pollen of P. filchnerae.  相似文献

 The shape of the flexural vibration wave of wooden beams at the first mode was detected using the transfer function. The dynamic modulus of elasticity (MOE) of beam sections of differing quality was estimated from the ratio of the curvature of the wave shape in this section to that of a clear beam. The results were as follows: (1) If a section with a lower dynamic MOE was introduced into a clear wooden beam, the curvature of the wave shape in that section became higher. (2) The ratio of the MOE and the reciprocal of the curvature ratio were highly correlated. (3) The MOE of a defect could be estimated, and the position of the defect could be determined accurately by examining the curvature of the flexural vibration wave shapes. ABSTRACT:   The effects of 80% methanol extracts from frozen samples of 41 macroalgae and one sea grass collected in the Ise-Shima region of Japan were investigated on histamine release from rat basophile leukemia cells (RBL-2H3) sensitized with antidinitrophenyl (DNP) IgE and stimulated with DNP-BSA. Of the 21 brown algae, five green and 15 red algae, and one sea grass tested, only extracts from seven brown algae suppressed histamine release from RBL cells, as determined by high-performance liquid chromatography (HPLC) analysis. When the cytotoxic effects of the seven brown algal extracts were investigated by Trypan blue staining, only Eisenia arborea and Sargassum thunbergii did not show cytotoxic effects. Therefore, we conclude that E. arborea and S. thunbergii may contain compounds that have antiallergic effects without inducing cell death.  相似文献

ABSTRACT:   Wild adult maturing and immature female Japanese flounder Paralichthys olivaceus were collected in June 2004 and January 2005, respectively, to clarify a possible role of gonadotropin-releasing hormones (GnRHs) in reproduction. Levels of salmon GnRH (sGnRH), chicken GnRH-II (cGnRH-II) and sea bream GnRH (sbGnRH) in the brain and pituitary were examined by time-resolved fluoroimmunoassay. Three forms of GnRHs were detected in the discrete brain at various levels. In the pituitary of both maturing and immature fish, sbGnRH was abundant together with a pronounced amount of sGnRH, whereas cGnRH-II was almost below the detectable limit. In maturing fish, levels of sbGnRH were high in the telencephalon, hypothalamus and pituitary, while levels of sbGnRH of immature fish were very low in these regions. These results indicate that sbGnRH is mainly responsible for gonadotropin secretion, and that sbGnRH in the anterior part of the brain is associated with gonadal maturation in the Japanese flounder.  相似文献

ABSTRACT:   To assess the effect of a seaweed mixture on lipid levels in serum as well as platelet aggregation in rats, Eisenia bicyclis ('Arame'), Hizikia fusiformis ('Hijiki') and Undaria pinnatifida sporophylls ('Mekabu'), all brown seaweeds, and Porphyra yezoensis ('Susabinori'), a red seaweed, were powdered and mixed in a ratio of 45:30:20:5 (w/w). When rats were fed a cholesterol-rich diet containing this mixture of seaweeds (9–10% w/w) for 28 days, serum total cholesterol, LDL-cholesterol, free cholesterol, and triglyceride levels declined significantly to 49.7%, 48.1%, 49.0% and 74.8%, respectively, of those of the control. Serum HDL-cholesterol, however, was unchanged. Though activated partial thromboplatin time, prothrombin time, antithrombin III activity, and fibrinogen levels in plasma were unchanged, the maximal ADP- and collagen-induced platelet aggregation decreased significantly to 89.0% and 85.5% control levels, respectively. These results indicate that this mixture of E. bicyclis , H. fusiformis , U. pinnatifida sporophylls, and P. yezoensis , is useful for the prevention of hyperlipidemia and thrombosis in rats.  相似文献

The occurrence of quartz particles having a platy morphology found in the white layers within the fossil forest deposits of Axel Heiberg Island, was further confirmed by optical and electron microscopic examinations. The platy quartz crystals were concentrated in the finer fractions. The δ 18O values of fractions enriched in platy quartz crystals ranged from +13.7 to +16.6 %0. These values were higher than those obtained for coarser fractions. The platy morphology and crystallographic character of the quartz particles concentrated in the finer size fractions are uncharacteristic both of aerosol derived material and of secondary silica. These findings and information available for the paleoenvironment of the study area favor a biogenic origin for the platy quartz. The oxygen-isotope data are consistent with this hypothesis.  相似文献

1. Inter-breed morphological comparisons were made among 11 breeds of Japanese native chickens (Gifujidori, Hinaidori, Shokoku, Totenko, Tomaru, Satsumadori, Shamo, Koshamo, Koeyoshi, Chabo and Nagoya), White Leghorn, broiler chickens (Chunky) and red junglefowl collected in the Philippines, based on results of direct measurements and analysis by computer vision techniques of the skull. 2. Analysis of direct measurements identified two groups of chicken: a small type that included the Chabo, Koshamo, red junglefowl, Gifujidori and Shokoku and a large type that included the remaining breeds studied. These groupings were made based on size determined both in the first (PC1) and second principal component (PC2). The greatest length of the cranium and condylobasal length greatly contributed to the morphological differences between these two groups. 3. Analysis by computer vision techniques, however, identified three groups: the Bantam group (which includes red junglefowl), Shokoku group and Shamo group. White Leghorn clustered within the Shokoku group while the broiler chicken belonged to the Shamo group. The region around the junction of the neural cranium and the visceral cranium contributed greatly to the morphological differences among breeds, both in the PC1 and PC2.  相似文献
