This study investigated the effect of pentoxifylline (PTX) and Basal Medium Eagle (BME) on frozen–thawed goat spermatozoa. Immediately after initial examination of ejaculated semen, samples were pooled and reexamined for quality. Then, samples were divided into eight equal aliquots and diluted with a basic tris-extender containing PTX (3, 6, 9 mM) and BME (5 mM) to reach a final concentration of 25 × 109 and frozen. After 24 hr, the samples were individually thawed at 37°C for 30 s and evaluated for different characteristics. Obtained post-thaw results from Computer-Assisted Sperm Analysis indicate using of 3 and 6 mM PTX led significantly to an improvement in total motility, progressive motility and velocity characteristics of spermatozoa, except the beat/cross frequency (BCF) which indicated statistically no differences (p > .05) among control and treatments. Diluents prepared with BME (5 mM) and PTX alone (3 and 6 mM) improved significantly the membrane integrity–functionality, acrosome integrity and also hyaluronidase activity. Regarding recovery rate, the results showed significantly (p < .05) higher values for diluents containing 3 and 6 mM PTX compared to other groups. Malondialdehyde concentration exhibited also a significant difference (p < .05) in diluents supplemented with 5 mM BME, 3, 6 and 9 mM PTX, and mixture of 3 mM PTX and 5 mM BME which illustrate a similarity for active mitochondria, apoptotic-like and dead spermatozoa. Finally, the ratio of sperm chromatin dispersion stained spermatozoa presented significant differences (p < .05) among treatments in which the diluents added PTX alone demonstrated significantly lower values than control and extenders containing the mixtures of BME and PTX. In conclusion, the observation in this study indicates using of 3 and 6 mM PTX and BME alone may improve significantly (p < .05) the quality of cryopreserved goat spermatozoa.
Growth stage effects on distribution of mineral nutrients or beneficial elements phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), sulfur (S), chloride (Cl), iron (Fe), zinc (Zn), manganese (Mn), copper (Cu), molybdenum (Mo), sodium (Na), silicon (Si) and nickel (Ni), and the elements bromine (Br), rubidium (Rb), strontium (Sr), barium (Ba), lanthanum (La), cerium (Ce), and uranium (U) in two barley (Hordeum vulgare L.) cultivars and how the distribution of these elements changed were determined during the 2006–2007 growing season in a field experiment. Barley plants were sampled from the field at shooting, heading, soft dough, hard dough and harvest stages, and mineral nutrients and other elements concentrations of spike, flag leaf, old leaf, and stem samples were determined by polarized energy dispersive X-ray fluorescence (PEDXRF). Distribution patterns varied considerably from element to element. At the end of the season much of the Ca, Mg, S, Si, Fe, Mn, Cu, Ni, Sr, Ba, La, Ce, and U were located in the spikes. However, much of the P, K, Zn, Cl, Na, Br, and Rb remained in the old leaves or stem.
The aim of this study was to evaluate the water and nitrogen use efficiency and some quantitative and qualitative characteristics of forage beet cultivars under the influence of different irrigation methods and nitrogen levels in two cropping years, 2017–18 and 2018–19, at Agricultural Research Station in Karaj, Iran. Experimental factors included the first factor with four irrigation methods (normal leakage, alternate furrow irrigation, fixed furrow irrigation, type (drip-strip)), the second factor was the amount of nitrogen fertilizer with three levels (150, 200 and 250?kg N ha?1) and the third factor included three forage beet cultivars (Sbsi052, Jamon and Kyros). Among irrigation treatments, alternate furrow irrigation and fixed furrow irrigation had the highest sugar content with 9.28% and 9.17%, respectively. The highest yield of digestible organic matter was obtained in leakage irrigation treatment, nitrogen fertilizer of 250?kg ha?1 and in Kyros at the rate of 19.45?t ha?1. The highest yield of root digestible dry matter, potassium, sodium and free nitrogen was observed in leakage irrigation treatment and consumption of 200?kg ha?1 nitrogen was observed in foreign cultivars. The highest crude protein was observed in alternate furrow irrigation conditions with a consumption of 200?kg ha?1 nitrogen in cultivar Sbsi052 at 13.08%. Leakage irrigation and type tape had the highest consumption efficiency and efficiency of nitrogen uptake with application of 150 and 200?kg ha?1 N, and the highest water use efficiency was also observed in leakage irrigation and type tape with application of 250?kg ha?1 N in domestic and foreign cultivars. The type irrigation method showed better quantitative and qualitative yield than the furrow irrigation methods.
Unidentified olive plants naturally grow in the Golestan province of Iran, on different soils and under climates spanning from sub-temperate to desert conditions, represented by single trees or groups of few trees. We collected samples from representative sites and analyzed them by simple sequence repeat markers in order to determine their identity and their relationships to prominent Iranian and Mediterranean reference cultivars. Population structure analysis separated these ecotypes from Mediterranean and, surprisingly, from all Iranian cultivars, the parentage test excluded their direct contribution as candidate parents or offspring of cultivars, and they also showed a high level of admixture. Their differentiation from cultivated olives may be attributed to different factors: they could represent wild plants or could derive from natural dissemination of ancestral cultivated trees. Their survival up to now may be due to the fact that most of them are grown on sacred sites such as necropolis. Anyhow, the adaptation to strong environmental stresses, and their fruit size and oil content make the olive Golestan ecotypes a valuable source of genetic variation previously uncharacterized and currently threatened with extinction.
Transgenic Korean rice plants containing the cry1Ab gene were developed for resistance against yellow stem borer (Scirpophaga incertulas, YSB). More than 100 independent transgenic lines from three Korean varieties (P-I, P-II and P-III) were generated. The amount
of Cry1Ab in transgenic T0 plants was as high as 2.88% of total soluble proteins. These levels were sufficient to cause 100% mortality of YSB larvae.
The majority of T1 transgenic lines originated from the varieties P-I and P-II followed a Mendelian fashion of segregation. Deviation from the
expected segregation ratio was observed in a small number of the transgenic lines of P-I and P-II origins. However, this deviation
was primarily observed in the P-III originated lines. Segregation analysis of the T1 generation indicated that 1–3 copies of the cry1Ab gene were integrated into the genome of the majority of the transgenic lines originating from varieties P-I and P-II. Stunted
and semi-fertile mutants were observed in some transgenic lines. These aberrations were either independent or closely linked
to the introduced cry1Ab gene loci in different transgenic lines. Reduction in GUS expression levels and loss of toxicity against YSB larvae were
found in some transgenic lines. The transgenic T3 and T4 lines causing 100% mortality of third instar YSB larvae in the lab were completely protected in the field. Analysis of important
yield components on nine selected transgenic lines indicated that stem length, panicle length, grain number per panicle, and
seed setting rates were reduced in transgenic plants compared to those in non-transgenic parental rice lines. Number of panicles
per cluster, however, was significantly higher in transgenic plants. The numerical value of the average yield was in general
greater in the controls than in all the transgenic lines, indicating some ‘yield drag’. Since some selected lines were highly
resistant to the YSB with good yielding potential, they offer effective potential for use in insect resistance management
programs.
Serum IgG, IgM and IgA antibody response in 20 cattle naturally infected with Mycobacterium paratuberculosis and in 15 non-infected cattle were measured by enzyme-linked immunosorbent assay. A strong IgG response was detected in 16 (80%) of the infected animals. Diagnostic levels of IgM were detectable in all of the infected animals as well as in 8 (53%) of the non-infected animals. Animals with paratuberculosis had a very weak specific serum IgA response and this appears to be of little value in detection of infection in these animals.
Fungal toxins in feed are leading issue in poultry industry causing a detrimental effect on the performance and health of poultry. The study was carried out to determine the incidence and concentration of the aflatoxins and their major producer Aspergillus flavus in home mix layer poultry feed in respect of seasonal variation throughout the year. A total of (n?= 204) home mix poultry layer feed samples were analyzed for the isolation of fungi. The isolates were initially screened through colony morphology and microscopic examination. However, aflatoxin concentration was determined by ELISA. Revealed results indicated that, the highest percentage of A. flavus was found during the months of June to August 50/54 (92.5%) followed by September to November 43/65 (66.1%), March to May 21/40 (52.5%), and December to February 18/45 (40%). As a whole, the incidence was recorded 132/204 (64.7%). Moreover, of the 132 samples, 41 (31%) were exceeded in respect of aflatoxin contamination from the legal limit (20 μg/kg) imposed by Food Drug Association (FDA). Statistically, the growth of A. flavus and aflatoxin production was found significantly different in respect of seasonal variation. As highest total viable fungal count (9.9?×?104 CFU/g) and aflatoxin level (72.27 μg/kg) were recorded during the months of June to August and lowest in December to February. Consequently, instantaneous essential control measures are demanded regarding appropriate storage and adequate drying in post-harvesting season. Along with surveillance plans and austere regulations for monitoring the aflatoxin contents for the wellbeing of consumers.
Summary The immune responses following vaccination and resistance to diseases were compared in male and female meat poultry breeders of the same flock. Female poultry breeders maintained antibody titres to Newcastle disease virus and infectious bronchitis virus up to the fifty-fifth day following vaccination, whereas those of the males declined significantly over the same period of time (P<0·05). In the same flock, outbreaks of Gumboro disease (60 to 62 days of age), coccidiosis (68 to 74 days of age) and aortic rupture (99 to 112 days of age) produced significantly higher losses in males. Following vaccination against fowl pox by the wing web method, 96·7% of females had a vaccine reaction (vaccine take) compared to none of the males. Immune injuries, following vaccination, were observed in 85% of the males compared to none of the females. The immune injuries included appearance of facial papules, vesicles, and reddish brown to black scabs.
Comparacion De La Inmunidad Y Resistencia A Enfermedades En Machos Y Hembras Reproductores Aviares En El Libano
Comparison De L'immunite Et De La Resistance Aux Maladies Chez Des Reproducteurs Males Et Femelles Dans Un Elevage De Volailles De Chair Au Liban
This study was done to evaluate the effects of increasing concentrations of CO2 (CC) on rapeseed. Pot experiments were done with three cultivars (Okapi, Zarfam and RGS003) of rapeseed (Brassica napus) for salinity tolerance. Four levels of salinity (0, 5, 10 and 15 dS m?1) were tested on the three cultivars at three CC (350, 700 and 1050 mmol L?1) at the greenhouse of Tarbiat Modares University, Iran, during the crop seasons of 2010 to 2011. Three CCs were considered as three environments and the other two treatments (salinity and cultivar) were tested within these environments in a complete block design arranged as a factorial. Results indicated that photosynthetic rates declined with increasing levels of salinity. Elevated CC significantly increased rates of photosynthesis. The highest CC reduced the impact of salinity on photosynthesis. Increased CC reduced the rate of transpiration, which had the effects of increasing rates of photosynthesis and water use efficiency (WUE); these effects increased vegetative growth and reduced the adverse effects of salinity. Increased CC and salinity reduced harvest index. WUE increased with CC increment, and decreased with salinity elevation.
