Zebrafish sperm cryopreservation is a fundamental methodology to manage and back-up valuable genetic resources like transgenic and mutant strains. Cryopreservation usually requires liquid nitrogen for storage, which is expensive and hazardous. Our objective was to evaluate if electric ultrafreezers (??150 °C) are a viable alternative for zebrafish sperm storage. Zebrafish sperm was cryopreserved in the same conditions (??20 °C/min), stored either in liquid nitrogen or in an ultrafreezer, and thawed after 1 week, 1 month, and 3 months. Sperm motility, membrane integrity, and fertilization ability were assessed. There were no significant differences in motility and hatching rate throughout storage time. Additionally, we aimed at understanding if cryopreservation directly in an ultrafreezer (??66 °C/min) could improve post-thaw sperm quality. Freezing at ??20 °C/min was performed as before, and compared to samples cryopreserved with a fast cooling rate by placing directly in an ultrafreezer (??66 °C/min). Sperm quality was assessed according to motility, viability, DNA fragmentation, and apoptosis (annexin V). The ??66 °C/min cooling rate showed significantly higher membrane and DNA integrity, and lower number of cells in late apoptosis in comparison to the other treatments. This study showed that zebrafish sperm cryopreservation and storage in an ultrafreezer system is possible and a fast cooling rate directly in ultrafreezer improves post-thaw sperm quality.  相似文献

The onset and progression of type II diabetes is closely related to environmental factors, in particular dietary habit. Moreover, the environmental exposures very early in life can influence the risk for development of type II diabetes later in life. In this study, we investigated pathophysiological changes in the pups of maternal Spontaneously Diabetic Torii (SDT) rats that were fed a high‐fat diet (HFD) throughout gestation and lactation. Maternal SDT rats were continued on HFD for 5 weeks, from day 8 of gestation to day 21 after birth, and biological analyses of the pups were performed from 2 to 22 weeks of age. Results of serum lipid levels in pups from dams fed HFD were higher than pups from dams fed a standard diet, and the onset of diabetes was significantly accelerated in pups from dams fed HFD. In pathological analyses, pups from dams fed HFD showed increases in liver weight and vacuolation of hepatic cells at 2 weeks of age. In conclusion, the metabolic disorder of lipids and glucose in SDT rats is closely related to the nutritional condition of dams during the periods of gestation and lactation.  相似文献

The vine bud moth, Theresimima ampellophaga (Bayle-Barelle 1808) (Lepidoptera: Zygaenidae; Procridinae), is a pest on grape vine, Vitis vinifera (L.), and on the ornamental vine Parthenocissus quinquefolia (L.) and other Parthenocissus species. Its recent distribution in Europe is relatively well known, but there is a paucity of locality data for Turkey. As a consequence, we began a large-scale investigation into its distribution and seasonal activity in this country in 2008. The main tools for these investigations were pheromone traps that were baited with the synthetic pheromone of the species, viz. (2R)-butyl (Z7)-tetradecenoate. As a result of field work carried out in the eastern Mediterranean region of Turkey in 2008 and 2009, Th. ampellophaga was recorded in five provinces, viz. Adana, Mersin, Hatay, Kahramanmara? and Kilis. The only province where the pest was not found was Gaziantep. Theresimima ampellophaga produces one or two generations per year, depending on the local conditions and other unknown factors. The moths of the first generation fly in June, while those of the second generation occur in August. To the best of our knowledge the present paper provides the first well-documented evidence of the occurrence of a second generation of the vine bud moth in Turkey.  相似文献

The aims of this study were to evaluate the efficiency of simple and complex extenders in prolonging the cold storage of sperm (Experiments 1 and 2) and to test the diluted‐cooled sperm in the best extender with regard to sperm quality parameters (Experiment 3) in the streaked prochilod, Prochilodus lineatus. In all the experiments, aliquots of 0.3 mL of sperm were diluted 1:10 in extenders and stored at 4–6 °C. Sperm diluted in simple extenders (NaCl and glucose solutions) yielded 0–26% sperm motility, whereas sperm diluted in complex extenders (BTS?, M III? and Androstar?) yielded 62–81% sperm motility on day 4 after cold storage. When Androstar? was further investigated, the following was observed on day 4: 53% motility with 94 s of duration; 47% live spermatozoa; 26–61% fertility rate; and 22–60% hatching rate. The use of Androstar? improves the sperm fertility of the streaked prochilod during a 4‐day storage period and can therefore be used to facilitate artificial reproduction.  相似文献

Mycoplasma pneumonia of swine (MPS) is caused by Mycoplasma hyopneumoniae (M.hp) and is a common chronic respiratory disease of pigs. Recently, a genetically selected variant of the Landrace pig (Miyagino L2) has a lower incidence of pulmonary MPS lesions. We investigated the pathological and immunological characteristics of MPS resistance in these pigs (n = 24) by comparing with the normal landrace pig (control: n = 24). The pathological MPS lung lesion score in MPS‐selected landrace pigs was significantly lower than in the control. The gene expression of interleukin (IL)‐12p40, which acts as a chemoattractant and a component of the bioactive cytokines IL‐12 and IL‐23, was significantly higher at the hilar lymph nodes, lung, and spleen in MPS‐selected landrace pigs than in control landrace pigs, and these were negatively correlated with the macroscopic MPS lung lesion score. In summary, we demonstrate that resistance against MPS in Miyagino L2 pigs is associated with IL‐12p40 up‐regulation, in comparison with normal landrace pigs without the MPS vaccine. In addition, a comparative study of macroscopic MPS lung lesions and IL‐12p40 gene expression in lung and hilar lymph nodes may lead to beneficial selection traits for the genetic selection for MPS resistance in pigs.  相似文献

Temporal changes in a hydrological system and riparian ecosystem were examined with reference to land-use conversion in order to clarify the linkages between these two systems. First, the hydrological system of the Toikanbetsu River basin was divided into three components that measure water retention, inundation and conveyance. Variation in the hydrological system was expressed as a basis of delineating the three components and estimating their functions. The rainfall-runoff system was also examined using a model which can predict responses of surface-, subsurface- and base flows on rainfall intensity. Second, areas and fragmentation of the riparian forests, maximum stream temperature in summer and amount of coarse woody debris (CWD) were selected as parameters indicating the condition of the riparian ecosystem. Temporal changes in stream temperature and amount of CWD were estimated using multiple regression analysis and analysis of variance, respectively. The results indicated that the hydrological system has been altered since the 1970s, increasing flood peaks by 1.5–2.5 times and shortening peak appearance by 7 hours. Riparian forests have been disappearing since the 1960s due to extensive development of agricultural lands and river channelization. The summer maximum stream temperature increased from 22 °C in 1947 to 28 °C at present. The amount of CWD should substantially decrease with river channelization and associated forest cutting. Fish favoring cool water, such as masu salmon, could survive in 1947 although they are forced to migrate to cooler forested upstream tributaries now. The ecological systems were closely related to and distinctly altered by land-use. Finally, we propose a new perspective for understanding the two interrelated systems. Riparian ecosystems can be restored by restoring water retention and inundation functions, which also reduce the flood hazard generated by elevated flood peaks.  相似文献
