Selection for high (H) and low (L) oxygen consumption (OC) as an indirect estimation of maintenance energy requirement was determined. Feed intake and body weight were measured and feed conversion ratio (FCR) of 4–8‐week‐old mice was calculated. Respiratory activity of hepatic mitochondria was measured at 12 weeks. Total feed intake (H: 103.74 g, L: 97.92 g, P < 0.01), daily feed intake (H: 3.70 g/day, L: 3.50 g/day, P < 0.01) and FCR (H: 18.79, L: 15.50, P < 0.01) were significantly different between lines. The line by sex interaction was significant for FCR. No line differences were observed in males; and the FCR of the H line was greater than in the L line in females. H line mice had the highest hepatic mitochondrial respiratory activity in state 2 (P < 0.01), the highest uncoupled respiratory rate of mitochondria in the presence of an uncoupling agent (P < 0.001), and the mitochondrial proton leak. The adenosine diphosphate/ O ratio was highest in the L line (P < 0.05). This suggests that the selection for high and low OC induced differences in basal mitochondrial respiration and basal metabolism, resulting in difference in FCR between H and L lines.  相似文献

ObjectiveTo investigate the effects of habituation and isolation on mechanical nociceptive thresholds in pigs at the pelvic limbs and at the tail.Study designProspective randomized multifactorial study.AnimalsThirty-two healthy castrated male (experiment 1), and 12 castrated male and 12 female (experiment 2) Danish Landrace × Yorkshire pigs, weighing 63.5 ± 0.8 kg and 55.4 ± 0.6 kg (the mean ± SD, experiment 1 and 2, respectively).MethodsMechanical nociceptive thresholds were quantified with a von Frey anesthesiometer applied to two distinct anatomical regions (tail and pelvic limbs). Pigs receiving the mechanical challenge in the pelvic limbs were tested inside a cage, whereas pigs exposed to stimuli at the tail region were tested in an open arena. For both experiments, the effect of familiarity to the procedure was evaluated by comparing thresholds of nociception in habituated versus naïve pigs. The presence of a companion animal was also evaluated in pigs receiving stimuli at the pelvic limbs.ResultsPigs tested inside the cage were affected by the habituation to the procedure as indicated by the increase in willingness and time spent by the animals in the test cage. This effect was reflected in the lower mechanical nociceptive thresholds (medians with 25–75 percentiles) recorded for familiar pigs compared with naïve animals [495 g (302–675) versus 745 g (479–1000), respectively; p = 0.026]. Mechanical nociceptive thresholds measured at the tail of the pigs in the open arena were not affected by the familiarity of the animals with the experimental procedure.Conclusions and clinical relevanceThe current results reiterate the value of habituation in research involving animal behaviour. Further characterization of the methodology is needed to allow its application in the evaluation of clinical conditions in pigs.  相似文献

Ceramic aggregates (Turface® and Profile®) are common soilless media components, but their sorption/desorption chemistry is poorly understood. We investigated: labile (readily desorb-able or readily plant-available) ion concentrations; the effect of rinsing and soaking pretreatments on labile ions; sorption of applied nutrients; and nutrient uptake from the aggregates by plants. Variability in labile ions was extremely high among bags of aggregates. Manganese, boron, magnesium, calcium, sulfur and potassium were most likely to desorb in excess for plants. Phosphorus, iron, copper and zinc were sorbed by the aggregates; only copper was found nearly deficient in plant tissue. Rinsing and soaking pretreatments adjusted labile ions to more suitable concentrations for plants. However, growth data suggested a worst-case scenario of high levels of labile ions may not be mitigated by these pretreatments. With frequent leaching after planting or where the aggregates are a minor component of media, excessive nutrient uptake would likely be limited.  相似文献

Knowledge about the role of microorganisms in P cycling at conditions of constant soil respiration rates and constant size of microbially bound P is lacking. To study the kinetics of microbial P uptake and cycling under such conditions, soils differing in biological activity were 33PO4 labelled by introducing a carrier-free tracer solution and incubated for 56 days. The 33PO4 incorporation into the fraction of microbial P releasable by chloroform treatment (Pchl) was assessed and the isotopic composition [=specific activity (SA); SA=33PO4/31PO4] of Pchl and soil solution P compared. Soils were taken from a 20-year-old field experiment including a non-fertilised control (NON), a minerally fertilised conventional (MIN) and two organic farming systems [bio-organic (ORG); bio-dynamic (DYN)]. Tracer P incorporation continuously increased during incubation in DYN, ORG and MIN soils. It decreased in the order DYN>ORG>MIN, with differences in 33PO4 uptake between the farming systems being higher than suggested by the differences in the amount of Pchl. In the P-deficient NON soil, the highest initial incorporation of tracer P was found, but no additional uptake could be detected thereafter. In all soils, the SA of Pchl converged to the SA of the soil solution with increasing time. Since Pchl remained almost constant during the experiment, the findings suggest an intensive uptake of P from the soil solution into Pchl and concomitant release of P back to the soil solution and, thus, a rapid cycling through Pchl. Intensive P cycling between Pchl and the soil solution was confirmed in an additional experiment where microbial activity was stimulated by glucose and N additions.  相似文献

A novel indole glucosinolate, 1,4-dimethoxyglucobrassicin (1,4-dimethoxyindol-3-ylmethylglucosinolate), was isolated as the desulfo derivative from roots of the P-type of Barbarea vulgaris ssp. arcuata, and its structure was determined by spectroscopy including 2D NMR spectroscopy. 4-Hydroxyglucobrassicin (4-hydroxyindol-3-ylmethylglucosinolate) was isolated as the desulfo derivative from green siliques (fruits) of Arabidopsis thaliana and identified by comparison of its (1)H NMR spectrum with the spectrum of the known desulfoglucosinolate from Brassica napus. The delayed elution of desulfo indole glucosinolates from the DEAE Sephadex column used in sample preparation was examined, and the diode-array UV spectra of desulfo indole glucosinolates were measured, to ensure a reliable determination of 1,4-dimethoxyglucobrassicin and 4-hydroxyglucobrassicin with the existing analysis method based on the HPLC of desulfoglucosinolates. 1,4-Dimethoxyglucobrassicin was not detected in 10 other Arabidopsis, Brassica, and Barbarea species, indicating an evolutionarily recent mutation in the indole glucosinolate biosynthesis in B. vulgaris ssp. arcuata type P.  相似文献

In a breeding programme where young potential breeding bulls are reared on performance test stations, selection based on own results can be carried out before test inseminations. Both beef and milk production traits are included in the total merit index used for selection, and estimates of genetic and phenotypic parameters of these traits are therefore of interest for an optimal construction of such indices. Data on first lactation milk records from the field and beef records of potential dairy breeding bulls from the Danish performance test stations were analysed in bivariate animal-sire models using the AI-REML algorithm. Genetic correlations of 0.16, 0.25 and 0.43 between feed intake capacity and protein yield were obtained for Red Danish (RD), Danish Black and White (DBW) and Danish Jersey (DJ), respectively. These correlations were significantly different from zero for the two populations (DBW and DJ). Genetic correlations around zero between feed efficiency and protein yield were obtained for all three populations. Genetic correlations of 0.44, 0.19 and 0.47 between average daily gain and protein yield were obtained for RD, DBW and DJ, respectively. The genetic correlations between protein yield and muscle area was close to zero for DBW, while it was -0.31 for RD. Selection index calculations indicate that indices composed of different beef performance traits can be used as early predictors for milk yield. Selection on such an index could increase the breeding value of the young bulls for milk production traits by 0.8-2.0% of the population mean.  相似文献

Abstract

Alternative selection strategies in the bull dam path were investigated to reduce the magnitude of bias. The study's objective was to examine the effect of alternative strategies under the assumption of particular biases. Bias in the evaluation of bull dam estimated breeding values (EBVs) causes an estimated reduction of 32% in their genetic superiority, compared with the theoretical potential. Biased evaluations caused selection of too many bull dams from later lactations. Two alternative strategies were developed to account for existing bias. Results suggested that the influence of bias could be reduced. The estimated improvement in selection response was 11–13% higher than current practice. Problems with biased EBVs in conventional breeding schemes are discussed and a method to counteract bias from future breeding schemes is presented.  相似文献

Cattle weight gain responses to seasonal weather variability are difficult to predict for rangelands because few long-term (>20 yr) studies have been conducted. However, an increased understanding of temperature and precipitation influences on cattle weight gains is needed to optimize stocking rates and reduce enterprise risk associated with climatic variability. Yearling steer weight gain data collected at the USDA-ARS High Plains Grasslands Research Station at light, moderate, and heavy stocking rates for 30 years (1982–2011) were used to examine the effects of spring (April–June) and summer (July–September) temperature and precipitation, as well as prior-growing-season (prior April–September) and fall/winter (October–March) precipitation, on beef production (kg · ha?1). At heavier stocking rates, steer production was more sensitive to seasonal weather variations. A novel finding was that temperature (relatively cool springs and warm summers) played a large predictive role on beef production. At heavier stocking rates, beef production was highest during years with cool, wet springs and warm, wet summers, corresponding to optimum growth conditions for this mixed C3–C4 plant community. The novelty and utility of these findings may increase the efficacy of stocking rate decision support tools. The parsimonious model structure presented here includes three-month seasonal clusters that are forecasted and freely available from the US National Oceanic and Atmospheric Administration up to a year in advance. These seasonal weather forecasts can provide ranchers with an increased predictive capacity to adjust stocking rates (in advance of the grazing season) according to predicted seasonal weather conditions, thereby reducing enterprise risk.  相似文献
