This paper presents the first report of multiple anthelmintic resistance in the gastrointestinal nematodes of goats and its possible contributory factors in an irrigated area (Pakistan). A total of 18 privately owned Beetal goat flocks were selected in order to determine the anthelmintic resistance against commonly used anthelmintics. Forty to 48 animals from each flock were selected according to their weight and egg count. The three anthelmintics viz., oxfendazole, levamisole and ivermectin, were given to three groups at manufacturer’s recommended dose while one group was kept as untreated control. Anthelmintic resistance was determined through faecal egg count reduction and egg hatch tests while assessment of the contributory factors of anthelmintic resistance was measured through the rural participatory approach. Faecal egg count reduction test revealed high prevalence of anthelmintic resistance (83.3%) and it was either single (levamisole) or multiple (oxfendazole and levamisole). Egg hatch test confirmed the resistance against oxfendazole as detected with faecal egg count reduction test. None of the goat flocks was resistant to ivermectin. Copro-cultures revealed that Haemonchus contortus, Trichostrongylus colubriformis and Teladorsagia circumcincta were the most common species exhibiting resistance to levamisole and oxfendazole. Step-wise logistic regression of the data on worm control practices revealed significant role of under-dosing, low-protein diets, healthcare supervision by the traditional healers and mass treatments.  相似文献

Tissue samples were collected at random from cattle (Bos taurus) and buffalo (Bubalus bubalis) from an abattoir of the district of Lahore and were analyzed for the presence of Mycobacterium avium subsp. paratuberculosis and Mycobacterium bovis through acid-fast staining and polymerase chain reaction (PCR). Body condition of animals and diarrhea were recorded. Most of the animals were emaciated. Diarrhea was noticed in 15.6% of buffaloes and 19.2% of cattle. Intestinal pathology was observed in 29% of buffaloes and 32.8% of cattle. Number of mesenteric lymph node (MLN) showing gross lesions was a bit higher (35.6%) in cattle than buffalo (31.2%). Acid-fast staining of tissue scraping smears revealed the presence of acid-fast bacilli (AFB) in 17.4% intestinal and 16.4% MLN tissue samples in buffalo, while in cattle 19.2% intestinal and 17.8% MLN were found positive for AFB. In buffaloes, PCR confirmed 12.8% intestinal and 12.4% MLN positive samples for M. avium subsp. paratuberculosis. However, in cattle, PCR analysis demonstrated 14.2% positive results for M. avium subsp. paratuberculosis in both MLN and intestinal tissue samples. PCR also confirmed M. bovis in 5.8% of cattle and 5% of buffalo MLN and intestinal tissues. PCR positive tissue samples for M. avium subsp. paratuberculosis were from those animals which were emaciated, having diarrhea, and severe gross lesions. AFB were also detected in tissue scraping smears of these animals. It is concluded that infection by various mycobacterium species can be differentiated by PCR, which is not possible by acid-fast staining technique.  相似文献

A pot experiment was conducted to evaluate the role of glycinebetaine (GB) in chromium (Cr) tolerance in mung bean (Vigna radiata L.) grown in Cr-stressed soil. Three concentrations of Cr (0, 250 and 500 µM) were tested with three (0, 50 and 100 mM) concentrations of foliar-applied GB. Cr alone led to a significant decrease in plant growth, biomass, and concentrations of chlorophyll a, b and carotenoids. Cr concentration and electrolyte leakage significantly increased in plants with increasing Cr levels in the soil. Lower Cr stress enhanced the activities of superoxide dismutase (SOD), peroxidases (POD) and catalase (CAT), while higher Cr concentrations decreased the activities of these enzymes. Foliar application of GB successfully alleviated toxic effects of Cr on mung bean and increased plant growth, biomass and chlorophyll contents under Cr stress. GB application reduced Cr accumulation and electrolyte leakage in plants and enhanced the activities of antioxidant enzymes in both shoots and roots as compared with Cr treatments alone. These findings suggest that foliar-applied GB alleviated Cr-induced oxidative stress in mung bean by reducing Cr uptake. The protective effect of GB against Cr stress varies with the concentrations of GB and Cr stress applied. Thus, further studies are still needed to specify the concentrations of GB required for detoxification of specific Cr concentrations under various climatic conditions.  相似文献

Two mustard (Brassica juncea L. Czern and Coss.) cultivars, Pusa Jai Kisan and SS2 differing in cadmium (Cd) tolerance were treated with 0, 25 and 50 umol L-1 Cd to study the physiological basis of difference in Cd tolerance. Cultivar SS2 (Cd sensitive) accumulated greater Cd in leaves than Pusa Jai Kisan (Cd tolerant). Further, SS2 also exhibited higher contents of thiobarbituric acid reactive substances (TBARS) and H2O2 and electrolyte leakage. However, the activities of antioxidant enzymes, catalase (CAT), ascorbate peroxidase (APX), dehydroascorbate reduetase (DHAR), and glutathione reductase (GR) were higher in Pusa Jai Kisan than those in SS2. Contrarily, the activity of superoxide dismutase (SOD) was higher in SS2 than that in Pusa Jai Kisan and was the greatest at 25 umol L-1 Cd. Treatment of 25 umol L-1 Cd induced the maximum activity of enzymes. However, the activity of GR increased up to 50 umol L-1 Cd in both the cultivars. The non-enzymatic antioxidants ascorbate (AsA) and glutathione (GSH) were higher in Pusa Jai Kisan than that in SS2, whereas dehydroascorbate (DHA) and oxidized glutathione (GSSG) were higher in SS2. Photosynthesis and growth were adversely and maximally decreased by 50 umol L-1 Cd treatment in both the cultivars, but SS2 exhibited greater reductions. The protection of photosynthesis and growth and lesser reduction in Pusa Jai Kisan were associated with its capacity to restrict accumulation of Cd in leaves resulting in lower level of TBARS and H2O2 and electrolyte leakage. Moreover, Pusa Jai Kisan exhibited efficient antioxidant metabolism for removal of Cd-induced reactive oxygen species.  相似文献

Poor quality of sandy loam soils ?is the main reason for low crop yield. Improvement of physicochemical properties of these soils is very challenging. Addition of organic sources may improve the soil properties. Therefore, this study investigated the adequacy of poultry-manure-compost (PMC) and pressmud-compost (PrMC) at 0 (control), 2, 4, 6, 8, and 10 t ha?1 for improving the physicochemical properties of sandy loam soil and maize performance. An increasing trend in most soil and crop traits was seen with increasing compost levels. For 10 t PMC ha?1, soil inorganic-N (512%), organic-carbon (78%), and water-holding capacity (65.36%) improved maximum. This resulted in the maximum mean crop growth rate (43.85%), stover yield (94%), grain protein (21%), and nitrogen use efficiency (30.6 kg kg?1). Contrarily, grain oil (?7%) was lowest at 10 t PMC ha?1. Consequently, 10 t PMC ha?1 could be much effective to improve the physicochemical properties of sandy loam soils and maize performance.  相似文献

It is crucial to advance the understanding of the soil carbon dioxide (CO2) flux and environmental factors for a better comprehension of carbon dynamics in subtropical ecosystems. Red soil, one of the typical agricultural soils in subtropical China, plays important roles in the global carbon budget due to their large potential to sequester C and replenish atmospheric C through soil CO2 flux. We examined the relationship between soil CO2 flux and environmental determinants in four different land use types of subtropical red soil-paddy (P), orchard (O), woodland (W) and upland (U) using static closed chamber method. Objectives were to evaluate the relationship of soil temperature, water-filled pore space (WFPS), and dissolved organic carbon (DOC) with the soil CO2 flux. Soil CO2 fluxes were measured on each site about every 14 days between 09:00 and 11:00 a.m. during 14-July 2004 to 25-April 2007 at the experimental station of Heshengqiao at Xianning, Hubei, China. Soil CO2 fluxes revealed seasonal fluctuations, with the tendency that maximum values occurred in summer, minimum in winter and intermediate values in spring and autumn except for paddy soil when it was submerged. Further, significant differences in soil CO2 fluxes were observed among the four soils, following the order of P > O > U  W. Average soil CO2 fluxes were estimated as 901 ± 114, 727 ± 55, 554 ± 22 and 533 ± 27 (±S.D.) g CO2 m−2 year−1 in paddy, orchard, upland and woodland soils, respectively. Variations in soil CO2 flux were related to soil temperature, WFPS, and dissolved organic carbon with a combined R2 of 0.49–0.75. Soil temperature was an important variable controlling 26–59% of soil CO2 flux variability. The interaction of soil temperature and WFPS could explain 31–60% of soil CO2 flux variations for all the land use types. We conclude that soil CO2 flux from red soil is under environmental controls, soil temperature being the main variable, which interact with WFPS and DOC to control the supply of readily mineralizable substrates.  相似文献

A field experiment was carried out to evaluate the feasibility of inoculating rice seedlings with biofertilizers (Azospirillum and Trichoderma) in order to reduce the use of chemical inorganic nitrogen (N) fertilizer on rice variety BU Dhan 1. The plant performances were better when 25% less inorganic N was applied with Trichoderma and combined application of Trichoderma and Azospirillum. Plants contained the highest chlorophyll concentrations when they were treated with 75% N + Trichoderma. Considering the yield attributes, 75% N + Trichoderma and 75% N + Trichoderma + Azospirillum performed similar to the control. The grain yield of rice was similar to the recommended dose even with 25% less N application. Application of Trichoderma resulted higher yield, followed by combined application with Azospirillum. Results revealed the greater scope of applying biofertilizer (Trichoderma) to supplement chemical N fertilizer with optimum yield of rice.  相似文献

Bioavailability and mobility of lead (Pb) in soils depend upon their partitioning between solution-solid phases and their further fractionation and redistribution among different solid-phase components. However, the dynamics of Pb in salt-affected (saline–sodic) Pb-contaminated (polluted) paddy soil need more exploration particularly under the influence of application of amendments at varying hydrological regimes and residence time. In this context, an incubation study was conducted to investigate the effect of application of three inorganic amendments (gypsum, rock phosphate and diammonium phosphate) on Pb fractions at two soil moisture regimes (flooding regime and 75% field capacity) and two incubation times (after 2 and 30 days) successively in non-saline/sodic and saline–sodic Pb-polluted paddy soils. After applied treatments, the concentration of Pb in five, i.e., exchangeable (F1), carbonate (F2), Fe–Mn oxide (F3), organic matter and sulfide bound (F4) and residual (F5) fractions, was assessed by sequential extraction. The results showed that the Pb spiked in the soils was significantly (P?≤?0.05) transformed from easily extractable (exchangeable and carbonate) fractions into less labile (Fe–Mn oxide, OM–S bound and residual) fractions. Among tested amendments, gypsum performed better in reducing the lability of Pb followed by DAP.  相似文献

BACKGROUND: The spotted bollworm Earias vittella (Fab.) is a serious pest of cotton and okra in Pakistan. Owing to persistent use of insecticides, this pest has developed resistance, especially to pyrethroids. The present studies aimed at determining the extent of resistance to pyrethroid, organophosphorus and new chemical insecticides in Pakistani populations of E. vittella. RESULTS: Field populations of E. vittella were monitored at Multan, Pakistan, from 1999 to 2007 for their resistance against six pyrethroid, four organophosphorus and six new chemical insecticides using a leaf‐dip bioassay. Of the pyrethroids, resistance was generally low to zeta‐cypermethrin and moderate to high or very high to cypermethrin, deltamethrin, esfenvalerate, bifenthrin and lambda‐cyhalothrin. Resistance to organophosphates chlorpyrifos, profenofos, triazophos and phoxim was recorded at very low to low levels. Among new chemicals, E. vittella had no or a very low resistance to spinosad, emamectin benzoate and methoxyfenozide, a very low to low resistance to abamectin, a very low to moderate resistance to indoxacarb and a moderate resistance to chlorfenapyr. CONCLUSION: The results indicate a lack of cross‐resistance between pyrethroid and organophosphorus insecticides in E. vittella. Rotation of insecticides showing no, very low or low resistance, but belonging to different insecticide classes with unrelated modes of action, may prevent or mitigate insecticide resistance in E. vittella. Cotton leaf curl disease (CLCuD) is a major threat to cotton production in Asia and Africa. Using marker-assisted breeding can be the best sustainable approach to tackle CLCuD. Identification of new QTLs in the indigenous cotton germplasm is necessary to combat CLCuD. The current study was designed to construct a genetic linkage map of bi-parental F2:F3 populations developed from highly tolerant MNH-886 and highly susceptible S-12 cotton cultivars. One hundred seven CLCuD-associated simple sequence repeat (SSR) marker alleles were identified as polymorphic and three new QTLs were found on chromosomes C11, C19 and C21. Two QTLs on chromosomes C11 and C19 were detected in both F2 and F3 populations in the region flanked by SSR markers CIR316 and BNL4094, and BNL285 and BNL3348, respectively. Whereas, one QTL on chromosome C21 was detected in the region flanked by SSR markers JESPR158 and JESPR135 in both F2 and F3 generations. The CLCuD-associated QTLs identified in this study can help fine-tune the molecular mapping of the QTLs on the cotton genome against CLCuD.  相似文献
