Stem rot symptoms caused by pectinolytic bacteria of Genus Pectobacterium and Genus Dickeya, which are commonly referred to as blackleg, strongly impact the quality of seed potato production in most European countries. Several biotic and abiotic factors, such as cultivar susceptibility, isolate aggressiveness, mother tuber infection density and a wide range of soil-related and climatic factors have been identified in the literature as having an effect on blackleg development. The aim of this study was to identify which biotic and/or abiotic factors are most critical to the development of blackleg in the field. In Switzerland, the predominant species have belonged to Genus Dickeya as far back as 1992, which is why this study only investigates blackleg symptoms induced by Dickeya isolates. Seven field trials, in which inoculated tubers were planted, were conducted during a 3-year period and the number of blackleg-diseased plants was counted. Multiple regression analysis was used in order to determine the factors that had the greatest impact on two different variables: (i) periods between emergence of the plant and disease outbreak and (ii) overall blackleg incidence throughout the growing season. The results of this analysis have revealed that environmental factors, such as evapotranspiration and soil moisture, explain about half of the variability in the number of days before disease outbreak, and the total number of diseased plants is widely dependent upon cultivar susceptibility and isolate aggressiveness.  相似文献

In managed landscapes, habitat structure is frequently manipulated through the creation of features such as tracks, hedges, and waterways. If predator and prey activity are concentrated around these features, levels of predation may be elevated in these landscapes. This issue is of particular importance when habitat structures are used to attract species of conservation concern. For example, the installation of linear waterways in wet grasslands is a common form of habitat management to benefit breeding waders and wader nests and foraging chicks tend to be aggregated around wet features. If predator activity is also focused around these features, and if their linearity increases the probability of prey being located, then the conservation benefits of this management technique may be eliminated. We explore predator movement in relation to the structure and complexity of linear wet features within a lowland wet grassland landscape. We examine patterns of nest and chick predation in lapwing (Vanellus vanellus) at the whole-site, between-field and within-field scales. Mammalian predators were responsible for the majority of nest predation. However, we found no evidence that mammalian predators used linear wet features disproportionately within the landscape, or that wet feature distribution influenced the probability of nest or chick predation. At the whole-site scale, nest predation rates were significantly higher in areas with greater predator presence and lowest where the number of breeding neighbours was high. Thus, predation levels were influenced by large-scale patterns of predator presence and lapwing density but not by the use of linear wet features as a habitat management tool. Managing predator impacts is therefore likely to require empirical assessments of local predator distribution and abundance in order to target measures effectively.  相似文献

Objectives To determine (1) the common types of canine splenic disorders, and the breeds affected, that are diagnosed by cytopathological and histopathological examination in Sydney, Australia and (2) the accuracy of cytopathological examination compared with histopathological examination for the diagnosis of canine splenic disorders. Design 69 cytopathological and 51 histopathological diagnoses of canine splenic disorders presented to the Veterinary Pathology Diagnostic Services, The University of Sydney during 2006 and 2007 were tabulated and analysed; 17 cases examined both cytopathologically and histopathologically during 2001–07 were also analysed. Results The most common cytopathological diagnoses were benign disorders of growth, vascular disturbances and necrosis (29%), followed by no abnormalities detectable (28%), malignant neoplasms (20%), equivocal diagnoses (20%) and inflammatory disorders (3%). The most common breeds were Kelpie crosses and mixed breeds. The most common histopathological diagnoses were benign disorders of growth, vascular disturbances and necrosis (49%), followed by malignant neoplasms (43%) and inflammatory disorders (8%). The most common breeds were German Shepherd Dogs, Boxers and Maltese Terriers. Cytopathological and histopathological diagnoses were in complete agreement in 59% of cases, partial agreement in 29% and disagreement in 12%. Conclusion Benign disorders of growth, vascular disturbances and necrosis were the most commonly diagnosed canine splenic disorders, both cytopathologically and histopathologically. Kelpie crosses presented most frequently for cytopathological examination. German Shepherd Dogs were the most common breed diagnosed histopathologically with haemangiosarcoma. Although cytopathological and histopathological splenic examinations are complementary for diagnosis, this study has shown a high correlation for complete and partial agreement between the two.  相似文献

Standardisation of cultural practises is one of the primary objectives to make the system ecologically sustainable and economically viable. In this context, the present study was conducted to optimise the time of sowing in relation to newly released wheat varieties under 4–6 year old poplar block plantation. Six widely grown wheat varieties (PBW 502, PBW 343, WH 542, PDW 274, PBW 509 and PBW 373) were intercultivated during three consecutive crop growth seasons at three times of sowing (mid November, late November and mid December) under block plantation of poplar (Populus deltoides Bartr.) clone G-48. The grain yield and nutrient uptake of wheat varieties was higher in open conditions than under the trees. The wheat variety PBW 502 out yielded the rest of wheat varieties over different sowing times. The highest grain yield was recorded when crop was sown during mid November over the 3 years. Thus adoption of PBW 502 under poplar plantation would substantially improve the overall productivity of the system without any additional input cost. All the varieties performed better when sown early compared to one-month delay. The various growth parameters like tiller height and number, spike length and 100 grain weight contributed significantly towards higher grain yield in early sown conditions (mid November) under poplar irrespective of its age of plantation. The nutrient uptake (N, P and K) by wheat straw and grain was higher in early sown crop with longer growth span than the late sown crop. The higher quantity of N, P and K was removed from soil by PBW 509, PBW 373 and PBW 343, respectively.  相似文献

Water-saving technologies that increase water productivity of rice are urgently needed to help farmers to cope with irrigation water scarcity. This study tested the ability of the ORYZA2000 model to simulate the effects of water management on rice growth, yield, water productivity (WP), components of the water balance, and soil water dynamics in north-west India. The model performed well as indicated by good agreement between simulated and measured values of grain yield, biomass, LAI, water balance components and soil water tension, for irrigation thresholds ranging from continuous flooding (CF) to 70 kPa soil water tension.Using weather data for 40 different rice seasons (1970-2009) at Ludhiana in Punjab, India, the model predicted that there is always some yield penalty when moving from CF to alternate wetting and drying (AWD). With an irrigation threshold of 10 kPa, the average yield penalty was 0.8 t ha−1 (9%) compared with CF, with 65% irrigation water saving, which increased to 79% at 70 kPa with a yield penalty of 25%. The irrigation water saving was primarily due to less drainage beyond the root zone with AWD compared to CF, with only a small reduction in evapotranspiration (ET) (mean 60 mm).There were tradeoffs between yield, irrigation amount and various measures of WP. While yield was maximum with CF, water productivity with respect to ET (WPET) was maximum (1.7 g kg−1) for irrigation thresholds of 0 (CF) to 20 kPa, and irrigation water productivity (WPI) increased to a maximum plateau (1.3 g kg−1) at thresholds ≥30 kPa.Because of the possibility of plant stress at critical stages known to be sensitive to water deficit (panicle initiation (PI) and flowering (FL)), treatments with additional irrigations were superimposed for 2 weeks at one or both of these stages within the 10, 20 and 30 kPa AWD treatments. Ponding for two weeks at FL was more effective in reducing the yield penalty with AWD than ponding at PI, but the biggest improvement was with ponding at both stages. This reduced the average yield loss from 9% (0.8 t ha−1) to 5% (0.5 t ha−1) for AWD with thresholds of 10 and 20 kPa. However, maximum WPI (1.1 g kg−1) was achieved with an irrigation threshold of 20 kPa combined with more frequent irrigation at FL only, but with a greater yield penalty (8%). Thus the optimum irrigation schedule depends on whether the objective is to maximise yield, WPET or WPI, which depends on whether land or water are most limiting. Furthermore, the optimum irrigation schedule to meet the short term needs of individual farmers may differ from that needed for sustainable water resource management.  相似文献

Labour and water scarcity in north west India are driving researchers and farmers to find alternative management strategies that will increase water productivity and reduce labour requirement while maintaining or increasing land productivity. A field experiment was done in Punjab, India, in 2008 and 2009 to compare water balance components and water productivity of dry seeded rice (DSR) and puddled transplanted rice (PTR). There were four irrigation schedules based on soil water tension (SWT) ranging from saturation (daily irrigation) to alternate wetting drying (AWD) with irrigation thresholds of 20, 40 and 70 kPa at 18–20 cm soil depth. There were large and significant declines in irrigation water input with AWD compared to daily irrigation in both establishment methods. The irrigation water savings were mainly due to reduced deep drainage, seepage and runoff, and to reduced ET in DSR. Within each irrigation treatment, deep drainage was much higher in DSR than in PTR, and more so in the second year (i.e. after 2 years without puddling). The irrigation input to daily irrigated DSR was similar to or higher than to daily irrigated PTR. However, within each AWD treatment, the irrigation input to DSR was less than to PTR, due to reduced seepage and runoff, mainly because all PTR treatments were continuously flooded for 2 weeks after transplanting. There was 30–50% irrigation water saving in DSR-20 kPa compared with PTR-20 kPa due to reduced seepage and runoff, which more than compensated for the increased deep drainage in DSR. Yields of PTR and DSR with daily irrigation and a 20 kPa irrigation threshold were similar each year. Thus irrigation and input water productivities (WPI and WPI+R) were highest with the 20 kPa irrigation threshold, and WPI of DSR-20 kPa was 30–50% higher than of PTR-20 kPa. There was a consistent trend for declining ET with decreasing frequency of irrigation, but there was no effect of establishment method on ET apart from higher ET in DSR than PTR with daily irrigation. Water productivity with respect to ET (WPET) was highest with a 20 kPa irrigation threshold, with similar values for DSR and PTR. An irrigation threshold of 20 kPa was the optimum in terms of maximising grain yield, WPI and WPI+R for both PTR and DSR. Dry seeded rice with the 20 kPa threshold outperformed PTR-20 kPa in terms of WPI through maintaining yield while reducing irrigation input by 30–50%.  相似文献

Histone deacetylase inhibitors (HDACis) can change the histone acetylation and significantly enhance the developmental competence of the pre‐implantation SCNT embryo. To select a proper histone deacetylase inhibitor to improve the success rate and potentially developmental ability of handmade cloning (HMC) embryos of miniature porcine, we compared the effect of two histone deacetylase inhibitors (SAHA vs. VPA) on HMC embryo development, their histone acetylation level and the expression level of relevant genes. The blastocyst rate and number of blastocyst cells of HMC embryos treated with SAHA (SAHA‐HMC) or VPA (VPA‐HMC) were significantly higher than those of control (Control‐HMC), respectively, but there were no significant difference between SAHA‐HMC and VPA‐HMC groups. In addition, the acetylation level (AcH4K8) of Control‐HMC and VPA‐HMC embryos at the blastocyst stage, respectively, was significantly lower than that of in vitro fertilized (IVF) and SAHA‐HMC embryos. However, the acetylation H4K8 of the blastocysts had no significant difference between SAHA‐HMC and the IVF groups. The SAHA‐HMC blastocysts indicated comparative expression levels of Oct4 and HDAC1 (histone deacetyltransferase gene) with those of IVF blastocysts. In contrast, the expression levels of Oct4 were lower and those of HDAC1 were higher in the VPA‐HMC and Control‐HMC blastocysts, respectively, compared to those of the IVF blastocysts. Our results demonstrated that the HMC embryos treated by SAHA could promote the pre‐implantation development and increase the levels of histone H4K8 acetylation and the expression of the OCT4 gene, yet decrease the expression of the HDAC1 gene to the comparable level of the IVF embryos. Our results proved that SAHA may be a better histone deacetylase inhibitor for porcine HMC compared to VPA, and furthermore, it may indicate that SAHA can effectively correct the abnormal histone acetylation during the HMC embryo development and subsequently improve the full‐term developmental potential of the HMC embryos after embryo transplantation.  相似文献

The sperm of European eel shows a high density and the time of spermatozoa motility is very short after activation with sea water. These characteristics make difficult the sperm handling and its quality assessment. Several diluents were previously described for the Japanese eel obtaining over 3 weeks’ conservation times under refrigeration, but they rendered bad results in the European species. In the present study, several diluents were developed taking as basis the P1 medium, and using different dilution ratios (1 : 50, 1 : 100) and two pH (6.5, 8.5). The effect of the addition of bovine serum albumin (BSA, 2% w/v) was also evaluated. At 24 h, undiluted samples already showed significant lower motility and viability than sperm samples diluted in the different media. The results for diluents with pH 6.5 and 8.5 were different. Spermatozoa diluted in media at pH 6.5 cannot be activated at 24 h, while samples diluted in the diluents with pH 8.5 and added with BSA did not show significant differences with respect to the fresh sperm motility until 48 h. The viability (percentage of alive cells) did not show differences until 1 week, independent of the dilution ratio. After 1 week, the motility was approximately 30% in the media containing BSA, which presented no differences for head size of the spermatozoa (perimeter and area) until 72 h and 1 week, respectively. In conclusion, the combination of one medium having similar physico‐chemical characteristics to the seminal plasma, including pH 8.5, and supplemented with BSA can be used in different dilution ratios for the sperm’s short‐term storage, preserving its motility capacity.  相似文献

The sperm‐mediated gene transfer method is applicable to transgenesis in many species that use spermatozoa for reproduction recently, which has been shown various results. In the current study, we show that transgenic porcine embryos can be efficiently produced by employing a simple transfection method that uses magnetic nanoparticles (MNPs). The complexes formed between plasmid DNA and MNPs were bounded on ejaculated boar spermatozoa at a higher efficiency compared to methods using DNA alone or lipofection. Using confocal microscopy, rhodamine fluorophore‐labelled MNPs were detected on external surfaces of the spermatozoa membrane, which were bounded on zona pellucida of in vitro maturated oocyte during in vitro fertilization. Electron microscopy revealed that clusters of MNPs were detected in inside of plasma membrane and nucleus of the spermatozoa head. Additionally, we found that magnetofected boar spermatozoa could be fertilized with oocytes in vitro and that the resulting gene of green fluorescent protein was detected in fertilized eggs by genomic PCR analysis. Taken together, these results suggest that MNPs can be used to efficiently introduce a transgene into embryo via spermatozoa.  相似文献
