The lacLM genes from Lactobacillus sakei Lb790, encoding a heterodimeric β-galactosidase that belongs to glycoside hydrolase family GH2, were cloned and heterologously expressed in Escherichia coli . Subsequently, the recombinant β-galactosidase LacLM was purified to apparent homogeneity and characterized. The enzyme is a β-galactosidase with narrow substrate specificity because o-nitrophenyl-β-D-galactopyranoside (oNPG) was efficiently hydrolyzed, whereas various structurally related oNP analogues were not. The K(m) and k(cat) values for oNPG and lactose were 0.6 mM and 180 s(-1) and 20 mM and 43 s(-1), respectively. The enzyme is inhibited competitively by its two end-products D-galactose and D-glucose (K(i) values of 180 and 475 mM, respectively). As judged by the ratio of the inhibition constant to the Michaelis constant, K(i)/K(m), this inhibition is only very moderate and much less pronounced than for other microbial β-galactosidases. β-Galactosidase from L. sakei possesses high transgalactosylation activity and was used for the synthesis of galacto-oligosaccharides (GalOS), employing lactose at a concentration of 215 g/L. The maximum GalOS yield was 41% (w/w) of total sugars at 77% lactose conversion and contained mainly non-lactose disaccharides, trisaccharides, and tetrasaccharides with approximately 38, 57, and 5% of total GalOS formed, respectively. The enzyme showed a strong preference for the formation of β-(1→6)-linked transgalactosylation products, whereas β-(1→3)-linked compounds were formed to a lesser extent and β-(1→4)-linked reaction products could not be detected. 相似文献

Asian turtles are overharvested for food, medicinal purposes and pets, and most species are now at risk of extinction (‘Asian turtle crisis’). As a result of high levels of trade, some species of Asian turtles are known to science mostly or only from animals found in trade. The Vietnamese box turtle (Cuora picturata) was described in 1998 and is known only from markets. This species is likely to be highly threatened, and identifying the origin of this “market species” is imperative if it is to be conserved. We used evidence from phylogeny, biogeography and trade patterns to focus field searches for this species, and in July 2010–January 2011 found it in the wild at three localities on the Langbian Plateau of southern Vietnam. This discovery provides the first opportunity to conserve the Vietnamese box turtle, and provides hope for determining the wild origin of other rare Asian turtles that remain known only from commercial trade. 相似文献

We have shown in pigs that oocytes denuded of cumulus cells at 24 h of in vitro maturation culture and subsequently matured for a total of 46 h (DO24 oocytes) have lower cytoplasmic maturity than those matured with cumulus cells for 46 h and then denuded (DO46 oocytes). In the present study, DO24 zona-free oocytes were fused with one (1C) or two (2C) cytoplasmic fragments produced by serial centrifugation ("centri-fusion") of DO46 oocytes (DO24+1C and DO24+2C oocytes, respectively). Groups of (1) DO46 (a control), (2) DO24, (3) DO24+1C and (4) DO24+2C oocytes were partheno-activated by an electrical pulse or fertilized in vitro and subsequently cultured for 6 days. In the fused groups, female pronucleus (FPN) formation rates were higher than that in the DO24 group after parthenogenetic activation (PA); however, the blastocyst rates were intermediate between those of the control and DO24 groups. After in vitro fertilization, the male pronucleus (MPN) formation rates in the fused groups were similar to that in the control group and higher than that in the DO24 group; the normal fertilization rate in the DO24+2C group was higher than that in the DO24 group and similar to that in the control group, resulting in significantly higher blastocyst rates in the DO24+2C and control groups than that in the DO24 group. These results suggest that centri-fusion using ooplasm from fully matured DO46 oocytes can offer a potentially novel approach for restoration of cytoplasmic maturity to oocytes with low developmental ability and subsequent improvement of fertilization and developmental competence. 相似文献

Since BSE testing of slaughtered cattle is obligatory in Japan, storage of ovaries at 15-20 C overnight in phosphate buffered saline has become a routine protocol in in vitro production (IVP) of cattle embryos. Ovary storage is known to reduce developmental competence of oocytes; however, its effects on oocyte gene expression have not been clarified yet. This study compared oocytes collected from stored slaughterhouse-derived ovaries with those collected by Ovum Pick-Up (OPU) in terms of the expression of 20 selected genes to determine if ovary storage affects cellular processes at the molecular level. Expression of mRNA in oocytes was assayed before and after in vitro maturation (IVM) by real-time quantitative PCR. Maternal mRNA levels of genes were investigated in 2-cell stage embryos obtained from slaughterhouse oocytes to assess their roles for blastocyst formation. In immature OPU oocytes, genes related to metabolism (GAPDH), transporters (GLUT8, ATP1A1) and stress resistance protein (HSP70) showed significantly higher expression compared with oocytes derived from stored ovaries. During IVM, the expression of GDF9, GLUT8, CTNNB1 and PMSB1 was significantly decreased irrespective of oocyte source. Two-cell stage embryos cleaving at 22-25 h after in vitro fertilization (IVF) showed a significantly higher blastocyst formation rate and ATP1A1 gene expression level compared with those cleaving at 27-30 h after IVF. Our results reveal that storage of ovaries alters mRNA levels in oocytes. Correlation of Na/K ATPase ATP1A1 expression in IVP embryos at the 2-cell and 8-cell stages with their developmental ability to the blastocyst stage may suggest the importance of maternal mRNA of this gene during blastulation in embryos derived from slaughterhouse oocytes. 相似文献

Abstract. The aim of this study was to investigate the effects of water submergence depth on radial oxygen loss (ROL), soil solution chemistry and rice growth performance in acid sulphate soils in southern Vietnam. ROL was measured in a solution culture. In a separate pot experiment the impact of water submergence depth on rice growth and soil solution chemistry was studied. Three submergence depths were used in the two experiments (5, 10 and 15 cm). ROL declined with submergence depth and was significantly greater in young roots (with no root hairs) than in older roots. In the pot experiment rice growth and soil solution chemistry were clearly affected by the submergence depth. During the first crop at 5 cm submergence, there was a significantly higher yield and a higher oxidation state (pe+pH) compared to 10 or 15 cm submergence. The Fe concentration was significantly greater at the 5 cm depth compared to the 10 or 15 cm depth. SO4 2– reduction was delayed at the 5 cm depth. Rice yield was c. 25% less at the 15 cm than at the 5 cm depth. During a second crop, there was a substantial SO4 2- reduction and H2 S formation and almost no significant effects of submergence depth on either soil solution chemistry or crop yield. In a field experiment with a dry-season rice crop, yield and Fe, Al and SO4 2– concentrations were higher at a shallow submergence depth than at greater depths in the same field, showing similar depth trends to those found during the first crop in the pot experiment. Farmers should be advised to use a shallow submergence depth and, if possible, avoid deep-rooted rice varieties. A conceptual model is suggested, which summarizes the relationships between ROL and soil solution chemistry. 相似文献

Dermanyssus gallinae, the poultry red mite, is an obligatory
blood-sucking ectoparasite. The genetic diversity of D. gallinae has been
examined in some countries, but so far not in Asian countries. Here, we sequenced a part
of the mitochondrial cytochrome oxidase subunit I (COI) and16S rRNA genes and nuclear
internal transcribed spacers (ITS) region in 239 mite samples collected from 40
prefectures throughout Japan. The COI and 16S rRNA nucleotide sequences were classified
into 28 and 26 haplotypes, respectively. In phylogenetic trees, the haplotypes clustered
into 2 haplogroups corresponding to haplogroups A and B, which were previously reported.
Haplogroups A and B were further subdivided into sub-haplogroups AJ1 and AJ2, and BJ1 and
BJ2, respectively. In both trees, the sequences of haplotypes in AJ1 and BJ2 were
relatively distant from those reported in other countries, while some sequences in AJ2 and
BJ1 were identical to those in Europe. In addition, the ITS sequences were classified into
two sequences, and both sequences were closely related to the sequences found in European
countries. These findings indicate a possibility of international oversea transmission of

Long-term nutrient management practices have lasting effects on the geographic distribution of soil microorganisms, loci of enhanced activity, and non-mobile nutrients such as phosphorus (P) to ultimately influence nutrient use efficiency by crops and edge-of-field losses. We determined the distribution of soil microbial biomass, phosphomonoesterases' activity, and P forms in a 10-ha no-till field that received annual additions of dairy manure at 0, 15, and 30 kg P ha?1 at the field scale for 16 consecutive years. The spatial structure of soil microbial indices and extractable P fractions were characterized based on their semivariance distributions for each manure treatment. The buildup in soil P occurred, although replacement was done at a crop removal rate. Manure additions resulted in overall mean increases in total labile P of 73 % and 156 % and alterations in the soil microbial ecosystem that depended on manure inorganic-to-enzyme-labile P composition in soils treated with 15 and 30 kg P ha?1, respectively. Distinct clusters of phosphate- and enzyme-labile organic P were observed within manure treatments, where accumulation of the latter forms was associated with high soil microbial biomass C and reduced acid phosphomonoesterase activity. The geographic variability highlighted the critical need for improving methods of field-scale application of manure and non-mobile nutrients. In addition, current soil testing methods that depend upon composite representative samples for estimating microbiological parameters linked to nutrient turnover and P requirements for optimal crop production should be modified to include site-specificity in sampling and interpretation approaches. 相似文献

Biofertilizer research for rice in Vietnam has focused on the isolation and selection of strains that can fix nitrogen, solubilize inorganic phosphates, stimulate plant growth, and breakdown soil organic matter. This paper assesses the consistent positive effect of BioGro on grain yield and agronomic parameters, including the rates and times for its application, the need for continued inoculation of crops grown in the same site, varietal differences, and nitrogen (N), phosphorus (P), and potassium (K) combinations on the effectiveness of BioGro. The commercial biofertilizer, BioGro, consists of four strains, one formerly considered as nitrogen fixing, Pseudomonas fluorescens, a soil yeast strain, Candida tropicalis is P-solubilizing, and two other bacilli, Bacillus amyloliquefaciens and Bacillus subtilis, potentially breaking down cellulose, protein, and starch. All four strains contribute to plant growth promoting rhizobacteria (PGPR) effect as shown by enhanced root growth. BioGro can be produced in local factories providing there is technical backup in the supply of starter culture and quality control of the final product. 相似文献

Thermal noise spectroscopy was used to measure the density and temperature of the main (cold) electron plasma population during 2 hours (1.5x10(5) kilometers perpendicular to the tail axis) around the point of closest approach of the International Cometary Explorer (ICE) to Comet Giacobini-Zinner. The time resolution was 18 seconds (370 kilometers) in the plasma tail and 54 seconds (1100 kilometers) elsewhere. Near the tail axis, the maximum plasma density was 670 per cubic centimeter and the temperature slightly above 1 electron volt. Away from the axis, the plasma density dropped to 100 per cubic centimeter (temperature, 2x 10(4) K) over 2000 kilometers, then decreased to 10 (1.5x 10(5)K) over 15,000 kilometers; outside that region (plasma tail), the density fluctuated between 10 and 30 per cubic centimeter and the temperature between 1x 10(5) and 4 x10(5) K. The relative density of the hot population rarely exceeded a few percent. The tail was highly asymmetrical and showed much structure. On the other antenna, shot noise was recorded from the plasma particle impacts on the spacecraft body. No evidence was found of grain impacts on the antennas or spacecraft in the plasma tail. This yields an upper limit for the dust flux or particle mass, indicating either fluxes or masses in the tail smaller than implied by the models or an anomalous grain structure. This seems to support earlier suggestions that these grains are featherlike. Outside the tail, and particularly near 10(5) kilometers from its axis, impulsive noises indicating plasma turbulence were observed. 相似文献

The Unified Radio and Plasma Wave (URAP) experiment has produced new observations of the Jupiter environment, owing to the unique capabilities of the instrument and the traversal of high Jovian latitudes. Broad-band continuum radio emission from Jupiter and in situ plasma waves have proved valuable in delineating the magnetospheric boundaries. Simultaneous measurements of electric and magnetic wave fields have yielded new evidence of whistler-mode radiation within the magnetosphere. Observations of aurorallike hiss provided evidence of a Jovian cusp. The source direction and polarization capabilities of URAP have demonstrated that the outer region of the lo plasma torus supported at least five separate radio sources that reoccurred during successive rotations with a measurable corotation lag. Thermal noise measurements of the lo torus densities yielded values in the densest portion that are similar to models suggested on the basis of Voyager observations of 13 years ago. The URAP measurements also suggest complex beaming and polarization characteristics of Jovian radio components. In addition, a new class of kilometer-wavelength striated Jovian bursts has been observed. 相似文献
