A simple, rapid, and highly sensitive ion pair liquid chromatographic method for the determination of albendazole sulfoxide, albendazole 2-aminosulfone, and albendazole sulfone, which constitute the marker residue of albendazole in animal tissues (muscle, fat, liver, and kidney), is described. Tissue samples were extracted with acetonitrile, and the extracts were partitioned, as ion pairs, into dichloromethane. The organic layer was evaporated to dryness, and the residue was reconstituted in phosphate buffer and extracted with ethyl acetate. Separation was carried out isocratically with a mobile phase containing both positively and negatively charged pairing ions. Detection was performed fluorometrically, with excitation and emission wavelengths set at 290 and 320 nm, respectively. Overall recoveries were better than 76%, and the overall relative standard deviation was better than 7.3% in all tissues examined. The limits of quantification were 20, 1, and 0.5 ng/g for sulfoxide, 2-aminosulfone, and sulfone metabolites, respectively. The method was successfully applied to determine residues in tissues of two sheep orally administered an albendazole formulation.  相似文献

The thermal stability of anthocyanin extract isolated from the dry calyces of Hibiscus sabdariffa L. was studied over the temperature range 60-90 degrees C in aqueous solutions in the presence or absence of beta-cyclodextrin (beta-CD). The results indicated that the thermal degradation of anthocyanins followed first-order reaction kinetics. The temperature-dependent degradation was adequately modeled by the Arrhenius equation, and the activation energy for the degradation of H. sabdariffa L. anthocyanins during heating was found to be approximately 54 kJ/mol. In the presence of beta-CD, anthocyanins degraded at a decreased rate, evidently due to their complexation with beta-CD, having the same activation energy. The formation of complexes in solution was confirmed by nuclear magnetic resonance studies of beta-CD solutions in the presence of the extract. Moreover, differential scanning calorimetry revealed that the inclusion complex of H. sabdariffa L. extract with beta-CD in the solid state was more stable against oxidation as compared to the free extract, as the complex remained intact at temperatures 100-250 degrees C where the free extract was oxidized. The results obtained clearly indicated that the presence of beta-CD improved the thermal stability of nutraceutical antioxidants present in H. sabdariffa L. extract, both in solution and in solid state.  相似文献

The total lipid content and fatty acid composition of the muscle tissue of tilapia (Oreochromis niloticus) and of hybrid red tilapia (Oreochromis sp.) from different culture systems and from the natural and artificial environment of Thailand were compared. Wild fish and fish reared under the most extensive conditions had a more favorable fatty acid profile for human consumption as they contained higher proportions of 18:3n-3, 20:5n-3, and 22:6n-3, higher n-3/n-6 PUFA ratios, and lower proportions of 18:2n-6. The muscle tissue of intensively cultured fish was characterized by increased fat deposition that was mainly saturated and monounsaturated fatty acids and 18:2n-6. It is undesirable for the consumer to reduce 20:5n-3 and 22:6n-3 in farmed tilapia and replace them with elevated 18:2n-6. It is recommended that the amount of 18:2n-6 in the feed of the intensively reared tilapia should be reduced by substituting vegetable oils rich in 18:2n-6 with oils rich in 18:1n-9 and/or 18:3n-3.  相似文献

The degradation and adsorption of the organophosphorus nematicide fosthiazate were investigated in nine soils with various physicochemical and biological characteristics. Fosthiazate was more persistent in acidic soils (pH <6), with half-life (t1/2) values ranging from 53.3 to 57.7 days, compared to soils with higher pH (pH >7), with t1/2 ranging from 14.1 to 20.7 days. Application of antibacterial and antifungal antibiotics to soil samples resulted in a significant inhibition of fosthiazate degradation only in two of the three acidic soils. In contrast, soil autoclaving resulted in doubling the t1/2 of fosthiazate in all studied soils, suggesting that both microbial and abiotic processes contribute to fosthiazate degradation. Statistical analysis indicated a significant negative correlation (P < 0.01) between soil pH and t1/2. Fosthiazate was generally weakly adsorbed with Freundlich adsorption coefficient (Kf) values ranging from 1.23 to 2.74 mL/g. Fosthiazate concentration was strongly correlated with soil organic matter content with higher Kf values in soils with higher organic matter content (P < 0.01). The mean t1/2 and Kf values derived from the laboratory studies were used to parametrize the FOCUS groundwater (GW) models PRZM, PELMO, PEARL, and MACRO for nematicide application in potato and tomato crops. Predicted environmental concentrations produced by the models PEARL and MACRO suggested a potential risk for GW in several scenarios, unlike PELMO and PRZM, which predicted low risk for GW. These findings suggest that the environmental fate of fosthiazate is strongly influenced by soil characteristics and that this nematicide should be used with care in acidic, light soils with low organic matter content.  相似文献

The protein kinase HOG1 in high osmolarity glycerol pathway is one of mitogen-activated protein kinases and plays an important role in fungi cellular responses to external stress and stimuli. In this study, AlHOG1, a yeast HOG1 homolog, was isolated from tobacco pathogenic fungus Alternaria longipes and its disrupted mutants were generated using homologous recombination. Compared with the wild-type strain, the mutants grew slower under Minimal-Media, produced less conidia on Potato-Dextrose-Agar and showed a retardation phenotype in conidial germination rate. Colony color observation indicated that AlHOG1 most likely negatively regulated melanin production under nutrients-deficient or osmotic stress. The same action mode of AlHOG1 on hyphal growth in response to certain carbon or nitrogen sources was speculated. Additionally, the disrupted strains were sensitive to osmotic and oxidative stress, and showed increased resistance to dicarboximide, phenylpyrrole and carbendazim fungicides. Meanwhile, results from the comparative analysis between the AlHOG1- and the AlHK1 (encoding a group III histidine kinase)-disrupted mutants showed that AlHOG1 mediated-pathway was partly regulated by the upstream AlHK1 kinase for cellular resistance to salts osmotic stress and that AlHK1 was a primary regulator for cellular sugar and fungicides stress adaptation and likely modulated both AlHOG1-mediated pathway and other unknown mechanisms. Noteworthily, AlHOG1 but not AlHK1 was confirmed to be associated with high concentration glycerol adaptation, which is the first report to our knowledge. Overall, our results indicated that AlHOG1 was involved in multiple physiological processes in A. longipes, including mycelial and conidial development, melanin production, nutrients sensing and multi-stress responses.  相似文献

Based on macroecological data, we test the hypothesis whether European tree species of temperate and boreal distribution maintain their water and nutrient supply in the more arid southern margin of their distribution range by shifting to more fertile soils with higher water storage than in their humid core distribution range (cf. soil compensatory effects). To answer this question, we gathered a large dataset with more than 200,000 plots that we related to summer aridity (SA), derived from WorldClim data, as well as soil available water capacity (AWC) and soil nutrient status, derived from the European soil database. The soil compensatory effects on tree species distribution were tested through generalized additive models. The hypothesis of soil compensatory effects on tree species distribution under limiting aridity was supported in terms of statistical significance and plausibility. Compared to a bioclimatic baseline model, inclusion of soil variables systematically improved the models’ goodness of fit. However, the relevance measured as the gain in predictive performance was small, with largest improvements for P. sylvestris, Q. petraea and A. alba. All studied species, except P. sylvestris, preferred high AWC under high SA. For F. sylvatica, P. abies and Q. petraea, the compensatory effect of soil AWC under high SA was even more pronounced on acidic soils. Soil compensatory effects might have decisive implications for tree species redistribution and forest management strategies under anthropogenic climate change. Therefore, soil compensatory effects deserve more intensive investigation, ideally, in studies combining different spatial scales to reduce the uncertainty associated with the precision of soil information.  相似文献
