A potentiometric sensor system based on potassium ion-selective electrodes was developed for agricultural purposes. Sensors were built using PVC ion-selective membranes over an inner solid contact prepared with graphite-epoxy composites. A copper plate was used as a reference electrode. A two-stage electronic circuit composed of current and voltage amplifiers was designed to interface the sensors to a distributed data acquisition system. Three ion-selective sensors and three off-the-shelf temperature sensors and their associated circuits were mounted in a PVC tube to set up a soil probe. The electronic controls were placed in an airtight box fixed at the upper part of the probe. The system was evaluated in the field, where the sensors presented sensibility within the range of 69-71 mV dec(-)(1). Extracts of soil samples were analyzed by a current flame photometry approach, and the results, compared with the probe measurements, showed a linear relationship (r (2) = 0.992 and 0.995, respectively, to 5 and 20 cm depths), which implies viability and instrumentation reliability for agricultural applications.  相似文献

Ranked among the top threats to conservation worldwide, infectious disease is of particular concern for wild canids because domestic dogs (Canis familiaris) may serve as sources and reservoirs of infection. On British Columbia's largely undeveloped but rapidly changing central and north coasts, little is known about diseases in wolves (Canis lupus) or other wildlife. However, several threats exist for transfer of diseases among unvaccinated dogs and wolves. To gain baseline data on infectious agents in this area, including those with zoonotic potential, we collected blood and stool samples from 107 dogs in 5 remote communities in May and September 2007. Serology revealed that the dogs had been exposed to canine parvovirus, canine distemper virus, Bordetella bronchiseptica, canine respiratory coronavirus, and Leptospira interrogans. No dogs showed evidence of exposure to Ehrlichia canis, Anaplasma phagocytophilum, Borrelia burgdorferi, Dirofilaria immitis, or Cryptococcus gattii. Of 75 stool samples, 31 contained at least 1 parasitic infection, including Taeniid tapeworms, the nematodes Toxocara canis and Toxascaris leonina, and the protozoans Isospora sp., Giardia sp., Cryptosporidium sp., and Sarcocystis sp. This work provides a sound baseline for future monitoring of infectious agents that could affect dogs, sympatric wild canids, other wildlife, and humans.  相似文献

During growing and flowering seasons for five years, comparisons were made between upper and lower leaf surfaces microflora of ragweed plants grown in three areas: a relatively non-contaminated site, a communal landfill, and a complex contaminated with heavy metals. Results indicated that phyllospheric microbial communities were qualitatively and quantitatively related to metal content in plant leaf, leaf surfaces and metal contaminated ecosystems where the plants grown. Results showed that there was a high positive correlation between phyllospheric microbial population densities and Cd, Ni, and Zn content in ragweed leaves, while it was low in presence of Cu. Comparatively, the correlation was at its highest in the case of aerobic heterotrophic bacterial population densities and more than yeasts and filamentous fungi communities. The most common diversities of metal resistant microorganisms were related to Pantoea agglomerans, Pseudomonas savastanoi, P. putida, Bacillus cereus, Corynebacterium striatum, Cryptococcus and Alternaria.  相似文献

The evolutionarily conserved serine-threonine kinase mammalian target of rapamycin (mTOR) plays a critical role in regulating many pathophysiological processes. Functional characterization of the mTOR signaling pathways, however, has been hampered by the paucity of known substrates. We used large-scale quantitative phosphoproteomics experiments to define the signaling networks downstream of mTORC1 and mTORC2. Characterization of one mTORC1 substrate, the growth factor receptor-bound protein 10 (Grb10), showed that mTORC1-mediated phosphorylation stabilized Grb10, leading to feedback inhibition of the phosphatidylinositol 3-kinase (PI3K) and extracellular signal-regulated, mitogen-activated protein kinase (ERK-MAPK) pathways. Grb10 expression is frequently down-regulated in various cancers, and loss of Grb10 and loss of the well-established tumor suppressor phosphatase PTEN appear to be mutually exclusive events, suggesting that Grb10 might be a tumor suppressor regulated by mTORC1.  相似文献

Bioassay-guided fractionation of the chloroform extract of Centaurea jacea L. afforded the isolation of cirsiliol, apigenin, hispidulin, eupatorin, isokaempferide, axillarin, centaureidin, 6-methoxykaempferol 3-methyl ether, trachelogenin, cnicin, 4'-acetylcnicin and three aliphatic glucose diesters, including the new natural product 1β-isobutanoyl-2-angeloyl-glucose. The structures of the compounds were established on the basis of spectroscopic analyses (UV, MS and NMR). All compounds were isolated for the first time from this species. The compounds were evaluated for their tumour cell growth inhibitory activities on HeLa, MCF-7 and A431 cells. Different types of secondary metabolites (flavonoids, sesquiterpenes) were found to be responsible for the antitumour effects of the extracts; the highest activity was exerted by centaureidin, in addition to moderately active compounds (cirsiliol, isokaempferide, apigenin, hispidulin, cnicin and 4'-acetylcnicin).  相似文献

From the biological point of view the value of autotrophy plant association is determined by the carbon fixation and the carbon cycle. Among the plant associations of Hungary, forest has the largest biological carbon fixation and carbon cycle.In general, the annual water cycle is the key factor in the organic material production of the Hungarian forests. The most intensive water consumption and organic material production take place from May till July, which period is named main water consumption and respectively main growing period. In Hungary the categories of the forest climate are characterized by main tree species and based on the characteristic meteorological data (Jaro and Tatraaljai, 1985). In Hungary the forest area covered by stand is 1,650,000 hm2. Beech forest climate covers 8% of the forest area, hornbeam-oak forest climate covers 22%, sessile oak-Turkey oak forest climate covers 48% and forest steppe climate covers 22%. Partly in the frame of ICP-Forests, the Department of Ecology in the Forest Research Institute carries out long term, complex ecophysiological investigations on several sample plots (so-called basic plots) throughout the whole country. The organic material production (growth), the nutrient and water cycle, the measurements of air pollutants and meteorological parameters, as well as chemical analyses are all part of the investigations. As a comparison the figure of two basic plots (Sopron-Piispokladany) shows the water cycles of a good growing beech stand in beech climate and a weak pedunculate oak stand in forest steppe climate in the hydrological year of 2001-2002. In the Hungarian forest 60%-70% of the precipitation is used for interception, evaporation, and in the vegetation season, for the transpiration both in beech and forest steppe climate. From other point of view, only 30%-40% of the open air precipitation infiltrates into the soil and can be utilized by the forest.  相似文献
