Salmonella hold considerable promise as vaccine delivery vectors for heterologous antigens in chickens. Such vaccines have the potential additional benefit of also controlling Salmonella infection in immunized birds. As a way of selecting attenuated strains with optimal immunogenic potential as antigen delivery vectors, this study screened 20 novel Salmonella Typhimurium vaccine strains, differing in mutations associated with delayed antigen synthesis and delayed attenuation, for their efficacy in controlling colonization by virulent Salmonella Typhimurium, as well as for their persistence in the intestine and the spleen. Marked differences were observed between strains in these characteristics, which provide the basis for selection for further study as vaccine vectors.  相似文献

A feeding study was conducted in the winter 2001 to determine the effects of feeding rotifers (Brachionus plicatilis) enriched with various levels of essential fatty acids on the growth and survival of haddock larvae (Melanogrammus aeglefinus). Rotifer enrichment treatments were: 1) mixed algae, 2) high DHA (docosahexaenoic acid, 22:6n-3), 3) high DHA and EPA (eicosapentaenoic acid, 20:5n-3), and 4) DHA, EPA, and AA (arachidonic acid, 20:4n-6). Larvae were fed rotifers enriched with the different treatments from days 1 to 16 post-hatch. From day 17 until 25 all treatment groups were fed rotifers reared on mixed algae and then weaned onto the International Council for Exploration of the SEA (ICES) Standard Reference Weaning diet (http://allserv.rug.ac.t/aquaculture/rend/rend.htm) over a five day period. The experiment was terminated on day 41 post-hatch. The enrichment treatments affected the fatty acid composition of the rotifers and correlated with the accumulation of these fatty acids in the haddock larvae. However, no significant differences in larval growth or survival to 40 days post hatch were detected, suggesting that all treatments provided the minimal essential fatty acid requirements for haddock.  相似文献

This study was designed to evaluate the effects of dietary supplementation with brewers yeast (Brewtech®) and nucleotides (Optimûn), either singularly or in combination, on red drum growth, body composition, stress responses and possible resistance to Amyloodinium ocellatum infection. Juvenile red drum (~1 g initially) fed practical fish‐meal‐based diets for 6 weeks had similar weight gain regardless of whether the diet was unsupplemented (basal) or supplemented with brewers yeast (2% of diet), nucleotides (Optimûn at 0.2% of diet) or both brewers yeast (2% of diet) and nucleotides (Optimûn at 0.2% of diet). Dietary effects on hepatosomatic index, intraperitoneal fat ratio or whole‐body composition were not significant, except that fish fed the singular nucleotide supplement had a significantly higher whole‐body lipid content compared with fish fed the basal diet. No significant dietary effects on cortisol response were observed after a 15 min confinement stress test perhaps because of the extreme variation among individual fish. Subsequently, in situ challenge by co‐habitation with A. ocellatum caused 100% mortality regardless of dietary treatment within a 48 h period. It is concluded that dietary supplementation of brewers yeast and nucleotides did not promote enhanced growth or improved cortisol response and resistance to uncontrolled amyloodiniosis in juvenile red drum, at least at the tested dosages.  相似文献

The length of the photoperiod has been implicated as affecting growth of sunshine bass. This study was done to determine if photoperiod length might affect insulin‐like growth factor‐I (IGF‐I), which is a major hormonal regulator of growth in fish. Growth, feed conversion, peritoneal fat content, and plasma concentrations of IGF‐I of sunshine bass were compared on fish held on a short (8 h) or a long (16 h) photoperiod, or after the fish were switched from one photoperiod to the other. Fish were fed daily to apparent satiation for up to 5 wk and were not fed for the last 2 wk of the experiment. Body weight and intraperitoneal fat content increased for the first 2 wk of the experiment and then remained steady for the remainder of the experiment, and feed consumption decreased from 3.5% during the first 2 wk to about 1.5% for the second 2 wk and finally to only 1% during the last week of feeding. Plasma IGF‐I concentrations fell steadily during the entire experiment and was at the lowest level during the last 2 wk when the fish were not fed. There was no consistent effect of the photoperiod in any of the conditions; however, feeding and IGF‐I concentrations were affected by fish density.  相似文献

Mitochondrial NAD(P)-dependent malic enzyme [EC 1.1.1.39, L-malate: NAD+ oxidoreductase (decarboxylating)] was purified from herring skeletal muscle to a specific activity of 8.2 mol/min/mg. The purification procedure involved chromatography on DEAE-cellulose, Red Agarose and a Sephacryl S-300 with a final recovery of 38% of enzyme activity. This enzyme catalyzes the oxidative decarboxylation of malate in the presence of either NAD or NADP in the presence of Mn2+. Some kinetic characteristics of this enzyme were determined. The pH optimum of activity is 7.0. ATP was shown to be a competitive inhibitor with malate. The inhibition by ATP displayed hyperbolic competitive kinetics with a Ki (ATP) of 0.28 mM in the presence of NAD and 0.75 mM in the presence of NADP. Fumarate reversed ATP inhibition.In vivo, regulation of NAD(P)-dependent malic enzyme might respond to changing levels of mitochondrial ATP and fumarate with the enzyme undergoing kinetic activation by an increase in the concentration of mitochondrial fumarate which could reverse enzyme inhibition by ATP.  相似文献

Background: The potential for acid-detergent insoluble ash (ADIA), alkaline-peroxide lignin (APL), and acid-detergent lignin (ADL) to predict fecal output (FO) and dry matter digestibility (DMD) by cattle offered bermudagrass [Cynodon doctylon (L) Pers.] hays of different qualities was evaluated. Eight ruminally cannulated cows (594 ± 35.5 kg) were allocated randomly to 4 hay diets: low (L), medium low (ML), medium high (MH), and high (H) crude protein (CP) concentration (79, 111,131, and 164 g CP/kg on a DM basis, respectively). Diets were offered in 3 periods with 2 diet replicates per period and were rotated across cows between periods. Cows were individually fed 20 g DM/kg of body weight in equal feedings at 08:00 and 16:00 h for a 10-d adaptation followed by a 5-d total fecal collection. Actual DM intake (DMI), DMD, and FO were determined based on hay offered, ort, and feces excreted. These components were then analyzed for ADL, APL, and ADIA concentration to determine marker recovery and marker-based estimates of FO and DMD. Results: Forage DMI was affected by diet (P = 0.02), and DMI from MH and H was greater (P 〈 0.05) than from L. Apparent DMD tended (P = 0.08) to differ among diets while FO (P = 0.20) was not affected by diet treatments. Average ADL recovery (1.16) was greater (P 〈 0.05) than that of ADIA (1.03) and APL (1.06), but ADIA and APL did not differ (P = 0.42). Estimates of FO and DMD derived using APL and ADIA were not different (P≥0.05) from total fecal collection while those using ADL differed (P 〈 0.05). There was no diet by marker interaction (P≥ 0.22) for either FO or DMD. Conclusion: Acid-detergent insoluble ash and APL accurately predicted FO and DMD of cattle fed bermudagrass hay of varying nutrient composition. In previous proof-of-concept work, it was shown that the use of treated coal mine water for rainbow trout (Oncorhynchus mykiss) culture in a cage was technically feasible, though only a 50-fish bioassay was grown and no work on production-related issues was conducted. To further advance the use of treated mine water, an under-utilized water resource throughout Mid Appalachia, work was conducted to assess the effects of using treated coal mine water for the intensive production of rainbow trout in a flow-through system. During this study, comprehensive water quality data were collected to supplement fish weight and length data taken during routine monthly sampling events. The 8000 fish grew well in the raceway system over the 9 months of production, where a feed conversion ratio of 1.4 and a condition factor of 5.1 × 10−4 were measured with stocking and harvest densities of 26.4 and 50.2 kg/m3, respectively. Further, total net production was 3275 kg (7220 lb) with 98.6% survival. Throughout the study, dissolved ion concentrations (Fe, Al, Mg, Ca, and SO4) often exceeded recommended tolerance limits. Further, elevated ammonia nitrogen concentrations generated from a component of the mine water-treatment process were identified as a potential limiting factor for aquaculture development. However, when the non-ideal effects of high ionic strength and the speciation of dissolved metal–ligand complexes were taken into account, the concentrations of free metal ions were within recommended tolerance limits.  相似文献
