Most of the cultivated species of citrus have narrow genetic basis. Relationships among species and cultivars are obscured by sexual compatibility, polyembryony, apomixis and a high incidence of somatic mutations. DNA analysis is crucial in genetic studies not only for citrus breeding programs but also for characterization of hybrids and species. In this paper, single nucleotide polymorphisms (SNPs) were investigated in 58 accessions of Citrus, hybrids and related genera. Genomic sequences of Pêra IAC sweet orange (Citrus sinensis L. Osbeck) were used for primer design and selection of sequence tagged sites (STSs) for identification of SNPs. Analysis of 36 STSs showed identical sequences among 40 of the 41 sweet orange accessions studied. However, these accessions were heterozygous for many SNPs. Ten selected STSs were analyzed in 17 additional accessions from 13 species and hybrids. Comparing to the Pêra IAC sweet orange accession, a total of 150 polymorphic nucleotides were identified and most of the alterations were transitions (52.7%). The greatest number of SNPs was observed in Poncirus trifoliata (L.) Raf. and the smallest in Ponkan mandarin (Citrus reticulata Blanco). At the intra-specific level, Baía Gigante (Citrus sinensis L. Osbeck) was the only sweet orange accession with a divergent SNPs genotype, which corroborates the hypothesis of a hybrid origin for this accession. Although the STSs analyzed represent randomly sampled genomic sequences, they provided consistent information about the level of polymorphism and showed the potential of SNPs markers for characterization and phylogenetic studies. 相似文献

Citrus breeding programs normally face several biological obstacles such as apomixis and polyembryony that result in a cumbersome
identification of hybrid seedlings. The main purpose of this work is to describe the combined use of visual selection based
on the leaf apex morphology and SSR analysis to differentiate hybrid from nucellar seedlings derived from the cross between
the ‘Murcott’ tangor [Citrus reticulata Blanco × Citrus sinensis (L.) Osb.] and ‘Pêra’ sweet orange [Citrus sinensis(L.) Osb.]. A new morphological variable named leaf apex morphometric index is also described as the quantitative basis of
the visual selection. The efficiency of visual selection of hybrids was tested under two growth conditions, seedlings germinated
in seedbeds and in plastic tubes. Putative hybrid seedlings were also confirmed through the analysis of simple sequence repeats
(SSR). The visual selection of hybrid seedlings resulted in an increase of 87.2% (p < 0.01) and 202.2% (p <0.001) in the number of correctly identified hybrids when compared to the method of random picking of seedlings in seedbeds
and plastic tubes,respectively. The results indicate that the combination of visual selection and SSR analysis for the identification
of hybrids derived from the cross of polyembryonic citrus cultivars will improve the accuracy of the selection,save time,
and reduce the costs involved in the use of molecular markers alone in citrus breeding programs.

Difficulties in obtaining new breeding lines of Brachiaria (Trin.) Griseb., an important forage grass in Brazil, are mostly related to differences in ploidy among the accessions, and
to apomixis, an asexual mode of reproduction. Usually, sexual accessions are diploid while apomicts are polyploid. Induced
tetraploids of Brachiaria brizantha (A. Rich.) Stapf have been successfully obtained and this paper presents the results of a study of their reproductive modes
and fertility. Despite frequent meiotic aberrations during microspore development, the induced tetraploids produced viable
pollen and produced progeny after controlled self-pollination. Similarly to the original diploid sexual progenitor, embryo
sacs of the Polygonum type with confirmed meiotic origin were present in the induced tetraploids suggesting chromosome doubling did not alter the
reproductive mode. The embryo sac of the Polygonum type was also observed in progenies obtained after self and open pollination. Nevertheless, embryo sacs of the Polygonum and the Panicum types within the same ovule were observed in some progenies obtained after open pollination, probably having resulted from
hybridization with tetraploid, apomictic plants. Indeed, the compatibility of the progeny with tetraploid, apomictic B. brizantha was confirmed by the formation of mature caryopses after controlled pollination. Evidence is presented that the induced tetraploids
and their progeny are sexual plants and that they are compatible with natural tetraploid B. brizantha. The induced tetraploids will be useful for analyses of apomictic inheritance as well as in the development of sexual tetraploid

Microcosm experiments on the behaviour of Se, Cr and Co were carried out with mangrove sediments from Sepetiba Bay, Brazil. Three 8-cm length sediment cores were covered with tidal water spiked with 75Se, 51Cr and 60Co to evaluate its behaviour within the sediments. Two cores retained almost all activities (99?C100?%) within the uppermost centimetre layer, while the third core presented a deeper penetration of all radiotracers, displaying a second activity peak at the 3?C4-cm depth interval, which evidenced benthic fauna bioturbation influence. This influence extended the diffusion into depths in which mangrove rhizosphere can retain the elements, suggesting increased retention efficiency. This mechanism of retention enhancement was proposed in addition to previous conceptual models describing trace elements behaviour in mangrove sediments. Increased bioturbation and rhizosphere development will probably increase this effect, while mangrove plant cover losses may promote a less efficient retention of elements recently diffused from tidal water. 相似文献

Citrus black spot (CBS) symptom expression on sweet oranges has been reported to be affected by the fruit developmental stage and inoculum concentration in greenhouse conditions. However, there is little information concerning the period in which the fruit is susceptible in commercial orchards. This study assessed the influence of fruit age and inoculum concentration of Phyllosticta citricarpa on CBS intensity and fruit drop in the field. Two field trials were conducted in commercial ‘Valencia’ orchards in São Paulo state, Brazil. Fruit were inoculated from October (petal fall stage) to July (c.6.5 cm diameter), with 101, 103 and 105 pycnidiospores/ml. CBS symptoms and fruit drop were observed in higher levels for fruit inoculated from October to February than from March to July. In both trials, when fruit were inoculated 10 times from October to July or single-inoculated in November or December, high CBS severities of 11%–22% were observed and the proportion of fruit remaining on the tree was 48%–77%; in contrast, noninoculated fruit had severity below 1.1% and 90% remained on the tree. Inoculations in November or December of green fruit with 1.5–3.0 cm diameter using 105 pycnidiospores/ml were associated with higher CBS intensity and fruit drop and shorter incubation period than inoculations with lower concentrations. This work provides a better understanding of fruit stages and P. citricarpa inoculum concentrations most related to symptom expression and losses under natural conditions and may be helpful for improvement of CBS management in the São Paulo citrus belt. 相似文献

Cancer mortality data from the National Center for Health Statistics, covering the period 1950 through 1978, were used to test a reported association between childhood leukemia and exposure to radioactive fallout from nuclear weapons tests in Nevada between 1951 and 1958. No pattern of temporal and geographic variation in risk supportive of the reported association was found. Comparison of these results with those presented in support of an association of risk with fallout suggests that the purported association merely reflects an anomalously low leukemia rate in southern Utah during the period 1944 to 1949. 相似文献

Humoral immune response of water buffalo naturally infected with Toxocara vitulorum was monitored using three different antigens of this parasite in serum and colostrum of buffalo cows and calves. Soluble extract (Ex) and excretory/secretory (ES) larval antigens and perienteric fluid antigen (Pe) of adult T. vitulorum were used to measure the antibody levels by an indirect ELISA. Serum of 7-12 buffalo cows for the first 365 days and colostrum of the same number of buffalo cows for the first 60 days of parturition, and serum of 8-10 buffalo calves for the first 365 days after birth were assayed. The ELISA detected antibodies against all three T. vitulorum antigens in the colostrum and serum of 100% of buffalo cows and calves examined. The highest antibody levels against Ex, ES and Pe antigens were detected in the buffalo cow sera during the perinatal period and were maintained at high levels through 300 days after parturition. On the other hand, colostrum antibody concentrations of all three antigens were highest on the first day post-parturition, but decreased sharply during the first 15 days. Concomitantly to the monitoring of immune response, the parasitic status of the calves was also evaluated. In calves, antibodies passively acquired were at the highest concentrations 24 h after birth and remained at high levels until 45 days coincidentally with the peak of T. vitulorum infection. The rejection of the worms by the calves occurred simultaneously with the decline of antibody levels, which reached their lowest levels between 76 and 150 days. Thereafter, probably because of the presence of adults/larvae stimulation, the calves acquired active immunity and the antibodies started to increase slightly in the serum and plateaued between the days 211 and 365. All three antigens were detected by the serum antibodies of buffalo calves; however, the concentration of anti-Pe antibody was higher than anti-EX and anti-ES, particularly after 90 days of age. By conclusion, the buffalo cows develop immunity and keep high levels of antibodies against T. vitulorum-Ex, ES and Pe antigens and these antibodies are transferred to their calves through the colostrum. This passively acquired immunity does not protect the calves against the acquisition of the infection, but these antibodies, passively or actively acquired, may have an important role during worm rejection by the calves and prevention of intestinal reinfection. 相似文献

A feeding trial was carried out where five protein concentrations [26%, 29%, 33%, 37%, and 41% crude protein (CP)] were fed to jundia fingerlings at two dietary energy concentrations [3200 and 3650 kcal metabolizable energy (ME)/kg diet] to establish the protein requirement for this species. Triplicate groups of 23 fish (average weight 1.52±0.34 g) were each stocked in 120-l aquaria and fed semipurified diets twice a day to apparent satiation. After 90 days, fish weight gain (WG), specific growth rate (SGR), feed efficiency (FE), protein efficiency ratio (PER), energy retention (ER), apparent net protein utilization (ANPU), feed consumption (% body weight) and body composition (crude protein, fat, and ash) were affected by diet composition. Weight gain, SGR, FE, ANPU and ER increased (P<0.05) as the dietary protein concentration increased up to 33% and 37%, for fish fed diets containing 3650 and 3200 kcal, respectively. A significant interaction between dietary protein and energy was observed for WG, SGR, and PER. Feed consumption was not affected by dietary energy concentration, but decreased with increasing dietary protein concentration (P<0.05). Body fat decreased as dietary protein increased at both energy concentrations, but was higher in fish fed the 3650 kcal diet. The opposite was observed for body protein, ER, and ANPU (P<0.05). Our findings demonstrate that jundia presents dietary protein sparing effect when the energy concentration increases from 3200 to 3650 kcal/kg. Jundia protein requirement, determined by the broken line method, is between 32.6% and 37.3% CP, depending on dietary energy concentration. 相似文献
