Two crossbred puppies exhibited a generalized tremor that was accentuated by excitement or voluntary movement. Hypermetria was also present. Neurohistologic examination showed a bilaterally symmetric spongy condition, predominantly of the gray matter in the brain and spinal cord. These two dogs were different, both in their clinical presentation and histologic appearance, from other reported cases of spongy degeneration. The clinicopathologic correlation between spongy degeneration and tremor is unclear. Spongy degeneration should be included in the differential diagnosis of tremor syndromes in puppies.  相似文献

A technique based on the use of specific primers for polymerase chain reaction (PCR) was developed for the identification of the stem and bulb nematode belonging to the Ditylenchus dipsaci species complex. The internal transcribed spacer region ITS1 and ITS2, the gene 5.8 S and part of genes 18 S and 26 S of twenty populations of the D. dipsaci species complex belonging to both D. dipsaci sensu stricto and Ditylenchus sp. B (corresponding to populations of giant individuals associated to Vicia faba) and three congeneric species were amplified with two universal ribosomal primers. PCR-amplified DNA samples were digested with five restriction enzymes in order to reveal some polymorphism allowing the identification of D. dipsaci populations associated with Fabaceae seeds. The polymorphism among species was confirmed by the sequencing of the PCR products. A primer (DdpS2) was designed in a region conserved in all populations of both D. dipsaci sensu stricto and D. sp. B studied in the present work. The other Anguinidae species (except a few species from Central Asia associated to Astereaceae and D. sp. G associated to Plantago maritima) differ in two to four nucleotides at the 3′ extremity of this region. This sequence portion coincides with a TspEI restriction site. In combination with a primer located in the ribosomal region, this first primer is a good candidate for identification by PCR of populations of the D. dipsaci species complex found in Fabaceae seeds. A second primer (DdpS1) was designed in a similar way and was specific to D. dipsaci sensu stricto. The utility of these two sets of primers is discussed against the background of quarantine regulation.  相似文献

The assessment of an early and reliable pregnancy diagnosis in sows is very important for limiting the number of non-productive days in sow herds. The aim of the present study was to investigate the accuracy of trans-abdominal ultrasound detection for early pregnancy diagnosis in sows under field conditions. Particular attention was paid to the time when a reliable diagnosis can be made, and to possible differences between two commonly used types of scanners. In total, 202 sows from two herds were tested daily with scanners A (linear transducer of 5 MHz) and B (sector transducer of 3.5 MHz) from day 16 until 25 after insemination. Ninety-three percentage of the sows were pregnant 36 days after insemination. Using ultrasound, the first pregnancy could be detected already 18 days after insemination. The sensitivity, specificity and overall accuracy were higher than 95% from day 24 (scanner A) or day 23 (scanner B) post-insemination onwards. The positive predictive values from day 19 onwards were consistently higher than 95% with both types of scanners, whereas the negative predictive values were consistently low for both scanners during the entire period. The sensitivity of testing tended to be slightly lower in sows with a litter size of <10 total born piglets, compared to sows with 10 or more piglets. In conclusion, pregnancy diagnosis in sows using trans-abdominal ultrasound testing can be assessed quickly and reliably under field conditions from day 23 of gestation onwards. Predictive values of positive test results were high in these herds, whereas those of negative test results were low. This implies that sows with a negative test result early in pregnancy should be retested later.  相似文献

The aim of this work was to measure the myeloperoxidase (MPO) concentration in bronchoalveolar lavage (BAL) fluid collected from horses with recurrent airway obstruction (RAO), both in crisis and in remission, as well as from healthy horses. Seven horses with RAO were exposed to moldy hay until the maximum change in pleural pressure was greater than 1.5 kPa. At that point, BAL was performed, and the total cell counts and percentages in the fluid were immediately determined. To measure the MPO concentration in BAL-fluid supernatant, we used a specific enzyme-linked immunosorbent assay with polyclonal antibodies against equine MPO. The tests were repeated on the horses with RAO after they had spent 2 mo on pasture. Six healthy horses serving as controls underwent the same tests. The absolute and relative neutrophil counts and the MPO concentration in the BAL fluid were significantly greater in the horses with an RAO crisis than in the control horses. After 2 mo on pasture, the horses that had been in RAO crisis were clinically normal, and their neutrophil counts and MPO levels in BAL fluid had significantly decreased; during remission their neutrophil counts were not significantly different from those in the healthy horses, but their MPO concentration remained significantly higher. This study showed that determining the MPO concentration in a horse's BAL fluid is technically possible and that during remission from RAO the concentration remains higher than normal. Thus, MPO may be a marker of neutrophil presence and activation in the lower airways.  相似文献

The objective of this study was to investigate whether intramuscular vaccination of healthy adult horses with a killed or a modified live equine herpesvirus type 1 (EHV-1) vaccine could induce transient positive PCR results in either blood or secretions collected on a nasopharyngeal swab. Four horses in each group received either a single killed or a modified-live vaccine intramuscularly. Two local commingled and 2 distant nonvaccinated controls were included for each group. All horses were observed daily for evidence of clinical abnormalities throughout the study periods. Blood and nasopharyngeal swabs were collected twice before vaccination and once weekly for 4 weeks after vaccination and submitted for PCR testing for EHV-1 by 2 independent laboratories using different real-time PCR methodologies. Serum samples collected from all horses on the vaccination day and 21 days later were tested for antibodies against EHV-1 using a serum neutralization test. Whereas the 2 vaccine strains tested positive in both EHV-1 PCR assays, nasopharyngeal swabs and whole blood collected from vaccinated and control horses had negative PCR test results for EHV-1 during the entire study period. Serum neutralization testing revealed a 2- to 4-fold increase in titers for all vaccinated horses, whereas titers in control horses were largely unchanged. The use of seropositive horses before immunization and the sampling frequency of 7 days may have prevented the occasional molecular detection of the vaccine virus in whole blood and nasopharyngeal secretions. However, the study results demonstrate that detection of EHV-1 DNA by PCR in vaccinated and unvaccinated healthy horses is not a common event.  相似文献

During the last decade, several computer assisted sperm analysis (CASA) systems have been validated for canine sperm quality assessment. Regarding the impressive possibilities of these systems, further research is required to determine which CASA measurements are of clinical importance in canine andrology. In the present study, the sperm quality parameters obtained by the Hamilton-Thorne Semen Analyser (Ceros 12.1; HTR) were correlated with the body weight and the age of the dogs. Moreover, the sperm quality parameters of dogs with a different breeding history were compared. The sperm-rich fraction was collected from 111 dogs of 50 different breeds, which were presented at our department. Immediately after collection, the concentration, the total sperm output (TSO) and 13 different sperm motility and velocity characteristics were measured by the HTR. The percentage of live spermatozoa and the spermatozoal morphology were examined on eosin/nigrosin stained smears. Based on their breeding history, the dogs were divided in three groups: 'fertile' (n = 60), 'subfertile' (n = 17) or 'not used for breeding' (n = 34). Significant (p < 0.05) correlations were established between the body weight of the dogs and the TSO (r = 0.245) and velocity curvilinear (VCL; r = -0.220), respectively. The age was negatively correlated with the percentage of normal spermatozoa (r = -0.203; p < 0.05). The correlations with all the other evaluated sperm parameters were low and not significant. Significant differences between the 'fertile' and the 'subfertile' group were found for all of the evaluated sperm quality parameters (except for BCF, LIN, STR and MEDIUM). In conclusion, dogs tend to produce ejaculates with a lower percentage of normal spermatozoa with increasing age and dogs with higher body weights produce ejaculates with a higher TSO and a lower VCL. Significantly poorer sperm characteristics were found for dogs with lower in vivo fertility results.  相似文献

Shedding patterns of 2 virulent (P-2208 and KC-152-D) and 1 attenuated (BUK) strains of pseudorabies virus (PRV) were determined in groups of intranasally inoculated feeder pigs. Clinical signs observed following inoculation with the P-2208 and KC-152-D strains included increase in rectal temperatures up to 42.2 C, anorexia, severe respiratory disturbance, and fatal CNS signs in 2 cases. Clinical signs in pigs inoculated with 7.2 X 10(7) median tissue culture infective dose (TCID50) of the BUK strain were limited to depression and a rise in rectal temperatures to 40.5 C for 3 to 4 days. Evaluation of the efficacy of the virus isolation method used showed that the presence on swabs of only 12.5 TCID50 of the P-2208 strain or 8.4 TCID50 of the BUK strain resulted in a 50% chance of virus recovery. Intranasal inoculations with 500 TCID50 of the P-2208 or KC-152-D strain did not result in synchronous infection of the whole group. Intranasal inoculations with 5,000 TCID50 of the KC-152-D strain or 50,000 TCID50 of the P-2208 strain resulted in continuous virus shedding in all pigs between postinoculation days (PID) 4 and 13 (KC-152-D strain) or 14 (P-2208 strain). Some of the pigs in these 2 groups further shed the P-2208 or KC-152-D strain in a continuous or discontinuous pattern up to PID 19 (P-2208 strain) or 20 (KC-152-D strain). The time of onset or the level of virus neutralizing antibody production in individual pigs was not found to have an influence on their shedding patterns.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

From the 62 swine abscesses examined, 56 yielded positive cultures, of which 38 cases were mixtures of facultative aerobes and obligate anaerobic bacteria (OAB). Only 6 species appeared to be sterile. In all specimens, which yielded OAB, VFA (C4-C6) could be detected by gas-liquid chromatography (GLC). False negatives were not recorded, and only one false positive was found, which might have been due to faulty culture technique, or to the fact that the metabolic markers may persist longer than the OAB themselves. Because current methods to isolate non-sporulating OAB are laborious and not available in most veterinary clinical situations and because there is a need to evaluate the importance of non-sporulating OAB in veterinary medicine, GLC seems to be a useful alternative to culture techniques for detecting the presence of OAB in infectious processes in animals.  相似文献

This study was conducted to compare the effects of a preventive in-feed medication programme using tilmicosin (Pulmotil 200 premix, Elanco Animal Health) at 200 p.p.m. with those of Mycoplasma hyopneumoniae (Mh) vaccination programme (Stellamune Mycoplasma, Pfizer Animal Health). A pig herd with chronic respiratory disease in which infection with Mh played an important role was selected, and a total of 204 piglets were randomly allocated to either the medication (P) or the vaccination (V) group. Pigs in the P group received medicated feed for 3 weeks after weaning (days 34-55), and for 2 weeks late in the nursery period (days 77-98). The piglets in the V group were vaccinated twice intramuscularly, at 4 and 22 days of age. The two groups were compared on the basis of average daily gain (ADG), feed conversion rate (FCR), additional curative medication days (CMD), overall mortality (major variables), a coughing index, pneumonia lesions, and serology against Mh, influenza H1N1 and influenza H3N2 viruses, Actinobacillus pleuropneumoniae (App) and porcine reproductive and respirator, syndrome virus (PRRSV) (minor variables). No significant differences (P > 0.05) were observed for ADG (555 g/day in P group; 567 g/day in V group), FCR (2.64 in P group; 2.41 in V group) and mortality rate (11% in P group; 7% in V group). The average number of additional curative medication days (CMD) per pig was significantly higher (P < 0.01) in the P group (1.5) than in the V group (0.58). At slaughter age, the serological results and the prevalence of macroscopic lung lesions were comparable in the two groups (P > 0.05). With the exception of CMD, the preventive use of tilmicosin at this swine farm was found to confer similar beneficial effects to Mh vaccination.  相似文献
