We evaluated the applicability of a simple passive sampling method to quantify HNO3(g) in ambient air. The method has the advantages of not only ease of operation but also low cost. A sampling velocity of 214 m day?1 was determined based on the concentration of HNO3(g) measured by the four-stage filter-pack method at nine sites located within a 250?×?250-km area in Japan. This sampling velocity was applied at sites located outside of area to verify the applicability and accuracy of the simple passive sampling method. The variation in the results for the application of the sampling velocity ranged from 0.39 to 0.95. The simple passive sampling method should be applied to sites with different meteorological conditions, and the obtained data should be used to obtain more significant information and/or to indicate the need for further developments in the methodology.  相似文献

In order to clarify the location of feeding centers in the ruminant brain, this study used a single-unit activity (SUA) recording electrode to investigate the existence of appetite-regulating neurons in the lateral hypothalamic area (LHA) in goats. Seven male Japanese Saanen goats were used in the experiment. The animals were fed twice daily, once in the morning (1000 to 1200) with 1.5 kg of roughly crushed alfalfa hay cubes, and once in the afternoon (1600 to 1800) with 200 g of commercial ground concentrate feed. The animals were allowed free access to drinking water. In this study, the animals were surgically operated on to position the recording electrode in the LHA. Recordings of SUA were carried out continuously over a 2.25-h period beginning 15 min prior to the commencement of morning feeding. The eating rates of crushed alfalfa hay cubes were highest 10 min after feeding commencement, but decreased sharply by the time 40 min had elapsed. The cumulative feed intake after the completion of the 2-h feeding period was 1164 +/- 38 g. The cumulative water intake upon the conclusion of the 2-h feeding period was 2422 +/- 107 mL. This study recorded 31 units, of which five showed a response to feeding and altered their firing rates. In response to a sharp increase in eating rates, all five units increased their firing rates to a level higher than that of prefeeding (P < 0.05). As the animals reached a level of satiety (eating rates declined to very low levels), firing of units I and II stopped completely, while the firing rates of units III, IV, and V decreased. Examination of a serial histological section confirmed that the five units in which changes in firing rates with feeding were observed were all located in the dorsolateral hypothalamic area close to the fornix. The LHA neurons recorded in this experiment characteristically showed neuronal activity increases at high levels of feeding, but decreases at low levels. The results suggest that there are cells located in the LHA of goats that are active in the physiological regulation of hay (dry forage) intake.  相似文献

The physical properties of sawdust including porosity, water retention, and water drainage were analyzed to prove its suitability for use as an artificial soil in the automatic decomposer-extinguisher (GADE) machine. The physical and chemical properties of residual sawdust from the GADE machine were also analyzed, the mechanical abrasion of sawdust in the GADE machine was tested, and the morphology of this residue was observed through a scanning electron microscope to investigate changes of these properties in the medium of decomposing garbage. Sawdust, which showed a lower specific gravity and larger porosity than soil, is considered capable of supplying air to bacteria. It was found that sawdust became worn from the operation of the machine. The spaces of residual sawdust were still observed, but water drainage decreased. The portion of hollocellulose in residual sawdust decreased, although the extractives in it increased. Results indicated that the capacity of sawdust to function as an artificial soil in the GADE machine was decreased owing not only to the destruction of sawdust grain but also to the adherence of products from decomposition, such that sawdust needed to be replaced every few months.Part of this report was presented at the 43rd annual meeting of the Japan Wood Research Society, Morioka, April 1993  相似文献

The authors described a novel submerged batch culture system that produced high levels of amylase by Aspergillus kawachii using whole barley (WB), the surface of which is covered by its husk. In this study, detailed analyses determining the amylase activities, residual sugars, fungal morphology and expression levels of genes were performed in a submerged culture using WB to address the mechanism underlying high amylase productivity in A. kawachii. High levels of glucoamylase and acid-stable u-amylase were produced in this culture, and expression levels of amylases, as well as glucose-repressive genes including high-affinity glucose transporter and peroxidase/catalase were also high. On the other hand, the morphology of mycelia was altered, with swollen, bulbous, multi-septum hyphae and conidiophores that normally form in a solid culture being partially generated. Furthermore, cell cycle and post-translational modification-related gene expression levels were altered, and were similar to those in the solid culture. These findings suggest that high amylase productivity in the submerged culture using WB is accompanied by both the up-regulation of amylase genes and activation of post-translational modifications due to fungal morphological changes being brought closer to those in the solid culture.  相似文献

Hermaphroditic organisms avoid inbreeding by a system of self-incompatibility (SI). A primitive chordate (ascidian) Ciona intestinalis is an example of such an organism, but the molecular mechanism underlying its SI system is not known. Here, we show that the SI system is governed by two gene loci that act cooperatively. Each locus contains a tightly linked pair of polycystin 1-related receptor (s-Themis) and fibrinogen-like ligand (v-Themis) genes, the latter of which is located in the first intron of s-Themis but transcribed in the opposite direction. These genes may encode male- and female-side self-recognition molecules. The SI system of C. intestinalis has a similar framework to that of flowering plants but utilizing different molecules.  相似文献

Continuous monitoring of concentrations of particulate matter and gaseous compounds for three years by a four-stage filter-pack method clarified the characteristics in the concentrations of chemical species in particulate matter and gaseous compounds in ambient air in Kobe, Japan. The amount of materials in blank filters was low enough for this method to be used for the monitoring of ambient air. Little or none of the hydrogen chloride was derived anthropogenically. The concentrations of both particulate sulfate and sulfur dioxide in spring and summer were significantly higher than those in autumn and winter. The concentration of gaseous nitric acid was high in summer, which suggests that active photochemical reactions in the atmosphere provided more nitrogen species in summer. The measured concentration products [HNO3][NH3] showed good agreement with the theoretical predictions with some exceptions. The total concentration of ammonium species showed no significant seasonal variation except that the concentration in winter was low. The concentration of nss-Ca2+ in spring was significantly higher than that in the other seasons, which is most likely associated with the so-called yellow-sand events. The concentrations of nss-K+ and nss-Ca2+ accounted for most of K+ and Ca2+, respectively, whereas that of nss-Mg2+ accounted for only 5% of Mg2+  相似文献

Among the chicken breeds, White Leghorn (WL) and White Plymouth Rock (WR) are major breeds and have different history in their establishments. Whole mitochondrial DNA of the breeds were sequenced in order to elucidate the genetic relationship between the breeds. The lengths of the two WL and two WR mitochondrial DNA were found to be 16 788 and 16 785 base pairs, respectively. When the DNA sequences were compared, the similarity was found to be 99.96% (six nucleotide differences). In addition, the present study conformed the existence of an extra nucleotide ‘C’ in NADH dehydrogenase subunit 3 (ND3) of the chicken mitochondrial DNA, which has been consistently observed in Galliformes.  相似文献

Viruses were isolated from leaves of plants of Aconitum species with symptoms such as mottling and yellowing in Hokkaido and Gunma prefectures in Japan. In spite of a relatively optimistic pre‐season forecast, the total return of adult sockeye salmon (Oncorhynchus nerka) to the Fraser River (British Columbia, Canada) in 2009 was the lowest recorded since quantitative records began in the late 1940s. A plausible mechanism is proposed that links a sequence of extreme oceanic and climatic events to poor marine survival. It began with record‐setting snow packs in the coastal mountain range during the winter of 2007 that led to the development of unprecedented oceanographic conditions in the spring of 2007 from Queen Charlotte Strait in central British Columbia to Southeast Alaska. When combined with equally extreme atmospheric anomalies in the region in the spring of 2007, with a winter wind regime persisting through July, a coastal surface ocean with characteristics that are known to be associated with lower marine survival was established. Most of the sockeye salmon that were expected to return to the Fraser River as adults in 2009 passed through this atypical ocean as juveniles on their migration to the open ocean in 2007. A trophic gauntlet hypothesis is proposed as a new paradigm to describe the oceanic environment faced by sockeye salmon after they emigrate northward from the Strait of Georgia. The hypothesis identifies a new type of high nutrient low chlorophyll region that can explain how oceanographic extremes at critical locations along the migration route beyond the Strait of Georgia can reduce marine survival in some years.  相似文献

ABSTRACT:   Artificially hatched Seriola species have the problem of malformation, mainly in their vertebrae, head, and mouth parts. To clarify the cause of vertebral malformation, the effects of hypoxia during embryogenesis on the induction of centrum defects was investigated in artificially hatched amberjack Seriola dumerili . Firstly, 7-somite stage embryos were exposed to waters of 0, 12.5, 25, 50, and 100% dissolved oxygen (DO) for 0.5, 1, 2, 3, 4, and 5 h to confirm the effective dose (DO concentration and duration of exposure) of hypoxia that induces somitic disturbances in newly hatched larvae. Exposure of embryos to 12.5% DO concentration for longer than 0.5 h induced somitic disturbances. Following this result, centrum defects in juveniles were investigated by an induction experiment with embryos exposed to 12.5% DO for 2 h at the gastrula, 1- or 2-somite, 10-somite, 15-somite, or heart beating stage. This experiment revealed that centrum defects were induced only during somitogenesis, and somitic disturbances were the premonitory symptom of centrum defects. These results indicate hypoxia during somitogenesis as a possible cause of centrum defects in amberjack.  相似文献
