We examined foliar endophyte frequencies in two native (Betula pendula and Betula pubescens) and three exotic (Betula ermanii, Betula platyphylla and Betula resinifera) birch species and their hybrids in Punkaharju, Finland. The most frequently isolated endophytic fungi in the experimental trees were Fusicladium betulae and Gnomonia setacea making up 80–90% of all endophyte infections. Total endophyte infection levels varied from 0.5 colony forming units (CFU)/cm2 in B. platyphylla to 8.6 CFU/cm2 in B. pubescens that had highest total infection levels of both examined endophyte species. The resistance of hybrids was generally very close to the more resistant parent (the only exception being Fusicladium in B. platyphylla × B. pendula hybrid) supporting the hypothesis that the resistance of birch hybrids to these fungi is genetically based and caused by dominant inheritance of resistance traits.  相似文献

The complete genomic DNA of a novel papillomavirus (PV) was isolated from a basosquamous carcinoma on the wing of an Egyptian fruit bat (Rousettus aegyptiacus). Initial short sequences of the E1 and L1 genes of this virus were retrieved by PCR with degenerate papillomavirus-specific primers, and the entire R. aegyptiacus papillomavirus type 1 (RaPV-1) DNA was then amplified by long template PCR, cloned and sequenced with a transposon insertion method. The RaPV-1 genome counts 7970 basepairs and contains the typical papillomavirus open reading frames (ORF) (E1, E2, E4, E6, E7, L1 and L2). Based on a concatenated alignment of the E1, E2, L1 and L2 open reading frames of RaPV-1 and 46 other human and animal papillomavirus type species, a neighbor-joining phylogenetic tree was constructed. This phylogenetic analysis shows that RaPV-1 has a close-to-root position in the papillomavirus evolutionary tree. Since RaPV-1 is only distantly related to other papillomaviruses (with maximally 50% nucleotide sequence identity across the L1 open reading frame), it cannot be assigned to one of the existing papillomavirus genera and therefore represents the first member of a novel, as yet unnamed, close-to-root papillomavirus genus. This is the first time a papillomavirus has been isolated and characterized from a member of the Chiroptera order.  相似文献

In the present experiment, the single and combined effects of elevated temperature and ozone (O(3)) on four silver birch genotypes (gt12, gt14, gt15 and gt25) were studied in an open-air field exposure design. Above- and below-ground biomass accumulation, stem growth and soil respiration were measured in 2008. In addition, a (13)C-labelling experiment was conducted with gt15 trees. After the second exposure season, elevated temperature increased silver birch above- and below-ground growth and soil respiration rates. However, some of these variables showed that the temperature effect was modified by tree genotype and prevailing O(3) level. For instance, in gt14 soil respiration was increased in elevated temperature alone (T) and in elevated O(3) and elevated temperature in combination (O(3) + T) treatments, but in other genotypes O(3) either partly (gt12) or totally nullified (gt25) temperature effects on soil respiration, or acted synergistically with temperature (gt15). Before leaf abscission, all genotypes had the largest leaf biomass in T and O(3) + T treatments, whereas at the end of the season temperature effects on leaf biomass depended on the prevailing O(3) level. Temperature increase thus delayed and O(3) accelerated leaf senescence, and in combination treatment O(3) reduced the temperature effect. Photosynthetic : non-photosynthetic tissue ratios (P : nP ratios) showed that elevated temperature increased foliage biomass relative to woody mass, particularly in gt14 and gt12, whereas O(3) and O(3) + T decreased it most clearly in gt25. O(3)-caused stem growth reductions were clearest in the fastest-growing gt14 and gt25, whereas mycorrhizal root growth and sporocarp production increased under O(3) in all genotypes. A labelling experiment showed that temperature increased tree total biomass and hence (13)C fixation in the foliage and roots and also label return was highest under elevated temperature. Ozone seemed to change tree (13)C allocation, as it decreased foliar (13)C excess amount, simultaneously increasing (13)C excess obtained from the soil. The present results suggest that warming has potential to increase silver birch growth and hence carbon (C) accumulation in tree biomass, but the final magnitude of this C sink strength is partly counteracted by temperature-induced increase in soil respiration rates and simultaneous O(3) stress. Silver birch populations' response to climate change will also largely depend on their genotype composition.  相似文献

Chromatophoromas are neoplasms that develop from the dermal pigment-bearing and light-reflecting cells (chromatophores) in the skin of reptiles, fish, and amphibians. Seventeen cutaneous chromatophoromas were identified from 851 bearded dragon submissions (2%) to a private diagnostic laboratory in a 15-y period. No sex predilection was found. Ages ranged from 9 mo to 11 y. Chromatophoromas most commonly were single, raised, variably pigmented masses or pigmented scales on the trunk, and less commonly the extremities or head. Microscopically, iridophoromas, melanophoromas, mixed chromatophoromas, and nonpigmented chromatophoromas were identified. Neoplasms were often ulcerated and invaded deep into the subcutis and muscle. Most commonly, nuclear atypia was mild-to-moderate, and mitotic count was low. Six neoplasms had abundant, periodic acid-Schiff–positive, mucinous stroma. Histologic examination was often adequate to diagnose chromatophoromas in bearded dragons given that 11 of 17 had some degree of pigmentation, although it was often scant. IHC for S100 and PNL2 could be helpful to diagnose poorly pigmented neoplasms. No lymphatic invasion or metastases at the time of excision were noted in any of the cases. Follow-up data were available for 6 cases, with no reports of recurrence or neoplasia-related death. Two cases had elevated mitotic counts and nuclear pleomorphism, which has been associated with metastasis in other reports of bearded dragon chromatophoromas. In general, it appears that aggressive surgical excision is often curative, but monitoring may be warranted for cases in which tumors had high mitotic count and nuclear pleomorphism.  相似文献

Opossums (Didelphis spp.) are the definitive host for the protozoan parasite Sarcocystis neurona, the causative agent of equine protozoal myeloencephalitis (EPM). Opossums shed sporocysts in feces that can be ingested by true intermediate hosts (cats, raccoons, skunks, armadillos and sea otters). Horses acquire the parasite by ingestion of feed or water contaminated by opossum feces. However, horses have been classified as aberrant intermediate hosts because the terminal asexual sarcocyst stage that is required for transmission to the definitive host has not been found in their tissues despite extensive efforts to search for them [Dubey, J.P., Lindsay, D.S., Saville, W.J., Reed, S.M., Granstrom, D.E., Speer, C.A., 2001b. A review of Sarcocystis neurona and equine protozoal myeloencephalitis (EPM). Vet. Parasitol. 95, 89-131]. In a 4-month-old filly with neurological disease consistent with EPM, we demonstrate schizonts in the brain and spinal cord and mature sarcocysts in the tongue and skeletal muscle, both with genetic and morphological characteristics of S. neurona. The histological and electron microscopic morphology of the schizonts and sarcocysts were identical to published features of S. neurona [Stanek, J.F., Dubey, J.P., Oglesbee, M.J., Reed, S.M., Lindsay, D.S., Capitini, L.A., Njoku, C.J., Vittitow, K.L., Saville, W.J., 2002. Life cycle of Sarcocystis neurona in its natural intermediate host, the raccoon, Procyon lotor. J. Parasitol. 88, 1151-1158]. DNA from schizonts and sarcocysts from this horse produced Sarcocystis specific 334bp PCR products [Tanhauser, S.M., Yowell, C.A., Cutler, T.J., Greiner, E.C., MacKay, R.J., Dame, J.B., 1999. Multiple DNA markers differentiate Sarcocystis neurona and Sarcocystis falcatula. J. Parasitol. 85, 221-228]. Restriction fragment length polymorphism (RFLP) analysis of these PCR products showed banding patterns characteristic of S. neurona. Sequencing, alignment and comparison of both schizont and sarcocyst DNA amplicons showed 100% identity. Although Koch's postulates have not been demonstrated in this case study, the finding of mature, intact S. neurona schizonts and sarcocysts in the tissues of this single horse strongly suggests that horses have the potential to act as intermediate hosts. Further studies are needed to demonstrate Koch's postulates with repeated transfer of S. neurona between opossums and horses.  相似文献

A 3‐year‐old cat from New Zealand developed three small raised non‐ulcerated plaques on the face. Serology detected antibodies against feline immunodeficiency virus (FIV). Histology of the plaque revealed epidermal hyperplasia with keratinocytes either distended with large blue‐grey cytoplasmic bodies or with shrunken nuclei surrounded by a clear halo. Papillomavirus (PV) antigen was detected immunohistochemically and feline viral plaque was diagnosed. Swabs were taken of both lesional and non‐lesional skin, and polymerase chain reactions were used to detect PV DNA. Three different PV DNA sequences were amplified, one from a Felis domesticus PV type 1 (FdPV‐1) previously amplified from a feline viral plaque, a second (FdPV‐JM) previously amplified from feline cutaneous squamous cell carcinomas, and a third FdPV‐MY that was not reported previously. All three sequences were amplified from swabs of both lesional and non‐lesional skin. These results extend the geographical range of FdPV‐1 outside North America and also demonstrate the ability of FdPV‐1 to asymptomatically infect feline skin. However, the detection of multiple PV sequences within both lesional and non‐lesional samples makes it difficult to determine whether or not any of the PVs caused feline viral plaque development in this cat. This is the first time PV DNA has been detected in a feline skin swab sample. Additionally, it is the first report of multiple PVs being detected in a single sample from a cat. This may suggest that FIV infection predisposes cats to cutaneous PV infection.  相似文献

We conducted two studies on how highways affect their adjacent habitats by sampling carabid beetles (Coleoptera, Carabidae) in patches of formerly continuous forest next to highways. (1) We sampled carabids at 14 highway intersections near Helsinki, Finland. Each intersection (constructed 2–40 years ago) had two forested patches to study: a remnant (0.5–37.4 ha) and, isolated from the remnant by an intersection lane, an islet (size 0.2–1.8 ha). Pitfall trap catch data (2301 carabids, 25 species) showed that remnants hosted higher catches of three carabid species, and slightly higher species richness, than islets (patch-size effect). Time since intersection construction had no apparent effect on carabids. Traffic volume along the intersection lane determined the assemblage structure of carabids in dry patches, and the abundance of a forest carabid Calathus micropterus. Compared to moist patches, drier patches hosted lower catches of four generalist species; they also had different assemblages of carabids (habitat-type effect). An interaction between patch size and habitat type for a forest generalist Pterostichus oblongopunctatus indicated that the patch-size effect was dependent on habitat type. (2) We examined possible dispersal of carabids among forested patches that were separated by highway lanes in Drenthe, the Netherlands. We released 2696 marked individuals of 10 species, and recaptured 376 using dry pitfall traps. We found no evidence for inter-patch movement for nine forest species, but 22 of 225 recaptured individuals of Poecilus versicolor, an eurytopic open-habitat species, had crossed the highway. Catches of seven forest species were also significantly lower in the road verges, compared to the adjacent forests. These two studies suggest that (i) decreasing patch size negatively affects forest-carabid catch and overall species richness, (ii) habitat type can affect the intensity of the patch-size effect, (iii) carabid assemblages of forest fragments vary with traffic volume (which may be linked with urbanization), (iv) forest carabids rarely cross highways, and (v) open habitats associated with road margins are dispersal barriers for forest carabids.  相似文献

Canine pituitary dwarfism in German Shepherd and related dog breeds has been reported to be associated with a 7-bp deletion mutation in intron 5 of the LHX3 gene. This mutation is transmitted as an autosomal recessive trait that results in dwarf dogs with significantly smaller stature and abnormal haircoat, and potentially early death. Phenotypically, affected adult dogs are proportionally dwarfs. These dwarfs also have a soft, woolly puppy coat that fails to transition into the typical adult hair coat, and marked hair loss occurs in some dogs. We report a similar manifestation of dwarfism in Tibetan Terriers with the same LHX3 mutation. Dwarf Tibetan Terrier puppies were born physically normal but failed to gain weight or to grow at the same rate as their normal littermates. The 7-bp deletion mutation of the LHX3 gene was identified in both alleles of 3 Tibetan Terrier dwarfs from 3 litters, which were biologically related. All parents of these dogs are carriers, confirming transmission of dwarfism in an autosomal recessive manner. Recognition and detection of this mutation will help in guiding future breeding plans to eventually eliminate this trait from Tibetan Terriers.  相似文献

Primary particulate matter is emitted directly into the atmosphere from various anthropogenic and natural sources such as power plants (combustion of fossil fuels) or forest fires. Secondary particles are formed by transformation of SO2, NOx, NH3, and VOC in the atmosphere. They both contribute to ambient particulate matter concentrations, which may have adverse effects on human health. Health hazards are caused by small particulate size, high number of especially fine (< 2.5 µm) and ultra-fine (< 0.1 µm) particles and/or their chemical composition. As part of an integrated assessment model developed at IIASA, a module on primary particulate matter (PM) emissions has been added to the existing SO2, NOx, NH3 and VOC sections. The module considers so far primary emissions of total suspended particles (TSP), PM10 and PM2.5 from aggregated stationary and mobile sources. A primary PM emission database has been established. Country specific emission factors for stationary sources have been calculated within the module using the ash content of solid fuels.  相似文献
