A series of random copolyesters having various compositions were synthesized by bulk copolymerization of bishydroxyethyl terephthalate (BHET) with 1,4-cyclohexane dimethanol (CHDM) or dimethyl isophthalate (DMI). CHDM and DMI content was less than 10 wt%. For the synthesized copolyesters, isothermal crystallization rate, melting behavior, and equilibrium temperature were investigated by calorimetry and by Avrami and Hoffman-Weeks equation. Crystalline lattice and morphology were studied by WAXD and SEM. Regardless of the composition, the value of the Avrami exponent was about 3, which indicates that crystallization mechanism of the copolyester was similar to those of PET homopolymer. Incoporation of CHDM or DMI units in PET backbone decreased the crystallization rate of the copolyesters. Surface free energy of copolyesters was evaluated using the newly proposed equation. The value of surface free energy was about 189×10−6 J2/m4 regardless of comonomer contents. This result is in good agreement with that of PET homopolymer.  相似文献

A 14‐wk feeding trial was carried out to evaluate the optimum dietary ascorbic acid (AA) level in juvenile sea cucumber, Apostichopus japonicus. Sea cucumbers averaging 1.49 ± 0.07 g (mean ± SD) were randomly distributed into 18 rectangular plastic tanks of 20 L capacity in a recirculating system (20 animals per tank). Six semipurified experimental diets were formulated to contain 0 (l ‐ascorbyl‐2‐monophosphate [AMP]; AMP0), 30 (AMP24), 60 (AMP48), 120 (AMP100), 240 (AMP206), and 1200 (AMP1045) mg AA/kg diet in the form of AMP using casein as the main protein source. Sea cucumbers were fed each of the six experimental diets in triplicate groups. At the end of 14 wk of feeding trial, weight gain (WG), specific growth rate (SGR), and feed efficiency (FE) of sea cucumbers fed AMP100, AMP206, and AMP1045 were significantly (P < 0.05) higher than those of animals fed AMP0, AMP24, and AMP48. However, there were no significant differences in WG, SGR, and FE among sea cucumbers fed AMP100, AMP206, and AMP1045 and among animals fed AMP0, AMP24, and AMP48. Whole‐body vitamin C concentration increased with AA content of the diets. Broken‐line analysis of WG showed an optimum dietary AA level of 105.3 mg AA/kg diet in sea cucumber. These results indicated that the optimum dietary vitamin C level in sea cucumber in the form of AMP could be greater than 100 mg AA/kg diet but less than 105.3 mg AA/kg diet.  相似文献

Eyestalk ablation is often used to induce ovarian development in female shrimp, but it possibly alters the animal’s hormonal balance, and affects its physiology and immune system. Therefore, this study was conducted to investigate alterations to the immune response during induced ovarian development by unilateral and bilateral ablation in Litopenaeus vannamei. Sub-adult female shrimp could be induced to undergo ovarian development over a 20-day period by both unilateral and bilateral eyestalk ablation; however, ovaries did not reach the final mature stage in which cortical rods are observed. In terms of immune response, total hemocyte count decreased significantly at 4 h after unilateral ablation, and at 20 days after bilateral ablation. After 20 days, expression in the hemocytes of the immune-related genes prophenoloxidase and peroxinectin in bilaterally ablated shrimp increased significantly, whereas that for penaeidin, for lipopolysaccharide- and β-glucan-binding protein, and for transglutaminase mRNA expression did not change significantly. The results of this study suggest that, while eyestalk ablation induces ovarian development, it may also bring about a hormonal imbalance that leads to immune-related genes being activated.  相似文献

ABSTRACT Potato early dying (PED), also known as Verticillium wilt, caused by Verticillium dahliae, is a seasonal yield-limiting disease of potato worldwide, and PED-resistant cultivars currently represent only a small percentage of potato production. In this study, we developed a real-time quantitative polymerase chain reaction (Q-PCR) approach to detect and quantify V. dahliae. The efficiency of the designed primer pair VertBt-F/VertBt-R, derived from the sequence of the beta-tubulin gene, was greater than 95% in monoplex Q-PCR and duplex (using Plexor technology) procedures with primers PotAct-F/PotAct-R, obtained from the sequence of the actin gene, designed for potato. As few as 148 fg of V. dahliae DNA were detected and quantified, which is equivalent to five nuclei. Q-PCR detected V. dahliae in naturally infected air-dried potato stems and fresh stems of inoculated plants. Spearman correlations indicated a high correlation (upward of 80%) between V. dahliae quantifications using Q-PCR and the currently used plating assays. Moreover, Q-PCR substantially reduced the variability compared with that observed in the plating assay, and allowed for the detection of V. dahliae in 10% of stem samples found to be pathogen free on the culture medium. The described Q-PCR approach should provide breeders with a more sensitive and less variable alternative to time-consuming plating assays to distinguish response of breeding lines to colonization by V. dahliae.  相似文献

Stem cell based cell therapies offer significant potential for the field of regenerative medicine. Human amniotic fluid stem cells (hAFSCs) are an attractive source for lineage-specific differentiated stem cell therapy since they have properties that are able to differentiate into cells representing all three germ layers. To better understand the fate and location of implanted hAFSCs, a means to monitor cells in living subjects is essential. Here, we showed that differentiated cells, such as neurogenic, endothelial, and myogenic cells, derived from hAFSCs can be effectively labeled by the FITC-incorporated silica-coated nanoparticles, MNPs@SiO2 (FITC), although the labeling efficacy and cytotoxicity were distinct depending on the differentiated cell type. In addition, we observed that MNPs@SiO2-labeled cells provided sufficient signals for detection by optical and confocal microscope imaging when transplanted into the mice. These results suggest that the fluorescent dye incorporated MNPs@SiO2 are a useful tool for the cell labeling and in vivo tracking of differentiated cells derived from hAFSCs.  相似文献

A large-scale outbreak of Akabane viral encephalomyelitis in cattle was reported in the southern part of Korea in 2010. Fifteen Akabane virus (AKAV) strains were isolated from the brain and spinal cord samples by using BHK-21 and/or HmLu-1 cells. To examine the genetic relationships and characteristics of the isolates, nucleotide sequences of the S, M, and L segments of the 15 isolates were determined and analyzed. Complete sequence analysis of the 15 AKAV isolates showed 99.9-100% amino acid identities, indicating that the 15 isolates originated from a single strain. The S and M RNA segments of a representative isolate (AKAV-7/SKR/2010) were also compared with the segments of representative reference sequences. This AKAV-7/SKR/2010 strain showed the highest identity with the Iriki and KM-1/Br/06 strains. Neighbor-joining phylogenetic trees of S and M RNA segments were constructed. Four representative AKAV isolates were classified into subgroup Ia, which contains the Iriki and KM-1/Br/06 strains recognized to cause encephalomyelitis in calves and adult cattle in Japan. Moreover, experimental intraperitoneal infection was performed using the AKAV-7/SKR/2010 and AKAV-17/SKR/2010 strains to assess pathogenesis in suckling mice. The 2 isolates, genetically related to the Iriki strain, were neurovirulent and caused neurological signs in suckling mice. In contrast, the 93FMX strain and the K0505 strain, related to the OBE-1 strain, were avirulent in mice. The present results indicate that these isolates most likely had originated from the Iriki strain and are closely related to the Iriki strain both genetically and pathogenically.  相似文献

Two miniature Schnauzer dogs with chronic pancreatitis were investigated. Both dogs showed systemic hypertension and increased concentrations of triglycerides and C-reactive protein. Abdominal radiography revealed cylindrical calcification in the retroperitoneum, and computed tomography (CT) showed extensive calcification of the abdominal and peripheral arteries in both dogs. Metastases and other dystrophic conditions that can cause arterial calcification were excluded based on the laboratory tests, and the dogs were diagnosed with atherosclerosis ante mortem. Atherosclerosis should be considered when extensive arterial calcification is observed on abdominal radiography or CT in miniature Schnauzers.  相似文献

X-chromosome inactivation (XCI) is an epigenetic process that equalizes expression of X-borne genes between male and female eutherians. This process is observed in early eutherian embryo development in a species-specific manner. Until recently, various pluripotent factors have been suggested to regulate the process of XCI by repressing XIST expression, which is the master inducer for XCI. Recent insights into the process and its regulation have been restricted in mouse species despite the evolutionary diversity of the process and molecular mechanism among the species. OCT4A is one of the represented pluripotent factors, the gate-keeper for maintaining pluripotency, and an XIST repressor. Therefore, in here, we examined the relation between OCT4A and X-linked genes in porcine preimplantation embryos. Three X-linked genes, XIST, LOC102165544, and RLIM, were selected in present study because their orthologues have been known to regulate XCI in mice. Expression levels of OCT4A were positively correlated with XIST and LOC102165544 in female blastocysts. Furthermore, overexpression of exogenous human OCT4A in cleaved parthenotes generated blastocysts with increased XIST expression levels. However, increased XIST expression was not observed when exogenous OCT4A was obtained from early blastocysts. These results suggest the possibility that OCT4A would be directly or indirectly involved in XIST expression in earlier stage porcine embryos rather than blastocysts.  相似文献

Transitional cell carcinoma (TCC) is the most common malignant tumor of the canine urinary tract. In this case study, a dog with metastatic urethral TCC was treated with sorafenib. The tumor expression levels of receptor tyrosine kinase genes, including VEGFR-1, VEGFR-2, PDGFR-α, PDGFR-β, ALK, EGFR, ErbB2, and B-RAF, were analyzed. VEGFR was overexpressed in tumor tissues compared to the normal tissues. Considering the high frequency of B-RAF mutation in canine urological tumors, the B-RAF gene was examined, and the B-RAF V595E mutation was detected in the tumor tissue. Therefore, the antitumor effect of sorafenib, a multi-tyrosine kinase inhibitor, on unresectable metastatic urethral TCC characterized by B-RAF V595E was evaluated and circulating cell-free tumor DNA (ctDNA) was assessed for monitoring the treatment response. After the initiation of oral sorafenib therapy (4 mg/kg/day escalated to 10 mg/kg/day), the dysuria was alleviated gradually, and the patient remained stable for 3 months. During that treatment period, the patient showed various levels of changes associated with B-RAF V595E mutation in ctDNA as evident from longitudinal plasma samples after initiation of sorafenib therapy. The findings of this study suggest that ctDNA may serve as a useful non-invasive tool for monitoring the treatment response to anticancer drugs.  相似文献
