Here we explore the significance of fungal diversity on ecosystem processes by testing whether microfungal ‘preferences’ for (i.e., host recurrence) different tropical leaf species increases the rate of decomposition. We used pairwise combinations of γ-irradiated litter of five tree species with cultures of two dominant microfungi derived from each plant in a microcosm experiment. The experiment was designed to test whether early leaf decomposition rates differed depending on relationships between the leaf litter from which the fungi were derived (i.e., the source plant) and the leaf substrata decomposed by these fungi in microcosms. Relationships tested were phylogenetic relatedness between the source and substratum leaves, and similarity in litter quality (lignin, N and P) between the source and substratum. We found a significant interaction between microfungi and leaf species (P<0.0001), and differences among the four classes of source–substratum relationships were highly significant (P=0.0004). Combinations in which fungal source leaves were of the same species or family as the substratum, or the fungal source resembled the substratum in quality had marginally faster decomposition than when the fungal source and substratum leaves were mismatched (i.e., unrelated and of dissimilar quality). In some microcosms, a basidiomycete contaminant had a strong additive effect on decomposition of Croton poecilanthus leaves resulting in faster decomposition than with microfungi alone (P<0.0001). Comparisons among leaf–microfungal combinations were made after the effect of the basidiomycete covariate was adjusted to zero. The data on microfungi suggest differential abundance in particular hosts, which contributes to species diversity of decomposer fungi in tropical forests, affects rates of decomposition.  相似文献

The iron-reducing activity of coffee beverages was determined by the ferric reducing antioxidant power (FRAP) assay. The influence on FRAP due to the degree of roasting (light, medium, and dark), species (Coffea arabica and Coffea robusta), and caffeine content (regular and decaffeinated) was investigated using ground and soluble coffee samples. The concentration of specific chlorogenic acids and caffeine in the beverages was determined by high-performance liquid chromatography and related to FRAP using Pearson correlation coefficients. All measurements were expressed per unit of soluble solids. Beverages prepared with ground coffee had, on average, 27% higher FRAP values than those prepared with soluble coffee (p < 0.05). In the former beverages, FRAP of C. robusta samples was significantly higher (on average, 50.3%) when compared to that of C. arabica samples, and FRAP values decreased with increasing degree of roasting (p < 0.05). A strong correlation (r > 0.91) was found between FRAP and the total content of chlorogenic acids, particularly that of the caffeoylquinic acid isomers. The iron-reducing activity of coffee beverages was not influenced by caffeine.  相似文献

Finkel-Biskis-Reilly murine sarcoma virus (FBR-MuSV) ubiquitously expressed (FAU) gene is down-regulated in human prostate, breast and ovarian cancers. Moreover, its dysregulation is associated with poor prognosis in breast cancer. Sponges (Porifera) are animals without tissues which branched off first from the common ancestor of all metazoans. A large majority of genes implicated in human cancers have their homologues in the sponge genome. Our study suggests that FAU gene from the sponge Suberites domuncula reflects characteristics of the FAU gene from the metazoan ancestor, which have changed only slightly during the course of animal evolution. We found pro-apoptotic activity of sponge FAU protein. The same as its human homologue, sponge FAU increases apoptosis in human HEK293T cells. This indicates that the biological functions of FAU, usually associated with “higher” metazoans, particularly in cancer etiology, possess a biochemical background established early in metazoan evolution. The ancestor of all animals possibly possessed FAU protein with the structure and function similar to evolutionarily more recent versions of the protein, even before the appearance of true tissues and the origin of tumors and metastasis. It provides an opportunity to use pre-bilaterian animals as a simpler model for studying complex interactions in human cancerogenesis.  相似文献

This study examined the impact of oxidative stress indicated by thiobarbituric acid reactive substances (TBARS) and protein carbonyl (PC), induced by intensive exercise and cadmium chloride (CdCl2) on ethoxyresorufin-O-deethylase (EROD) activity in juvenile carp (Cyprinus carpio). In the first experiment, fish were divided into three groups: (1) control, (2) carp exposed to intensive exercise, and (3) carp that was not exercised but previously, as well as carp in group 2, received single dose of 3-methylcholantrene (3-MC). The third and sixth day fish were sacrificed and the measurements were conducted. In the second experiment, fish were divided into (1) control, (2) carp in water containing CdCl2, and (3) carp in dechlorinated tap water (2 and 3 received single dose of 3-MC on the seventh day after exposure to CdCl2). The carp were killed 6?days later and livers were excised for biochemical analyses. In the first experiment, on the sixth day after treatment with 3-MC, results show statistically significant increase in EROD activity in non-exercised carp, while that increase in carp exposed to intensive exercise was significantly lower. Three days after exposure to 3-MC, statistically significant increase in TBARS was observed in both exercised and non-exercised carp. Six days after exposure to 3-MC, PC levels were significantly higher in exercised carp. Pretreatment with CdCl2, in the second experiment, caused oxidative stress and reduction of EROD activity. Results show linkage between expression of EROD activity and oxidative stress biomarkers and possible influence of oxidative stress on the cell membrane structures and consequently on EROD activity.  相似文献

The effects of different simulated rainfall regimes on water relations, growth, and biomass production and allocation of five provenances of Cedrela fissilis, C. saltensis and C. balansae were assessed in a pot-grown seedling experiment in greenhouse under uncontrolled conditions. Four simulated rainfall regimes were applied: 600 mm year?1 (severe water deficit), 800 mm year?1 (mild water deficit), 1,000 mm year?1 (mild wet), and 1,200 mm year?1 (well-watered) used as control. Provenances were compared in terms of water relations (midday leaf water potential, Ψ md and relative water content, RWC), growth (root, shoot and leaf length, neck diameter), growth rate (shoot- and neck growth rate, SGR and NGR), biomass production (root, shoot and leaf dry weight), and biomass allocation (root:shoot ratio, RSR). Multivariate analyses revealed that water relations differed significantly under all simulated rainfall treatments by means of several variables throughout time. Effects of species × treatment, and provenance nested within species × treatment interactions significantly were important. SGR and RSR also were important to explain differences in Ψ md and RWC between treatments and provenances. Broadly C. balansae and C. saltensis provenances seem to display a better performance than C. fissilis provenances under severe water deficit only. However, despite differences between provenances, it was not possible to clearly separate them according to their origin, based on water relations and growth performance. Even though the results of this study performed on pot-grown seedlings are based on relatively few observations and require additional confirmation, they allow an opportunity for speculation on the factors that are related to intraspecific variations that are likely to occur during the growth of seedlings from different geographical sites in field condition.  相似文献

Chloroplastic and cytoplasmic alfalfa (Medicago sativa) leaf protein concentrates were tested as dietary replacements for fish meal in practical diets for tilapia fingerlings. Each concentrate was used to replace 15, 25, 35, 45 and 55% of the fish meal protein in diets containing 40% protein. Diets containing cytoplasmic leaf protein gave the best results, with growth rates higher than those obtained with a fish meal-based diet when the plant protein replaced up to 35% of the fish meal protein in the diet; higher inclusion levels of plant protein depressed growth. No mortalities were recorded during the 9-week test period with any of the experimental diets. It was concluded that the alfalfa leaf protein could be included at levels of up to 35% of the dietary protein in feeds for tilapia.  相似文献

Infection of American and European chestnuts with the chestnut blight fungus Cryphonectria parasitica results in the formation of cankers, lesions caused by the growth of mycelia within bark tissue of the host plant. Infection of the fungus with Cryphonectria hypovirus 1 (CHV‐1) results in conversion of the mycelial phenotype from virulent to hypovirulent, thus allowing production of callus around cankers as a reaction by infected trees, rendering active into inactive cankers. In this study, we sampled one USA and six European chestnut stands and assessed frequency of hypovirulent C. parasitica and diversity of vegetative compatibility (vc) types present in calluses and randomly sampled cankers. Callused cankers on C. dentata at West Salem in the USA yielded significantly more hypovirulent C. parasitica isolates compared with four sampled populations on C. sativa, while all six sampled European populations did not show any statistically significant differences among themselves. We observed no correlation between hypovirulence frequencies in randomly sampled cankers and calluses, as well as no correlation of C. parasitica vc type diversity in calluses and residential populations of the fungus. Furthermore, even though we have observed calluses with more than one vc type, they do not occur regularly. Even when present in C. parasitica populations with high vc type diversity, no more than three different vc types were observed in a single callus.  相似文献

The effect of the drying temperature on the volatile components and sensory acceptance of the Roselle (Hibiscus sabdariffa) extract in powder was investigated. The Roselle extraction was carried out by maceration with 7 L of 30% ethanol (v/v), 560 g of fresh Roselle calyces for 168 h. The Roselle extracts were spray dried at different temperatures 150, 160, 170, 180, 190, 200 and 210 °C, giving different outlet values about yield and final moisture. The volatile compounds in Roselle extract and dried samples were performed using needles of solid phase microextraction (SPME) and gas chromatography/mass spectrometry (GC/MS HP-5890). Twenty volatile compounds were identified in the extracts among them terpenoids, esters, hydrocarbons and aldehydes. Fourteen volatile compounds were identified in the powder sample, but only ten were present in the Roselle extract. This indicates that some compounds were lost and some others were generated due to a degradation process. An acceptability sensory analysis showed that the best powder sample was the Roselle extract dehydrated using temperature between 190 °C and 200 °C (p < 0.05). There was not statistically significant difference in the pH of Roselle extracts ranging from 3.4 to 3.9. It was concluded that the spray drying temperature of the Roselle extracts has an effect on the volatile compounds losses.  相似文献

Structural characteristics of starches have been important to determine their physicochemical and functional properties. Solubilization procedures were tested to find a higher solubilization percentage and thereafter to study the structural characteristics of amylose and amylopectin. Size‐exclusion chromatography with refractive index (SEC‐RI) system using a pullulan standard curve was tested to study the amylose molar mass. Also, a microbatch system using a MALLS detector was used to determine the molar mass and gyration radius of starch and amylopectin. Microwave heating produced higher solubility percentages than autoclaving, and there was a difference between both starches. The sample solubilized with microwave heating presented higher molar mass and gyration radius values than autoclave samples, showing that this process for structural studies provided information representative of the initial starch sample. When starch components were separated, amylose showed lower purity than amylopectin. Lower purity was obtained for amylose separated from barley starch, but no difference was obtained for purity of amylopectin separated from both starches. Barley amylopectin had a higher solubility percentage than maize amylopectin. Molar mass of barley amylose was 1.03 × 105 g/mol and for maize of 2.25 × 105 g/mol. Molar mass values of amylopectin separated from both starches were lower than the starch counterparts, although the same solubilization procedure (microwave heating) was used. The difference might be due to depolymerization during separation of starch components.  相似文献
