The dynamics of acetanilide pesticide interactions with organic matter (OM) surrogates were examined using nuclear magnetic resonance (NMR) spectroscopy. Differences in the relative changes in (13)C and (1)H spin-lattice relaxation times (T(1)) were measured at multiple molecular sites of metolachlor and the probe compound acetanilide to identify interaction sites and/or surfaces between the molecules and dissolved and colloidal OM surrogates. The decrease in T(1) at specific sites of acetanilide molecules was a function of the OM used and its concentration. High-affinity interactions at nonaromatic sites of metolachlor and acetanilide were observed with cellulose, chitin, and collagen, but interactions with lignin occurred with less site specificity and involved both aromatic and nonaromatic sites of the molecules. Changes in relaxation were compared to calculated and experimentally determined binding coefficients (K(oc)). The T(1) relaxation of the aromatic sites of acetanilides showed better relations with K(oc) than the nonaromatic sites. This study shows that NMR relaxation measurements can identify the high-affinity molecular interaction sites of acetanilides to OM surrogates.  相似文献

The results provided evidence to support the following statements: (a) the assemblage pattern is not homogeneously distributed across the four Mediterranean ecoregions studied (biotic gradients hypothesis); and (b) the assemblage pattern does not change significantly when the information is aggregated to higher taxonomic levels (taxonomic sufficiency hypothesis).
  • 3. Surrogate taxonomic categories higher than species, such as genus and family, can be used to reveal the multivariate pattern of the coralligenous assemblages.

Landscape connectivity, defined as the degree to which the landscape facilitates or impedes movement among resource patches, has been considered to be a key issue for biodiversity conservation. However, the use of landscape connectivity measurements has been strongly criticised due to uncertainties in the methods used and the lack of validation. Moreover, measurements are typically restricted to the population level, whereas management is generally carried out at the community level. Here, we used satellite imagery and network metrics to predict the landscape connectivity at community level for semi-natural herbaceous patches in an urban area near Paris (France). We tested different measurement methods, both taking into account and ignoring the spatial heterogeneity of matrix resistance estimated by the normalised difference vegetation index (NDVI), and quantifying the link strength between patches with the shortest path and flow metrics. We assessed the fit of these connectivity predictions with empirical data on plant communities embedded in an urban matrix. Our results indicate that the best fit with the empirical data is obtained when the connectivity is estimated with the flow metric and takes into account the matrix heterogeneity. Overall, our study helps to estimate the landscape connectivity of urban areas and makes recommendations for ways in which we might optimise landscape planning with respect to conservation of urban biodiversity.  相似文献

This study evaluated, for the first time, the selection of antibiotic resistance in fecal Escherichia coli, a potential reservoir of genes of resistance, during the prolonged exposure to fluoroquinolones after the implantation of a local drug delivery system (LDDS) in a swine model. Fourteen pigs were randomly assigned to group IM (5 mg/kg/day of intramuscular enrofloxacin--EFX) or LD (surgical implantation of EFX-polymethyl-methacrylate peri-femoral implants). Blood samples were collected daily for determination of plasma EFX and ciprofloxacin (CFX) concentrations. Fecal samples were collected daily to determine the E. coli counts and the susceptibility patterns of its isolates as evaluated by antibiotic disk diffusion tests. In both groups, EFX administration significantly reduced the bacterial counts after 2 days. During recolonization, the bacterial counts remained lower than baseline in group IM but not significantly, and almost reached pre-treatment levels in group LD. Susceptibility to EFX, CFX, and nalidixic acid of recolonizing E. coli in LD pigs slightly decreased but remained within the limit of "susceptible" isolates. In contrast, quinolone susceptibility of recolonizing E. coli in IM pigs dropped dramatically (P < 0.0001). In addition, intramuscular exposure to fluoroquinolones significantly decreased the susceptibility of E. coli to ampicillin and trimethoprim-sulfamethoxazole (P < 0.05). In conclusion, the use of a dosing regimen that minimized the intestinal output of fluoroquinolones also minimized the selection of resistance to several classes of antibiotics. This could represent another advantage of LDDS usage compared to long-lasting systemic administration of fluoroquinolones.  相似文献

Though the ovine stifle is commonly used to study osteoarthritis, there is limited information about the age‐related morphometric changes of the tidemark. The objective of this study was to document the number of tidemarks in the stifle of research sheep without clinical signs of osteoarthritis and of various ages (n = 80). Articular cartilage of the medial and lateral tibial condyles and of the medial and lateral femoral condyles was assessed by histology: (a) to count the number of tidemark; and (b) to assess the OARSI (Osteoarthritis Research Society International) score for structural changes of cartilage. The number of tidemarks varied between anatomical regions, respectively, from 4.2 in the medial femoral condyle to 5.0 in the lateral tibial condyle. The axial part showed a significant higher number of tidemarks than the abaxial part, for all regions except the medial tibial condyle. Whilst the tidemark count strongly correlated with age (Spearman's correlation coefficient = 0.70; 95% confidence interval (95% CI): 0.67–0.73; p < 0.0001), the OARSI score was weakly correlated with age in our cohort of sheep (Spearman's correlation coefficient = 0.25; 95% CI: 0.19–0.30; p < 0.0001). Interestingly, no tidemark was seen in the three animals aged 6 months. Our data indicate that the number of tidemarks increases with age and vary with anatomical region. The regional variation also revealed a higher number of tidemarks in the tibia than in the femur. This could be attributed to the local variation in cartilage response to strain and to the difference in chondrocyte biology and density.  相似文献
