Three hundred and seven genotypes belonging to four cultivated and one wild species of Capsicum were screened against pepper leaf curl virus (PepLCV) causing devastating leaf curl disease of chilli (Capsicum annuum). Initial screening was done under field conditions based on coefficient of infection (CI), disease reaction to each genotype was assigned. Subsequently, selfed progenies of eight symptom-less and highly resistant lines were challenged by viruliferous white fly under glasshouse conditions, out of which only three genotypes, viz. GKC-29, BS-35 and EC-497636 showed no symptom. Using scion and root stalk of susceptible genotype (Pusa Jwala), these three putative symptom-less genotypes were further challenged by grafting and alternate grafting. The resistant reactions of GKC-29, BS-35, EC-497636 were confirmed because even after 50 days of successful grafting/alternate grafting, no viral symptom appeared on all the grafted plants of these genotypes. When subjected to PCR amplification with degenerate primers deigned to detect gemnivirus like PepLCV, the three symptom-less genotypes did not show any amplification, suggesting that the resistant reaction in three identified symptom-less resistant sources was because of the absence of viral genome and they are not symptom-less carrier.  相似文献

Equine babesiosis, a tick transmitted haemoprotozoan disease caused by Theileria equi is globally distributed and responsible for heavy economic losses to the equine husbandry. Equids reared in endemic areas usually pick up infection at an early age and become immune tolerant throughout their life span. We studied the level of passively transferred antibodies in neonate foals born from pre-immuned mares. Latently T. equi infected pre-immuned pony and donkey mares (three each) were selected and T. equi antibody titres in neonates was monitored till 90 days post foaling (DPF) by applying Dot-ELISA on sequentially collected serum samples from foals and their dams. A very high antibody titre was observed in pre-immuned pony and donkey mares. The maximum antibody of 1:60 to 1:80 was observed in pony's and donkey's foal on 2–16 and 2–10 DPF, respectively and thereafter it declined to less than 1:20 on 63–77 and 56–63 DPF. Simultaneously parasite carrying status in neonate foals and their dam was also monitored by applying PCR on blood samples. We could demonstrate PCR amplification in dam's blood samples while no amplification was recorded in neonate's blood samples. This study indicated that new-born foals were born naïve and passively transferred immunity was transitory which wanes after 63–77 DPF.  相似文献

In the present study, a novel antigenic protein expressed in the piroplasm stage of Theileria orientalis was characterized. A 4,707 bp genomic fragment amplified by PCR contained two open reading frames (ORFs). The deduced amino acid sequence of the first ORF showed significantly high similarlity to the ubiquitin carboxy terminal hydrolases/proteases while the second ORF (To ORF2) showed homology to several surface antigens of plasmodia. To ORF2 was expressed to determine whether the protein product is expressed by the parasite. In western blot analysis, bovine antiserum from a T. orientalis-infected calf recognized the recombinant protein containing a C-terminal part of the ORF expressed by baculovirus system. Western blot analysis with the anti-To ORF2 mouse serum recognized a 48 kDa protein in T. orientalis piroplasm lysates. Indirect immunofluorescence antibody test by confocal scanning laser microscopic analysis showed that antisera against the recombinant protein recognized T. orientalis piroplasm in the infected erythrocyte. The results from this study indicate that To ORF2 protein is expressed at the piroplasm stage and is immunogenic. This novel antigenic To ORF2 protein could be exploited for vaccine development against bovine piroplasmosis.  相似文献

Juri is a biodiversity-rich primary forest in Bangladesh, which remains ecologically unexplored. We identified tree species and examined the richness, alpha(a)diversity and floristic similarity patterns within the identified communities. Vegetation and environmental data were sampled in 120(0.04 ha) study plots. Tree communities were delimited by two-way indicator species analysis(TWINSPAN). In total, 78 tree species of 35 families and58 genera were identified. TWINSPAN identified six tree communities: A—Tricalysia singularis; B—Kydia calycina-Castanopsis tribuloides; C—Polyalthia simiarum-Duabanga grandiflora; D—Ficus roxburghii; E—Artocarpus lacucha; F—Artocarpus lacucha. Mean richness, Shannon and Gini-Simpson indices were highest for the Polyalthia simiarum-Duabanga grandiflora community, while Ficus roxburghii showed lowest diversity. Significant differences(p = 0.05) in three diversity indices were recorded between Polyalthia simiarum-Duabanga grandiflora and Ficus roxburghii. Tree compositional similarity was greatest between Kydia calycina-Castanopsis tribuloides and Polyalthia simiarum-Duabanga grandiflora(0.712).  相似文献

Iron (Fe) is one of the essential micronutrient plays a crucial role in redox systems in cells and in various enzyme. The Fe-deficient plants have lower redox reaction and enzymatic activity. Intensive chemical fertilizer resulted in depletions of micronutrients especially Fe from the soil. To understand the dynamics of Fe under long-term fertilizer experiment the present study was initiated during 1972 at experimental farm of College of Agriculture CSK HPKV, Palampur in randomized block design with eleven treatments replicated thrice. The soil of the experimental area was silty loam, acidic in reaction and taxonomically classified as “Typic Hapludalfs.” Surface (0–20 cm) and subsurface (20–40 cm) soil samples collected after the harvest of maize (Kharif, 2008). The continuous use of chemical fertilizers and amendments for the last 36 years in maize-wheat system in an acid Alfisol brought out marked depletion in the pools of Fe as compared to buffer plots. All the pools of Fe were noticeably higher in farmyard manure (FYM) amended plots compared to zero-fertilized plots. Although, residual fraction was found to be the most dominant fraction but organically bound and exchangeable forms were found to play major role in the nutrient supply and crop productivity and nutrients’ uptake. Highest productivity of maize (2008) and wheat (2008–2009) was recorded under 100% nitrogen, phosphorus and potassium (NPK) + FYM treatment. FYM and lime treatments resulted in significantly higher uptake of all the nutrients by both the crops compared to other treatments. Organically bound fraction was found to have highest significant and positive correlation with yield and nutrient uptake of maize and wheat crops. Further, regression analysis studies too revealed that organic form was the most important pool contributing towards the variation in the above parameters. While, exchangeable and organically bound forms contributed significantly towards diethylene triamine pentaacetic acid (DTPA)-extractable Fe.  相似文献

Recent advances in desert afforestation underlines its viability and importance in combating global warming and acidification. In this paper, the inter-relation between afforestation, global warming and acid rain has been analyzed. Numerical simulations indicate that afforestation of deserts has distinct advantage as carbon sink and as an important factor for changing microclimate of the region rather than a source of energy. Acidic deposition may well be utilised as fertiliser in nutrient deficit soil of tropical arid areas. However, past trends and projections of acidic deposition in arid areas adjacent to Thar deserts indicate an early efforts are required to cap the opportunity. Delays may contribute towards more incidences of failures.  相似文献

The effects of increasing levels of metals (10 and 20 mg of Cr kg-1 and 25 and 50 mg of Cd, Pb, and Ni kg-1 soil) and arbuscular mycorrhizal (AM) fungi Glomus intraradices on the yield, chemical composition of volatile oil, and metal accumulation in sweet basil (Ocimum basilicum L.) were investigated in a pot experiment. The shoot yield, content of essential oil, and root yield of sweet basil were increased by the application of low dose of Cd, Pb, and Ni as compared to control. The application of high level of metals had deleterious effect on the yield. In soil with low dose of metal applied, AM fungi inoculation significantly enhanced the metal concentration in shoots and had adverse effect on the yield, whereas in soil with high dose of metal applied, AM fungal inoculation reduced the metal concentration in shoot and had beneficial effect on the yield. The content of linalool in basil oil was decreased and that of methyl chavicol was increased by the application of Cr, Cd, and Pb in soil as compared to control. Similarly, the level of linalool and methyl chavicol was decreased and that of methyl eugenol was increased by the application of Ni as compared to control. However, AM fungal inoculation led to maintain the content of linalool, methyl chavicol, and methyl eugenol in volatile oil, which were either increased or decreased by the application of metals. We conclude that the AM–sweet basil symbiosis could be used as a novel approach to enhance the yield and maintain the quality of volatile oil of sweet basil under metal-contaminated soils.  相似文献
