利用BIOLOG和磷脂脂肪酸(PLFA)技术,研究了受足尾铜矿废水影响下的日本西那须地区渡良濑游水地东、西两侧土壤生物学特性、微生物群落结构及功能变化.结果表明:东侧(对照区)土壤全铜含量为45.07mg/kg,有效态铜含量为0.42 mg/kg,同西侧相比,平均降低了19.32%和89.95%.土壤呼吸速率主要受土壤有效态铜、土壤有机质和C/N的影响,随土层深度增加,土壤微生物量氮和溶解性氮都迅速减小,东侧土壤具有较强的氮矿化能力,渡良濑游水地东、西两侧0-5 cm土层土壤的微生物群落生理代谢剖面在培养48 h后出现明显变化;土壤微生物群落的平均颜色变化值(AWCD)随培养时间的延长,趋于平缓增加.在48 h时培养后,西侧位点土壤的AWCD值与东侧相比,分别下降了40%,34%和16%;除B位点样品在20-40 cm土层深度的AWCD略高于其它3个位点外,其余样品的AWCD无明显差异.渡良濑游水地东侧土壤微生物对单一碳源底物的利用能力较高,土壤微生物群落利用碳源的功能多样性也较高.渡良濑游水地西侧区域C位点土壤的格兰仕阳性菌、格兰仕阴性菌、放线菌数量均显著高于东侧A位点,但渡良濑游水地东侧区域土壤3大类群微生物中细菌和放线菌比例更高.在渡良濑游水地自然恢复30多年后,东、西两侧土壤微生物群落结构与功能仍存在较明显的差异.  相似文献

This study examined the effects of contact with wood on the living human body using a physiological index and subjective evaluation. Consecutive blood pressure measurements were used as the physiological index, and sensory evaluation using the semantic differential (SD) method was used for subjective evaluation. Consideration was also given to cases in which materials were cooled and heated as well as kept at room temperature, to eliminate the effects of heat flux due to differences in thermal conductivity between wood and other materials. It was found that contact with wood produced coarse/natural sensations, with no associated increase in systolic blood pressure. Contact with cold wood created subjectively dangerous/uncomfortable but still coarse/natural sensations, also with no associated increase in blood pressure; therefore, there was no correspondence between subjective evaluation and physiological responses. Contact with aluminum kept at room temperature and cold acrylic plastic created flat/artificial and dangerous/uncomfortable sensations, with an associated significant increase in blood pressure; thus, there was a close correlation between subjective evaluation and physiological responses. It was therefore concluded that contact with wood, unlike artificial materials such as aluminum, induces no physiological stress even when kept at room temperature or cooled. Thirty‐four Holstein dry cows and 16 lactating cows were used in balance trials to identify the effects of feeding and animal performance on nitrogen (N), phosphorus (P) and potassium (K) excretion by dairy cows, and to develop prediction models for these excreta. Orchard grass silage, corn silage, alfalfa silage or timothy hay were offered to dry cows. Orchard grass silage or alfalfa silage, and concentrates were offered to lactating cows. In the statistical analysis, the independent variables were bodyweight (kg), dry matter (DM) intake (kg/day), milk yield (for lactating cows only, kg/day), water intake (free water plus water in feed, kg/day), intake (g/day) of N, P and K and dietary contents (% of DM) of crude protein, P and K. The dependent variables were N, P and K excretion (g/day) in feces and urine. In both dry and lactating cows, intake of N, P and K had large effects on corresponding excretion. The results indicated that a decrease in the intake of N, P and K could decrease the corresponding excretion. Further research by path analysis showed that K intake positively affected urinary N excretion in dry cows indirectly, through water intake and urine volume.  相似文献

To investigate the pathological role of staphylococcal enterotoxins (SEs) and toxic shock syndrome toxin-1 (TSST-1) in bovine mastitis, the production of SEs and TSST-1 was investigated in staphylococci isolated from 120 mammary gland secretions (MGS, 51 from no clinical sign-mammary glands and 69 from staphylococcal mastitic-mammary glands) collected from dairy farms where staphylococcal mastitis frequently occurred in Miyagi and Yamagata prefectures from 1997 to 1998. Concentrations of these toxins and specific antibody titers in each MGS were also measured. Furthermore, SEC and TSST-1 were inoculated into lactating mammary glands and inflammatory responses were analyzed. A high percentage of staphylococci including Staphylococcus aureus and coagulase-negative staphylococci isolated from both no clinical sign- and mastitic-MGS produced both SEC and/or TSST-1. The concentration of SEC increased with the severity of the mastitis, and was significantly higher (P<0.05) in acute mastitic-than in no clinical signs-MGS. Titers of specific antibodies to TSST-1 in MGS were significantly higher (P<0.05) than those to SEC, regardless of whether or not the cows were lactating or mastitic. Specific antibodies purified from MGS neutralized each toxin in vitro. A significant increase (P < 0.05) in somatic cell counts was induced by the intramammary inoculation of SEC but not TSST-1. These findings indicated that SEC rather than TSST-1 plays an important role in the pathology of staphylococcal bovine mastitis. The inflammatory activity of TSST-1 was probably neutralized by specific antibodies in MGS.  相似文献

OBJECTIVE: To evaluate the effects of a high-protein diet versus dietary supplementation with ammonium chloride (NH4Cl) on struvite crystal formation in the urine of clinically normal cats by measuring the urine concentration of hydrochloric acid (HCl)-insoluble sediment, urine pH, struvite activity product (SAP), number of struvite crystals in urine, and urine volume. ANIMALS: 23 healthy adult cats. PROCEDURE: Urine was fractionated by centrifugation with subsequent extraction of the sediment with 1 N HCl (study 1). Diets containing either 29% crude protein or 55% crude protein were fed to cats in a crossover trial of 3 weeks/period (study 2). Diets supplemented with either sodium chloride (NaCl) or NH4Cl were fed, by use of a 3 x 3 Latin-square design with 3 wk/period (study 3). In studies 2 and 3, urine samples were collected for the last 7 days of each period. RESULTS: The HCl-insoluble sediment contained Tamm-Horsfall glycoprotein (THP; study 1). The high-protein diet (study 2) and dietary supplementation with NH4Cl (study 3) resulted in a decrease in urine pH, SAP, and the number of struvite crystals in urine. However, the high-protein diet decreased urine concentrations of HCl-insoluble sediment containing THP (study 2), in contrast to the NH4Cl supplementation that increased urine volume without a significant effect on the urine concentration of the HCl-insoluble sediment (study 3). CONCLUSIONS AND CLINICAL RELEVANCE: Our results indicate that compared with dietary supplementation with NH4Cl, the high-protein diet is preferable as a urine acidifier for the prevention of struvite crystal formation in clinically normal cats.  相似文献

The inhibitory effects of 102 methanol extracts of 40 mycelia, 9 culture fluids, and 53 fruiting bodies of 40 strains of Ganoderma lucidum on 5-reductase were investigated. The methanol extract of the fruiting body of each strain was found to show the strongest 5-reductase inhibitory activity among the extracts tested.  相似文献

Comparative virulence of blue-stain fungi isolated from Japanese red pine   总被引:2,自引:0,他引:2  
 We made inoculations to determine the comparative virulence of blue-stain fungi isolated from Japanese red pine (Pinus densiflora Sieb. & Zucc.). Based on the symptoms caused by each fungus, the most virulent fungus was Leptographium wingfieldii. Also, Ophiostoma minus was relatively virulent while lesions induced by some species did not differ from those in the control treatment. Based on the results, we concluded that the pathogenicity of the blue-stain fungi varied widely, and thus their impact on the host most likely varies too. Relationships between tree mortality and bark beetle infestation onAbies veitchii at the wave-regenerated forest in Mt. Asahi, Okuchichibu area were investigated. Most of the firs with green needles and newly developed current year’s shoots in the dieback zone were heavily infested by bark beetles before the death of the trees. After heavy infestation of beetles, about half of the infested firs died within the year, and the other half died in the next year. When the species composition of bark beetles and associated ophiostomatoid fungi were investigated in Mt. Asahi and also at a typical wave-regenerated forest in Mt. Shimagare, Yatsugatake area,Cryphalus montanus andC. piceae were dominant beetle species for Mt. Asahi and Mt. Shimagare, respectively.Ophiostoma subalpinum andO. europhioides were dominant fungal species at both wave-generated forests. BecauseO. subalpinum was detected more frequently from deeper areas of sapwood thanO. europhioides, it was suggested that the fungal species may accelerate the death of stressed firs in wave-regenerated forests.
