Abstract

Many studies have evidenced the negative effects of synthetic fertilizers toward the environment and also the crop products itself. Hence, the improvement of organic fertilizers is necessary to resolve this problem. Seaweeds or marine macroalgae are considered to be an excellent natural resource in different aspects of agricultural fields. Brown macroalgae Turbinaria murayana (TM) is often found in Indonesian coastal areas with high bioavailability throughout the year. This study evaluated the potentials of TM as biofertilizer in tomato plants (Lycopersicum esculentum Mill.). Evaluation of phytohormones present in TM crude extract was conducted with high performance liquid chromatography (HPLC). Macro and micronutrients available in TM crude extract were determined by atomic absorbance spectroscopy (AAS). A concentration of 4% TM crude extract was applied to tomato plants. The vegetative properties of the treated tomatoes were observed such as growth and yield. Current results evidenced the presence of growth-promoting factors (kinetin, gibberellin and ABA) and essential elements (N, P, K, Ca, Fe and Mn) in TM crude extract. Application of 4% TM extract to tomato plants significantly increased vegetative growth compared to untreated plants; plant height, leaf number and branch number, approximately 58.4%, 87.5% and 200%, respectively. Similar results were also found in the response of generative growth. Application of 4% liquid extract significantly improved flower and fruit number compared to control; 53.6% and 125%, respectively. These results suggest that TM crude extract is a potential source for developing organic fertilizers.  相似文献

This study evaluated the possible use of the fish SSN-1 cell line to investigate the development of Macrobrachium rosenbergii nodavirus (MrNV). Cells were incubated with viral particles and cytopathic effects were observed. De novo synthesis of viral capsid proteins was shown by immuno-fluorescence labelling and a sandwich ELISA test. Viral genomic replication was demonstrated by RT-PCR using primers specific to RNA-1 as well as by quantitative RT-PCR (RT-qPCR). Using electron microscopy, only a few empty particles were observed and attempts to isolate complete infectious particles or to re-infect healthy cells (second passage) were unsuccessful. As complete viral particles were rarely observed, it appeared that defaults in MrNV virogenesis might arise resulting in the formation of scarce and non-infectious particles. SSN-1 cells were found to be partially permissive to MrNV infection that induced cell lysis, but key elements for viral infection were lacking such as regulatory factors for gene replication or post-translational modifications.  相似文献

We investigated the feasibility of using several fast-growing tropical or subtropical hardwood species for timber production by measuring key wood qualities in relationship to the high rates of lateral growth. The trees tested were sampled from even-aged plantations of Acacia mangium, A. auriculiformis, hybrid Acacia (A. mangium × A. auriculiformis), Eucalyptus grandis, E. globulus, and Paraserianthes falcataria (Solomon and Java origin) that had already reached commercial harvesting age. The released strain of the surface growth stress (RS), xylem density (XD), microfibril angle (MFA), and fiber length (FL) were measured at the outermost part of the xylem at breast height in each tree. Results were then compared to the lateral growth rate (radius/age) at breast height, which provides a relative indicator of the amount of tree growth per year. Our findings indicated that RS was constant, regardless of lateral growth rate in each species. Similar results were observed for XD, MFA, and FL, with a few exceptions, suggesting that high growth rates do not intrinsically affect the wood properties of fast-growing tropical or subtropical species that have reached harvesting age. However, special attention must be paid to patterns of xylem maturation when developing plantations of such species.  相似文献

Koi herpesvirus specifically infects and causes mass mortality on koi and carp, resulting in severe economic losses. In this study, we presented the efficacy of KHV DNA vaccine administration by immersion method on Cyprinus carpio. Two different immersion densities of fish were applied, namely 800 fish L?1 and 1200 fish L?1. Thirty‐day‐old common carp juveniles were immersed for 30 min in the water containing 1.3 × 108 CFU mL?1 of heat‐killed Escherichia coli carrying DNA vaccine encoding glycoprotein‐25, and without vaccination treatment as controls. The challenge test was performed at 30 days post vaccination by injecting 0.1 mL KHV filtrate (10?3 of dilution rate). The result showed that higher relative per cent survival of KHV‐challenged fish was obtained in 800 fish L?1 (< 0.05). Furthermore, significant specific antibody anti‐KHV response (< 0.05) was detected on 28 and 36 days post vaccination in 800 and 1200 fish L?1, respectively, compared to the controls there was no specific antibody detected. In conclusion, the KHV DNA vaccine could provide good protection in common carp against KHV infection, which has practical applications in aquaculture practices.  相似文献

The cocoa industry in Sulawesi, the main region of cocoa production in Indonesia, is threatened by destructive diseases, including vascular-streak dieback (VSD) caused by the basidiomycete Oncobasidium theobromae and stem canker and Phytophthora pod rot (PPR) or black pod, caused by Phytophthora palmivora. Using the considerable genetic diversity of cocoa on farms, host resistance was identified and tested with the participation of farmers. Forty-nine local and international cocoa selections with promising resistance characteristics (as well as susceptible controls) were side-grafted onto mature cocoa in a replicated trial with single-tree plots. Developing grafts were assessed in the dry season for severity of VSD infection, scored from 0 (no infection) to 4 (graft death). All of the 49 clones in the trial became infected with VSD in at least some replicates. Average severity varied from 0.2 to 1.6. Potential VSD-resistance was found in eight clones, including DRC 15, KA2 106 and a local Sulawesi selection, VSD2Ldg. Some of the most susceptible clones were local Sulawesi selections from areas with a history of little or no VSD. Thirty-four pod-bearing clones were evaluated over a 2-year period for yield, quality and resistance to natural infections of PPR. Cumulative PPR incidence for all clones was 22% but varied from 8.6 to 43% among clones. Clones with less than 15% PPR incidence were designated as resistant, including DRC 16 and local Sulawesi selections, Aryadi 1, Aryadi 3 and VSD1Ldg. Scavina 12 was moderately resistant in the trial with a PPR incidence of 23%. Cumulative incidences of the mirid, Helopeltis spp., determined in the same evaluation period, indicated that DRC16 was the most susceptible clone with an incidence of 52% in ripe pods and 23% in immature pods. In comparison, KKM4 showed evidence of resistance to Helopeltis spp., with incidences of 34 and 0.8% in ripe and immature pods, respectively. The impact of diseases and pests (including cocoa pod borer) on bean losses and bean quality varied between clones but generally the bean size (or bean count) was affected more than the fat content or shell content.  相似文献

Severe mosaic, yellowing and stunting symptoms were observed on petunia (Petunia hybrida L.) growing in pots at NBRI and in various gardens of Lucknow, India. The association of Cucumber mosaic virus (CMV) with the mosaic disease was detected based on positive bioassay on susceptible hosts, isometric cored virus particles of ~28?nm during electron microscopic observations in leaf dip preparations and positive amplification of expected size (~650?bp) during RT-PCR using coat protein gene specific primers. Further, the complete RNA 3 genomic fragment of virus isolate was amplified by RT-PCR using RNA 3 specific primers. The obtained amplicons of ~2.2 Kb were cloned and sequenced. The analysis of sequence data of RNA 3 revealed highest sequence identities (96%) with several CMV strains which belong to subgroup IB. The virus isolate also showed closest phylogenetic relationships with banana strain of CMV of subgroup IB (Acc. EF178298) reported from India. To the best of our knowledge, we report the first molecular characterization of CMV strain of subgroup IB causing severe mosaic disease on petunia in India.  相似文献

The impact of four coniferous tree species and their corresponding soil factors on N transformation rates and presence of ammonia-oxidising bacteria (AOB) was studied in an acid pine forest soil (Appelscha, The Netherlands). Pine soil had a relatively low net nitrification rate, while spruce, fir and larch soils showed high net nitrification rates. 16S rRNA and amoA sequences were only found in soils with high nitrification rates and belonged solely to Nitrosospira cluster 2. We conclude that tree species, possibly through their effects on soil C/N ratios, determines the presence of Nitrosospira cluster 2. Whenever AOB are present, however, the AOB community composition appears to be similar.  相似文献

White tail disease of the farmed freshwater prawn, Macrobrachium rosenbergii, is the cause of mortalities in the French West Indies, China and India. Two different sized particles, both developing in the cytoplasm of target cells, are found associated with diseased animals. These two viruses were separated, purified and subsequently characterized. The larger one, called MrNV, is icosahedral in shape and 27 nm in diameter. Its genome is composed of two fragments of linear single-stranded RNA (ss-RNA), of 2.9 and 1.3 kb, respectively and its capsids exhibited a single polypeptide of 43 kDa. These characteristics and the partial sequence of a cloned fragment of RNA-1 suggest this agent is a member of the family Nodaviridae, but with differences from both the genera Alphanodavirus and Betanodavirus. The smaller virus, named XSV, is icosahedral in shape, 15 nm in diameter, possesses a linear ss-RNA genome of about 0.9 kb, and its capsid exhibits two polypeptides of 16 and 17 kDa, respectively. The relationships between these two viruses remain unknown.  相似文献
