Congenital vitelline duct anomalies other than Meckel’s diverticulum are rare in animals. A cyst of approximately 8 mm in diameter was observed on the antimesenteric surface of the ileal serosa in a 10-week-old female Crl:CD(SD) rat. Microscopically, the cyst closely resembled the ileum, but it did not communicate with the ileal lumen. We diagnosed this case as a vitelline cyst derived from the vitelline duct based on the location where it developed and its histological behavior. In rats, only Meckel’s diverticulum has been reported with a congenital anomaly of the vitelline duct, and no other spontaneous anomalies including a vitelline cyst have been reported. This case may be the first report concerning a vitelline cyst in the rat ileum.  相似文献

Consideration of the shortened fattening period seems to be worthwhile for the realization of profitable beef production. In this study, change of fatty acid composition of the lumbar longissimus during the final stage of fattening was investigated in Japanese Black cattle. Each of 110 fattening animals was sampled three times: the initial two samples were taken by biopsy (25.7 months and 27.5 months on average) and the final one was from carcasses (29.9 months on average). Preliminary analysis indicated that removing muscle tissues from the constant body position of the living animals should be essential for sampling. Average monounsaturated fatty acids (MUFA) at three sampling points were 58.1%, 58.5% and 60.5%, and the differences of the third sampling with the first and second samplings were significant. Both in steers and heifers, MUFA also increased as the fattening stage proceeded, and MUFA of the heifers at all the sampling points were significantly higher than those of the steers. The increasing rate of MUFA rose from 0.21 percentage points (pp)/month at period 1 (from the first sampling to the second sampling) to 0.84 pp/month at period 2 (from the second sampling to the slaughter).  相似文献

Free amino acids are important components of tastants and flavor precursors in meat. To clarify the correlation between muscle fiber type and free amino acids, we determined the concentrations of various free amino acids and dipeptides in samples of different muscle tissues (n = 21), collected from 26‐month‐old Japanese Black steers (n = 3) at 2 days postmortem. The proportions of the myosin heavy chain (MyHC), slow (MyHC1) and fast (MyHC2) isoforms were determined by sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE). The contents of free amino acids and dipeptides were measured by high performance liquid chromatography (HPLC). The MyHC isoform composition varied among the tissue samples. The MyHC1 proportion ranged from 6.9% ± 3.9% to 83.3% ± 16.7%. We confirmed that there was a strong positive correlation between MyHC1 composition and total free amino acid concentrations, including those for two dipeptides. Among the 31 measured free amino acids and dipeptides, 11 showed significant positive correlations and five showed significant negative correlations with MyHC1 composition. These results suggest that a high MyHC1 content induces high free amino acid contents in bovine muscles possibly because of greater oxidative metabolism. This high level of free amino acids could contribute to the intense flavor of meat that is rich in slow‐twitch fibers.  相似文献

Typical skeletal muscles are composed of mixed muscle fiber types, which are classified as slow-twitch (type I) and fast-twitch (type II) fibers, whereas pectoralis major muscles (PMs) in broiler chickens are 100% composed of type IIb fast-twitch fibers. Since metabolic properties differ among muscle fiber types, the combination of muscle fiber types is involved in physiological functions and pathological conditions in skeletal muscles. In this study, using serial block-face scanning electron microscopy, we compared three-dimensional (3D) mitochondrial properties in type IIb fibers in broiler PMs and those in type I fibers of broiler gastrocnemius muscles (GMs) heterogeneously composed of slow- and fast-twitch muscle fibers. In type I fibers in the GMs, elongated mitochondria with numerous interconnections to form a substantial network among myofibrils were observed. Along with lipid droplets sandwiched by mitochondria, these features are an adaptation to effective oxidative respiration and constant oxidative damage in slow-twitch muscle fibers. In contrast, type IIb fibers in the PMs showed small and ellipsoid-shaped mitochondria with few interconnections and no lipid droplets, forming a sparse network. The mitochondrial spatial network comprises of active mitochondrial dynamics to reduce mitochondrial damage; therefore, type IIb fibers possess physiologically low capacity to maintain mitochondrial wellness due to static mitochondrial dynamics. Based on 3D mitochondrial properties, we discussed the contrasting physiological functions between type I and IIb fibers and proposed a high contractile power and low stress resistance as unique physiological properties of broiler PMs.  相似文献

Scallops (family Pectinidae) comprise species of high commercial value, supporting both commercial fisheries and mariculture activities. Accurate and reliable molecular methods for the species level identification are of outstanding utility for taxonomic and food authentication surveys. The mitochondrial 16S rRNA gene has been used to design species‐specific primers for identification of different bivalve species. However, the low interspecific variability at the 3′ end of this gene has limited its utility and only few scallop species have been assessed. In this study, we used the high variable 5′ end of the 16S gene to develop a novel decaplex PCR assay that enabled a fast and accurate identification of eight commercially important scallop species in a single PCR reaction. A total of 285 individuals including fresh and manufactured samples from eight different processed presentations from 11 different scallop species were collected representing diverse locations around the world. Our assay accurately identified all the analysed samples at the species level. Furthermore, to enhance the utility of our assay, the PCR product amplified by the family specific primer set that was utilized as positive control was also used for the identification of unknown (non‐target) scallop species by DNA sequencing analysis. In its present form, our multiplex PCR method can be of great utility for different types of studies involving scallop species and for research institutes and governmental agencies that regulate seafood authentication around the world.  相似文献

Laboratory experiments were firstly conducted to determine the possibility of modification of the collapsible pot for blue swimming crab Portunus pelagicus by designing appropriate escape vents in terms of shape, position, and size to improve size selectivity by reducing the catch of immature crabs. In laboratory observations, nearly square-shaped vents of 35 mm height and 45 mm length, located at the lower part of the side panel, showed the best performance to allow the escape of immature-size crabs by side-crawling escape behavior through the vents, with L 50% selection carapace length (CL) of 39.1, 44.4, and 48.7 mm CL for 40, 45, and 50 mm vent length, respectively. Comparative fishing trials between conventional and vented pots with vents of 35 × 45 mm size were conducted to examine the crab size and species selectivity. Use of the vented pots was found to reduce the number of immature crabs in the catch from 70.5% to 11.0% on average, while not affecting the catch efficiency of mature-size crabs. The vented pots also had a selective effect for reducing the catch of bycatch species in comparative fishing trials.  相似文献

The effect of temperature on the swimming performance of jack mackerel Trachurus japonicus was examined in a flume tank by measuring the swimming endurance time and heart rate. The lower swimming performance was observed at 10°C (the lowest temperature tested), manifesting as the shortest endurance time and the slowest maximum sustained speed. ECG measurements of the heart rate under free-swimming conditions at zero flow velocity revealed a temperature effect, with 25.3 beats/min observed at 10°C, 38.9 at 15°C, and 67.2 at 22°C. The heart rate also increased with swimming speed to maximum levels of 60, 125, and 208 beats/min, respectively, at these three temperatures. Heart rate recovery times measured after the fish had been swimming at prolonged speed tended to increase with temperature, while a negative correlation resulting in relatively short recovery times was observed after swimming at close to the burst swimming speed at each water temperature.  相似文献

Morphological changes in the cytoskeleton, nuclei, and vacuoles were monitored during the cell death of short-lived ray tracheids in the conifer Pinus densiflora. After formation of the dentate thickenings that occurred at the final stage of formation of cell walls, organelles started to disappear in differentiating ray tracheids. First, the microtubules and vacuoles disappeared. Then actin filaments disappeared in the differentiating ray tracheids adjacent to ray tracheids that lacked nuclei, and, finally, the nuclei disappeared. These features indicate that cell death in ray tracheids might differ from the programmed cell death of tracheary elements that has been studied in vitro in the Zinnia culture system.
