A reversed passive latex agglutination test (RPLA) using anti-canine haemoglobin (Hb) antibody was developed for detecting bleeding in the lower digestive organs in dogs, and its applicability as a simple test for faecal occult blood was assessed. In Ouchterlony's gel immunodiffusion test, the anti-canine Hb antibody used to sensitize the latex reacted with canine Hb but not with Hbs, plasmas or meat extracts from pigs, goats, sheep, cattle, horses or chickens, or with fish extracts. Using latex sensitized with 50 µg/mg of anti-canine Hb IgG antibody, the lowest limit of detection for canine Hb was 21 µg/ml, and the latex reacted negatively with all test specimens other than canine Hb. In an in vitro experiment with a mixture of canine faeces and erythrocytes, the antigenicity of the Hb was found to undergo only very slight changes even when the specimens were allowed to stand for 12 h at room temperature. Hb could not be detected by RPLA in any of four successive faecal samples from three experimental dogs after infusion of autologous blood (5, 3 or 1 ml) into the stomach. In 3 other experimental dogs given an infusion of autologous blood (5, 3 or 1 ml) into the ascending colon, the presence of Hb was confirmed by RPLA in all four successive faecal samples obtained from those which received 5 or 3 ml of blood and in all except that obtained following the first defecation from the animal which had received 1 ml of blood.  相似文献

Pollen of garden varieties and species of Delphinium that was cryopreserved at –30 °C after 3 hours of air drying and storage on silica gel, had high germination and pollen tube growth in vitroeven after 180-day storage. On the other hand, pollen stored at 25 °C showed a marked decrease in the germination rate within 10 days. The best in vitro germination of Delphinium pollen was on a 1% water agar containing 15% sucrose and 0.005 to 0.01% boric acid and at 15 to 20 °C. Field pollination with the cryopreserved pollen showed higher fruit and seed set than pollination with pollen stored at 25 °C, and was not significantly different from fresh pollen. We examined the effects of heavy-ion bombardment on mutagenesis in sweet pepper (Capsicum annuum L.). Dose–response studies indicated that 10 Gy irradiation of 12C or 20Ne ions on dry seeds is suitable for inducing mutations in plants. From 20Ne-irradiated M1 plants, putative mutants included two dwarf plants and one plant whose pericarp was yellow were isolated. Phenotypes of their M2 progeny were similar to those of the M1 plants and did not segregate. F1 plants resulting from reciprocal crosses between the mutants and wild-type plants showed the wild-type phenotype, but phenotypes of F2 and BC1F1 segregated at 1:3 (mutant:wild) and 1:1, respectively. These crossing experiments indicate that the three mutants have monogenic recessive mutations in nuclear genes. In light of these data, we discuss the effectiveness of using heavy-ion bombardment to mutate sweet peppers.  相似文献

Observations of air pollutants were conducted at Chengshantu in January 1996 to clarify the extent of trans-boundary pollution from the Asian continent. A nested air quality prediction modeling system (NAQPMS), which included parameters on emission, transport, diffusion, deposition and transformation of sulfur oxides, was performed to compile sulfur concentrations over the observation site. The model calculation reproduced the observed variations of sulfur oxides and sulfate well, although the model calculations could not reproduce the extensively low sulfate concentrations. Using nesting improves the ability of the modeling system to capture peak episodes seen in the observations. Calculation of the origins of deposited sulfur in each country with the NAQPMS shows that the long-range transport of sulfur is very serious. For Japan, 49% of sulfur deposition was from other countries during the campaign period.  相似文献

A series of crocetin glycosides (crocins) are the main pigment of the stigmas of saffron (Crocussativus L.) and the fruits of gardenia (Gardenia jasminoides Ellis). Although numerous studies have demonstrated that crocetin and crocins have a variety of biological functions, the metabolism of dietary crocetin and crocins remains unknown. In the present study, we investigated the intestinal absorption of orally administered crocetin and crocins in mice. Orally administered crocetin was rapidly absorbed into the blood circulation and was present in plasma as an intact free form and as glucuronide conjugates (crocetin-monoglucuronide and -diglucuronide). Crocetin and its glucuronide conjugates were also found in crocins-administered mouse plasma, whereas intact crocins (glycoside forms) were not detected. These results indicate that orally administered crocins are hydrolyzed to crocetin before or during intestinal absorption, and absorbed crocetin is partly metabolized to mono- and diglucuronide conjugates.  相似文献

To prevent health problems of humans exposed to dioxin, it is important to enhance the fecal excretion of dioxin stored in the body. The effects of seaweed such as wakame, hiziki, and kombu on the gastrointestinal absorption and reabsorption of 17 types of polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated dibenzofuran (PCDF) congeners was investigated in Wistar rats. Rats were fed 4 g of the basal diet or a seaweed diet containing PCDD and PCDF standard solution [233 ng of toxic equivalents (TEQ)/kg of body weight] once during the experiment period. In the group fed the 10% wakame diet, the levels of fecal excretion of PCDD and PCDF congeners were higher (p < 0.01) from days 1 to 5 by 2.8-fold for 2,3,7,8-TCDD, by 4.0-fold for 1,2,3,7,8-pentaCDD, by 3.4-fold for 1,2,3,4,7,8-hexaCDD, by 3.2-fold for 1,2,3,6,7,8-hexaCDD, by 2.5-fold for 1,2,3,7,8,9-hexaCDD, by 1.7-fold for 1,2,3,4,6,7,8-heptaCDD, by 1.1-fold for 1,2,3,4,6,7,8,9-octaCDD, by 3.0-fold for 2,3,7,8-tetraCDF, by 3.7-fold for 1,2,3,7,8-pentaCDF, by 3.7-fold for 2,3,4,7,8-pentaCDF, by 3.2-fold for 1,2,3,4,7,8-hexaCDF, by 3.0-fold for 1,2,3,6,7,8-hexaCDF, by 3.2-fold for 1,2,3,7,8,9-hexaCDF, by 2.9-fold for 2,3,4,6,7,8-hexaCDF, by 1.6-fold for 1,2,3,4,6,7,8-heptaCDF, by 2.2-fold for 1,2,3,4,7,8,9-heptaCDF, and by 1.2-fold for 1,2,3,4,6,7,8,9-octaCDF than those of the basal group, respectively. Rats were fed 4 g of the basal diet containing PCDD and PCDF standard solution (2991 ng of TEQ/kg of body weight) once on day 1 and then place on the basal diet for 7 days. After 1 week, the rats were fed either the basal diet or seaweed diet from days 8 to 35. In the group fed the 10% wakame diet, the levels of fecal excretion of PCDD and PCDF congeners were higher (p < 0.01 or p < 0.05) during the period from days 8 to 35 by 1.7-fold for 2,3,7,8-TCDD, by 1.8-fold for 1,2,3,7,8-pentaCDD, by 2.0-fold for 1,2,3,4,7,8-hexaCDD, by 1.9-fold for 1,2,3,6,7,8-hexaCDD, by 1.6-fold for 1,2,3,7,8,9-hexaCDD, by 1.5-fold for 1,2,3,4,6,7,8-heptaCDD, by 2.0-fold for 2,3,4,7,8-pentaCDF, by 2.1-fold for 1,2,3,4,7,8-hexaCDF, by 1.9-fold for 1,2,3,6,7,8-hexaCDF, by 1.7-fold for 2,3,4,6,7,8-hexaCDF, by 1.5-fold for 1,2,3,4,6,7,8-heptaCDF, and by 1.9-fold for 1,2,3,4,7,8,9-heptaCDF than those of the basal group, respectively. These findings suggest that the administration of seaweed such as wakame is efficient in preventing the absorption and reabsorption of dioxin from the gastrointestinal tract and might be useful in treatment of humans exposed to dioxin.  相似文献

To obtain basic information about enzymatic deterioration of buckwheat flour, triacylglycerol lipase (LIP; EC 3.1.1.3) was purified from buckwheat seed. The LIP consisted of two isozymes, LIP I and LIP II, and they were purified with purification folds of 60 and 143 with final specific activities of 0.108 and 0.727 mumol of fatty acid released per minute per milligram of protein at 30 degrees C using triolein as a substrate. Molecular weights were estimated to be 150 (LIP I) and 28.4 kDa (LIP I) by gel filtration and 171 (LIP I) and 26.5 kDa (LIP II) by SDS-PAGE. Optimal pHs of LIP activities were 3.0 (LIP I) and 6.0 (Lip II) using triolein as a substrate. Both LIP I and II reacted in the acidic pH range. Optimal temperatures were 30 (LIP I) and 40 degrees C (LIP II), and both LIP I and II were stable below 30 degrees C when p-nitrophenyl-laurate was used as a substrate. However, they were inactivated above 60 degrees C. On the other hand, when triolein was used as a substrate, optimal temperatures were 30 degrees C for both LIP I and II, and they retained 40% of their activity after a 4 h incubation of enzymes at 70 degrees C. LIP I and II had higher activity against triolein than monoolein or tri/monopalmitin. Most of the LIP activity was distributed in the embryo.  相似文献

A 28-yr-old female California sea lion (Zalophus californianus) in a commercial aquarium developed an ulcerated lingual tumor and died. Necropsy revealed a moderately differentiated squamous cell carcinoma. Immunohistochemical staining and electron microscopic studies revealed that the tumor cells were strongly positive with anti-keratin-cytokeratin antibody and had abundant tonofibrils and desmosomes. The neoplasm had metastasized to a mandibular lymph node.  相似文献

Bovine herpesvirus 1 (BHV-1) isolates from respiratory tract and from vagina of bovine in Japan were analyzed by SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and the DNA restriction endonuclease cleavage pattern, and compared with European BHV-1 strains. Both protein profile and DNA cleavaged pattern of BHV-1 isolates from respiratory tract were the same as those of European infectious bovine rhinotracheitis (IBR) virus, whereas the protein profile and DNA cleavage patterns of one isolate (M1) from vagina was the same as those of the European infectious pustular vulvovaginitis (IPV) virus. The facts indicate that IPV virus has existed in Japan.  相似文献

Establishment of a disposal plan for carcasses in advance is important for prevention of epidemics. A disposal plan for contaminated goods such as poultry carcasses infected with highly pathogenic avian influenza (HPAI) virus was studied in Hyogo Prefecture, Japan. We investigated all poultry farms with over 1,000 birds for their locations, species and numbers of birds, structure and size of poultry facilities and land spaces of the farms. Moreover, we judged whether they could dispose of all the carcasses at their farms. In 2005, 5.5 million layers and 2.7 million broilers were being kept. If HPAI had broken out, 44.0% of the farmers could bury all the carcasses, and 65.6% could compost them at their farms. However, 23.4% could not dispose of them except by burning them at incineration facilities. We decided to choose burning first for rapid disposal as long as the virus was not a pandemic type.  相似文献
