Incubation and pot experiments were conducted to investigate the impact of commercially distributed biofertilizers (effective microorganisms [EM], BIOSTIMULATOR, BACTOFIL‐A, and BACTOFIL‐B) on soil microbial‐biomass content and activity, net N mineralization in soil, and growth of Lolium perenne. According to the manufacturers, the products tested are based on microbial inoculants or organic growth stimulants, and are supposed to influence soil microbial properties and improve soil conditions, organic‐matter decomposition, and plant growth. In the incubation experiment (40 d, 20.6°C, 50% maximum water‐holding capacity), EM was repeatedly applied to soil together with different organic amendments (nonamended, chopped straw, and lupine seed meal). Under the experimental conditions of this study, no or only marginal effects of EM on organic C, total N, and mineral N in soil could be observed. In soil treatments without any organic amendment, EM suspension slightly enhanced microbial activity measured as soil CO2 evolution. In soil with easily degradable plant residues (lupine seed meal), EM suspension had a suppressive effect on microbial biomass. However, comparisons with sterilized EM and molasses as the main additive in EM suspension showed that any effect of EM could be explained as a pure substrate effect without the influence of added living organisms. In the pot experiment with Lolium perenne (air‐conditioned greenhouse cabin, 87 d, 16.8°C, 130 klxh d–1 light quantity), the products EM, BIOSTIMULATOR, BACTOFIL‐A, and BACTOFIL‐B were tested in soil with growing plants. The products were repeatedly applied for a period of 42 d. Within this study, no effects of the different biofertilizers on mineral N in soil were detectable. There were clear suppressive effects of all tested biofertilizers on microbial‐biomass content and activity. Comparisons with sterilized suspensions showed that the effects were not due to living microorganisms in the suspensions, but could be traced back to substrate‐induced processes.  相似文献

Leptospirosis is the most widespread zoonosis worldwide and is caused by serovars of pathogenic Leptospira species. The understanding of leptospiral pathogenesis lags far behind that of many other bacterial pathogens. Current research is thus directed at identification of leptospiral virulence factors. Saprophytic Leptospira species are environmental organisms that never cause disease. Comparative genomics of pathogens and saprophytes has allowed the identification of more than 900 genes unique to either Leptospira interrogans or Leptospira borgpetersenii; these genes potentially encode virulence-associated proteins. However, genes of unknown function are over-represented in this subset of pathogen-specific genes, accounting for 80% and 60% of open reading frames, respectively. This finding, together with the absence of virulence factor homologues among the proteins of known function, suggests that Leptospira possesses unique virulence mechanisms. Whole genome microarray studies have identified genes whose expression is differentially regulated under a range of simulated in vivo conditions, such as physiological temperature and osmolarity, low iron levels, and the presence of serum. The subset of genes identified by these studies is likely to include virulence factors. However, most such genes encode proteins of unknown function, consistent with the hypothesis that leptospiral virulence genes do not have homologues in other bacterial species. The recent development of mutagenesis systems for pathogenic Leptospira spp. has allowed the screening of defined mutants for attenuation of virulence in animal infection models and has identified definitively for the first time a range of virulence factors, including lipopolysaccharide, flagella, heme oxygenase, and the OmpA-family protein, Loa22. Interestingly, inactivation of a number of genes hypothesised to encode virulence factors based on in vitro virulence-associated properties did not result in attenuation of virulence, suggesting a degree of functional redundancy in leptospiral pathogenic mechanisms.  相似文献

A survey is given of methods to characterize the lowest three levels of starch structural features: individual chains, branched molecules, and the arrangement of branched molecules in a sample (e.g., crystalline and amorphous lamellae of starch granules in grain). The survey also covers ways of treating the results so as to understand starch structure–property correlations: for example, the structural characteristics that control the rate of digestion of a starch‐containing food. A number of studies are then examined not only to show how these techniques have been used to discover correlations between these structural characteristics and properties of importance but also to deduce reasonable causal explanations for the correlations. An overview of problems that have not yet been solved in each of these starch structural levels is also given. The applications of these characterization methods have considerable potential as tools to choose and process native starches with improved functional properties for human food, animal feed, and industrial uses, including biomaterials.  相似文献

Substantial progress was made in the last decade in understanding the diversity in Dioscorea species. However, most of the studies so far concentrated in the ‘yam belt in Africa’. We present a genetic diversity and structure analysis among commercial and local varieties of water yam (Dioscorea alata L.) in Brazil using microsatellite and morphological markers. Twelve microsatellite primers were used to generate DNA profiles of 72 local varieties and 17 commercial accessions of water yam collected in four different regions in Brazil. Also, four morphological traits were evaluated on individual plants under field conditions. The morphological characterization showed considerable diversity. High polymorphism was found with 100 % polymorphism observed for 11 primers and a discriminating power value of 0.92, on average. We did not observe a strong population structure among sampling regions, while most of the genetic diversity was concentrated within regions (95.9 %). Analysis of the relationship between accessions did not clearly separate the local and commercial genotypes. However, the molecular analyses of D. alata showed high intraspecific diversity in local accessions from different regions in Brazil, with the highest Shannon index value (H′ = 0.41) for the Southeast region. These results confirmed an admixture of accessions in all sampling regions, consistent with the lack of a significant correlation between geographic and genetic distances, suggesting that farmers exchanged water yam materials extensively. The genetic diversity can be explained by the result of a continuous exchange of accessions by farmers throughout Brazil.  相似文献

Several national parks on the African continent have been fenced to mitigate human–wildlife conflict, but the effectiveness of different fencing strategies seems to vary between regions. Here, we assessed the effectiveness of electro-fencing for restricting movement patterns of several wild ungulates in Akagera National Park (NP), Rwanda. Following the turmoil of the Rwandan genocide and civil war (1991–1995), large numbers of returning war refugees brought about an increasing pressure on land use, after which considerable parts of the western Akagera NP and adjacent Mutara Game Reserve were degazetted, followed by the construction of an electric fence in 2013 and a helicopter-based game-drive of ungulates into the modern park. We conducted distance sampling of wildlife and domestic livestock in the Uruhita plain – now no longer part of Akagera NP but bordering the modern Akagera NP to the west – for extended periods of time (2010–2015) before and after fencing. For abundant species population densities could be calculated, whereas for other species encounter frequencies are reported. We report on continuously high cattle densities, and increasing encounter frequencies of sheep/goats in the Uruhita plain. We found decreasing densities of zebra (Equus quagga) and decreasing encounter frequencies of waterbuck (Kobus ellipsiprymnus), stagnant encounter frequencies in the case of topi (Damaliscus lunatus jimela) and stagnant densities of impala (Aepyceros melampus). The latter species can jump fences, and we argue that competitive release and reduced predation may be the prime factors leading impala to (temporarily) exploit grasslands outside the modern national park.  相似文献

Phytosterols have been shown to reduce cholesterol absorption in humans. Supplementing phytosterols in fat-free formulations, however, has yielded controversial results. In the present study, we investigated the effect of supplementing test meals with different fat-free phytosterol products on cholesterol incorporation into mixed micelles during simulated digestion and accumulation of micellar cholesterol by Caco-2 cells: control orange juice (OJ), orange juice supplemented with either multivitamin/multimineral tablets (MVT), multivitamin/multimineral tablets containing phytosterols (MVT+P), and phytosterol powder (PP). These combinations were added to Ensure-based test meals and spiked with cholesterol of natural isotopic composition or 13C2-cholesterol to differentiate external from endogenous cholesterol. After simulated gastric/small intestinal digestion, micelle fractions were analyzed for cholesterol enzymatically (n = 6-20/product) and by high-performance liquid chromatography-tandem mass spectrometry (n = 12/product) and added to Caco-2 cells to determine the accumulation of 13C2-cholesterol (n = 10-24/product). As compared to OJ, PP and MVT+P significantly decreased cholesterol micellarization (determined enzymatically) by 70 +/- 39 (mean +/- SD) and 70 +/- 39%, respectively (P < 0.001, Bonferroni). The stable isotope experiments revealed that both PP and MVT+P reduced cholesterol micellarization [by 25 +/- 12 (P = 0.055) and 21 +/- 8% (P = 0.020), respectively, Fisher's protected LSD test] and Caco-2 cell accumulation (by 28 +/- 8 and 10 +/- 8%, respectively; P < 0.010, Bonferroni). OJ+P did not inhibit micellarization or accumulation of cholesterol by Caco-2 cells. This study shows that fat-free phytosterol-containing products can significantly inhibit cholesterol micellarization and Caco-2 cell bioaccessibility, albeit to different extents depending on individual formulations. This is most likely explained by inhibition of cholesterol micellarization.  相似文献

By amplifying the melanocortin type 1 receptor from the woolly mammoth, we can report the complete nucleotide sequence of a nuclear-encoded gene from an extinct species. We found two alleles and show that one allele produces a functional protein whereas the other one encodes a protein with strongly reduced activity. This finding suggests that mammoths may have been polymorphic in coat color, with both dark- and light-haired individuals co-occurring.  相似文献

The aim of the study was to determine the effects of a single social isolation (4 h) of piglets on immediate changes in stress hormones and immune responses at 7, 21 or 35 days of age. This social stressor caused an increase in plasma ACTH and cortisol concentrations and a decrease in plasma TNF-α. The percentage of CD8+ cells increased and the CD4+ cell percentage decreased, resulting in decreasing CD4+/CD8+ ratio. The observed changes were consistent for all days studied. Further, the isolation treatment resulted in diminished LPS-stimulated IL-1β and IL-10 production in whole-blood cultures. In isolated piglets, positive correlations were estimated between changes in percentage of CD8+ cells and cortisol, and negative ones between changes in plasma TNF-α and culture supernatant IL-1β with ACTH and cortisol. The data suggest that psychosocial stress in neonatal pigs induced immune alterations to maintain adaptive stability, but reflecting also negative emotions experienced by this treatment.  相似文献

The climatic changes on earth may have serious implications for the carbon (C) cycle in the terrestrial Arctic throughout the 21st century. Arctic vegetation takes up carbon dioxide (CO2) from the atmosphere producing biomass. In a cold and often moist soil environment, dead organic matter is preferentially preserved as soil organic matter (SOM) due to the inhibition of decomposition processes. However, viable soil microbes exhale huge amounts of CO2 and methane (CH4) annually. Hence, Arctic ecosystems exhibit annual fluxes of both carbon‐based (CO2 and CH4) greenhouse gases (GHGs) that are in an order of magnitude of millions of tons. Rising Arctic temperatures lead to the degradation of much of today's permafrost in the long run. As a result, large quantities of frozen SOM may become available for decomposers, and GHGs that are entrapped in permafrost may be released. At the same time, warming tends to stimulate the growth, development, and reproduction of many Arctic plants, at least transiently. The present northward migration of boreal shrubs and trees into southern tundra areas may be amplified by that, increasing the ecosystems' gross primary production and, thus, their C sequestration. On the other hand, rising temperatures boost SOM decomposition and microbial respiration rates. In general, soil temperature and soil moisture are key environmental variables to control the intensity of aerobic and anaerobic respiration by microbes, and autotrophic respiration by plants. On the basis of published data on Arctic CO2 and CH4 fluxes, the calculations on the terrestrial C‐based Arctic GHG balance made in this review reveal a current annual GHG exchange that ranges between a weak storage of ≤ 225 Tg CO2 equivalent (eq.) y–1 and a huge release of ≤ 1990 Tg CO2 eq. y–1. Hence, the Arctic GHG balance does apparently already contribute positively to the climatic changes at present. Regarding the future, the relative development of the uptake and release of CO2 and CH4 by northern ecosystems is fundamental to the overall GHG status of the Arctic under scenarios of continued climate change.  相似文献

An imbalance between formation and detoxification of oxygen radicals leads to oxidant stress that may increase in more intense oxidative metabolism caused by a high intake of metabolizable energy to provide metabolic intermediates for the milk synthesis and secretion. This hypothesis was tested using dairy cows and the concentration of hydroperoxides in lipids (LHP) extracted from circulative lipoprotein particles of low and very low density (LDL and VLDL/chylomicrons) as oxidant stress indicator. The particles were prepared by ultracentrifugation of serum obtained by coccygeal bleeding (13 cows, 1. parity, n=8 and 2. parity, n=5, lactation stage, 53 +/- 1.4 days post partum) and purified by precipitation. Concentrations of LHP-LDL/mg Lipoprotein correlated significantly with daily milk yield (r = 0.73, P = 0.004) or daily milk energy output (r = 0.77, P = 0.003) in contrast to LHP of VLDL/chylomicron particles. Thus, some evidence was obtained for an almost linear, positive relationship between milk productivity and oxidant stress occurring in LDL.  相似文献
