Objective

1) To determine the pharmacokinetics of tramadol hydrochloride and its active metabolite, O-desmethyltramadol (M1), after administration through different routes in female and male C57Bl/6 mice; 2) to evaluate the stability of tramadol solutions; and 3) to identify a suitable dose regimen for prospective clinical analgesia in B6 mice.

Study design

Prospective, randomized, blinded, parallel design.

Animals

A total of 18 male and 18 female C57Bl/6 mice (20–30 g).

Methods

Mice were administered 25 mg kg?1 tramadol as a bolus [intravenously (IV), intraperitoneally (IP), subcutaneously (SQ), orally per gavage (OSgavage)] over 25 hours [orally in drinking water (OSwater) or Syrspend SF (OSSyrsp)]. Venous blood was sampled at six predetermined time points over 4 to 31 hours, depending on administration route, to determine tramadol and M1 plasma concentrations (liquid chromatography and tandem mass spectrometry detection). Pharmacokinetic parameters were described using a noncompartmental model. The stability of tramadol in water (acidified and untreated) and Syrspend SF (0.20 mg mL?1) at ambient conditions for 1 week was evaluated.

Results

After all administration routes, Cmax was >100 ng mL?1 for tramadol and >40 ng mL?1 for M1 (reported analgesic ranges in man) followed by short half-lives (2–6 hours). The mean tramadol plasma concentration after self-administration remained >100 ng mL?1 throughout consumption time. M1 was found in the OSSyrs group only at 7 hours, whereas it was detectable in OSwater throughout administration. Tramadol had low oral bioavailability (26%). Short-lasting side effects were observed only after IV administration. Water and Syrspend SF solutions were stable for 1 week.

Conclusions and clinical relevance

1) At the dose administered, high plasma concentrations of tramadol and M1 were obtained, with half-life depending on the administration route. 2) Plasma levels were stable over self-consumption time. 3) Solutions were stable for 1 week at ambient conditions.  相似文献

Thermophilic Campylobacter spp. and Salmonella spp. are zoonotic bacteria, commonly harbored in the enteric tract of avian species. This survey aimed at verifying the presence of these microorganisms in a heterogeneous Italian population of pet birds and captive birds of prey (e.g., Passeriformes, Psittaciformes, Accipitriformes, Falconiformes, and Strigiformes) that live in close contact with humans. A total of 151 individuals were tested for thermophilic Campylobacter spp. and 444 for Salmonella spp. Six C. jejuni and one S. Livingstone were isolated from birds of prey: 4 Accipitriformes (2 Buteo regalis, 2 Parabuteo unicinctus), 1 Falconiformes (Falco peregrinus) and 1 Strigiformes (Strix aluco) were positive for C. jejuni, and 1 Falconiformes (Falco peregrinus) for S. Livingstone. Five C. jejuni strains were genotyped by Multilocus Sequence Typing (MLST) and identified as ST 2116, assigned to ST-353 CC. Results of this study suggest that these microorganisms are not common inhabitants of the digestive tract of pet birds and captive birds of prey, but occasional findings. However, as potential reservoirs of zoonotic bacteria, pet birds should be monitored in order to preserve human health.  相似文献

Prevalence of anti-Neospora caninum antibodies was measured in serum samples randomly collected from dairy (40 cows from four farms) and beef cattle (120 animals from 12 farms) from the municipality of Santarém, Pará State, Brazil, calculated by using the Win Episcope 2.0 statistical program. The presence of anti-N. caninum antibodies was determined by indirect immunofluorescence-antibody test with a cut-off value of 1:100. We found that 13 farms (81.25%) showed infection rates above 10%, which indicates widespread distribution of N. caninum in the region. The frequency per animal was 19%. No difference was observed between the prevalence values in dairy and beef animals or between farms, which was probably due to the small number of dairy farms examined. The results confirm, for the first time, the presence of anti-N. caninum antibodies in cattle from Pará State and the necessity to further investigate the epidemiology of N. caninum in the Amazon region.  相似文献

The aim of this study was to evaluate whether a normal yeast flora is present in the intestine of healthy foals and whether establishing yeast flora or yeast overgrowth might be associated with foal-heat diarrhea. Among the sampled foals, 49% showed foal-heat diarrhea and 28.3% were positive for yeasts. Candida famata was isolated 9 times; Geotrichum sp., 6; Saccharomyces cerevisiae, 5; Cryptococcus laurentii, 4; Rhodotorula sp., 3; Zygosaccharomyces sp., 2; and Candida colliculosa, Candida globosa, and Candida pelliculosa 1 time, respectively. The colony-forming unit ranged from 1 to 10 in all positive cultures. Fifteen of 92 (16.3%) foals with heat diarrhea and 11 of 92 (11.9%) asymptomatic animals yielded positive samples for yeasts. None of the animals shed the same yeast more than 2 times consecutively, and yeasts were found in pure or mixed culture. Equine gastrointestinal tract did not appear to be colonized by yeasts during the first 15 days of life, and yeasts are not involved in foal-heat diarrhea.  相似文献

The goal of this study was to quantify the effect of a traditional Italian conditioning method (CM). Six Standardbred yearlings were kept in standardized conditions for 140 days. From T1 (day 1) to T10 (day 10), the horses were hitched for 30 minutes/day to accustom them to a cart. From T11 to T70, they were jogged or exercised for 30 minutes/day at a slow trot (4.4 ± 0.3 m/second) and from T71 to T140 for 40 minutes/day (5 m/second ± 0.3 m/second). Every 15 days, the horses performed fast exercise for a distance of 5,000 m with increased speeds. Blood samples were collected before and after daily training (DT) every 21 days (P1…P7), to measure the following variables: erythrocytes (RBC), hemoglobin (Hb), packed cell volume (PCV), mean corpuscular hemoglobin concentration (MCHC), mean corpuscular hemoglobin (MCH), mean corpuscular volume (MCV), total protein (Pt), albumin, aspartate amino transferase (AST), alkaline phosphatase (ALP), creatine kinase (CK), lactate dehydrogenase (LDH), urea, creatinine, glucose, triglycerides, and blood lactate. A three-step standardized exercise test (SET) also was performed. Analysis of variance was used to study differences during the training. Tukey's post-hoc test was used for statistical multiple comparison (P ≤ 0.05 and 0.01). Lactate level did not show an increase over 4 mmol/L in all periods. Glucose was higher at P1 than P7. There was no correlation between method of conditioning and the level of CK, LDH, or AST. Conditioning method induced an increase in triglycerides for the change in exercise metabolism supply. According to SET, conditioning method induced an increase of SVO2max (speed [m/second] reached to a theoretical maximal oxygen consumption), V4 (speed [m/second] reached at a lactate content of 4 mmol/L), V200 (speed [m/second] at 200 beats/minute) and HR4 (heart rate [beats/minute] at 4 mmol/L of lactate). We concluded that the Italian conditioning method used in this study improved the fitness of horses.  相似文献

BACKGROUND

Incompatible insect technique (IIT) is a population suppression approach based on the release of males with manipulated Wolbachia infection inducing egg inviability in wild females. We here present results of multiple field releases of incompatible ARwP males carried out in 2019 in a 2.7-ha green area within urban Rome (Italy) to assess the effect on Aedes albopictus egg viability. Data are compared with results obtained in 2018, when the approach was tested for the first time in Europe.

RESULTS

An average of 4674 ARwP males were released weekly for 7 weeks, resulting in a mean ARwP:wild male ratio of 1.1:1 (versus 0.7:1 in 2018). Egg-viability dynamics in ovitraps significantly varied between treated and control sites, with an estimated overall reduction of 35% (versus 15% in 2018). The estimated proportion of females classified as mated with ARwP males was 41.8% and the viability rate of eggs laid by these females (9.5%) was on average significantly lower than that of females only mated with wild males (87.8%); however, high variability in fertility was observed. Values of ARwP male competitiveness were 0.36 and 0.73 based on the overall viability rate of eggs in ovitraps and on female fertility, respectively; thus, well above the conventional 0.2 threshold for an effective suppressive impact in the field.

CONCLUSIONS

Results further support the potential of IIT as a tool to contribute to Ae. albopictus control in the urban context, stressing the need for larger field trials to evaluate the cost-efficacy of the approach in temperate regions. © 2023 The Authors. Nodular trombiculinosis has been reported in Brazil in chickens [Torres, S., Braga, W., 1939. Apolonia tigipioensis, g. e sp. n. (Trombiculinae) parasito de Gallus gallus dom. Chave para determinação de gêneros. Boletim da S.A.I.C. 4, 37–44] and humans [Carneiro, L.S., 1952. Uma nova acaríase humana—Contribuição ao seu estudo. Imprensa Industrial, Recife. Faculdade de Medicina da Universidade do Recife, Tese Livre Docência, p. 56]. In this report, a juvenile ostrich and a house sparrow, both originating from a riverside property in the town of Petrolina in the state of Pernambuco, presented 87 and eight nodules, respectively, on various locations of their bodies. Physical expression of the nodules liberated parasites that were morphologically identified as mites from the family Trombiculidae. The mites were further identified as Apolonia tigipioensis by the presence of an elongated body form and transversely striated, three pairs of long legs each with seven segments, primary coxae with a single seta, each tarsus terminating with three claws, and a scutum with an anteromedian projection and paired anteromedian setae. Histopathologic examination of skin biopsies from these birds, stained with hematoxilin–eosin, revealed acute parasitic cystic lymphoplasmacytic dermatitis.  相似文献

Nine Chlamydia suis isolates, obtained from pigs with conjunctivitis, were molecularly characterized by ompA sequencing and their in vitro susceptibility to six cathelicidin peptides (SMAP-29, BAC-7, BMAP-27, BMAP-27, BMAP-28, PG-1, LL-37) determined in cell culture. SMAP-29 was the most active peptide, reducing the intracellular inclusion number by > or =50% at a concentration of 10 microg/ml (3 microM) in six of the nine isolates tested. Three molecularly identical isolates were insensitive at a concentration as high as 80 microg/ml (25 microM). Of the remaining cathelicidin peptides tested, BAC-7 and BMAP-27 were active against six C. suis isolates at a concentration of 80 microg/ml (25 and 26 microM, respectively). Cathelicidins LL-37 and PG-1 did not show any anti-chlamydial activity at 80 microg/ml.  相似文献
