A new Integrated Pest Management program based on the combination of synthetic pesticide with a GRAS(generally recognized as safe)-classified sanitizer for the control of Botrytis cinerea in field conditions was described. The aim behind this research was to determine whether the use of this mixture would enhance the efficiency of pesticides while decreasing the recommended dose. Naturally infected tomato plants, grown in the greenhouse, were treated with two commonly used fungicides SWITCH(Syngenta, Switzerland) and SIGNUM(BASF, Germany) each alone or combined with a commercially available organic sanitizer PERACLEAN?5(Evonik Industries, Germany). A total of 27 treatments were tested consisting of three concentrations of synthetic fungicide(×1, ×1/2 and ×1/4 of the recommended dose) either applied separately or combined with three concentrations of the tested sanitizer(0.5, 1 and 1.5%). The control efficacy achieved by the fungicides applied alone ranged between 0 and 66.7% while all fungicide-sanitizer mixtures resulted in up to 70% control of grey mould. The treatment that provides the maximum control of B. cinerea was the result mixture of ×1/4 of the recommended concentration of SWITCH(15 g L~(–1)) with 0.5% of PERACLEAN~?5. This combination suppressed 85% of grey mold infections while decreasing the usually used amount of this pesticide by 75%, reducing therefore the well known negative impacts of chemical pesticides on environment and consumers health.  相似文献

Receptor tyrosine kinases are key regulators of cellular growth and proliferation. Dysregulations of receptor tyrosine kinases in cancer cells may promote tumorigenesis by multiple mechanisms including enhanced cell survival and inhibition of cell death. Araguspongines represent a group of macrocyclic oxaquinolizidine alkaloids isolated from the marine sponge Xestospongia species. This study evaluated the anticancer activity of the known oxaquinolizidine alkaloids araguspongines A, C, K and L, and xestospongin B against breast cancer cells. Araguspongine C inhibited the proliferation of multiple breast cancer cell lines in vitro in a dose-dependent manner. Interestingly, araguspongine C-induced autophagic cell death in HER2-overexpressing BT-474 breast cancer cells was characterized by vacuole formation and upregulation of autophagy markers including LC3A/B, Atg3, Atg7, and Atg16L. Araguspongine C-induced autophagy was associated with suppression of c-Met and HER2 receptor tyrosine kinase activation. Further in-silico docking studies and cell-free Z-LYTE assays indicated the potential of direct interaction between araguspongine C and the receptor tyrosine kinases c-Met and HER2 at their kinase domains. Remarkably, araguspongine C treatment resulted in the suppression of PI3K/Akt/mTOR signaling cascade in breast cancer cells undergoing autophagy. Induction of autophagic death in BT-474 cells was also associated with decreased levels of inositol 1,4,5-trisphosphate receptor upon treatment with effective concentration of araguspongine C. In conclusion, results of this study are the first to reveal the potential of araguspongine C as an inhibitor to receptor tyrosine kinases resulting in the induction of autophagic cell death in breast cancer cells.  相似文献

To test the effect of insulin on renal perfusion and the participation of NO and PG as mediators of this response, renal blood flow (RBF) was measured in sheep (n = 8) implanted with ultrasonic flow probes around renal arteries and with a systemic arterial pressure (SAP, n = 4) telemetry device. Three protocols were performed: 1) RBF and SAP were recorded (0800 to 1800 h) in fed and fasted sheep, with the latter receiving intravenous (i.v.) infusions (0.5 mL/min) of insulin at 2 or 6 mU/(kg·min); 2) fasted sheep received i.v. infusions of either an inhibitor of NO synthesis (N(G)-nitro-L-arginine methyl ester, L-NAME) alone [0.22 mg/(kg·min), 1000 to 1200 h] or L-NAME (1000 to 1200 h) + insulin during the second hour (6 mU/(kg·min), 1100 to 1200 h); and 3) the same protocol was followed as in protocol 2, substituting L-NAME with ketoprofen [0.2 mg/(kg·min)], a cyclooxygenase inhibitor. In all protocols, plasma insulin and glucose were determined. During insulin administration, euglycemia was maintained and hypokalemia was prevented by infusing glucose and KCl solutions. After the onset of meals, a long-lasting 18% increase in RBF and a 48% insulin increase were observed (P < 0.05), without changes in SAP. Low- and high-dose insulin infusions increased RBF by 19 and 40%, respectively (P < 0.05). As after meals, the increases in RBF lasted longer than the insulin increase (P < 0.05). The L-NAME infusion decreased RBF by 15% (P < 0.05); when insulin was added, RBF increased to preinfusion values. Ketoprofen decreased RBF by 9% (P < 0.05); when insulin was added, RBF increased to 13% above preinfusion values (P < 0.05). In no case was a modification in SAP or glucose noted during the RBF changes. In conclusion, insulin infusion mimics the meal-dependent increase in RBF, independent of SAP, and lasts longer than the blood insulin plateau. The RBF increase induced by insulin was only partially prevented by L-NAME. Ketoprofen failed to prevent the insulin-dependent RBF increase. Both facts suggested that complementary vasodilatatory agents accounted for the insulin effect on sheep renal hemodynamics.  相似文献

Particulate matter is emitted to the atmosphere from variousenergy conversion processes, mainly from the electric powerand the transportation sectors. Recent studies and peer reviews conducted by international organizations have indicated the adverse health and economic effects of the accumulation of these particulates in the environment.In the absence of any data related to the levels of air pollution in the Beirut environment, this study was initiated with the objective of conducting a preliminary investigation that includes air sampling and the X-ray fluorescence analysis of the samples. Emphasis is placed on the impact of the transport sector on the dissemination of different pollutants and in particular lead. The results show high levels of particulates resulting from different sources and high concentrations of lead within the city ofBeirut. A direct correlation between the particulate constituents and street activities is identified. The results indicate the seriousness of the air pollution problem that the transport sector can inflict on the society if appropriate mitigative measures are not implemented, especially in urbanized regions where traffic flow is consistently heavy. Mitigation options applicable to Lebanon and possibly to other countries of similar economic and social structure are recommended.  相似文献

Several studies investigated the usage of spirulina (Arthrospira platensis) in aquaculture as a functional dietary additive or plant proteins replacers; however, more efforts still needed to test its nano form inclusion in aquafeed. In this study, Nile tilapia (Oreochromis niloticus) fed dietary spirulina (Aplatensis) nanoparticles (SNP) at 0 (control), 0.1, 1 and 10 g/kg diet while the fifth diet was mixed with 10 g/kg of the normal form of Aplatensis as a positive control. The final weight (FW), weight gain (WG) and feed conversion ratio of fish fed SNP at 10 g/kg displayed improved values comparing with the other groups (p < 0.05). Further, fish fed 1 g SNP had higher FW and WG than the control and 0.1 g SNP and 10 g normal Aplatensis. The incorporation of SNP at 1 and 10 g/kg in Nile tilapia diets significantly (p < 0.05) enhanced total protein, albumin, globulin, lowered the values of serum glucose and activities of ALT and AST compared with control and spirulina groups. The SOD, CAT, GPx, lysozyme and respiratory burst activities were meaningfully promoted (p < 0.05) in fish received 1 and 10 g/kg diet concerning the control and spirulina groups. Nile tilapia challenged with Aeromonas hydrophila showed congestion of the kidney, liver and spleen tissues while SNP alleviated the inflammation induced by Ahydrophila. Different levels of SNP-incorporated group showed lower cumulative mortality than the other groups. In conclusion, the obtained results illustrated that spirulina in nano form is recommended to enhance Nile tilapia well-being.  相似文献

The higher-order assembly of chromatin imposes structural organization on the genetic information of eukaryotes and is thought to be largely determined by posttranslational modification of histone tails. Here, we study a 20-kilobase silent domain at the mating-type region of fission yeast as a model for heterochromatin formation. We find that, although histone H3 methylated at lysine 9 (H3 Lys9) directly recruits heterochromatin protein Swi6/HP1, the critical determinant for H3 Lys9 methylation to spread in cis and to be inherited through mitosis and meiosis is Swi6 itself. We demonstrate that a centromere-homologous repeat (cenH) present at the silent mating-type region is sufficient for heterochromatin formation at an ectopic site, and that its repressive capacity is mediated by components of the RNA interference (RNAi) machinery. Moreover, cenH and the RNAi machinery cooperate to nucleate heterochromatin assembly at the endogenous mat locus but are dispensable for its subsequent inheritance. This work defines sequential requirements for the initiation and propagation of regional heterochromatic domains.  相似文献

The present work is undertaken mainly for genotyping of kappa-casein gene in buffalo bulls. DNA was extracted from the frozen semen of nine buffalo bulls used in artificial insemination in addition to 40 blood samples of male buffalo calves for identification and genotyping of kappa-casein gene by PCR-RFLP assay. Different restriction endonucleases, including HindIII, HinfI and Taq1 were used. The PCR product of the primer specific for K2 and KY gives the specific band at size 379 and 280 bp respectively. Digestion of 379 bp fragment K2 by restriction endonuclease HindIII generated two fragments of 156 and 223 bp. For fragment 280 bp kappa-casein KY, the above mentioned endonuclease generated two fragments of 180 and 100 bp. Digestion of the 379 bp PCR product with HinfI resulted two restriction fragments of 288 and 91 bp. On the other hand, the amplified DNA (379 bp) from buffalo remained undigested by Taq1 restriction enzyme.In conclusion, the PCR-RFLP technique demonstrated that bulls were monomorphic for the kappa-casein gene; they possess only allele B in homozygosis form.  相似文献
