The tip of a projectile point made of mastodon bone is embedded in a rib of a single disarticulated mastodon at the Manis site in the state of Washington. Radiocarbon dating and DNA analysis show that the rib is associated with the other remains and dates to 13,800 years ago. Thus, osseous projectile points, common to the Beringian Upper Paleolithic and Clovis, were made and used during pre-Clovis times in North America. The Manis site, combined with evidence of mammoth hunting at sites in Wisconsin, provides evidence that people were hunting proboscideans at least two millennia before Clovis.  相似文献

Conformational dynamics play a key role in enzyme catalysis. Although protein motions have clear implications for ligand flux, a role for dynamics in the chemical step of enzyme catalysis has not been clearly established. We generated a mutant of Escherichia coli dihydrofolate reductase that abrogates millisecond-time-scale fluctuations in the enzyme active site without perturbing its structural and electrostatic preorganization. This dynamic knockout severely impairs hydride transfer. Thus, we have found a link between conformational fluctuations on the millisecond time scale and the chemical step of an enzymatic reaction, with broad implications for our understanding of enzyme mechanisms and for design of novel protein catalysts.  相似文献

During courtship flights, males of some hummingbird species produce diverse sounds with tail feathers of varying shapes. We show that these sounds are produced by air flowing past a feather, causing it to aeroelastically flutter and generate flutter-induced sound. Scanning laser doppler vibrometery and high-speed video of individual feathers of different sizes and shapes in a wind tunnel revealed multiple vibratory modes that produce a range of acoustic frequencies and harmonic structures. Neighboring feathers can be aerodynamically coupled and flutter either at the same frequency, resulting in sympathetic vibrations that increase loudness, or at different frequencies, resulting in audible interaction frequencies. Aeroelastic flutter is intrinsic to stiff airfoils such as feathers and thus explains tonal sounds that are common in bird flight.  相似文献

Aflatoxin contamination is one of the main factors affecting peanut seed quality. One of the strategies to decrease the risk of peanut aflatoxin contamination is the use of genotypes with resistance to Aspergillus infection. This laboratory study reports the resistance to Aspergillus infection and aflatoxin contamination of six peanut genotypes inoculated with 21 Aspergillus isolates obtained from the peanut production region of Cordoba, Argentina. The resistance was investigated in the seed coat and cotyledons of three resistant genotypes (J11, PI 337394, and PI 337409) and three breeding lines (Manfredi 68, Colorado Irradiado, and Florman INTA) developed at the Instituto Nacional de Tecnologia Agropecuaria (INTA), Manfredi Experimental Station, Cordoba, Argentina. Resistance to fungal colonization and aflatoxin contamination was found to be associated with seed coat integrity in the PI 337394, PI 337409, and J11 genotypes, whereas the INTA breeding lines such as Colorado Irradiado showed a moderate resistance and the Manfredi 68 and Florman INTA genotypes the least resistance. Furthermore, another type of resistance associated with cotyledons was found only in the PI 337394 genotype.  相似文献

The adhesion of methyl ricinoleate droplets to cells of the yeast Yarrowia lipolytica was investigated. A new cytometric method, relying on the double staining of fatty globules with Nile Red and of cells with Calcofluor, enabled us to quantify methyl ricinoleate droplet adhesion to cells precultured on a hydrophilic or on a hydrophobic carbon source. In this last case, droplet adsorption was enhanced and a MATS (microbial adhesion to solvents) test revealed that this increase was due to Lewis acid-base interactions and not to an increase in the hydrophobic properties of the cell surface. These preliminary results demonstrate that the developed cytometric method is promising for various applications concerning the study of interactions between microorganisms and an emulsified hydrophobic substrates.  相似文献

A polymerase chain reaction (PCR) assay using primers SsF and SsR designed from the internal transcribed spacer (ITS) regions ofSpongospora subterranea f. sp.subterranea was developed for the specific identification and quantification ofS. subterranea. These primers amplified a 434 bp product from DNA ofS. subterranea spore balls, but not from DNA of healthy potato, common scab tuber, and taxonomically related plasmodiophorids. This PCR assay was successfully used for the detection ofS. subterranea in naturally infected symptomatic and asymptomatic potato tubers.Spongospora subterranea in other infected symptomless host plants was detected by PCR. The PCR assay was modified with improved soil DNA extraction methods to detectS. subterranea in soil. The assay was sensitive, and one spore ball per gram of soil could be detected. Following the design of a heterologous competitor DNA template from the sequence of λDNA, a competitive PCR assay for the quantification ofS. subterranea in soil was developed and provided accurate quantification in the range of 1 to 104 spore balls per 0.25 g of soil. In a preliminary survey of naturally infested field soil samples, spore ball concentrations were estimated to vary from ca 0 to 3600 spore balls per 0.25-g soil sample by this competitive PCR assay. The spore ball levels were compared with the powdery scab disease incidence of potatoes in these fields, and a correlationship between spore ball levels and subsequent disease incidence was found. The PCR assays developed in this investigation can be routinely used to detect and quantifyS. subterranea in diseased plant tissue, asymptomatic plant tissue, and infested soil.  相似文献

Current flu vaccines provide only limited coverage against seasonal strains of influenza viruses. The identification of V(H)1-69 antibodies that broadly neutralize almost all influenza A group 1 viruses constituted a breakthrough in the influenza field. Here, we report the isolation and characterization of a human monoclonal antibody CR8020 with broad neutralizing activity against most group 2 viruses, including H3N2 and H7N7, which cause severe human infection. The crystal structure of Fab CR8020 with the 1968 pandemic H3 hemagglutinin (HA) reveals a highly conserved epitope in the HA stalk distinct from the epitope recognized by the V(H)1-69 group 1 antibodies. Thus, a cocktail of two antibodies may be sufficient to neutralize most influenza A subtypes and, hence, enable development of a universal flu vaccine and broad-spectrum antibody therapies.  相似文献

There is increasing interest worldwide in using tree harvesting biomass as an energy source. Bark retained on logs is commonly used as an energy source, but is generally removed from eucalypt logs during harvest. In order to evaluate the potential use of eucalypt bark as fuel, there is a need for information on the productivity and cost implications of retaining eucalypt bark during harvest operations. The study examined the impact of retaining bark on logs on the productivity and costs of a whole?tree to roadside harvesting system in a short?rotation Eucalyptus nitens plantation in Australia being harvested for pulp logs. Trees were felled and bunched with a feller?buncher in spring, then left infield for four weeks to promote bark adhesion and reduce bark loss. A skidder extracted the trees to roadside where a processor processed them to predominantly 10?m logs. Machine productivities were calculated from estimated tree and log volumes and cycle times recorded from video recordings. The feller?buncher's productivity (65 m3 PMH0?1) was less than expected as it appeared to be underpowered to handle the larger trees on the study site. The skidder's productivity (56 m3 PMH0?1) was comparable to those reported in studies under similar conditions and with bark retained. The roadside processor's productivity (25 m3 PMH0?1) was lower than expected. This was believed to result from the operator separately stacking 10?m and 5?m logs, and the lower feed speed resulting from slippage due to the reduced feed roller pressure used in the study to reduce bark loss. Future research could identify feed rollers that increase feed speed while retaining bark. Harvest system costs (AUD18 GMt?1) were similar to those reported for a eucalypt roadside processing trial where bark was removed. These results suggest that retaining bark on the logs at roadside did not affect the harvesting system's productivity or costs.  相似文献
