The current experiment was conducted to find out the optimal conditions for mass rearing and developmental changes of Cyclosainsulana. The lab. conditions were maintained at(27±2)℃ and(65±5)% RH. The clear perplex cages and natural diet consisting of the aphids, larvae of the house fly and larvae of drosophila were used for rearing. C. insulana took(123.12±7.26) days to develop from eggs to adults passing through eight instars under prevailing vivo conditions. The eggs were greenish white in color with average size of 0.57 mm ±0.034. The eggs spent(7.52±1.64) days in emergence. Maximum number of spiderlings survived at the 5th instar(84%) and minimum at the 1st instar(34%). The measurements of different body parts including the total body length, cephalothorax and pedipalps of the both male and female C. insulana were done with the help of micrometer and presented as mean±SD. The measurements varied in the each developing instar. It was concluded that spiders were difficult to rear in the lab. conditions and each developing stage which was regarded as instars showed variations in size colors and body characteristics.  相似文献

A phenol-water extract of Listeria monocytogenes virulent strains 9-125 (serotype 4b) was purified by 3 cycles of ultracentrifugation. The purified extract reacted positively in Limulus amoebocyte lysate assay at a concentration of 1 microgram/ml. This value was 1,000 times higher than that for Salmonella abortus equi lipopolysaccharide. The phenol extract was toxic to chicken embryos (median lethal dose was 40.5 micrograms) and contained carbohydrates (heptose, hexose, hexosamine, methylpentose, 2-keto-3-deoxyoctanate, dideoxyhexose), lipid, 16 amino acids in the protein moiety, glucosamine, galactosamine, phosphorus, and ribonucleic acid.  相似文献

Synthetic seed technology is an emerging and broadly used technique in the field of plant biotechnology to conserve economically important plants. In the present study, nodal segments of Capparis decidua were entrapped in calcium alginate gel matrix to produce firm and uniform synthetic seeds. 3% sodium alginate and 75 mM calcium chloride were found best for encapsulation. Among all the concentrations and combinations of thidiazuron (TDZ) either singly or with indole -3- acetic acid (IAA) augmented in Murashige and Skoog medium used, TDZ (5.0 µM) + IAA (0.5 µM) was found most effective in conversion of synthetic seeds into plantlets as 79% plantlets were developed on this combination with 13.2 ± 0.87 shoots and 5.5 ± 0.40 cm shoot length after 8 weeks of culture. Further, synthetic seeds stored at low temperature (4 °C) can retain their viability up to 4 weeks and showed maximum conversion rate (93%) into plantlets, when placed back to regeneration medium. Root formation was also occurred in the same regeneration medium and roots were healthy. Plantlets were successfully hardened in culture room in plastic cups filled with sterile vermiculite and after 4 weeks, they were transferred to greenhouse where they exhibited normal growth with 80% survival. Growth parameters were evaluated in micropropagated plants and compared with the seedlings of same age. Effect of different days of acclimatization were also recorded on various physio-biochemical activities and showed a positive response that can be interpreted as better protection mechanism of micropropagated plants against the stress possibly generated due to reactive oxygen species when transferred to ex vitro environment.  相似文献

Drought is a major factor limiting crop production worldwide. Barley is a well‐adapted cereal that is largely grown on dry marginal land where water and salinity are the most prevalent environmental stresses. This study was carried out to investigate the effects of drought stress and subsequent recovery on growth, photosynthetic activity, water relations, osmotic adjustment (OA), and solute accumulation of wild (Hordeum maritimum) and cultivated barley (H. vulgare L.). In a pot experiment, 60 d old seedlings were subjected to drought stress for 0, 7, 14, 21, or 28 d, and then re‐watered to recover for up to 21 d. Plants were harvested at the end of each of these drought/recovery treatments. Drought significantly reduced fresh and dry weights at the whole‐plant level, photosynthetic activities, and solute and water potentials, while increasing leaf Na+ and K+ concentrations. The adverse effects of drought on growth were more marked in cultivated barley than in wild barley and the reverse was true for photosynthetic activities. During recovery, all wild barley seedlings completely recovered. For cultivated barley seedlings, rehydration had a beneficial effect on growth and photosynthesis, independent of treatment duration, but complete recovery did not occur. The reduction in leaf solute potential at full turgor in drought‐stressed barley, relative to the control, suggests active OA which was more significant in wild barley than in cultivated barley. OA was mainly due to the accumulation of inorganic (K+ in cultivated barley and Na+ in wild barley) and organic (soluble sugars and proline) solutes. The results suggest that OA is an important component of the drought‐stress adaptation mechanism in wild barley, but is not sufficient to contribute to drought tolerance in cultivated barley. In the latter species, the results show that even short periods (as little as 7 d) of water deficit stress had considerable long‐term effects on plant growth.  相似文献

A total of 70 sheep and 330 goats were selected randomly. All the animals were kept under same housing and management conditions. Serum samples were collected from all the animals and tested for the presence of antibodies against Peste des petits ruminants (PPR) virus using competitive ELISA (cELISA). All the animals were found negative showing percentage inhibition (PI) values <50. The animals were vaccinated against PPR with Nig/75/1 strain vaccine of PPR Serum samples were collected from randomly selected 12 sheep and 30 goats at 10, 30 and 45 days post-vaccination. The samples were subjected to cELISA to determine the presence of antibodies against PPRV. The samples with PI >50 were considered as sero-positive. The sheep found positive at 10, 30 and 45 days post-vaccination were 1(8.3%), 7(58.3%) and 12(100%) respectively. In case of goats 3(10.0%), 29(96.6%) and 27(90.0%) animals gave positive results at 10, 30 and 45 days post-vaccination respectively. Mean PI values in sheep at 10, 30 and 45 days post-vaccination were recorded as 37, 65 and 91 respectively, whereas in goats these values were 43, 78 and 86 respectively.  相似文献

This project was conducted to investigate the comparative efficiency of competitive ELISA (cELISA), standard Agar Gel Immunodiffusion Test (AGID) and Precipitinogen Inhibition Test (PIT) for the diagnosis of Peste des Petits Ruminants (PPR) in Pakistan. To deal with this, serum samples from 198 sheep and 82 goats were collected from three different government livestock farms and all the samples were run simultaneously with the three serological tests. The samples found positive for PPR antibodies through cELISA, AGID and PIT were 96 (34.2%), 60 (21.4%) and 72 (25.7%), respectively. Kappa statistics were applied to evaluate the concordance between the laboratory-based test (cELISA) and field-based tests (AGID and PIT). Kappa statistics scores for cELISA versus AGID and PIT were 0.6343 (95% Confidence Interval CI 0.5231–0.7456) and 0.7134 (95% Confidence Interval CI 0.5987–0.8281), respectively, which indicate a “substantial” agreement between cELISA and AGID and “significant” agreement between cELISA and PIT. AGID and PIT revealed relative diagnostic sensitivities with cELISA of 59.3% and 69.7% and relative diagnostic specificities of 98.3% and 97.2%, respectively. The data suggested that for mass screening and control of PPR, these serological tests proved practical in the absence of cELISA since they have high relative diagnostic specificities and a satisfactory relative diagnostic sensitivities.  相似文献

This report describes in vitro shoot induction and plant regeneration from nodal segments of Balanites aegyptiaca on Murashige and Skoog (MS) medium fortified with 6-benzyladenine (BA), thidiazuron (TDZ) and kinetin (Kin) (0.5–20.0 μM). MS medium supplemented with BA (12.5 μM) was the most effective in inducing bud break and growth and also in initiating multiple shoot proliferation. However, the optimal level of TDZ supplementation to the culture medium was 5.0 μM. Shoot cultures were established by repeatedly subculturing the original nodal explants on the same medium. Highest number of shoots (11.5 ± 0.7) and shoot length (5.0 ± 0.2 cm) were achieved when cultures were subcultured on MS medium supplemented with 12.5 μM BA and 1.0 μM α-naphthalene acetic acid (NAA). The shoots regenerated from TDZ supplemented medium when subcultured to hormone free MS basal medium considerably increased the rate of shoot multiplication and shoot length by the end of fifth subculture. Rooting of the shoots was achieved on MS medium augmented with 1.0 μM indole-3-butyric acid (IBA) plus 0.5% activated charcoal followed by their transfer to half strength MS basal medium. The in vitro raised plantlets with well developed shoots and roots were successfully established in earthen pots containing garden soil and were grown in greenhouse with 70% survival rate. The results of this study provide the first successful report on in vitro direct plant regeneration of B. aegyptiaca.  相似文献

Successful rearing of larval fish requires culture conditions and feeding strategies matching the ontogenetic status of larvae. This study describes the external morphology and development of organs and structures involved in the feeding process (i.e. sensorial organs, mouthparts and digestive system) from hatching until first feeding in Pacific red snapper. Hatching occurred 26 h after fertilization at 26°C and total length (TL) was 2.45 ± 0.08 mm. The larvae showed an undifferentiated eye and digestive tract. At 48 hah, TL was 3.31 ± 0.12 mm. Yolk and oil globule were still present. The mouth was still closed, but the Meckel's, quadrate, hyoid and hyomandibular cartilages were present. The retina was formed by 5 layers, and a thin layer of pigment epithelium was observed in the outer nuclear layer (ONL). At 70 hah, TL was 3.44 ± 0.22 mm. A remnant of oil globule was still present. The mouth and anus were open. At 93 hah, the number of cones in the ONL of the retina have increased and there was more pigment in the pigment epithelial layer. A joint between Meckel's and the quadrate cartilage and also a joint between the hyomandibular cartilage and the skull were present. The presence of live feed was detected in the digestive tract of these larvae. Based on these observations, the Pacific red snapper larvae is functional to start ingesting live feed between the 3rd and 4th day after hatching.  相似文献
