In this work, ergosterol and ergocalciferol were identified for the first time in hop. In addition, in this article, a simple and reliable analytical methodology for analysis of these compounds in different commercial forms of hop is presented. The performance of the method was assessed by the evaluation of parameters such as absolute recovery (higher than 70%), repeatability (lower than 3 %), linearity ( r(2) > 0.9988) and limits of detection (ranging from 0.034 for ergocalciferol to 0.058 mg/L for ergosterol) and quantification (ranging from 0.113 for ergocalciferol to 0.195 mg/L for ergosterol). On the basis of standard additions applied with the optimized procedure and high-performance liquid chromatography with diode array detection, it appears that the Nugget hop plant (crop 2006) contains 1.84 +/- 0.09 microg/g of ergosterol and 1.95 +/- 0.05 microg/g of ergocalciferol. The identity of the compounds was confirmed by high-performance liquid chromatography/electrospray ionization tandem mass spectrometry in the positive ion mode. The presence of ergosterol here reported should have great potential for the assessment of hop as related to the fungal contamination proportion and hence the quality of this raw material. 相似文献
The flavonoids present in passion fruit (Passiflora edulis) leaves were identified by a high-performance liquid chromatography-diode array detection-tandem mass spectrometry (HPLC-DAD-MS/MS) method. Sixteen apigenin or luteolin derivatives were characterized, which included four mono-C-glycosyl, eight O-glycosyl- C-glycosyl, and four O-glycosyl derivatives. With the exceptions of C-hexosyl luteolin and C-hexosyl apigenin, all the compounds exhibited a deoxyhexose moiety. Moreover, the uncommon C-deoxyhexosyl derivatives of luteolin and apigenin have been identified for first time in P. edulis by HPLC-DAD-MS/MS. The antioxidative capacity of passion fruit leaves was checked against DPPH radical and several reactive oxygen species (superoxide radical, hydroxyl radical, and hypochlorous acid), revealing it to be concentration-dependent, although a pro-oxidant effect was noticed for hydroxyl radical. 相似文献
This case report describes a highly pruritic, papular and crusted dermatitis affecting the dorsum of a dog and microscopically diagnosed as dermatitis induced by Straelensia cynotis. Histologically, each papule was characterized by a dilated hair follicle with marked pseudoepitheliomatous hyperplasia, perifollicular mucinosis and an abundant mononuclear infiltration by plasma and mast cells. Each dilated follicle contained a larval mite. A therapy including systemic ivermectins and oral antibiotic therapy was initiated, but the clinical response was poor. To the best of the authors' knowledge, this is the first report of straelensiosis in Portugal, and the second in Europe. 相似文献
Natural resistance against brucellosis in cattle is linked to the Nramp1 gene, which encodes a divalent cation transporter that localizes in the phagolysosome membrane in macrophages. Nramp1 gene in mouse plays a critical role in innate immunity favoring bacterial killing by macrophages in addition to its influence on adaptative immunity. Polymorphisms at the bovine Nramp1 3' untranslated region (3'UTR), detectable by Single Strand Conformational Analysis (SSCA), are associated with natural resistance against brucellosis. Such polymorphisms are associated with variation in the number of GT repeats. This study compared the frequency of Nramp1 3'UTR polymorphisms between Zebu and European bovine breeds. Eighty-one Holsteins (Bos taurus taurus) and 167 Zebu (Bos taurus indicus), including the following breeds: Nelore (n=95), Guzerá (n=37), and Gir (n=35), totaling 248 pure breed cattle studied. DNA extraction was performed using the guanidium protocol and genotyping was performed by SSCA. DNA from cattle considered genotypically resistant to brucellosis resulted in a single band (homozygous) with 175bp, corresponding to the 3'UTR with 13 GT pairs (GT13), whereas DNA from genotypically susceptible cattle generated one single band with 177bp (homozygous GT14) or double bands with both 175 and 177bp, or 175 and 179bp (heterozygous GT13/GT14 or GT13/GT15, respectively). A marked difference in the frequency of alleles was detected between the Zebu and Holstein cattle. Holsteins had an extremely homogeneous genotype, with 100% of the individuals with a GT13 genotype. In sharp contrast the Nelore breed had the most heterogeneous genotype with four allelic combinations, namely, homozygous GT13, homozygous GT14, heterozygous GT13/GT14, and heterozygous GT13/GT15. When the Zebu breeds were compared to each other, the only significant difference observed was the frequencies of the genotypes GT13 and GT14 between the Nelore and Guzerá breeds. The knowledge of allelic frequencies in different breeds of cattle may prove to be very useful in the future for planning breeding strategies for selection of resistant cattle. 相似文献
The utility of benzoxazinones and some of their synthetic derivatives in the search for new leads for herbicide model development has been widely discussed. As the benzoxazinone skeleton contains three different potential areas for functionalization (C-2, N-4, and aromatic protons H-5, H-6, H-7, and H-8), and the first two have already been optimized, the main objective of this work was the substitution of aromatic protons for different substituent types and the study of the effects of the prepared chemicals on selected standard target species (STS) and weeds. Thus, different combinations of aromatic substituents, including methoxy, methoxycarbonyl, fluorine, chlorine, and trifluoromethyl, were introduced at different positions. Phytotoxicity results were successfully correlated with steric and electronic molecular parameters, the resulting molecular volume (V) and dipole moment (mu) being the most influential ones. Halogenations at C-6 and fluorination at C-7 were the most successful modifications. Compounds 6-fluoro-(2H)-1,4-benzoxazin-3(4H)-one (6F-D-DIBOA), 7-fluoro-(2H)-1,4-benzoxazin-3(4H)-one (7F-D-DIBOA), and 6-chloro-(2H)-1,4-benzoxazin-3(4H)-one (6Cl-D-DIBOA) had the highest phytotoxic activities. The dose-response profiles on wheat and two of its most common weeds (Lolium rigidum Gaud. and Avena fatua L.) were compared by means of a proposed selectivity index, which displayed 7F-D-DIBOA as the most selective of the tested benzoxazinones. 相似文献
The phenolic compounds of Rumex induratus leaves were determined by HPLC-DAD-MS/MS-ESI. The results revealed a profile composed of 19 compounds: caffeoyl-hexoside, two p-coumaroyl-hexoside isomers, feruloyl-hexoside, sinapoyl-hexoside, 6-C-hexosyl-quercetin, 8-C-hexosyl-luteolin, 6-C-hexosyl-luteolin, 6-C-hexosyl-apigenin, 3-O-hexosyl-quercetin, 3-O-rutinosyl-quercetin, 7-O-hexosyl-diosmetin, 3-O-rutinosyl-isorhamnetin, 7-O-(acetyl)-pento-hexosyl-diosmetin, 6-C-hexosyl-genkwanin, and four unidentified O-glycosyl-C-glycosylflavones. The quantification of the identified phenolics by HPLC-DAD showed 6-C-hexosyl-luteolin as the main compound. Organic acid composition was determined by HPLC-UV, revealing a high content of oxalic acid. R. induratus was also investigated for its capacity to act as a scavenger of DPPH and superoxide radicals. Good antioxidative results were obtained against both radicals. Unlike other species of the genus Rumex, R. induratus did not present any anthraquinone derivative. 相似文献
The Neotropics harbour the greatest diversity of freshwater fish on Earth. Despite recent advances in characterizing the fish fauna, the total number of species, distributional range, evolution and ecological traits remain uncertain. Thus, we quantify shortfalls in the knowledge of taxonomy (Linnean shortfall), geographic distribution (Wallacean shortfall), evolutionary relationships (Darwinian shortfall) and feeding habits (Raunkiæran shortfall) of Auchenipteridae driftwood catfishes, one of the most representative groups of the Siluriformes family in the Neotropics. We find a steep increase in the historical accumulation of valid species over time, suggesting that 45% of the total number predicted remains to be described. Auchenipterids also remain under‐collected; only 45% of the ecoregions and less than 3% of the one‐degree grid cells covering the Neotropics are reasonably sampled. The topologies of recent phylogenies are more similar to each other than former ones, showing a tendency towards a robust phylogenetic hypothesis for this family. Current knowledge on feeding habits is biased towards a few genera and species and is still expanding with every new published study. Our study highlights specific knowledge gaps that need to be addressed: a considerable number of Auchenipteridae species remain to be described; and most of valid species lack reliable information on their geographic distribution and feeding habitat. It implies that research on fish systematic biology needs to advance and it will require a concerted effort of taxonomists, ecologists and biogeographers to reduce these gaps. 相似文献
This study aimed at assessing the effects of the dietary tryptophan (Trp) supplementation on growth and feed utilization, brain serotonin content, and expression of selected liver genes (involved in the liver serotonin pathway, protein synthesis degradation, and antioxidant activity) in zebrafish. A growth trial was conducted with zebrafish juveniles fed five experimental isoproteic (40%DM) and isolipidic (8%DM) fishmeal-based diets containing graded levels of Trp: a Trp-non-supplemented diet (diet Trp0, with 0.22% Trp) and four Trp-supplemented diets containing 2–16 times higher Trp content (diets Trp2, Trp4, Trp8, and Trp16 with 0.40, 0.91, 2.02, and 3.34% Trp, respectively). Diets were tested in quadruplicate, with fish being fed twice a day, 6 days a week for 6 weeks to apparent visual satiation. At the end of the trial, growth performance and feed utilization were assessed, and fish from all experimental groups were sampled for whole-body composition analysis. In addition, fish fed low (Trp0), medium (Trp4), and high (Trp16) Trp diets were also sampled for analysis of brain serotonin content and liver gene expression. Tested tryptophan levels did not influence growth performance nor feed intake. However, values of energy and nitrogen retention as well as body energy content indicate a better feed utilization with diets containing around 0.9% and 2.0% DM Trp. Brain serotonin content increased with increasing dietary tryptophan levels. In addition, regarding liver genes, dietary treatment had a modulatory effect on the expression of Htr1aa and Htr2cl1 genes (encoding for serotonin receptors), TPH1a gene (encoding for tryptophan hydroxylase, the rate-limiting enzyme in the synthesis of serotonin from tryptophan), TOR gene (involved in protein synthesis), and Keap1 gene (involved in antioxidant responses).
Many soil microorganisms are able to transform insoluble forms of phosphorus to an accessible soluble form, contributing to plant nutrition as plant growth-promoting microorganisms (PGPM). The objective of this work was to isolate, screen and evaluate the phosphate solubilization activity of microorganisms in maize rhizosphere soil to manage soil microbial communities and to select potential microbial inoculants. Forty-five of the best isolates from 371 colonies were isolated from rhizosphere soil of maize grown in an oxisol of the Cerrado Biome with P deficiency. These microorganisms were selected based on the solubilization efficiency of inorganic and organic phosphate sources in a modified Pikovskaya's liquid medium culture containing sodium phytate (phytic acid), soybean lecithin, aluminum phosphate (AlPO4), and tricalcium phosphate (Ca3(PO4)2). The isolates were identified based on nucleotide sequence data from the 16S ribosomal DNA (rDNA) for bacteria and actinobacteria and internal transcribed spacer (ITS) rDNA for fungi. Bacteria produced the greatest solubilization in medium containing tricalcium phosphate. Strains B17 and B5, identified as Bacillus sp. and Burkholderia sp., respectively, were the most effective, mobilizing 67% and 58.5% of the total P (Ca3(PO4)2) after 10 days, and were isolated from the rhizosphere of the P efficient L3 maize genotype, under P stress. The fungal population was the most effective in solubilizing P sources of aluminum, phytate, and lecithin. A greater diversity of P-solubilizing microorganisms was observed in the rhizosphere of the P efficient maize genotypes suggesting that the P efficiency in these cultivars may be related to the potential to enhance microbial interactions of P-solubilizing microorganisms. 相似文献
