True-to-type clonal fidelity is one of the most important pre-requisites in micropropagation of crop species. Genetic fidelity of in vitro raised 45 plants of gerbera (Gerbera jamesonii Bolus) derived from three different explants, viz., capitulum, leaf and shoot tips, was assessed by 32 ISSR markers, for their genetic stability. Out of 32 ISSR markers, 15 markers produced clear, distinct and scorable bands with an average of 5.47 bands per marker. The markers designed from AG motif amplified more number of bands. The markers anchored at 3′ ends produced high number of consistent bands than unanchored markers. Fifteen ISSR markers generated a total of 3773 bands, out of which 3770 were monomorphic among all the clones. The Jaccard's similarity coefficient revealed that out of 45 clones derived from different explants, 44 were grouped into a single large cluster alongwith the mother plant with a similarity coefficient value of 1.00, whereas one clone (C38) remained ungrouped. The clones derived from capitulum and shoot tip explants did not show any genetic variation, whereas, one of the leaf-derived clones exhibited some degree of variation. 相似文献
A study was conducted to examine the effect of nutritional stress on the development of the thymus, bursa, and pancreas of 7-to-14-day-old commercial meat-type chickens. One group of 7-day-old chickens was given access to food for only 30 minutes daily for 7 days. The birds were necropsied, and the thymus, pancreas, and bursa were compared with those of the control chickens fed ad libitum and necropsied at 7 and 14 days of age. The thymuses from birds on the restricted diet were atrophied (0.45 +/- 0.08 g) and congested compared with the thymuses from chickens fed ad libitum (1.32 +/- 0.31 g). The bursae from chickens on a restricted diet were also smaller (0.26 +/- 0.08 g) than the bursae from birds with free access to food (0.74 +/- 0.11 g). The restricted diet did not appear to cause any gross or histological pancreatic changes. The thymic lesions produced by nutritional stress were similar to those observed in the runting/stunting syndrome. 相似文献
A case study focusing on the socio-economic and ecological aspects of a forest-based village, Debrasai, in the Singhbhum District of Bihar, India was undertaken in 1980 to evolve village-specific community forestry package programmes that could bring stability to the village both economically as well as ecologically. The village extending over 365.07 ha is inhabited by 361 people comprising 195 males and 166 females in 81 households. The villagers are by and large dependent on agriculture and forestry for their livelihood. The largest proportion of the land in the village is under agriculture whereas forests, in a highly degraded state, occupy only 5.7% of the total area. The villagers exploit the protected forests in their vicinity removing nearly 350.9 mt of fuelwood, 22.8 mt of small timber and 393.4 mt of fodder annually. The village has a manpower potential of 40 035 man days a year. Agriculture is the main source of employment at the moment absorbing 14 266 man days. The non-agricultural sector provides employment to an extent of 4433 man days. Fresh employment opportunities are needed to absorb nearly 53% of man days of the village at present unused. Three land-resource, skill-based package programmes, a tasar silk culture, an energy plantation, and dairying have been suggested to generate employment opportunities and to meet the basic needs of the villagers. About 115 ha of the hitherto neglected land resource has been located in the village to sustain such community forestry (CF) programmes. Tasar culture suggested on 35 ha will alone generate employment to an extent of 18 690 man days from the fourth year of tasar silk plantation. A plantation, to provide energy and fodder, on 25 and 30 ha respectively is expected to yield fuelwood, timber and fodder to more than satisfy village needs. A dairy has been recommended as it will generate income, provide nutrition and promote gobar gas production — an alternative energy source to fuelwood. The cost—benefit analysis of these projects indicates their economic feasibility with the cost likely to be recovered in 4.5 years. Debrasai being an agricultural village, a land-inequity compensatory scheme has been suggested for CF project net benefit sharing, ensuring reasonable benefit flows to the landless and marginal farmers. 相似文献
Intensive use of chemical herbicides and pesticides has raised serious concern about their effect on non-target organisms, plants, human bodies and the environment. Hence there is a continuous search for benign, harmless, non-hazardous and eco-friendly technology to control the growth of weeds which compete with crops for water, nutrients, light and space; thus reducing crop yield significantly. An attempt was made to control the weeds through smothering. This controls the weeds by cutting off light from the photosynthetic portion of weed plants by using black polyethylene sheets (LPDE film) which inhibit the top growth. Experiments to control the weeds in a lowland rice field through smothering were conducted in the high rainfall coastal region of eastern India wth eight treatments including smothering and conventional methods like hand/mechanical weeding and herbicides, etc. The treatment of smothering for one week + puddling + transplanting proved to be better than other conventional methods and may replace herbicide and hand/mechanical weeding totally with a better rice yield. 相似文献
Detached wheat ears (15 days post-anthesis) were cultured in liquid nutrient medium containing different sucrose concentrations (2. 6 and 10%). The volume of solution taken up was highest from 2% sucrose solution and decreased with increasing sucrose concentration. Grain development in terms of grain weight, glumes and rachis weight, nitrogen and protein content showed a linear increase in these variables over 11 days. Dry weight per grain increased with the sucrose level in the medium and the reduced uptake of solution did not decrease the moisture percentage of the grain. The culture of ears decreased the solution pH more with 2% than with 10% sucrose. Addition of 0.5% chloramphenicol to the culture solution had no adverse effect on grain weight, rather it prevented contamination and maintained a higher pH of the solution. 相似文献
Neonatal calf mortality is a major concern to livestock sector worldwide. Neonatal calf diarrhoea (NCD), an acute severe condition causes morbidity and mortality in calves. Amongst various pathogens involved in NCD, E. coli is considered as one of the major causes. The study was targeted to characterize E. coli isolates from neonatal calves for diarrhoeagenic Escherichia coli (DEC) types (pathotyping), antimicrobial resistance (AMR) profiling and to correlate with epidemiological parameters. From neonates, a total of 113 faecal samples were collected, out of that 308, lactose fermenting colonies were confirmed as E. coli. Pathotypable isolates (12.3%) were represented by STEC (6.1%), EPEC (2.9%), ETEC (1.9%), EAEC (0.9%) and EHEC (0.3%). Occurrence of STEC was more in non-diarrhoeic calves, whereas ETEC was observed more in diarrhoeic calves. EPEC occurrence was observed in both diarrhoeic and non-diarrhoeic calves. Fishers extract test showed no significant association for occurrence of DEC types to type of dairies, health status, species, breed, age and sex of neonatal calves. Two hundred and eighty isolates were tested for antimicrobial susceptibility. The isolates showed maximum resistance towards ampicillin (55.4%) followed by tetracycline (54.3%), while minimum resistance was observed towards meropenem (2.5%). Multidrug resistant E. coli isolates were found to be 139 (49.6%), and Extended-spectrum beta-lactamase (ESBL) producers were 120 (42.9%). DEC pathotypes like STEC, ETEC, EHEC and EAEC that are also multidrug resistant present in neonatal calves have zoonotic potential and hence are of public health significance.
A field study was conducted for six years (1981–1986) on sandy loam soil on intercropping hedgerows of Leucaena leucocephala (Lam) de Wit. with three field crops viz. maize (Zea mays L), black gram (Vigna mungo L) and cluster bean (Cyamopsis tetragonoloba L Taub.). In treatments 1 and 2 Leucaena hedges were planted as pure crops at close (25 cm × 75 cm) and wide (25 cm × 375 cm) spacings. In treatments 3, 4 and 5 the
three field crops were intercropped between the hedgerows of Leucaena at the wide spacing, and in treatments 6, 7 and 8 the field crops were raised as pure crops. Leucaena was topped to 75 cm each time it attained a height of 175 cm.
The pure crop of Leucaena at close spacing produced an average, over the six years, of 34 t ha−1a−1 of green fodder and 9.4 t ha−1a−1 of air dry fuelwood. The Leucaena at wide spacing produced 18.9 t ha−1a−1 of green fodder and 6.3 t ha−1a−1 of fuelwood. Intercropping with field crops decreased the yield of green fodder and fuelwood. The yield of all the field
crops was less when raised as intercrops than as pure crops.
Mean maximum net returns were obtained from intercrops of Leucaena and cluster bean (Rs 3540 ha−1a−1) which were significantly higher than the returns from pure crop of Leucaena at wide spacing but similar to the returns from pure crops of cluster bean. Leucaena with maize (Rs 3273 ha−1a−1) and black gram (Rs 3125 ha−1a−1) gave significantly higher net returns over pure crops of Leucaena at wide spacing, maize and black gram.
