An intergeneric hybrid between Brassica napus (AC, n = 19) and Raphanus raphanistrum (Rr, n = 9) was developed using the sequential ovary–ovule culture. Morphological, molecular, cytological studies and fluorescent genomic in situ hybridization were conducted to establish the hybridity and genomic relatedness. F1 hybrid plants were male sterile and morphologically intermediate between the two parents. Cytological analysis of the hybrid plants revealed 10II + 8I as the predominant meiotic configuration with the mean bivalent frequency of 8.83. A maximum of 13II were observed in 8.24% of the pollen mother cells (PMCs). GISH studies facilitated the identification of two allopairs, confirming homoeologous pairing between A/C and Rr genomes. This may have significant implications in terms of transgene flow and possible introgression into Rraphanistrum, which is a common canola weed in Canada and Australia. The intergeneric hybrid Bnapus × Rraphanistrum so produced also has the potential to be used as a bridging species for the transfer of desirable genes to amphiploid crop Brassica species carrying A/C genomes.  相似文献

This study compared endometrial cytology vis‐a‐vis uterine fluid cytology for assessment of uterine health in clinically normal and subclinical endometritis (SE)‐affected buffaloes. Uterine fluid samples and endometrial samples were collected from the buffaloes (n = 38) at oestrus using blue sheath and cytobrush, respectively. The smears were stained with Field stain for 3 minutes, and a minimum of 400 cells were counted in each smear for determination of the percentage of polymorphonuclear (PMN) leucocyte. The incidence of subclinical endometritis, based on the cytobrush cytology, was 23.08%. The correlation between cytobrush cytology with uterine fluid cytology was positive and significant (r = .37; p = .02). The ratio of PMN leucocyte in cytobrush cytology to uterine fluid cytology was 1:2.4. ROC analysis revealed that the threshold value of 6.16% PMN leucocyte in uterine fluid cytology showed a diagnostic sensitivity and specificity of 100% in differentiating normal from SE‐affected buffaloes. In conclusion, collection of uterine fluid was easier compared to collection of endometrial samples using cytobrush and the percentage of PMN leucocyte in uterine fluid cytology can be used as a tool for diagnosis of subclinical endometritis in buffaloes.  相似文献

Non‐genetic factors influencing functional longevity and the heritability of the trait were estimated in South African Holsteins using a piecewise Weibull proportional hazards model. Data consisted of records of 161,222 of daughters of 2,051 sires calving between 1995 and 2013. The reference model included fixed time‐independent age at first calving and time‐dependent interactions involving lactation number, region, season and age of calving, within‐herd class of milk production, fat and protein content, class of annual variation in herd size and the random herd–year effect. Random sire and maternal grandsire effects were added to the model to estimate genetic parameters. The within‐lactation Weibull baseline hazards were assumed to change at 0, 270, 380 days and at drying date. Within‐herd milk production class had the largest contribution to the relative risk of culling. Relative culling risk increased with lower protein and fat per cent production classes and late age at first calving. Cows in large shrinking herds also had high relative risk of culling. The estimate of the sire genetic variance was 0.0472 ± 0.0017 giving a theoretical heritability estimate of 0.11 in the complete absence of censoring. Genetic trends indicated an overall decrease in functional longevity of 0.014 standard deviation from 1995 to 2007. There are opportunities for including the trait in the breeding objective for South African Holstein cattle.  相似文献

Ultraviolet laser microprobe analyses of a calcium-aluminum-rich inclusion (CAI) from the Allende meteorite suggest that a line with a slope of exactly 1.00 on a plot of delta17O against delta18O represents the primitive oxygen isotope reservoir of the early solar nebula. Most meteorites are enriched in 17O and 18O relative to this line, and their oxygen isotope ratios can be explained by mass fractionation or isotope exchange initiating from the primitive reservoir. These data establish a link between the oxygen isotopic composition of the abundant ordinary chondrites and the primitive 16O-rich component of CAIs.  相似文献

Recently, the Kunjin strain of West Nile virus (WNVKUN) has been detected using qRT‐PCR in belly skin lesions of farmed juvenile saltwater crocodiles. This follows an established association between similar lesions and West Nile virus in American alligators. The lesions present as cutaneous lymphohistiocytic aggregates in the dermal layers of both species. While these lesion do not create an obvious defect on the live crocodile, upon tanning the lesion area collapses and does not uptake the dye evenly, thus reducing its aesthetic appeal. As a result, skins are being rejected jeopardising the economic viability of the Australian crocodile industry. Over 50 skin lesions have since been confirmed as WNVKUN‐positive and preliminary evidence of lesion restructuring is presented. Horizontal transmission of WNVKUN by mosquitoes is well‐established but other transmission routes, such as ingestion and cloacal shedding, need further evaluation. An infection trial is currently underway to ensure WNVKUN is the causative agent of these skin lesions.  相似文献

An online survey on the state of existing dairy data, dairy improvement infrastructure and human capacity in sub-Saharan Africa (SSA) was undertaken with the aim of assessing whether the state of existing animal recording, dairy improvement methods and key issues facing dairy production together with means of addressing the issues differ among countries and regions of SSA. Respondents comprised experts and practitioners in livestock production and genetic resources from research institutes, animal breeding companies, universities, non-governmental organisations and government agricultural ministries. The main dairy farming system in which the respondents were involved was mixed crop-livestock system (30.2%), and this was mainly practised in the private land tenure system (46.3%). Data were analysed using linear model and paired Student t test in R software package. Respondents identified key issues affecting dairy production as poor genetic assessment of imported exotic breeds and crosses in Africa (62.3%), fluctuations in milk prices within both the formal and informal markets (50.9%), no comprehensive sire ranking systems (39.6%), housing and health management regimes which adversely affect milk yield (32.1%), poor market networks for dairy products (25.5%), poor feeding (13.3%), inadequate genetic technologies (9.4%) and poor animal performance recording systems (9.4%). Respondents emphasised the need for updated breeding policies, sire ranking systems, adequate farm management systems, capacity building, across-country collaborations and joint genetic assessments of dairy breeds found in sub-Saharan Africa. The current situation of dairy production though similar for the different countries, differed in order of emphasis and magnitude across the countries and regions in sub-Saharan Africa.

In a field experiment with soil compaction by tractor traffic on a loam soil, the denitrification rate (using the C2H2 inhibition method), the soil structure, and the wheat yield were investigated. Tractor traffic on wet soil (> – 50 mbar matric potential) reduced the pore volume, doubled the percentage of large aggregates (> 20 mm), reduced the wheat yield by about 25%, and increased the N-loss through denitrification by a factor of 3–4. Neither of these parameters were affected by tractor traffic at low soil moisture content. The weight of the tractor (1800 kg vs 4800 kg) did not significantly alter the effect of compaction on the measured parameters. There was a factor of 2–6 between the measured denitrification rate in compacted and that in uncompacted soil, and this factor showed little dependence on the average activity level on each date of measurement. Accumulated values for the measured denitrification during 75 days (May 23-August 9) were 3–5 kg N ha–1 in uncompacted soil and 15–20 kg N ha–1 in soil which was compacted in wet condition.  相似文献

Optically gated recording and nonvolatile readout in a digital volume holographic data storage system that uses a pair of mutually incoherent light sources during recording and only one for readout were demonstrated recently. This approach used stoichiometric lithium niobate, which after post-growth processing gave rise to an at least two orders of magnitude improvement in sensitivity over the best materials reported previously. It is also shown that by adding certain dopants (iron and manganese) to near-stoichiometric lithium niobate, the dark storage time and gating efficiency can be increased compared with previous work. The underlying physical mechanisms of gated recording and the effectiveness of the gating process responsible for this manifold improved performance are discussed, and bipolarons and small polarons are identified as the responsible photorefractive species.  相似文献

Laboratory studies show that the reaction of short-lived O2(B3Sigmau) molecules (lifetime approximately 10 picoseconds) with N2 and the photodissociation of the N2:O2 dimer produce NOx in the stratosphere at a rate comparable to the oxidation of N2O by O(1D). This finding implies the existence of unidentified NOX sinks in the stratosphere. The NO2 observed in this experiment is isotopically heavy with a large 15N/14N enhancement. However, photodissociation of this NO2 unexpectedly produced NO molecules with a low 15N/14N ratio. The diurnal odd-nitrogen cycle in the stratosphere will be marked by a complex isotope signature that will be imprinted on the halogen and HOX catalytic cycles.  相似文献
