Oral squamous cell carcinoma (OSCC) is an aggressive and treatment‐resistant malignancy in both feline and human patients. Recent work has demonstrated aberrant expression of fatty acid synthase (FASN) and an increased capacity for lipogenesis in human OSCC and other cancers. In human OSCC, inhibition of FASN decreased cell viability and growth in vitro, and diminished tumour growth and metastasis in murine preclinical models. This study aimed to characterize FASN as a therapeutic target in feline OSCC. Immunohistochemistry revealed high FASN expression in primary feline OSCC tumours, and FASN expression was detected in OSCC cell lines (3 feline and 3 human) by immunoblotting and quantitative real‐time‐polymerase chain reaction (qRT‐PCR). Orlistat, a FASN inhibitor, substantially reduced cell viability in both feline and human OSCC lines, although feline cell lines consistently displayed higher sensitivity to the drug. FASN mRNA expression among cell lines mirrored sensitivity to orlistat, with feline cell lines expressing higher levels of FASN. Consistent with this observation, diminished sensitivity to orlistat treatment and decreased FASN mRNA expression were observed in feline OSCC cells following incubation under hypoxic conditions. Treatment with orlistat did not potentiate sensitivity to carboplatin in the cell lines investigated; instead, combinations of the 2 drugs resulted in additive to antagonistic effects. Our results suggest that FASN inhibition is a viable therapeutic target for feline OSCC. Furthermore, cats may serve as a spontaneous large animal model for human oral cancer, although differences in the regulation of lipogenesis between these 2 species require further investigation.  相似文献

Two batches of experimental media containing 10% milk whey (sweet or sour) and 1% bovine serum supported the growth of BHK-21 cells in serial passages. The cell yield was greater than 2×106/ml at 48 h. The cells were susceptible to inoculation with foot and mouth disease (FMD) virus type A and the virus titres obtained were comparable with those in Eagle's MEM-G with 10% serum. The use of milk whey could reduce the consumption of serum required for the growth of FMD virus in BHK-21 cells by up to 90%.  相似文献

The effects of a commercial lactate salt formulation—containing potassium lactate (KL) and potassium acetate (KA)—and liquid smoke (LS) on the growth of selected lactic acid bacteria (LAB; Carnobacterium maltaromaticum, Carnobacterium inhibens, Lactococcus lactis, Lactobacillus curvatus, and Enterococcus faecalis), fish spoilage bacteria (Photobacterium phosphoreum, Pseudomonas putida, and Vibrio vulnificus), and Listeria innocua, were examined in a tryptic soy broth model system at 20ºC based on BioScreen data. The most pronounced inhibition effect on growth of bacteria was seen in the presence of 6% KL + 0.4% KA, used either combined with LS or alone. Only a minor inhibition effect on growth was found in the presence of LS alone. The only exception was Lactobacillus curvatus, which grew quite well in the presence of LS compared to control medium. The growth of Vibrio vulnificus was prevented in 6% KL + 0.4% KA, and significantly inhibited in the presence of 3% KL + 0.2% KA. When V. vulnificus was grown in NaCl, KA, and KL + KA, it was observed that KL + KA had a better inhibition effect than sodium salt within the same concentration range.  相似文献

Biochar (BC) application to agricultural soil has been proposed as an effective countermeasure to mitigate climate change. A laboratory incubation experiment was carried out to gain insight into the effectiveness of BC on methane (CH4) consumption in a tropical clayey vertisol. Except for the control treatment, BC of two different sizes (<0.25 or 0.25–2.00 mm) was mixed with vermicompost (VC), poultry manure (PM) or farmyard manure (FYM). BC and organic amendment were added to soil at 10% w/w and 80 kg N/ha, respectively. BC increased CH4 consumption rate, k, in soil, irrespective of organic amendment type. The CH4 consumption potential of soil was greater with the smaller size BC (<0.25 mm). Of the three organic amendments, VC exhibited the highest k (0.105) followed by FYM (0.093) and PM (0.072). BC (<0.25 mm) + PM was the most effective of the organic amendments in enhancing CH4 consumption (k = 0.242). The lag phase varied between 7.3 day (control) and 1.0 day (soil + VC). Results revealed that there was a significant (P < 0.0001) effect of organic amendment and BC on CH4 consumption, CO2 production and microbial abundance. Cumulative CO2 production (mg/g soil) varied between 2.15 (control) and 8.77 (soil + PM + BC < 0.25 mm). Pearson's correlation analysis showed significant correlation between CH4 consumption and methanotrophs abundance (P < 0.001). The study shows that BC enhanced CH4 consumption potential in agricultural land on a tropical vertisol, particularly using the smaller size (<0.25 mm), and could be an effective strategy to mitigate atmospheric CH4.  相似文献

Here, we report the karyotypes and mean haploid idiograms of the ten Asian Corchorus species (2n = 2x = 14). Chromosomes were small, with a mean chromosome length of 2.30 μm. The largest chromosome was recorded in C. pseudo-olitorius (3.50 μm) and the shortest in C. pseudocapsularis (1.60 μm). The karyotypes of the two cultivated species (C. capsularis and C. olitorius) and C. pseudo-olitorius were the most diverse and specialized, whereas those of C. depressus and C. trilocularis were the least diverse. C. fascicularis had the most asymmetrical and C. urticifolius the most symmetrical karyotypes. An increase in genome size was accompanied by increasing karyotype diversity in terms of morphologically distinct chromosome types and interchromosomal asymmetry, with uneven distribution of additional DNA throughout the karyotype. A positive correlation between interchromosomal asymmetry and dispersion index suggested that size differences between chromosomes were mainly associated with karyotype asymmetry. Karyotypes of the Corchorus species became progressively asymmetrical in the course of evolution. Relationships among the ten Corchorus species were defined by using a neighbor-joining tree inferred from inter-simple sequence repeat data. C. fascicularis and C. pseudocapsularis, with shorter karyotypes and smaller genomes, were closely related to C. pseudo-olitorius and C. capsularis, respectively, which were characterized by relatively longer karyotypes and larger genomes. However, the two cultivated species with different levels of interchromosomal asymmetries, dispersion indices and genome sizes were distantly related. Taking this molecular evidence into consideration, we have discussed chromosomal evolution in relation to karyological data including genome size.  相似文献

Consequent to the interest in converting degraded lands for cultivation of biofuel crops, concerns have been expressed about greenhouse gas (GHG) emissions resulting from changes in soil‐carbon (C) stock following land conversions. A literature‐based study was undertaken for estimating the magnitude of emission of GHGs, particularly carbon dioxide (CO2), following an assessment of the extent and causes of land degradation and the nature of CO2 emission from soils. The study estimated the potential for CO2 emission resulting from changes in soil‐carbon stock following land conversions, using oil palm (Elaeis guineensis Jacq.) as a case study. The analysis indicated that, overall, the magnitude of CO2 emission resulting from changes in soil C stock per se following opening up of degraded land would be low compared with other potential sources of CO2 emission. However, lack of data on critical aspects such as baseline soil C status was a limitation of the study. Soil respiration is the single best measure of GHG emission from soils. Fixation of C in additional biomass will compensate, over time, for C loss through soil respiration following a change in land use or land management, unless such changes involve conversion of existing large C stocks. Therefore, any net CO2 emission from soils resulting from changes in soil C stock following opening up of degraded land is likely to be a short‐term phenomenon. The aim of this study was to determine whether processed karanj (Pongamia glabra) oil seed cake can be used as a supplement to partially replace soybean meal (SBM). Male lambs (n = 24) of uniform body weight (12.88 ± 0.15 kg) were equally allotted at random to a SBM‐based control (CON) and three test concentrate mixtures, containing detoxified solvent extracted karanj cake (SKC) using three processing methods: water washing (WW), 2.5% lime (LM) and 0.4% binder (BN) treatment. The processed SKC replaced 50% nitrogen of SBM of CON. The respective concentrate mixtures were fed along with ad libitum chaffed oat (Avena sativa) straw for 196 days. Dry matter intake was significantly (p < 0.01) lower on WW, LM and BN. Apparent digestibility coefficient of nutrients was comparable, except for total carbohydrates, which was significantly (p < 0.01) lower in LM and BN. Total gain, average daily gain and feed: gain ratio was comparable (p > 0.05) between the CON and WW diets but significantly lower in LM and BN groups. Yield of greasy wool was lower (p < 0.05) in BN group. Comparable dry matter and nutrient (crude protein and total digestible nutrients) conversion efficiency was observed on CON and WW diet but the lambs on the LM and BN diets exhibited lower (p < 0.01) conversion efficiency. It is concluded that SKC after water washing could replace 50% of SBM nitrogen in protein supplementation.  相似文献

In Arunachal Pradesh and other sub-Himalayan areas of India, accidental consumption of Senecio plants by yaks is often fatal as the plant contains toxic alkaloids like Seneciophylline. The present investigation was undertaken to demonstrate the pro-oxidant effects of an ethanolic extract of Seneciochrysanthemoides (S-EtOH). S-EtOH impaired viability in macrophages, the IC50 being 13.8 ± 1.11 μg/mL. The effect of S-EtOH (1 μg/mL) on generation of reactive oxygen species (ROS) in macrophages was measured by flow cytometry using 2′,7′-dichlorofluorescein diacetate (H2DCFDA) where it caused a significant increase in the mean fluorescence channel (MFC) from 8.55 ± 0.03 to 47.32 ± 2.25 (p < 0.001). S-EtOH also effected a 3.8-fold increase in extracellular nitric oxide (NO) generation from 4.90 ± 0.72 μM to 18.79 ± 0.32 μM (p < 0.001), a 2.2-fold increase in intracellular NO production, the MFC increasing from 14.95 ± 0.48 to 33.34 ± 1.66 (p < 0.001), and concomitantly depleted non protein thiols as analyzed by flow cytometry using mercury orange, with a reduction in MFC from 632.5 ± 49.44 to 407.4 ± 12.61 (p < 0.01). Additionally, S-EtOH (14 μg/mL, 24 h) caused apoptosis as evident by increased Annexin V binding and terminal deoxynucleotidyl transferase mediated dUTP DNA nick end labeling. Taken together, the cytotoxicity of S-EtOH can be partly attributed to its capacity to inflict oxidative damage via generation of both reactive oxygen and nitrogen species culminating in apoptosis.  相似文献
