Two different wild duck species common in Chile and neighboring countries, Chiloe wigeon (Anas sibilatrix) and cinnamon teal (Anas cyanoptera), were intranasally inoculated with 10(6) mean embryo infective dose (EID50) of the H7N3 low pathogenicity (LP) avian influenza virus (AIV) (A/chicken/Chile/176822/02) or high pathogenicity (HP) AIV (A/chicken/Chile/ 184240-1/02), in order to study the infectivity and pathobiology of these viruses. None of the virus-inoculated ducks had clinical signs or died, but most seroconverted by 14 days postinoculation (DPI), indicating a productive virus infection. Both LPAIV and HPAIV were isolated from oral swabs from two of six Chiloe wigeons and from oral and/or cloacal swabs from all five of the cinnamon teal at 2 DPI. Both LPAIV and HPAIV were efficiently transmitted to cinnamon teal contacts but not to Chiloe wigeon contacts. This study demonstrates that the cinnamon teal and Chiloe wigeons were susceptible to infection with both Chilean H7N3 LPAIV and HPAIV, but only the cinnamon teal showed contact transmission of the virus between birds, suggesting that the cinnamon teal has the potential to be a reservoir for these viruses, especially the LPAIV, as was demonstrated in 2001 with isolation of a genetically related H7N3 LPAIV strain in a cinnamon teal in Bolivia. This study was to investigate dynamic and evolution of PRRSV in a seed-stock farm by monitoring PRRSV status from 11 June 2009 to 4 August 2010. For laboratory test, around 18-24 umbilical cords from farrowed sows and 5-95 sera from nursery and grow/finish pigs were submitted around every 2 weeks interval during the study. The submitted samples were tested for PRRSV using IDEXX PRRS 2XR ELISA kit, RT-nested PCR. The PRRSV-positive samples were further sequences based on ORF5 and analyzed using MEGA 3.1 program and Beast 1.5.4 package. The surveyed farm was first infected with type II PRRSV but it was infected newly with type I PRRSV of unknown origin, showing rapid substitution to type I PRRSV as a dominant strain in 2 weeks. The type I PRRSV was first detected from umbilical cord of a farrowed sow in 12 January 2010, and secondly from nursery pigs in 26 January 2010. Although sudden increase of mean S/P ratio was found in grow/finish pigs around 2 months earlier than first type I PRRSV detection, no type I PRRSV viremia was found. Thirty three ORF5 full sequences from 14 type II to 19 type I PRRSVs were obtained chronologically in this farm and the genetic characteristics and evolution rates of those sequences were analyzed. The substitution rates (/site/day) of two types were 4.03×10(-5) (type I), 3.09×10(-5) (type II), respectively, which was more frequent than previous reports. The calculated divergence time of type I PRRSV was consistent with the time when the sudden elevation of serum IgG in grow/finish barn was first observed. This study provided fundamental data for type I PRRSV dynamic in a previously type II PRRSV-infected farm and suggested grow/finisher barn could be a primary site for PRRSV introduction. 相似文献

A wide variety of veterinary antibiotics (VAs) has been detected in environmental water samples, and this is of potential environmental concern due to their adverse effects. In particular, the potential for development of antibiotic-resistant bacteria has raised social concerns leading to intensive investigation regarding the influence of antibiotics on human and ecosystem health. One of the main sources of antibiotic effluence to the environment is livestock manures that often contain elevated levels of VAs that survive normal digestive procedures following medication in animal husbandry because unlike human waste, waste generated on farms does not undergo tertiary wastewater treatment, and consequently, the concentration of antibiotics entering the environment is expected to be larger from farming practices. Animal feed is often supplemented with VAs to promote growth and parasite resistance in the medicated animals, and this practice typically resulted in higher use of VAs and consequential excretion from livestock through urine and feces. The excretion rate varied depending on the type of VA used with around 75, 90, and 50?C100% being excreted for chlortetracycline, sulfamethazine, and tyolsin, respectively. The excreted VAs that initially present in livestock manures were degraded more than 90% when proper composting practice was used, and hence, this can be employed as a management strategy to decrease VA environmental loads. The reduction of VA concentrations during composting was mainly attributed to abiotic processes rather than biotic degradation. The VAs released to soils by the application of manure and manure-based composts can be degraded or inactivated to various degrees through abiotic process such as adsorption to soil components. Depending on the antibiotic species and soil properties, residues can be transferred to groundwater and surface water through leaching and runoff and can potentially be taken up by plants. 相似文献

The ultimate goal of our study is to establish thin-layer chromatography (TLC) as a quick and simple method for identifying the type of refined petroleum products present in the environmental media. As a preliminary step, TLC chromatograms of different petroleum products, including gasoline, kerosene, and diesel, were characterized and compared. Methanol was determined as the optimum carrier solution in TLC analysis. The spherical-shaped TLC chromatogram of gasoline showed the longest migration distance, and thus the highest retardation factor (R f) of 0.91. This was followed by that of kerosene (0.63) with an elliptical-shaped, and diesel (0.24) with an elongated trapezoid-shaped chromatogram. R f of kerosene and diesel increased with the dilution factor, while gasoline showed a constant value. Additionally, it was observed that the TLC chromatograms of oils produced the same peak pattern with the corresponding petroleum products in gas chromatography (GC). A mixed sample of kerosene and diesel presented a triangular shaped chromatogram, underlining the need to consider the shape of chromatogram in addition to the R f value, as an indicator of the petroleum type. The findings indicate that TLC has a huge potential to be used as a quick and reliable method for identifying the type of refined petroleum products in the environmental media. 相似文献

genes conditioning resistance to location-specific isolates of the blast pathogen and thereby to identify linked markers to
resistance gene for isolate KI-313 collected in Korea, RFLP markers previously reported to be linked to major blast resistance
genes in different rice germplasm and other markers mapped to nearby regions were surveyed for polymorphism between a resistant
(`Suweon 365') and a susceptible (`Chucheongbyeo') parent. Linkage associations of the RFLP markers with the resistance gene
were verified using an F2 and F3 segregating population of known blast reaction. RFLP analysis showed that Pi-18 was located near the end of chromosome 11, linked to a single copy clone RZ536 at a distance of 5.4 centiMorgans (cM) and
that this gene was different from Pi-1(t). An allelism test revealed that this gene was also different from Pi-k. Currently, a combination of RAPD and microsatellite primers is being employed to find additional markers in this region.
Tightly linked DNA markers will facilitate selection for resistant genotypes in breeding programs and provide the basis for
map based cloning of this new blast resistance gene.

Aging is a major risk factor for many chronic diseases, such as cancer, cardiovascular disease, and diabetes. The exact mechanisms underlying the aging process are not fully elucidated. However, a growing body of evidence suggests that several pathways, such as sirtuin, AMP-activated protein kinase, insulin-like growth factor, autophagy, and nuclear factor erythroid 2-related factor 2 play critical roles in regulating aging. Furthermore, genetic or dietary interventions of these pathways can extend lifespan by delaying the aging process. Seaweeds are a food source rich in many nutrients, including fibers, polyunsaturated fatty acids, vitamins, minerals, and other bioactive compounds. The health benefits of seaweeds include, but are not limited to, antioxidant, anti-inflammatory, and anti-obese activities. Interestingly, a body of studies shows that some seaweed-derived extracts or isolated compounds, can modulate these aging-regulating pathways or even extend lifespans of various animal models. However, few such studies have been conducted on higher animals or even humans. In this review, we focused on potential anti-aging bioactive substances in seaweeds that have been studied in cells and animals mainly based on their anti-aging cellular and molecular mechanisms. 相似文献

Salinity is one of the major abiotic stresses that severely effects rice production throughout the world. Previously, there have not been many studies which focused on studying diversity among rice germplasm based on specific physiological traits for salt tolerance. Our diversity study was based on physiological traits such as Na+ concentration, K+ concentration, K+/Na+ ratio, osmotic potential, and biomass which are major components determining salt tolerance. This study has systematically analyzed phenotypic data of 191 germplasms in two different salt concentrations apart from the control. The current study identified salt tolerant germplasms based on their response to a single physiological trait as well as a combination of different physiological traits. Some of the germplasm identified outperformed known salt tolerant cultivar Pokkali. This study identifies correlation among various physiological traits. The salt tolerant germplasms can be taken forward into developing better varieties by conventional breeding and exploring genes for salt tolerance. 相似文献
