This study was carried out to determine the pretreatment effect of proton beam irradiation on germination and pasting properties of starches in two rices. Mature and healthy seeds irradiated with 10 doses (0, 50, 100, 200, 300, 400, 500, 600, 700, and 800 Gy) for determination of LD50 and characteristics of germination were recorded at 14 days after irradiation. The rice seeds irradiated with five doses (0, 50, 100, 200, and 300 Gy) were used to evaluate the irradiation effects of pasting properties of starches. It showed that a lower survival rate in germinated rice seeds was observed above 300 Gy showing 31 and 35% in Ilpum and Hanmaum, respectively. The higher plant height and root length were also recorded in 50 and 100 Gy. Amylose content in proton beam irradiated with 50, 100, 200, and 300 Gy was significantly decreased in two rice cultivars. Peak viscosity, hot peak viscosity, cooling peak viscosity, and setback viscosity decreased with increasing proton beam dose levels. The degree of crystallinity was significantly increased with increasing proton beam dose levels. Consequently, it might be deduced that proton beam irradiation causes changes of starch properties affecting crystalline regions of starch granules, especially at high dosage irradiation.  相似文献

The expression of the major histocompatibility complex (MHC) classical class I genes is important for the adaptive immune response to target virus-infected cells and cancer cells. The up-regulation of the MHC is achieved by hormonal/cytokine signals including IFN-γ-inducible elements. The swine leukocyte antigen (SLA), the MHC class I region of pigs, consists of the duplicated classical class I genes, SLA-1, SLA-2 and SLA-3, but the molecular mechanisms involved in their up-regulation after T cell stimulation have not been fully elucidated. In order to better understand some of the putative regulatory mechanisms of SLA class I gene expression in activated T cells, we examined the coordinated expression of the SLA classical class I, IFN-γ and interferon regulatory factor-1 (IRF-1) genes in the peripheral blood mononuclear cells (PBMCs) of SLA homozygous Clawn miniature swine stimulated for 72h with either IFN-γ or an enterotoxin produced by Staphylococcus aureus. This enterotoxin, toxic shock syndrome-1 (TSST-1), is known to act as a superantigen (sAG) to activate the T cells in various vertebrate species. We showed by using mAbs and flow cytometry that the CD4(+)CD25(+) cell number of swine PBMCs was also increased by TSST-1 and to a lesser degree by IFN-γ. Time course analyses of the expression of the IFN-γ, IRF-1 and the three classical class I genes, SLA-1, SLA-2, and SLA-3, in PBMCs by quantitative real-time PCR revealed a transitory response to TSST-1 or IFN-γ stimulation. The IFN-γ mRNA levels in the PBMCs were continuously up-regulated over the first 48h by TSST-1 or IFN-γ. In contrast, SLA class I expression moderately increased at 24h and then decreased to a baseline level or less at 72h of IFN-γ or TSST-1 stimulation. The three classical SLA class I genes showed similar expression kinetics, although SLA-3 mRNA level was consistently lower than those of SLA-1 and -2. The expression of IRF-1, a modulator of SLA expression, showed similar kinetics to those of the three classical SLA class I genes. The expression profiles detected by flow cytometry of the SLA molecules on the cell surface of PBMCs were maintained at a consistently high level during cell stimulation with either TSST-1 or IFN-γ, which was distinct from the kinetics of mRNA expression. These results showed that miniature swine SLA class I mRNA expression was effectively and equally up-regulated among the three loci and coordinately with IRF-1 gene expression after stimulation of T cell activation by sAG or IFN-γ.  相似文献

To investigate the potential transmission of subtype H3 influenza virus to cats, a serological survey was carried out in South Korea. Serum samples (n = 1027) were obtained from 809 pet cats and 218 domesticated cats living in urban colonies (D-cats) from 2008 to 2010, and tested using an influenza anti-nucleoprotein (NP)-specific enzyme-linked immunosorbent assay (ELISA) and the haemagglutination inhibition (HI) test, which was recommended by the World Organization for Animal Health. Anti-influenza virus antibodies were detected in 3.12% and 2.43% of cat sera tested using the NP-specific ELISA and HI test, respectively. Anti-H3 antibodies were also identified when the HI assay was used for influenza virus serotyping. These data may indicate the sporadic transmission of subtype H3 influenza virus from other infected species to cats in South Korea.  相似文献

Genome‐wide association mapping for complex traits in cattle populations is a powerful, but expensive, selection tool. The DNA pooling technique can potentially reduce the cost of genome‐wide association studies. However, in DNA pooling design, the additional variance generated by pooling‐specific errors must be taken into account. Therefore, this study aimed to investigate factors such as: (i) the accuracy of allele frequency estimation; (ii) the magnitude of errors in pooling construction and in the array; and (iii) the effect of the number of replicate arrays on P‐values estimated by a genome‐wide association study. Results showed that the Illumina correction method is the most effective method to correct the allele frequency estimation; pooling errors, especially array variance, should be taken into account in DNA pooling design; and the risk of a type I error can be reduced by using at least two replicate arrays. These results indicate the practical capability and cost‐effectiveness of pool‐based genome‐wide association studies using the BovineSNP50 array in a cattle population.  相似文献

Natural regeneration by strip-clearcut has rarely been used for restocking of evergreen broad-leaved forests in the East Asia. To contribute to a better understanding of the likely reasons for either the successful recovery of these forests or failure, the early response of stand structure and tree species diversity to strip-clearcut were studied in Okinawa Island, southwest Japan. The 4,000 m2 study area was divided into 10 adjacent sections of equal area with a 10 m × 10 m sampling plot in the center, of which five sections were strip-clearcut while the remaining five sections uncut. Eight-year secondary succession following strip-clearcut showed that the density of woody stems, tree species diversity increased significantly not only in the regenerating stands but also in the residual stands compared to the primary stands. The primary dominant target species, Castanopsis sieboldii, continued to dominate both the regenerating and residual stands although colonizing species invaded abundantly in the stands, occupying approximately one-third of total tree species in each treatment. The community similarity analyses showed that the species composition were similar between the primary stands and the regenerating stands or residual stands reflected the high similarity amongst the primary stands and the regenerating stands or the residual stands. The similar stand structure, high tree species diversity and greater community similarity after strip-clearcut provided no evidence of forest degeneration, suggesting that the regenerating stand might gradually develop into stand similar to that prior to strip-clearcut.  相似文献

ABSTRACT

Sugarcane smut, caused by Sporisorium scitamineum, is one of the most important sugarcane diseases in Japan. Wild sugarcane, Saccharum spontaneum, is known to be a key breeding material to obtain high-yielding clones. In this study, we sought to identify Japanese wild sugarcane accessions with high resistance to smut. Thirty wild sugarcanes and three sugarcane cultivars were tested by the pinprick method. The results of the inoculation tests aided in identifying wild sugarcanes with high resistance to smut disease, namely JW90, Iriomote8, and Iriomote15. After screening the germplasm, progeny distribution of smut resistance from the inoculation test and dry matter productivity in the smut disease-free field were compared. The highly resistant wild sugarcane accession had a much better impact on progeny distribution of smut resistance compared with the susceptible accession. No relationship was found between smut resistance and dry matter productivity in both populations.  相似文献

This study aimed to investigate the efficacy of banana peel extract as a possible substitute for the environmentally unfavorable sodium dithionite in cellulosic fiber dyeing with indigo. Antioxidant activity and total sugar content of the banana peel extract were measured. Composition of the extract was investigated by GC/MS analysis. Its reducing power for synthetic indigo was evaluated by reduction potential measurements and dyeing tests. It was shown that the studied extract could be applied successfully in the reduction of indigo in spite of their lower reducing power when compared with that of sodium dithionite. In dyeing study, sodium dithionite showed its superiority in term of reducing power over the studied extract but the studied extract was superior stability of reduction to sodium dithionite. So, it can be concluded that banana peel extract could be a biodegradable and sustainable alternative to sodium dithionite as a reducing agent in indigo dyeing processes.  相似文献

We analyzed alcoholic extracts of herbs possessing anti-neosporal activity against Neospora (N.) caninum. To identify the chemical components of Sophora (S.) flavescens and Torilis (T.) japonica associated with anti-neosporal activity, specific fractions were isolated by high-performance liquid chromatography (HPLC). In vitro activity of the fractions against N. caninum was then assessed. Gas chromatography/mass spectrometry (GC/MS) was used to identify and quantify specific anti-neosporal molecules in the herbal extracts. Almost all HPLC fractions of S. flavescens and T. japonica had higher levels of anti-neosporal activity compared to the not treated control. Active constituents of the extracts were sophoridane, furosardonin A, and tetraisopropylidene-cyclobutane in S. flavescens; 5,17-β-dihydroxy-de-A-estra-5,7,9,14-tetraene, furanodiene, and 9,12-octadecadienoic acid (Z,Z)-(CAS,1) in T. japonica.  相似文献

The hippocampus is affected by various stimuli that include hyperglycemia, depression, and ischemia. Calcium-binding proteins (CaBPs) have protective roles in the response to such stimuli. However, little is known about the expression of CaBPs under diabetic conditions. This study was conducted to examine alterations in the physiological parameters with type 1 diabetes induced with streptozotocin (STZ) as well as time-dependent changes in the expression of two CaBPs changes of were being evaluated. Rats treated with STZ (70 mg/kg) had high blood glucose levels (>21.4 mmol/L) along with increased food intake and water consumption volumes compared to the sham controls. In contrast, body weight of the animals treated with STZ was significantly reduced compared to the sham group. CB-specific immunoreactivity was generally increased in the hippocampal CA1 region and granule cell layer of the dentate gyrus (DG) 2 weeks after STZ treatment, but decreased thereafter in these regions. In contrast, the number of PV-immunoreactive neurons and fibers was unchanged in the hippocampus and DG 2 weeks after STZ treatment. However, this number subsequently decreased over time. These results suggest that CB and PV expression is lowest 3 weeks after STZ administration, and these deficits lead to disturbances in calcium homeostasis.  相似文献
