ABSTRACT: A new abalone postlarval culture system, Stott's abalone postlarval production system (SAPPS), is being developed as an alternative method for settling larvae and on-growing postlarval abalone. SAPPS was tested in trial 1 using two commercially available artificial diets, Adam and Amos (Adam; supplied by Adam and Amos, Mt Barker, SA, Australia) and Cosmo (Cos; supplied by Cosmo, Matsuyama, Japan), against the diatom biofilm method (Diatom). In trial 2, SAPPS was tested against the previous production system (PPS, used by Stott et al. 2002). Larvae were induced to metamorphose and the resulting postlarvae were on-grown in 10-L flow-through tanks for 28 days in their respective treatments. The final survival of postlarvae was significantly higher ( P < 0.05) in the SAPPS-Cos group (56.7 ± 11.15%) than in the Diatom (9.4 ± 2.7%) and SAPPS-Adam groups (8.5 ± 1.1%). The final length of postlarvae in SAPPS-Cos (1065 ± 73 µm) was significantly higher ( P < 0.05) than those in the other two treatments (average of 841–883 µm). In trial 2 the metamorphosis rate was improved by using Spirulina platensis. The final length of postlarvae in SAPPS (1449–1471 µm) was significantly higher ( P < 0.05) than PPS (1065–1075 µm) and Diatom (993 µm). There is potential for SAPPS to be used as an alternative to the current diatom method in the culture of postlarval abalone. 相似文献

Gassericin A, a bacteriocin produced by Lactobacillus gasseri LA39, has a cyclic structure linking N‐ and C‐terminal amino acids. Gassericin A was expressed in Escherichia coli JM109 as a biotinylated fusion protein on the basis of the DNA sequence of mature bacteriocin. A positive clone accumulated the bacteriocin, with no activity, as a soluble fusion protein in the cytoplasm. After release of an N‐terminal tag with factor Xa protease, gassericin A was converted into an active peptide having N‐ and C‐termini. The total amount of purified bacteriocins (expressed and native) was 480 µg/L and 370 µg/L, respectively. However, the specific activity of expressed gassericin A was 15 AU/mg lower than that of native bacteriocin (2600 AU/mg). Although the actual Mr (molecular weight) of the expressed bacteriocin should be 5666, the peptide showed the same mobility (Mr 3800) in sodium dodecylsulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE) as native cyclic gassericin A, suggesting that the expressed peptide retains compact folding of the molecule similar to that of native gassericin A. 相似文献

A method for visualizing the sugar content in the flesh of melons was developed. This method was based on the sugar absorption band in the near-infrared (NIR) region to avoid bias caused by the color information of a sample. NIR spectroscopic analysis revealed that each of the two second-derivative absorbances at 874 and 902 nm had a high correlation with the sugar content of melons. A high-resolution cooled charged couple device camera with band-pass filters, which included the above two wavelengths, was used to capture the spectral absorption image of a half-cut melon. A color distribution map of the sugar content on the surface of the melon was constructed by applying the NIR spectroscopy theory to each pixel of the acquired images. 相似文献

(D-1) and 2-glycidyl-4-hydroxy-5,8-dimethoxy naphthalene (D-2)] were evaluated. Compound I showed higher 1,1-diphenyl-2-picrylhydrazyl
(DPPH) radical scavenging activity and reducing power than D-1 and D-2. However, D-1 and D-2 exhibited slightly stronger 2,2-azino-bis
(3-ethylbenzothiazoline-6-sulfonic acid)+ (ABTS+) radical scavenging activity than Compound I. Compound I also exhibited stronger cytotoxic activity than D-1 and D-2 against
the growth of HT-29 colon cancer cells. The results supported the hypothesis that methylation of naphthalene derivatives slightly

Assisted reproductive techniques (ARTs), such as artificial insemination, in vitro fertilization, and cryopreservation of gametes/zygotes, have been developed to improve breeding and reproduction of livestock, and for the treatment of human infertility. Their widespread use has contributed to improvements in human health and welfare. However, in dogs, only artificial insemination using frozen semen is readily available as an ART to improve breeding and control genetic diversity. A recent priority in sperm cryopreservation is the development of alternatives to egg yolk, which is widely used as a component of the sperm extender. Egg yolk can vary in composition among batches and is prone to contamination by animal pathogens. The latter can be a problem for international exchange of cryopreserved semen. Low-density lipoprotein and skim milk are promising candidates for use as extenders, to ensure fertility after artificial insemination. Although not tested for its effects on fertility following artificial insemination, polyvinyl alcohol may also be a useful alternative to egg yolk as an extender. The development of cryopreservation techniques for canine embryos lags behind that for other mammals, including humans. However, given the success of non-surgical embryo transfer in 2011, studies have sought to refine this approach for practical use. Research on sperm cryopreservation has yielded satisfactory results. However, investigation of other approaches, such as cryopreservation of oocytes and gonadal tissues, remains insufficient. Techniques for the efficient induction of estrus may aid in the development of successful canine ARTs. 相似文献

In the present study, we assessed small fishes as potential feed fishes with the lowest mercury levels. The mercury levels
of four small pelagic fishes and a benthic fish from the Pacific Ocean and the Sea of Japan, i.e., spotted chub mackerel Scomber australasicus, chub mackerel Scomber japonicus, horse mackerel Trachurus trachurus, round scad Decapterus punctatus, and bastard halibut Paralichthys olivaceus, were determined. Total mercury levels in fish muscle were measured using a reduction vaporizing atomic absorption method.
There were no significant correlations between body weight and mercury levels for these species. However, significant differences
were observed in the mercury levels of these species between the two catch locations, with fish caught in the Pacific Ocean
having higher mercury levels. This difference may be due to the Pacific Ring of Fire, a highly seismically and volcanically
active zone, which is located in the Pacific Ocean. Preference for these fishes for use in aquaculture should refer to the

Hereditary causes of variation in the vertebral number of chum salmon were analyzed to estimate heritability for populations in the Chitose and Shikiu Rivers, Hokkaido Island, northern Japan. A total of 70 families were produced by diallel cross mating and then heritability was estimated using two statistical tests: a two-way ANOVA and a parent-offspring regression. Heritability estimates ranged from 0.26 to 1.91 and showed a strong correlation between the values from the two different tests. Estimates from female components were substantially higher than those from male components, which can be attributed to the maternal effect. Heritability in the Shikiu River population was shown to be higher than that in the Chitose River population. These results indicate that the heritability of vertebral number in chum salmon is generally high, and also that vertebral number is controlled by hereditary causes. 相似文献
