The present study examined sex differences and the involvement of gonadal hormones in stress responses caused by road transportation in Shiba goats. In experiment 1, we investigated the stress responses of males and females to transportation. Plasma levels of cortisol (Cor) significantly increased during 1 h of transportation, and those in females were significantly higher than those in males. Plasma glucose (Glu) and free fatty acid (FFA) levels also increased similarly in both females and males by transportation, and there were no sex differences. Food intake following transportation decreased only in males compared with that in the basal session, in which the animals were not transported. Experiment 2 examined the involvement of gonadal hormones in stress responses to transportation using castrated males. Goats were given cholesterol (Cho), 5α‐dihydrotestosterone (DHT) or 17β‐estradiol (Es). The plasma Cor levels increased during transportation regardless of hormone treatment, and those in DHT treated goats were significantly lower than those in Cho or Es treated animals. Plasma Glu and FFA levels also increased during transportation, regardless of hormone treatment, and there were no differences between treatments. Food intake following transportation was significantly lower than that in the basal session only in goats given DHT. In conclusion, gender affects Cor secretion that is increased by transportation and the decrease of food intake following transportation in Shiba goats, and the major cause of these differences is androgen.  相似文献

A female Japanese Black cow showed hemorrhage under the mucous membrane of the eye conjunctiva and the vagina, and thrombocytopenia in peripheral blood. The clinical manifestations repeated spontaneously during long-term observation. The clinical findings were stable except for severe periodic bleeding. Prothrombin time, activated partial thromboplastin time and fibrinogen levels were normal. The anti-platelet antibody in the serum was analyzed by indirect platelet coagulation tests. Because the platelets in the micro-plate wells of the subject series showed coagulation up to 1:128, it was judged that anti-platelet antibody was present in the serum. A number of megakaryocytes were recognized in smear samples obtained from bone marrow of the scapula immediately after euthanasia. Therefore, this case was diagnosed as idiopathic thrombocytopenia.  相似文献

ORP150 is a hypoxic stress-induced protein located in the endoplasmic reticulum. Transgenic mice overexpressing ORP150 (ORP-Tg) exhibit vacuolar degeneration in the heart. To determine whether vacuolization is present in skeletal muscle, we pathologically examined ORP-Tg mice. After 60 days of age, severe vacuolization was found in the soleus muscles of the hind legs of the ORP-Tg mice. Immunohistochemical staining of ORP150 revealed co-localization of ORP150 and vacuolization in the affected cells. Electron microscopy revealed a marked increase in the number of rough-surfaced endoplasmic reticula (rER) and distention of the cisterna. These findings suggest that overexpression of ORP150 causes accumulation of ORP150 in the rER, resulting in vacuolar degeneration in the skeletal muscle of ORP-Tg mice.  相似文献

A 9‐month‐old intact crossbred female cat was presented with jaundice, intermittent anorexia and lethargy, increased hepatic enzyme activities, and hyperammonemia. Abdominal ultrasound and computed tomographic examinations determined that the liver had a rounded and irregular margin, and histopathological examination identified excessive accumulation of copper hepatocytes in the liver. Concentrations of both blood and urine copper were higher than in healthy cats. The patient responded well to treatment with penicillamine. Clinicopathological abnormalities and clinical signs improved within 2 months, and the patient was alive for >9 months after starting treatment. Genetic examination determined that the patient and its littermate had a single‐nucleotide variation (SNV, p. T1297R) that impaired the function of the ATP7B gene product; the gene that is mutated in patients with Wilson's disease (WD). Hepatic copper accumulation was believed to be associated with the SNV of the ATP7B gene, and the patient had a genetic disorder of copper metabolism equivalent to WD in humans.  相似文献

A 7‐year‐old neutered female domestic shorthaired cat born in Poland and then moved to Japan presented to the local clinic with recent onset of convulsive cluster seizures and status epilepticus. Magnetic resonance imaging revealed bilateral swelling of the hippocampus with T2 hyperintensity and contrast enhancing image, suggesting hippocampal necrosis. The cat completely recovered after treatment with antiepileptic drugs (AED) and administration of prednisolone (1 mg/kg PO q24h for 4 days and tapered). However, cluster seizures reoccurred and developed into status epilepticus despite increasing doses of AED. Although the convulsions were resolved by other AEDs, stupor and renal failure developed, and the cat was euthanized. Pathological findings were consistent with hippocampal necrosis. Immunological analysis for leucine‐rich glioma inactivated 1 (LGI1) autoantibodies was negative, but antibodies against DCC (deleted in colorectal carcinoma) known as netrin‐1 receptor were found. This report describes a case of feline autoimmune limbic encephalitis and hippocampal necrosis that were presumably associated with DCC autoantibodies.  相似文献

We investigated the effect of rice grain conservation methods on feed intake, milk production, blood metabolites, and rumen fermentation in dairy cows. Raw rice grain was dried before crushing (DRY), ensiled after crushing (ENS‐A), or ensiled before crushing (ENS‐B). Twelve multiparous Holstein cows were used in a replicated 3 × 3 Latin square design with three dietary treatments comprising ad libitum access to one of three total mixed rations (TMRs; containing DRY, ENS‐A, or ENS‐B at 17% of dietary dry matter) plus a standard allowance of 2.0 kg/day of dairy concentrates. The dietary treatments did not affect the feed intake, milk yield, or milk composition. The selected blood constituents were not influenced by the rice conservation method. The ruminal lactic acid and total volatile fatty acid (VFA) concentrations and the VFA proportion in the cows were not influenced by the rice conservation method. These results demonstrate that the rice grain conservation method has little impact on lactation performance when cows are fed a TMR containing 17% treated rice grain (dry matter basis).  相似文献

This study examined the mitogenic response of bovine peripheral T lymphocytes to leptin, a pleiotropic hormone regulating food intake and energy expenditure. Leptin alone slightly suppressed proliferation of T lymphocytes in the presence of concanavalin A (ConA). Leptin also inhibited proliferation of T lymphocytes induced by anti-CD3 antibody. ConA treatment activated some protein kinases, including p44/p42(MAPK) and Akt/PKB, while anti-CD3 antibody treatment increased mRNA expression of suppressor of cytokine signalling (SOCS) 3, interferon (IFN)gamma, interleukin (IL) 2 and IL4 in T lymphocytes. Leptin alone increased only SOCS3 mRNA expression. Simultaneous treatment with mitogens and leptin enhanced IFNgamma mRNA expression but decreased IL2 mRNA expression, without any synergistic effect on phosphorylation of protein kinases or mRNA expression of SOCS3 and IL4. These results suggest that leptin modulates bovine T lymphocyte functions.  相似文献

To improve the welfare of caged laying hens, a pecking device made of stones was introduced on the cage floor. Twenty‐four White Leghorn hens aged 15 months were divided into four groups: single‐housed hens with device, single‐housed control hens, pair‐housed hens with device and pair‐housed control hens. Hens housed with the device pecked at various pecking objects less often than control hens. Agonistic behavior was also lower in hens with the device than in hens without the device, implied a possibility of improvement in quality of pecking stimuli with the device. Not only time spent pecking, but also quality of pecking might be important to fill their need for stimulation. Both single‐ and pair‐housed hens more often pecked at the device in the evening. Response to various pecking objects also showed that pecking behaviors were most frequently expressed in the evening. Increased foraging at dusk is a well‐known habit; therefore, the increase in pecking behavior in the evening might reflect the hens' general circadian rhythm. These results indicate that the device made of stones could promote some instinctive behavior. Enhancement of behavioral repertories and reduced agonistic behavior with the pecking device might improve the welfare of caged laying hens.  相似文献
