Background:

Microglial cells act as the sentinel of the central nervous system .They are involved in neuroprotection but are highly implicated in neurodegeneration of the aging brain. When over-activated, microglia release pro-inflammatory factors, such as nitric oxide (NO) and cytokines, which are critical in eliciting neuroinflammatory responses associated with neurodegenerative diseases. This study examined whether bromelain, the pineapple-derived extract, may exert an anti-inflammatory effect in primary microglia and may be neuroprotective by regulating microglial activation.

Methods:

Following the isolation of neonatal rat primary microglial cells, the activation profile of microglia was investigated by studying the effects of bromelain (5, 10, 20, and 30 µg/ml) on the levels of NO, inducible nitric oxide synthase (iNOS), and nuclear factor kappa B (NF-κB) in microglia treated with lipopolysaccharide (LPS) (1 µg/ml). Data were analyzed using Student''s t-test. P values less than 0.05 were considered to be statistically significant, compared with the LPS-treated group without bromelain.

Results:

Results showed that pretreatment of rat primary microglia with bromelain, decreased the production of NO induced by LPS (1 µg/ml) treatment in a dose-dependent manner. Bromelain (30 µg/ml) also significantly reduced the expression of iNOS at mRNA level and NF-κB at protein level. Moreover, the study of mitochondrial activity in microglia indicated that bromelain had no cytotoxicity at any of the applied doses, suggesting that the anti-inflammatory effects of bromelain are not due to cell death.

Conclusion:

Bromelain can be of potential use as an agent for alleviation of symptoms in neurodegenerative diseases.  相似文献

The influence of high-pressure treatment (HPT) (200-800 MPa, 5 or 20 min, at 20 degrees C) on the rheological properties of solutions of amidated low-methoxyl pectin (LMP) and its mixtures with micellar casein (MC) has been investigated in the presence and absence of sucrose. The storage modulus G' of LMP gels containing 0-55 wt % sucrose and 0.1-1 wt % LMP was found to increase significantly following HPT at >or=400 MPa. Various concentrations of LMP in the presence of different amounts of MC (0.5-12 wt %) showed contrasting types of rheological behavior. In the presence of a low concentration of LMP (<0.3 wt %), HPT was found to induce a sol-gel transformation at relatively high LMP/MC molar ratios (<4 wt % MC), to reduce values of G' and the loss modulus G' ' at intermediate LMP/MC ratios (4-10 wt % MC), and to increase the values of G' and G' ' at low LMP/MC ratios (>10 wt % MC). In contrast, in the presence of a higher amount of LMP (>0.5 wt %), it was observed that HPT enhances the values of both the storage and the loss moduli over the whole range of MC concentrations.  相似文献

Two shortcomings of the non-numeric United States Soil Laboratory (USSL) water quality designation process are, first, the uncertainty involved with assigning samples to the bordering discrete classes is not considered and, secondly, it is difficult to map. One solution to these may lie in the use of a Mamdani fuzzy inference system (MFIS). The main aims of this study were (1) to compare the MFIS and USSL approach to classifying groundwater quality for irrigation and (2) to explore the spatial variability of groundwater quality for irrigation in the Marvdasht aquifer using the MFIS output. For this purpose, 49 agricultural wells were sampled and their sodium adsorption ratio and electrical conductivity were determined. In 81% of cases, the MFIS led to the same class as USSL and in 92.8% of cases agreed with USSL classification. In only 2 out of 49 samples did the MFIS and USSL strongly disagree. The comparison showed that the MFIS method is more acceptable, reliable, and logical in the classification of water quality for irrigation purposes than other methods. The output map of the MFIS scores indicates that the upper part of Marvdasht aquifer has better quality water for irrigation.  相似文献

A novel kinetic-spectrophotometry method is developed for the determination of trace amounts of silver nanoparticles (AgNPs) in groundwater samples based on their catalytic effect on the oxidation of indigo carmine by hexacyanoferrate (III) in acetate-acetic acid medium. The absorbance was measured at 612?nm with the fixed-time method. The optimization of the operating conditions regarding concentrations of the reagents and interferences was investigated. The calibration curve is linear over the concentration range 4.0?C300???g?mL?1 of AgNPs with good precision and accuracy. The proposed method is a highly sensitive, selective, and relatively rapid assay of AgNPs at trace level. The method was applied to the determination of AgNPs in groundwater and a synthetic sample.  相似文献

This paper describes the embryonic development of the common cutworm Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) which is divided into eight embryonic stages, through variations in the egg's contents. Experiments were conducted on the eggs ofS. litura under laboratory conditions. The highest concentration of the protein (min.-max.: 32-! 40 lag/egg) and the lowest of the carbohydrate (min.-max.: 3.0-44.0 lag/egg) were obtained; similarly, the DNA content was smaller (min.-max.: 1.0-38.2 lag/egg) than the RNA (min.-max.: 9.2-360.0 lag/egg) throughout the embryogenesis. Three peaks of ecdysteroid titre were observed which were responsible for appearance of particular events during embryogenesis. The last highest peak of 400_+33.4 lag/egg was appeared at 84 h after oviposition when larva was ready to hatch. Therefore, it is concluded that eight embryonic stages in S. litura were characterized by fluctuation in the egg's contents, which were responsible for gradual-progressive embryonic development. The egg-larval development did not show a normal distribution of embryonic developmental-time.  相似文献

It has been shown that pomegranate fruit powder (PFP) has the potential to decrease urea-nitrogen (N) mineralization and improve grain yield of maize, but the mechanisms of this effect are not fully understood. Therefore, the objective of this study was to investigate these mechanisms. For this purpose, an incubation and a pot experiment were conducted, in which four levels of PFP coating equivalent to 0%, 30%, 70%, and 100% (w/w) of urea were studied. PFP had very little effect on nitrification (1.4–2.9%). Mineral N and nitrite (NO2?)–N decreased (23% and 38%, respectively) with increasing PFP application, probably due to N-immobilization or urea hydrolysis inhibition. Biomass and grain yield of maize increased with increasing phosphorus (P)/N ratio of leaf and stem, and decreasing P/N ratio of grain, indicating soil-P-mobilization and soil-N-immobilization during vegetative growth and the inverse, during grain filling by PFP. According to these results, PFP can be introduced as an urea-N immobilizer and soil-P mobilizer.  相似文献

Using pedotransfer functions (PTF) is a useful way for field capacity (FC) and permanent wilting point (PWP) prediction. The aim of this study was to model PTF to estimate FC and PWP using regression tree (RT) and stepwise multiple linear regressions (SMLR). For this purpose, 165 and 45 soil samples from UNSODA and HYPRES datasets were used for development and validation of new PTFs, respectively. %Clay, geometric mean diameter (dg), and bulk density (BD) were selected as predictor variables due to the highest correlation and lowest multicollinearity. The results showed that clay percentage with W* = 0.89 and dg with W* = ?0.57 were the most effective variables to predict PWP and FC, respectively. The RT method had a better performance (R2 = 0.80, ME = ?0.002 cm3cm?3, RMSE = 0.05 cm3cm?3 for FC and R2 = 0.85, ME = 0.003 cm3cm?3, RMSE = 0.03 cm3 cm?3 for PWP) than SMLR in estimation of FC and PWP.  相似文献

In this study, a new Co3O4 nanoparticles (NPs) coated with sodium dodecyl sulphate (SDS) is developed for preconcentration of trace amounts of cadmium ions (Cd 2+) as a prior step to its determination by flame atomic absorption spectrometry (FAAS). The effects of various parameters, including pH of sample solution, amount of sorbent, flow rates of solution and eluent, sample volume, type, and least amount of the eluent for elution of the Cd 2+ from Co3O4 NPs were studied and optimized. Experimental conditions for effective separation of trace levels of the Cd 2+were optimized with respect to different experimental parameters in Column method. Under the best experimental conditions, the calibration curve was linear in the range of 1.0–500.0 ng.mL?1 of cadmium (Cd) with R2 = 0.999. The detection limit was 0.4ng.mL?1 in the original solution (3Sb/m) and the relative standard deviation for eight replicate determination of 0.1µg.mL?1 Cd was ±2.1%. The method was validated by the analysis of a certified reference material with the results being in agreement with those quoted by manufactures. The developed method was successfully applied to the extraction and determination of Cd in water and food samples with satisfactory results.  相似文献

Wireworms, the larvae of click beetle species Agriotes lineatus, Aobscurus and Asputator are serious pests for several field crops. They are considered severe pests of potato tubers and the damages caused by them can be resulted a significant loss especially in organic crop production. Since synthetic insecticides are prohibited in Organic Farming; biological control methods have to be used in organic crop production. In the current study naturalis Beauveria bassianaa strain ATCC 74040 was used both under laboratory and field conditions using wheat and potato crops as a food source respectively. Fungus showed a significant mortality in high number of wireworms boxes (50%) compared to low number of wireworms (17%) and untreated boxes (13%) respectively. However, seed germination showed no effect in all three categories. Field data have shown mixed results when B. bassiana has been applied in a variety of application methods. Maximum infestation (3.99%) was recorded in untreated plots; while significantly lower damage (1.11%) was recorded in whole surface treated plots. During first year all the treatments were significantly different from each other; however, no significant differences were noted between furrows and whole surface applications but were different from control treatment during second year of experimentation. The results showed that the biological control of wireworms by using entomopathogenic fungi, such as Bbassiana is a promising target specific option without disturbing the other communities in the soil.  相似文献

Hexaploid bread wheat (Triticum aestivum L. ssp. aestivum) is assumed to have originated by natural hybridization between cultivated tetraploid Triticum turgidum L. and wild diploid Aegilops tauschii Coss. This scenario is broadly accepted, but very little is known about the ecological aspects of bread wheat evolution. In this study, we examined whether T. turgidum cultivation still is associated with weedy Ae. tauschii in today’s Middle Eastern agroecosystems. We surveyed current distributions of T. turgidum and Ae. tauschii in northern Iran and searched for sites where these two species coexist. Ae. tauschii occurred widely in the study area, whereas cultivated T. turgidum had a narrow distribution range. Traditional durum wheat (T. turgidum ssp. durum (Desf.) Husn.) cultivation associated with weedy Ae. tauschii was observed in the Alamut and Deylaman-Barrehsar districts of the central Alborz Mountain region. The results of our field survey showed that the T. turgidumAe. tauschii association hypothesized in the theory of bread wheat evolution still exists in the area where bread wheat probably evolved.  相似文献
