ABSTRACT: Many of the known fish herpesviruses have important aquaculture species as their natural host, and may cause serious disease and mortality. Anguillid herpesvirus 1 (AngHV-1) causes a hemorrhagic disease in European eel, Anguilla anguilla. Despite their importance, fundamental molecular knowledge on fish herpesviruses is still limited. In this study we describe the identification and localization of the structural proteins of AngHV-1. Purified virions were fractionated into a capsid-tegument and an envelope fraction, and premature capsids were isolated from infected cells. Proteins were extracted by different methods and identified by mass spectrometry. A total of 40 structural proteins were identified, of which 7 could be assigned to the capsid, 11 to the envelope, and 22 to the tegument. The identification and localization of these proteins allowed functional predictions. Our findings include the identification of the putative capsid triplex protein 1, the predominant tegument protein, and the major antigenic envelope proteins. Eighteen of the 40 AngHV-1 structural proteins had sequence homologues in related Cyprinid herpesvirus 3 (CyHV-3). Conservation of fish herpesvirus structural genes seemed to be high for the capsid proteins, limited for the tegument proteins, and low for the envelope proteins. The identification and localization of the structural proteins of AngHV-1 in this study adds to the fundamental knowledge of members of the Alloherpesviridae family, especially of the Cyprinivirus genus.  相似文献

This study measured the effect on glutathione peroxidase (GSH-Px) and selenium (Se) in whole blood and plasma associated with subcutaneous Se injections in beef heifers fed organic or inorganic Se. Heifers (n = 120) were randomly divided into 2 groups, 1 of which received subcutaneous Se injections. Both groups were given the same total mixed ration with 3 mg of organic or inorganic Se daily. Until week 2, heifers that had received Se injections showed higher concentrations of plasma Se and GSH-Px and whole blood Se (P < 0.001) than those having had no injections. Concentrations of plasma Se and GSH-Px were higher in the group receiving organic Se than the group receiving inorganic Se. Whole blood GSH-Px concentrations increased significantly (P < 0.001) throughout a 12-week period but were not affected by Se source. Combination of Se injections and supplementation could help maintain normal Se and GSH-Px blood status in beef heifers during the first few weeks in the feedlot.  相似文献

Recombinant adenoviral vectors have emerged as an attractive system for veterinary vaccines development. However, for poultry vaccination a very important criterion for an ideal vaccine is its low cost. The objective of this study was to test the ability of chicken CD154 to enhance the immunogenicity of an adenoviral vector-based vaccine against avian influenza virus in order to reduce the amount of antigen required to induce an effective immune response in avian. Chickens were vaccinated with three different doses of adenoviral vectors encoding either HA (AdHA), or HA fused to extracellular domain chicken's CD154 (AdHACD). Hemagglutination inhibition (HI) assay and relative quantification of IFN-γ showed that the adenoviral vector encoding for the chimeric antigen is able to elicit an improved humoral and cellular immune response, which demonstrated that CD154 can be used as a molecular adjuvant allowing to reduce in about 50-fold the amount of adenoviral vector vaccine required to induce an effective immune response.  相似文献

Successful adoption of a One Health approach could have far-reaching impacts on poverty alleviation, health and food security, particularly in developing countries through integrated control of neglected zoonoses. However, the practical implementation of this approach presents many challenges. Anna Okello and colleagues argue that, for effective implementation, lessons learned and 'best practice' must be led by national and regional stakeholders drawn from a variety of disciplines. Coxiella burnetii is the causal agent of Q fever, a worldwide spread zoonosis. Prevention of C. burnetii shedding in cattle is critical to control the spread of the pathogen between animals, and from animals to humans. Vaccination with a phase 1 vaccine has been shown to be effective in preventing shedding when implemented in still susceptible animals, even in infected cattle herds. The identification of these animals (dairy cows and nulliparous females) as targets for vaccination consequently is crucial. Hygiene measures conventionally also are implemented, but their relative impact on C. burnetii diffusion remains unknown. The objectives of this study therefore were to (i) describe the distribution of the within-herd apparent seroprevalence among cows and nulliparous females and (ii) to explore the association between management practices and herd characteristics on the one hand, and these seroprevalences on the other. In a sample of 100 naturally and clinically infected dairy herds, blood samples were taken systematically from all nulliparous females (older than 12 months) and cows, and serologically tested. Information on herd characteristics and management practices were collected through a questionnaire filled in by each farmer. The variation in within-herd seroprevalence among cows and the risk for a herd of having at least one seropositive nulliparous female were investigated using multivariate (linear and logistic respectively) regression models. Median within-herd seroprevalence was 0.32 (Q1=0.22; Q3=0.43). We observed a low to null (median=0.01; Q1=0; Q3=0.10) within-herd seroprevalence in nulliparous females contrary to a high value (median=0.42) and variability (Q1=0.28; Q3=0.56) in cows. Only a few herd characteristics and management practices were found to be related to seroprevalence. Within-herd seroprevalence in cows was found to be significantly (P<0.10) higher in herds (i) with a number of cows<46, (ii) with seasonal calving, and (iii) with grazing or contact through the fence with other ruminant herds. The risk of having at least one seropositive nulliparous female was increased in herds (i) with seasonal calving and (ii) where the foetus and/or the placenta of aborted cows were not systematically removed. Our findings support, in addition to the implementation of high level of hygiene measures, the relevance of vaccination (at least in nulliparous females) as a method to control the spread of C. burnetii within an infected herd, as vaccination is effective in susceptible animals and given that nulliparous females are mostly not infected even in infected herds.  相似文献

The early curative uses of antimicrobial drugs such as fluoroquinolones before the onset of symptoms in veterinary medicine may be regarded as irrational antibiotic consumption. However, it should be stressed that in early curative antimicrobial treatment as in metaphylaxis, the bacterial burden at the infection site is often very low, and so the rapid eradication of the bacterial population could result. We investigated the impact of early versus later curative administrations of 1 or 40 mg/kg of marbofloxacin on the survival of mice, the eradication of the targeted pathogen and the selection of resistant bacteria in a mouse lung infection with Pasteurella multocida. In this model, for a given marbofloxacin dose, the clinical and bacteriological outcomes were better, and the selection of resistance less frequent, for the early rather than for the late treatment. Moreover, the early administration of 1mg/kg led to better clinical and similar bacteriological (eradication and selection of resistance) outcomes than the late administration of 40 mg/kg marbofloxacin. Our results suggest that the optimal doses for the animals' cure could be lower when administered early during the time course of the infection than when administered after the disease outbreak. As the main argument against early treatments such as metaphylaxis is the possible enhancement of resistance at the gut level, further studies should assess if lower doses of antibiotic administered to all the animals of a herd could have less impact on the commensal digestive flora than higher doses only administered to animals showing clinical symptoms.  相似文献

Maize (Zea mays L. ssp. mays) was domesticated from one wild species ancestor, the Balsas teosinte (Zea mays ssp. parviglumis) about 9000 years ago. Higher levels of gene diversity are found in teosinte taxa compared to maize, following domestication and selection bottlenecks. Diversity in maize can be increased via gene flow from teosinte, which has certainly occurred from various taxa, but the rate of flow from different teosinte taxa and the final impact on maize evolution has been difficult to measure. One hundred populations from six Zea taxa, both domesticated (maize) and wild (teosinte), including domesticated landraces from Asia, Africa, and the Americas, were genotyped with 17 SSR markers using 15 individuals per population. Overall levels of diversity were high, and populations could be distinguished based on markers. Relationships between populations followed most published reports, or can now help resolve previously conflicting reports. Gene flow into maize from different teosinte groups, and gene flow between different teosintes, was estimated. Evidence for contributions from the Balsas teosintes and from Chalco teosintes (Z. mays ssp. mexicana) to the maize gene pool was found, as well as from Chalco into ssp. mexicana race ??Durango?? and Z. mays ssp. huehuetenengensis. These contributions are almost certainly the result of post-domestication (and ongoing) exchanges. This information must give more impetus to in situ conservation of teosinte species, and use of these teosintes to continue to direct the evolution of maize, especially in response to new diseases, insect pests, and other biotic and abiotic stresses.  相似文献

Electrodialysis with ultrafiltration membrane (EDUF) is a technology based on the separation of molecules according to their charge and molecular mass. Some works have already successfully demonstrated the recovery of bioactive peptide fractions. However, the impact of ultrafiltration membrane (UFM) material, used in the EDUF system, on the peptide migration has never been studied. Consequently, the objectives of this work were (1) to evaluate the effect of two different UFM materials on the selective separation of peptides from a snow crab byproduct hydrolysate by electrodialysis with ultrafiltration membranes and (2) to determine the effect of UFM material on their potential fouling by peptides. It appeared that, after 6 h of EDUF separation using polyether sulfone (PES) and cellulose acetate (CA) UFM, peptides with low molecular weights ranging from 300 to 700 Da represented the most abundant population in the KCl1 (compartment located near the anode for the recovery of anionic/acid peptide fractions) and KCl2 (compartment located near the cathode for the recovery of cationic/basic peptide fractions) permeates. Peptides with molecular weights ranging from 700 to 900 Da did not migrate during the EDUF treatment. Moreover, only CA UFM allowed the recovery of high molecular weight molecules (900-20000 Da) in both KCl compartments. Peptides desorbed from PES and CA UFM after 6 h of EDUF separation had low molecular weights and belonged mainly to the 600-700 Da molecular weight range. These peptides represented a low proportion of the peptides initially present in the snow crab byproduct hydrolysate with individual molecular weight range proportions from 1.52 ± 0.31 to 10.2 ± 2.32%.  相似文献

Gastrointestinal microbiota impact host's biological activities, including digestion of indigestible feed components, energy harvest, and immunity. In this study, fecal microbiota of high body weight (HW) and low body weight (LW) growing pigs at 103 days of age were compared. Principal coordinates analysis separated the HW and LW groups into two clusters, indicating their potential differences between microbial community composition. Although the abundances of two major phyla, Firmicutes and Bacteroidetes, did not significantly differ between the HW and LW groups, some genera showed significant differences. Among them, Peptococcus and Eubacterium exhibited strong positive correlations with body weight (BW) and average daily gain (ADG) (Rho > 0.40), whereas Treponema, Desulfovibrio, Parabacteroides, and Ruminococcaceae_unclassified exhibited strong negative correlations with BW and ADG (Rho < ?0.40). Based on these results, the structure of intestinal microbiota may affect growth traits in pigs through host–microbe interactions. Further in‐depth studies will provide insights into how best to reshape host–microbe interactions in pigs and other animals as well.  相似文献

In livestock, improving maternal reactivity towards the litter is an important issue in breeding strategies to promote production and animal welfare. As of yet, no studies have investigated the within-breed genetic variation of maternal reactivity in sheep. The objective of this study was to estimate the genetic parameters of maternal reactivity traits. A total of 1,095 primiparous and 1,441 multiparous Romane ewes were phenotyped 24 hr postlambing using a behavioural test (arena test, AT) over a 10-year experimental period. The test consisted of three successive phases evaluating the ewe's attraction to her litter, reactivity to separation from her litter, and reactivity to a conflict between attraction to her litter and avoidance of a motionless human. The ewes were reared exclusively on rangelands (South of France) and lambed outdoors in the spring. High-pitched bleating and low-pitched bleating in the AT were mostly highly heritable (0.39‒0.46). Heritabilities were moderate for proximity to the litter in the presence of a human (0.27) and low for locomotion and vigilance in the AT (0.09‒0.15). The measurements of a given behaviour in the three phases of the AT were highly genetically correlated. Few genetic correlations were found between the different behavioural traits in the AT, the highest correlations being between high-pitched bleating and low-pitched bleating (−0.43 to −0.77). In conclusion, our findings demonstrate moderate-to-high heritability for maternal reactivity traits. These traits could be included in genetic selection schemes to enhance maternal attachment provided there is no unfavourable link with other production traits.  相似文献
