Intramolecular gluconic acid esterification reactions led to the formation of two lactones, gamma- and delta-gluconolactone (glucono-1,4-lactone and glucono-1,5-lactone). The presence of the first has not yet been reported in must or wine. These lactones are in equilibrium with gluconic acid, gamma- and delta-gluconolactone representing, respectively, 5.8 and 4.1% of the acid level. Correlations between must SO(2) binding power, gluconic acid, and consequently its lactones are shown. The SO(2) affinity of a mixture containing this acid and gamma- and delta-gluconolactone was determined, and gluconic acid appeared to be indirectly responsible for approximately 8% of the bindable SO(2) in musts from botrytized grapes.  相似文献

The contribution of dimethyl sulfide (DMS) to the aroma of Syrah and Grenache Noir wines from the Rhone Valley of France was investigated by sensory analysis, and its levels in these wines were measured. The potential DMS in the corresponding grapes and wines, susceptible to release during wine aging, was evaluated. Free DMS and potential DMS assessed by a heat-alkaline treatment were measured in grape juices and wines by SPME-GC-MS using methods previously reported and slightly modified. A relationship between potential DMS from grapes and the total DMS levels in wine was demonstrated. Furthermore, a linear regression between the ratio of free DMS levels to these total DMS levels in wine and time of storage was found. Free and potential DMS levels in grapes and wines depended on grape variety, vintage, and vine location. DMS imparted a noticeable and complex contribution to the aroma of the wines investigated, depending on the mode of sensory perception used, either before or after glass swirling. It significantly enhanced the fruity notes of the wines, and additional truffle and black olive notes.  相似文献

A thin-layer chromatographic (TLC) screening method was developed for the detection of ochratoxin A (OTA) in green coffee at a control level of 10 microg/kg. The method is based on extraction of OTA with a mixture of phosphoric acid and dichloromethane, purification by liquid-liquid partition into sodium hydrogen carbonate, separation by normal-phase TLC, and detection by visual estimation of fluorescence intensity under a UV lamp at 366 nm. The method was validated by performing replicate analyses of uncontaminated green coffee material spiked at 3 different levels of OTA (5, 10, and 20 microg/kg), and also by comparing results obtained on a series of test trial green coffees naturally contaminated with OTA (range 0.2 to 136.7 microg/kg) with those measured by a quantitative immunoaffinity/HPLC method. The agreement between the two methods was excellent, and neither false positive nor false negative results were recorded. This screening method is rapid, simple, robust, and very cheap, which makes it particularly well adapted for implementation in coffee-producing countries.  相似文献

Previously, we have presented a method for quantifying beta-carotene bioavailability based on analysis in serum, following administration of (13)C-labeled beta-carotene. Because stool samples can be collected noninvasively, we have now extended the method to measure the bioavailability based on measurements in feces. An extraction method was developed to enable measurement of concentrations and degree of isotopic enrichment of retinol, retinyl palmitate and carotenoids in feces. Relative bioavailability of beta-carotene from pumpkin (n = 6) was found to be 1.8 times (interval, 0.6, 5.5) greater than that from spinach (n = 8), based on data from feces compared with 1.7 times (interval, 0.9, 3.1) based on data from serum.  相似文献

Understanding and controlling structural and physical changes in meat during cooking is of prime importance. Nuclear magnetic resonance imaging (MRI) is a noninvasive, nondestructive tool that can be used to characterize certain properties and structures both locally and dynamically. Here we show the possibilities offered by MRI for the in situ dynamic imaging of the connective network during the cooking of meat to monitor deformations between 20 and 75 °C. A novel device was used to heat the sample in an MR imager. An MRI sequence was developed to contrast the connective tissue and the muscle fibers during heating. The temperature distribution in the sample was numerically simulated to link structural modifications and water transfer to temperature values. The contraction of myofibrillar and collagen networks was observed at 42 °C, and water began to migrate toward the interfascicular space at 40 °C. These observations are consistent with literature results obtained using destructive and/or nonlocalized methods. This new approach allows the simultaneous monitoring of local deformation and water transfer, changes in muscle structure and thermal history.  相似文献

Model systems composed of tristearin in solid state and tricaprin in liquid state with different solid-fat content (SFC) and storage time have been investigated by relaxation NMR and NMR diffusometry. The T(2) relaxation of the tricaprin in the melt exhibited a bimodal distribution as previously observed. The SFC had a major effect on the T(2) relaxation. This effect was explained according to the fast diffusive exchange model in porous media. According to this model the changes in T(2) relaxation as a function of the SFC and storage time were explained by the decrease of the surface-to-volume ratio of the crystal induced by Ostwald ripening. The diffusion coefficient D of the tricaprin in the melt decreased for higher SFC. Since no significant variation of D was observed for different diffusion time, D reflected the long-range connectivity and the tortuosity was calculated. During storage the diffusion coefficient remained constant.  相似文献

The various ecosystem functions of soil organic matter (SOM) depend on both its quantity and stability. Numerous fractionation techniques have been developed to characterize SOM stability, and thermal analysis techniques have shown promising results to describe the complete continuum of SOM in whole soil samples. However, the potential link between SOM thermal stability and biological or chemical stability has not yet been adequately explored. The objective of this study was to compare conventional chemical and biological methods used to characterize SOM stability with results obtained by thermal analysis techniques. Surface soil samples were collected from four North American grassland sites along a continental mean annual temperature gradient, each with a native and cultivated land use. Soil organic C concentrations ranged from 6.8 to 33 g C kg−1 soil. Soils were incubated for 588 days at 35 °C, and C mineralization rates were determined periodically throughout the incubation by measuring CO2 concentration using an infrared gas analyzer (IRGA) to calculate biological indices of SOM stability. Hot-water extractable organic C (HWEOC) contents were determined before and after incubation as chemical indices. Finally, samples from before and after incubation were analyzed by simultaneous thermal analysis (i.e., thermogravimetry (TG) and differential scanning calorimetry (DSC)) to determine thermal indices of SOM stability. Long-term incubation resulted in the mineralization of up to 33% of initial soil C. The number of days required to respire 5% of initial soil organic carbon (SOC), ranged from 27 to 115 days, and is proposed as a standardized biological index of SOM stability. The number of days was greater for cultivated soils compared to soils under native vegetation, and generally decreased with increasing site mean annual temperature. HWEOC (as % of initial SOC) did not show consistent responses to land use, but was significantly lower after long-term incubation. Energy density (J mg−1 OM) was greater for soils under native vegetation compared to cultivated soils, and long-term incubation also decreased energy density. The temperatures at which half of the mass loss or energy release occurred typically showed larger responses to land use change than to incubation. Strong correlations demonstrated a link between the thermal and biogeochemical stability of SOM, but the interpretation of the thermal behavior of SOM in bulk soil samples remains equivocal because of the role the mineral component and organo-mineral interactions.  相似文献

Many river revitalization projects are currently under development, but we still lack proper assessments of their effects on biodiversity, especially regarding non-aquatic vertebrates. We analysed the progressive changes in habitat mosaics, and the demographic response of two rare species of wading birds along a Swiss section of the Rhone river over a period of three decades. During the period preceding revitalization, the diversity of the habitat mosaic had been steadily declining. This impoverishment could however be reverted after 1994, when the revitalization was initiated: in 2006, the re-established habitat matrix was more diverse than at any time during the previous 27 years. The formerly channelized stream underwent radical transformation as it mutated into a heterogeneous braided river boosting pioneer riverine habitats. This has led the two wading bird species to establish significantly more territories than before the revitalization operations. Population size of Actitis hypoleucos and Charadrius dubius increased by 83% and 20%, respectively, between 1995 and 2007. During the same period, A. hypoleucos declined dramatically in Switzerland as a whole, whilst C. dubius showed no distinct population trend. The response of the two waders may have resulted from an increase in the number of lateral arms of the river, where the availability of productive temporary ponds provides excellent foraging opportunities. A significant proportion of these habitats was created by a targeted extraction of sediments with the objective to promote biodiversity while maintaining economic activity and improving security against floods. This suggests that gravel exploitation is not incompatible with efficient river restoration.  相似文献

Effects of soil and atmospheric drought on whole-tree transpiration (E(T)), leaf water potential (Ψ(L)) and whole-tree hydraulic conductance (K(T)) were investigated in mature rubber trees (Hevea brasiliensis, clone RRIM 600) during the full canopy stage in the rainy season in a drought-prone area of northeast Thailand. Under well-watered soil conditions, transpiration was tightly regulated in response to high evaporative demand, i.e., above reference evapotranspiration (ET(0)) ~2.2 mm day(-1) or maximum vapor pressure deficit ~1.8 kPa.
