OBJECTIVE: To evaluate, by use of population pharmacokinetics, the disposition of marbofloxacin in the aqueous humor after IV administration in dogs and identify its potential usefulness in the prophylaxis and treatment of intraocular infection. ANIMALS: 63 dogs. METHODS: Dogs received a single dose of marbofloxacin (2 mg x kg(-1), IV) at various time intervals before cataract surgery. Aqueous humor and blood samples were collected at the beginning of surgery. Marbofloxacin concentrations were measured by high-pressure liquid chromatography. Data were analyzed with a nonlinear mixed-effect model and, by use of population pharmacokinetic parameters, the time course of aqueous humor concentration was simulated for single doses of 3, 4, and 5.5 mg x kg(-1) IV. Pharmacodynamic surrogate markers and measured aqueous humor concentrations were used to predict in vivo antimicrobial activity. RESULTS: A maximum marbofloxacin concentration of 0.41 +/- 0.17 microg x mL(-1) was reached in the aqueous humor 3.5 hours after IV administration. In the post-distributive phase, marbofloxacin disappeared from aqueous humor with a half-life of 780 minutes. The percentage penetration into the aqueous humor was 38%. Predictors of antimicrobial effects of marbofloxacin (2 mg x kg(-1), IV) indicated that growth of the enterobacteriaceae and certain staphylococcal species would be inhibited in the aqueous humor. Marbofloxacin administered IV at a dose of 5.5 mg x kg(-1) would be predicted to inhibit growth of Pseudomonas aeruginosa and all strains of staphylococci but would not eradicate streptococcal infections. CONCLUSIONS AND CLINICAL RELEVANCE: Marbofloxacin administered IV can penetrate the aqueous humor of canine eyes and may be suitable for prophylaxis or treatment of certain anterior chamber infections.  相似文献

Biological weapons are considered as mass destruction and terror weapons. Terrorism including bioterrorism is the major threat in the future conflicts for our nations. The aim of bioterrorism is more related to the potential disorganisation of the society than to the lethal effects of the agents used. The dramatic consequences cannot be discarded, especially if contagious agents such viral are used. The preparation of specific defence measures is a major challenge for our countries.

The knowledge acquired from the struggle against natural infectious diseases and recent events are essential to improve behaviours to face the biological weapon threats. The defence attitude is based on the anticipation of the threat, the management of the victims, and the restoration of the operational capabilities. This global defence attitude implies six important functions: (i) alert, (ii) detection and diagnosis, (iii) availability of pharmaceutical countermeasures such as vaccine, sera and anti-infectious medicine and products, (iv) medical management of victims, (v) training and information, (vi) research and development.

Passive and active immunoprevention and immuntherapy belong to the approaches discussed in the context of bioterrorism countermeasures. Further researches might be focused on these topics.  相似文献

We investigated the phenotype of the T cells (CD4+ and CD8+) that produced Th1 (IFN-gamma) and Th2 cytokines (IL-4 and IL-10) during the firsttwo weeks of experimental fasciolosis in rats. We also followed the kinetics of the cytokine and proliferative responses of hepatic mononuclear cells (HMNC) over the same period. We found that HMNC were more numerous in the infected animals than in the controls. The percentage of CD4+ cells increased significantly after infection, whereas the percentage of CD8+ cells did not change. Moreover, the frequency of the cells producing (CP) cytokine changed after infection. The frequency of CP IFN-gamma on 7 days postinfection (pi) was similar to that in control animals. However, the frequency of CP IFN-gamma was clearly lower on day 14 pi, whereas the frequency of CP IL-4 and CP IL-10 had increased. The CP IL-10-were mostly CD4+. Mitogenic stimulation (phorbol myristate acetate/ionomycin) of HMNC led to an increase in the amounts of the Th2 cytokines in the supernatant on days 7 and 14 pi, with the increase more pronounced on day 14. In contrast, IFN-gamma levels also increased by day 7 pi but then decreased to below control levels by day 14. In addition, HMNC proliferation in response to mitogen followed a similar pattern to IFN-gamma production. These findings suggested that, during the first 2 weeks of infection, F hepatica induced a transient ThO cytokine profile followed by downregulation of the cellular response and the induction of a Th2 cytokine profile.  相似文献

The in situ distribution of NK cells in rat liver during the first 28 days of an experimental infection with F hepatica was investigated. NK cells were distributed homogeneously throughout the hepatic parenchyma in uninfected animals. The total number of hepatic mononuclear cells increased significantly following infection, but the proportion of NK cells did not change. After infection, these cells were found around the portal space, around the centrolobular vein, in the periportal fibrosis and in the band of collagen. However, no NK cells could be detected in or around the granuloma during infection. The frequency of both I L-2- and IFNgamma-producing NK cells was higher on day 7 postinfection (pi) but only the percentage of IFNgamma -CD161+ subsets remained elevated thereafter, whereas the percentage of both IL-2+CD161+ and IL-4+CD161+ subsets returned to the baseline. The number of CD161+IL10+ cells did not change significantly. These results suggest that NK cells could be another source for the early production of IFNgamma but provide no evidence that these cells are involved in early events associated with granuloma formation.  相似文献

Ethoxylates of rapeseed oil and of methylated rapeseed oil were synthesized and tested as adjuvants for 2,4-D and phenmedipham. Provided they had less than 6 units of ethylene oxide (EO), 1.0 to 10 g litre(-1) ethoxylates in water induced droplet spreading on barley leaves. In an acetone-based medium all derivatives strongly promoted the foliar uptake of 2,4-D, with no clear influence of the ethoxylation degree. In the same medium there was a negative influence of ethoxylate chain length on the foliar uptake of phenmedipham. In a water-based medium, phenmedipham applied with rapeseed oil emulsified with ethoxylated (20 EO) rapeseed oil displayed uptake rates close to a commercial preparation. The same was true for phenmedipham applied with ethoxylated (2 EO) methylated rapeseed oil. In bioassays, phenmedipham prepared with methylated rapeseed oil emulsified with ethoxylated (20 EO) rapeseed oil was as efficacious on barley as a commercial formulation. The same was true for phenmedipham prepared with ethoxylated (2 EO) methylated rapeseed oil. However, neither rapeseed oil nor methylated rapeseed oil emulsified with ethoxylated (2 EO) methylated rapeseed oil conferred good efficacy to phenmedipham. Hence, ethoxylated rapeseed oil derivatives are promising adjuvants or formulants for herbicides.  相似文献

Summary The pressure membrane and pressure plate techniques were used to establish the moisture content-water potential (M-) relationship of red pine (Pinus resinosa Ait.) sapwood in desorption above the fiber saturation point. The moisture content-water potential relationship is required for the development of a model of drying considering the gradient of water potential as the driving force of moisture in wood. This relationship was established at 18, 56 and 85 °C for radial desorption. The results obtained demonstrate that water potential increases with temperature T at a given moisture content M. There is no significant variation of /T with temperature. Also, there is no plateau at intermediate moisture contents as was the case for the M- relationship of aspen sapwood established in a previous work. The effective integral and differential pore size distributions inferred from the M- relationship are also presented. The largest proportion of effective pore openings was found for a radius of 0.2 m. Medium density fibreboard (MDF) was produced from fibres treated with maleated polypropylene wax. The objectives of this study were to investigate the effect of fibre treatment with maleated polypropylene wax on the advancing and receding contact angle and wicking of MDF panels by the Wilhelmy plate method; to verify the chemical reactions occurring between fibres and maleated polypropylene wax by infrared spectroscopy, and to verify if there are relationships with the water absorption of panels. Three maleated polypropylene wax contents (0, 3, 5%) and two resin types (urea–formaldehyde, UF and melamine–urea–formaldehyde, MUF) were considered in this study. Our results showed that the treatment increased the advancing contact angle of panels bonded with UF resin. The receding contact angles of MDF panels produced from treated fibres with 5% maleated polypropylene wax content were also increased by the treatment. Also, the wicking was reduced by the treatment independent of the maleated polypropylene wax content. Reductions of the wicking were 59% for panels bonded with UF resin and 73% for panels bonded with MUF. Relationships between the wicking and water absorption were observed. Chemical analysis realized by infrared spectroscopy did not detect the presence of an esterification reaction between wood fibres and anhydride groups of the maleated polypropylene wax. This suggests, therefore, that an esterification reaction did not take place or that chemical modifications were so small that they are not visible by infrared spectroscopy.  相似文献

The radial pattern of both maximum ring density and ring area of 36 black spruce trees were used to determine the transition age from juvenile wood to mature wood. The data were obtained by X-ray densitometry and both segmented linear and polynomial regressions were used to point out the age of the juvenile wood boundary. Three stand densities (1,790, 2,700 and 3,400 stems/ha) and three sampling heights (2.4, 5.1 and 7.8 m) were studied. Although maximum ring density and ring area presented similar radial patterns, they gave two significantly different results of transition ages. The maximum ring density over-estimated the transition age (17.6 years) in contrast to ring area (14 years). The results show that the transition from juvenile wood to mature wood occurred after 12 years at 7.8 m (versus 13.1 years at a height of 5.1 m, and 17.6 years at 2.4 m). Although transition age occurred later in the high stand density group (21 years), the difference was not significant between the three stand density groups. Nevertheless, transition age remains difficult to determine since no standard definition exists. The transition occurs over years, and most probably a transition wood exists between juvenile wood and mature wood. Estimation of the juvenile wood proportion in volume shows that it remains constant along the stem and it increases with stand density.  相似文献

Market-oriented production of gardening crops and cassava (Manihot utilissima) in the dry season is an increasingly frequent practice in Ségou, Mali. Traditionally, these crops are protected from roaming livestock with the help of dead fences. In order to provide a sustainable alternative to dead fences, the International Centre for Research in Agroforestry (ICRAF) began promoting the use of live fences, living trees planted closely together around a field plot. This study was conducted with the first farmers to use these live fences. These farmers expressed satisfaction with the protection offered by the trees and their ability to provide a variety of medicinal, economic, and food products. The form of land tenure, the social status of farmers within their families, and the availability of labour seem to be important factors in the decision to test the live fence. This raises questions about the accessibility of this technique and its possible contribution to the social and economic differentiation of its users. The amount of UF resin penetration into fibers, used for the production of medium density fiberboard (MDF), is unknown. To evaluate the relationship between resin viscosity and resin penetration depth, an experimental procedure involving confocal laser scanning microscopy (CLSM) and a Toluidine Blue O staining system was performed. The results indicate that CLSM in combination with a Toluidine Blue O staining system is a good way to characterize UF resin penetration into wood fibers. The main penetration direction is toward the fiber lumen. For wet fibers, whose moisture content is about 88%, the effect of resin viscosity with a range of 80 cps – 340 cps on penetration is very similar, with all adhesives reaching or almost reaching the fiber lumen after 60 min at room temperature. For MDF industrial samples, the highest depth of penetration of the adhesive was attained in the second dryer stage. After the second dryer stage, the resin penetration into the fiber did not increase.  相似文献
